The present paper addresses an approximate analytical model for contribution of an arbitrarily oriented circular crack into effective elastic compliance of a transversely isotropic material. We numerically examine the bounds of applicability of the hypothesis that change in elastic potential due to an arbitrarily oriented circular crack in a transversely-isotropic material can be approximated by the change calculated for a certain isotropic environment. In particular, we obtained that the error of such an approximation is less than 20% if the extent of anisotropy is moderate – the ratio of Young’s moduli in transverse and in-plane directions is less than 1.87. The obtained result can be used for development of a simple model for microcracked transversely-isotropic materials with mild-to-moderate extent of anisotropy.  相似文献

Transversely-isotropic material with an arbitrarily oriented penny-shaped crack is considered. We calculate fourth-rank compliance contribution tensor of the crack and second-rank crack opening displacement tensor and examine their dependence on crack orientation. It is shown that this dependence for the crack opening displacement tensor is negligible if transverse isotropy has elliptic character, i.e. if material symmetry can be described in terms of a certain second rank tensor.  相似文献

Quantum dot (QD) labeling combined with fluorescence lifetime imaging microscopy is proposed as a powerful transduction technique for the detection of DNA hybridization events. Fluorescence lifetime analysis of DNA microarray spots of hybridized QD labeled target indicated a characteristic lifetime value of 18.8 ns, compared to 13.3 ns obtained for spots of free QD solution, revealing that QD labels are sensitive to the spot microenvironment. Additionally, time gated detection was shown to improve the microarray image contrast ratio by 1.8, achieving femtomolar target sensitivity. Finally, lifetime multiplexing based on Qdot525 and Alexa430 was demonstrated using a single excitation-detection readout channel.  相似文献

Centrosomes and microtubules play crucial roles during cell division and differentiation. Spermatogenesis is a useful system for studying centrosomal function since it involves both mitosis and meiosis, and also transformation of the centriole into the sperm basal body. Centrosomin is a protein localized to the mitotic centrosomes in Drosophila melanogaster. We have found a novel isoform of centrosomin expressed during spermatogenesis. Additionally, an anticentrosomin antibody labels both the mitotic and meiotic centrosomes as well as the basal body. Mutational analysis shows that centrosomin is required for spindle organization during meiosis and for organization of the sperm axoneme. These results suggest that centrosomin is a necessary component of the meiotic centrosomes and the spermatid basal body.  相似文献

The influence of variable durations of contralateral acoustic stimulation on the suppression of click-evoked otoacoustic emissions was investigated in order to determine whether olivocochlear efferent fibers are equally effective whatever the acoustical stimulation duration or if they show fatigue. The suppression effect was measured for contralateral stimulus durations ranging from 10 to 180 s prior to the onset of otoacoustic emission recording, and continuing throughout the recording time (60 s). A Monte Carlo study was conducted using simulated dichotomous data to determine the effects of guessing on Rasch item fit statistics (weighted total, unweighted total, and unweighted between fit statistics) and the Logit Residual Index (LRI). The data were simulated using 100 items, 100 persons, three levels of guessing (0%, 25%, and 50%), and two item difficulty distributions (normal and uniform). The results of the study indicated that no significant differences were found between the mean Rasch item fit statistics for each distribution type as the probability of guessing the correct answer increased. The mean item scores differed significantly with uniformly distributed item difficulties, but not normally distributed item difficulties. The LRI was more sensitive to large positive item misfit values associated with the unweighted total fit statistic than to similar values associated with the weighted total fit or unweighted between fit statistics. The greatest magnitude of change in LRI values (negative) was observed when the unweighted total fit statistic had large positive values greater than 2.4. The LRI statistic was most useful in identifying the linear trend in the residuals for each item, thereby indicating differences in ability groups, i.e. differential item functioning.  相似文献

BACKGROUND: Nonspecific bronchial provocation tests may be simplified by the use of hand-held devices to deliver methacholine. OBJECTIVE: To study the feasibility of using a metered-dose inhaler (MDI) to administer methacholine in bronchial provocation tests, and the ability of such a device to diagnose bronchial hyperresponsiveness (BHR) accurately. METHODS: In an open randomized crossover pilot study, we compared the provocative dose that induces a 20% fall in FEV1 (PD20 FEV1) obtained with the methacholine MDI with that obtained using a conventional nebulizer in 20 hyperresponsive and 20 nonhyperresponsive subjects. The MDI delivers 400 doses of 100 microg of methacholine, and was used via a spacer. Bronchial hyperresponsiveness (BHR) was defined as a PD20 FEV1 <2,000 microg with the conventional test using the nebulizer. The tests were performed in each subject in a randomized order, 1 to 7 days apart. RESULTS: Of the subjects who had a nebulizer PD20 FEV1 <2,000 microg, all but one had an MDI PD20 FEV1 <800 microg. When 800 microg was taken as the threshold for the diagnosis of BHR with the MDI test, the accuracy of this test to diagnose BHR was 97.5%, and the two tests were highly concordant for the diagnosis of BHR (Pearson chi2, 36.19; p<0.0001). CONCLUSION: A hand-held device may be suitable for delivery of methacholine during bronchial provocation tests, if these results are confirmed in large samples.  相似文献

Pilot-scale constructed wetlands were used to treat water contaminated by polycyclic aromatic hydrocarbons (PAHs), particularly fluoranthene, and the possible role of fungi present in these ecosystems was investigated. A total of 40 fungal species (24 genera) were isolated and identified from samples (gravel and sediments) from a contaminated wetland and a control wetland. All of them were assayed for their ability to remove anthracene (AC) and fluoranthene (FA) from liquid medium. FA was degraded efficiently by 33 species while only 2 species were able to remove AC over 70%. A selection of 10 strains of micromycetes belonging to various taxonomic groups was further investigated for FA and AC degradation, toxicity assays and phenoloxidases (POx) detection. Interesting and not previously reported species were revealed (Absidia cylindrospora, Cladosporium sphaerospermum, and Ulocladium chartarum). They were all able to highly degrade the PAH-model compounds chosen. An interesting inducibility was noted for Ulocladium chartarum. Degradative ability of fungi was not related to their extracellular POx activity. This study may contribute to the improvement of constructed wetlands for water treatment, which may be enriched in efficient fungi.  相似文献

Rhizobia are soil bacteria that are able to form symbiosis with plant hosts of the legume family. These associations result in the formation of organs, called nodules in which bacteria fix atmospheric nitrogen to the benefit of the plant. Most of our knowledge on the metabolism and the physiology of the bacteria during symbiosis derives from studying roots nodules of terrestrial plants. Here we used a proteomics approach to investigate the bacterial physiology of photosynthetic Bradyrhizobium sp. ORS278 during the symbiotic process with the semi aquatical plant Aeschynomene indica that forms root and stem nodules. We analyzed the proteomes of bacteria extracted from each type of nodule. First, we analyzed the bacteroid proteome at two different time points and found only minor variation between the bacterial proteomes of 2-week- and 3-week-old nodules. High conservation of the bacteroid proteome was also found when comparing stem nodules and root nodules. Among the stem nodule specific proteins were those related to the phototrophic ability of Bradyrhizobium sp. ORS278. Furthermore, we compared our data with those obtained during an extensive genetic screen previously published. The symbiotic role of four candidate genes which corresponding proteins were found massively produced in the nodules but not identified during this screening was examined. Mutant analysis suggested that in addition to the EtfAB system, the fixA locus is required for symbiotic efficiency.  相似文献
