Scombridae species, such as tunas and mackerels, often do not spawn in land-based fish tanks without hormone treatment. To induce spawning in various fishes, a gonadotropin-releasing hormone analogue (GnRHa) is often administered by pellet implantation. Noninvasive administration is desired to induce spawning in scombrids that are sensitive to handling stress. Spawning induction by oral administration has been reported in several fishes, yet this method has not been put into practice in the aquaculture industry since a considerable amount of GnRHa is needed. Utilization of peptide synthesizers is widespread, and antigen-grade GnRHa (AgGnRHa) produced by a custom-peptide supplier is approximately 100-fold cheaper than conventional reagent-grade GnRHa (RgGnRHa), although the purity of AgGnRHa is lower. Here, we confirmed that the spawning induction potency of AgGnRHa was similar to that of RgGnRHa by pellet implantation in blue mackerel Scomber australasicus. Oral administration of AgGnRHa [6.0 mg/kg body weight (BW) per day] showed an equivalent ability to induce spawning of the mackerel as pellet implantation (0.1 mg/kg BW). We could also induce spawning of eastern little tuna Euthynnus affinis by oral administration of the AgGnRHa. Further, the obtained eggs showed higher survival. Thus, the oral delivery of AgGnRHa could be a powerful tool to induce spawning in Scombridae.  相似文献

Sericin was investigated as an alternative to fetal bovine serum (FBS) for bovine embryo culture. In vitro matured oocytes were developed using 0.05%, 0.1% or 0.15% sericin. The developmental rate, cryosurvival rate and blastulation time of these embryos were compared with those of embryos developed using 5% FBS. The number of lipid droplets was compared among the blastocysts developed using 5% FBS, using 0.05% sericin and in vivo. The rate of cleavage and blastocyst formation was similar among all groups. Blastulation occurred significantly earlier in the embryos developed using 5% FBS than in those developed using sericin at any concentration (P < 0.05). At 72 h after thawing, the cryosurvival rate of the blastocysts developed using 5% FBS and 0.05% sericin were significantly higher compared with those developed using 0.1% and 0.15% sericin (P < 0.05). The blastocysts developed using 0.05% sericin and in vivo produced a significantly fewer number of medium and large lipid droplets than those developed using 5% FBS. These results suggest that the blastocysts developed using 0.05% sericin show characteristics similar to those of the blastocysts developed in vivo and that the use of sericin as an alternative to FBS is feasible.  相似文献

Phytoplasma suspected symptoms of little leaf, flat stem, witches’ broom and leaf yellowing were recorded on the four legume species, cowpea (Vigna unguiculata (L.) Walp.), pigeon pea (Cajanus cajan (L.) Millsp.), lentil (Lens culinaris Medikus) and mung bean (Vigna radiata (L.) Wilczek) in the states of Delhi, Uttar Pradesh (UP) and Kerala from 2014 to 2016. DNA specific fragments of approximately 1.3 kb were amplified from symptomatic samples of cowpea, pigeon pea, lentil and mung bean in nested PCR assays by using two sets of universal phytoplasma nested specific primers P1/P7 followed by 3Far/3Rev. No DNA amplifications were observed in any of the non-symptomatic legume samples with same primer pairs. Pair wise sequence comparison, phylogeny and virtual RFLP analysis of 16S rDNA sequences of the four legume species confirmed the association of four different groups and subgroups of phytoplasmas in the present study. The mung bean witches’ broom at Delhi was identified to be associated with strain related to ‘Ca. P. aurantifolia’ (16SrII-D), pigeon pea little leaf at Faizabad, UP with strain related to ‘Ca. P. phoenicium’ (16SrIX-C), lentil witches’ broom at Faizabad, UP with ‘Ca. P. trifolii’ (16SrVI-D) and cow pea flat stem disease at Kerala with ‘Ca. P. cynodontis’ (16SrXIV-A). Association of ‘Ca. P. cynodontis' (16SrXIV-A) infecting cowpea, ‘Ca. P. trifolii’ (16SrVI-D) in lentil and phytoplasmas strain related to ‘Ca. P. phoenicium’ (16SrIX-C) infecting pigeon pea are the new reports to the world.  相似文献

