Polyion complexation between basic fibroblast growth factor (bFGF) and gelatin was studied by the turbidity change of mixed solution, heparin high performance liquid affinity chromatography (HPLAC), and isoelectric electrophoresis. When an aqueous solution of acidic gelatin with an isoelectric point (IEP) of 5.0 was mixed with that of bFGF, the turbidity of the mixed solution increased with time, whereas basic gelatin with and IEP of 9.0 did not cause any solution turbidity. A maximum turbidity of the mixed bFGF and acidic gelatin solution was observed around a bFGF/gelatin molar ratio of 1.0, irrespective of the gelatin concentration and solution temperature. The solution turbidity decreased with an increase in the ionic strength of the mixed solution. Complexation of bFGF with acidic gelatin was slower than that with poly(acrylic acid) probably because of the lower density of gelatin negative charge than that of poly(acrylic acid). HPLAC study revealed that complexation of bFGF with the acidic gelatin reduced the affinity of bFGF for heparin, in contrast to the basic gelatin, although the extent became smaller with the increasing ionic strength of the solution. An electrophoretic experiment showed that the IEP of bFGF shifted to a lower value after its gelatin complexation. These findings indicate that an electrostatic interaction is the main driving force for the complexation between acidic gelatin and basic bFGF.  相似文献

This short-term longitudinal study examined the associations between cross-racial/ethnic friendships and relative changes in forms of peer victimization or peer support and the roles of classroom diversity and sociometric status (i.e., social preference) in these associations. A total of 444 children (age range: 9–10 years) from racially/ethnically diverse elementary schools participated in this study. Results demonstrated that cross-racial/ethnic friendships (but not same-racial/ethnic friendships) uniquely predicted relative decreases in relational victimization. Further, classroom diversity moderated the relations of cross-racial/ethnic friendships with relative decreases in physical victimization and relative increases in peer support, such that these relations were stronger for children in highly diverse classrooms. Finally, social preference mediated the association between cross-racial/ethnic friendships and relative decreases in relational victimization. The associations among cross-racial/ethnic friendships, same-racial/ethnic friendships, social experiences with peers, and classroom diversity are discussed. In order to propose a simplified simulation method using finite element (FE) model for predicting deformation and residual stress generated by multi-pass butt welding, a series of experiments and numerical analyses were carried out. 3-pass butt welding of steel plates was simulated by the thermal elasto-plastic analysis with shell elements and with solid elements respectively. A heat input model for considering the temperature distribution in the thickness direction in shell elements was proposed. The validity of the heat input model was verified by comparing analytical results with experimental results or other analytical results using solid elements. Furthermore, the effectiveness for saving computing time by using shell elements was confirmed from the comparison with the case using solid elements. It was confirmed that the welding out-of-plane deformation and residual stress could be predicted with high accuracy by the proposed method. The computing time was around 14% of that by the precise model with solid elements.  相似文献

We present multilayer formation via spinodal decomposition in rutile TiO2-VO2 (TVO) epitaxial films on sapphire substrates. (001)- and (101)-oriented TVO solid-solution films are grown epitaxially on TiO2/Al2O3 using a pulsed laser deposition technique and annealed inside the spinodal region. X-ray diffraction measurements and scanning transmission electron microscopy (STEM) observations show that the films are phase-separated along the [001] direction and lamellar structures are formed in a parallel or slanted direction to the sapphire substrates depending on the film orientation. The results indicate the multilayer formation via spinodal decomposition in the TVO films. STEM investigations also reveal a relatively high Ti concentration in the decomposed phases, reflecting the influence of lattice deformation on the phase decomposition in the films. Our work shows that spinodal decomposition is a promising approach for the formation of a multilayer structure in TVO films and helps deepen understanding the spinodal decomposition in TVO system.  相似文献

The objective of this research was to investigate the difference between chicken and beef in the interaction of actomyosin (myosin B) with microbial transglutaminase (MTG). The gel strength of myosin B was improved in both species and was significantly greater in beef than in chicken (P < 0.01). The degree of protein viscosity and the ε(γ-glutamyl)lysine (G–L) content were significantly higher in beef than in chicken (P < 0.01). Myosin heavy chain (MHC) bands visualized by SDS–PAGE revealed that the same proteins in various meat species vary in their size and structure. Scanning electron microscope images (SEMI) revealed that myosin B in both species was polymerized, and formed multi-projection structures of G–L; surprisingly, more of these structures were found in beef than in chicken. It is possible that the proteins in chicken are folded into a strand shape that tightly encases a considerable number of glutamine and lysine residues, whereas MTG substrate cannot couple glutamine and lysine. This suggests that the reactivity of MTG is dependent on the residual amino acids present on the surface of myosin B in meat. Some protein components (peptides with long reiterated methylene groups attached) joined by disulfide bonds (cysteine) in chicken samples were inhibitory and reduced MTG activity. SEMI also suggested that all MTG-dependent mega-structures of protein molecules generated in chicken and beef may vary greatly in size, configuration and complexity after treatment with MTG. We concluded that the optimal cross-links in myosin B induced by MTG are heterogeneous in chicken and beef.  相似文献