Biological resources are the basic infrastructure of bioscience research. Rice (Oryza sativa L.) is a good experimental model for research in cereal crops and monocots and includes important genetic materials used in breeding. The availability of genetic materials, including mutants, is important for rice research. In addition, Oryza species are attractive to researchers for both finding useful genes for breeding and for understanding the mechanism of genome evolution that enables wild plants to adapt to their own habitats. NBRP-RICE contributes to rice research by promoting the usage of genetic materials, especially wild Oryza accessions and mutant lines. Our activity includes collection, preservation and distribution of those materials and the provision of basic information on them, such as morphological and physiological traits and genomic information. In this review paper, we introduce the activities of NBRP-RICE and our database, Oryzabase, which facilitates the access to NBRP-RICE resources and their genomic sequences as well as the current situation of wild Oryza genome sequencing efforts by NBRP-RICE and other institutes.  相似文献

A feeding experiment was conducted to determine the optimal formulation level of algae meal, which is rich in docosahexaenoic acid (DHA), in a non‐fish meal diet. Six iso‐nitrogenous (450 g/kg) and iso‐lipidic (130 g/kg) experimental diets were prepared. The control diet was formulated with fish meal (400 g/kg), fish oil (60 g/kg), plant protein sources (220 g/kg) and rapeseed oil (50 g/kg). Plant protein sources (soy protein concentrate, soybean meal and corn gluten meal), rapeseed and fish oil were formulated in the second diet (NFM + FO). In the third diet, fish oil of the NFM + FO diet was replaced by rapeseed oil (NFM + NFO) and designated as the negative control. In the other three diets, rapeseed oil in the NFM + NFO diet was replaced with algae meal (Schizochytrium sp. powder) at 50 g/kg, 100 g/kg and 150 g/kg (AM5, AM10 and AM15, respectively). Triplicate groups of juvenile red sea bream (8.8 g) were fed the experimental diets for 12 weeks near satiation. The growth was lowest in the fish fed NFM + NFO diet. This was improved by the formulation of algae meal, which reached the growth level of the NFM + FO group in the AM10 group. The lipid content of the whole fish body in the NFM + NFO group was significantly lower than those of other groups. The fatty acid profile showed significant differences among dietary treatments. DHA content in total and polar lipids of the whole body and liver was highest in the AM10 and AM15 groups. These results reconfirm that microalgae are a suitable lipid source for the replacement of dietary fish oil for marine fish, and the optimal level was estimated as 50 g/kg?100 g/kg in diet.  相似文献

A feeding experiment was conducted to develop non‐fish meal and non‐fish oil diet for red seabream by using plant protein source and Schizochytrium meal which is rich in 22:6n‐3 (DHA). Three iso‐nitrogenous and iso‐lipidic experimental diets were prepared (CP 41.2% ± 0.4%, CL 16.4% ± 1%). Control diet contained both fish meal (40%) and fish oil (6%). In the second diet, fish meal was replaced by plant meals (soy protein concentrate, soybean meal, corn gluten meal) [FO]. In the third diet, fish meal and fish oil were replaced by algae meal (Schizochytrium sp. powder) and plant proteins [AO]. Duplicated groups of juvenile red seabream (8.8 g ± 1.5) were fed the experimental diets for 12 weeks to near satiation. There was no statistical difference among treatment in specific growth rate. Feed conversion ratio of AO diet group was higher than that of control. In wet basis, whole body protein level was significantly higher in AO diet than FO group while lipid content was lower than control group. In fatty acid profile, AO group had significantly lower 18:4n‐3, 20:4n‐3, 22:5n‐3 and 20:5n‐3 (EPA) level, but significantly higher 18:3n‐3 and DHA level than the other two diet fed fish. The results might suggest that further developments in microalgae diet offer a promising lipid source of n‐3 PUFA as essential fatty acid on marine fish. And it showed possibility to develop non‐fish meal and non‐fish oil feed for marine aquaculture fish by using microalgae.  相似文献