Hormone therapy in the form of androgen deprivation is a major treatment for advanced prostate cancer. However, if such therapy is overly prolonged, tumour cells may become resistant to this treatment and result in recurrent fatal disease. Long-term hormone deprivation also is associated with side effects poorly tolerated by patients. In contrast, intermittent hormone therapy with alternating on- and off-treatment periods is a possible clinical strategy to delay progression to hormone-refractory disease with the advantage of reduced side effects during the off-treatment periods. In this paper, we first overview previous studies on mathematical modelling of prostate tumour growth under intermittent hormone therapy. The model is categorized into a hybrid dynamical system because switching between on-treatment and off-treatment intervals is treated in addition to continuous dynamics of tumour growth. Next, we present an extended model of stochastic differential equations and examine how well the model is able to capture the characteristics of authentic serum prostate-specific antigen (PSA) data. We also highlight recent advances in time-series analysis and prediction of changes in serum PSA concentrations. Finally, we discuss practical issues to be considered towards establishment of mathematical model-based tailor-made medicine, which defines how to realize personalized hormone therapy for individual patients based on monitored serum PSA levels.  相似文献

A hard-x-ray telescope is successfully produced for balloon observations by making use of depth-graded multilayers, or so-called supermirrors, with platinum-carbon (Pt/C) layer pairs. It consists of four quadrant units assembled in an optical configuration with a diameter of 40 cm and a focal length of 8 m. Each quadrant is made of 510 pieces of coaxially and confocally aligned supermirrors that significantly enhance the sensitivity in an energy range of 20-40 keV. The configuration of the telescope is similar to the x-ray telescope onboard Astro-E, but with a longer focal length. The reflectivity of supermirrors is of the order of 40% in the energy range concerned at a grazing angle of 0.2 deg. The effective area of a fully assembled telescope is 50 cm2 at 30 keV. The angular resolution is 2.37 arc min at half-power diameter 8.0 keV. The field of view is 12.6 arc min in the hard-x-ray region, depending somewhat on x-ray energies. We discuss these characteristics, taking into account the figure errors of reflectors and their optical alignment in the telescope assembly. This hard-x-ray telescope is unanimously afforded in the International Focusing Optics Collaboration for muCrab Sensitivity balloon experiment.  相似文献

An acoustic fiber sensor for measurement of ultrasonic waves, which used the approximate Raman-Nath diffraction effect where light diffraction waves were generated in an optical fiber by strain due to the ultrasonic waves, was proposed and examined. In order to characterize the acoustic fiber sensor as a basic study, measurements of low-frequency ultrasonic waves in water were examined using a step index fiber operating as a detection sensor. The results showed that characteristics of detected signals agreed with the theoretical prediction based on Fraunhofer diffraction. This indicates that our proposed fiber sensor can be used for the detection of low-frequency ultrasonic waves as well as the transmission of light diffraction signals.  相似文献

Almost 30 years have passed since a term 'tissue engineering' was created to represent a new concept that focuses on regeneration of neotissues from cells with the support of biomaterials and growth factors. This interdisciplinary engineering has attracted much attention as a new therapeutic means that may overcome the drawbacks involved in the current artificial organs and organ transplantation that have been also aiming at replacing lost or severely damaged tissues or organs. However, the tissues regenerated by this tissue engineering and widely applied to patients are still very limited, including skin, bone, cartilage, capillary and periodontal tissues. What are the reasons for such slow advances in clinical applications of tissue engineering? This article gives the brief overview on the current tissue engineering, covering the fundamentals and applications. The fundamentals of tissue engineering involve the cell sources, scaffolds for cell expansion and differentiation and carriers for growth factors. Animal and human trials are the major part of the applications. Based on these results, some critical problems to be resolved for the advances of tissue engineering are addressed from the engineering point of view, emphasizing the close collaboration between medical doctors and biomaterials scientists.  相似文献
