Among 11 subtypes of heterotrimeric G-protein gamma-subunit, gamma1 (rod), gamma8 (cone) and gamma11 are modified with farnesyl while the others are modified with geranylgeranyl at the C-terminus. To understand the role of specific isoprenylation (farnesylation) of retinal transducin, we examined how and to what extent the type of isoprenyl group affects transducin-beta gamma (beta1 gamma1) functions such as interactions with membranes, Galpha/receptor, and effectors. To this end, the C-terminal farnesylation signal sequence (CVIS) of gamma1 was replaced by a geranylgeranylation signal (CVIL), and the resultant mutant (S74L) or wild-type (WT) gamma1 was coexpressed with beta1 in the baculovirus-Tn5 insect cell system. Both gamma1WT and gamma1S74L expressed as a beta gamma complex were mixtures modified with farnesyl and geranylgeranyl groups. The ratio of farnesyl to geranylgeranyl in preparations of beta1 gamma1WT and beta1 gamma1S74L purified from the Tn5 cell membrane fraction was about 1:2 and 1:6, respectively. These two forms of recombinant beta1 gamma1 and retinal beta1 gamma1 were different in their abilities to associate with rod outer segment membranes with the following rank order: beta1 gamma1S74L > beta1 gamma1WT > retinal beta1 gamma1. Functionally, beta1 gamma1S74L was the most potent to promote pertussis toxin-catalyzed ADP ribosylation of transducin-alpha (Talpha), to stimulate metarhodopsin II-catalyzed GTPgammaS-binding reaction to Talpha and to modulate adenylyl cyclase and phospholipase C activities. All of the beta1 gamma1 functions absolutely required the isoprenylation of the gamma-subunit. As for the interaction with Goalpha and adenylyl cyclase, predominantly geranylgeranylated beta1 gamma1S74L was less effective than geranylgeranylated beta1 gamma2 purified from bovine brain. These results demonstrate that the properties of Gbeta gamma are strongly affected by the type of functionally indispensable isoprenylation in addition to the amino acid sequence of Ggamma. Cross‐linked polyethylene (XLPE) has been widely adopted as insulating material for high‐voltage power cables up to 500 kV. Further improvement of electrical and thermal properties on insulating material is required in order to increase cable operation efficiency. Therefore, the development of novel insulating material possessing high thermal properties will be necessary. Recent progress of catalysis technology contributes to obtain new polymeric materials which may be applied to electrical insulation. The authors investigated the basic properties of newly developed stereoregular syndiotactic polypropylene (s‐PP) which is synthesized with homogeneous metallocene catalyst. Though recycling of cross‐linked polymers such as conventionally used XLPE may be difficult because of their poor heat deformation, the s‐PP which is not cross‐linked must be suitable for recycling. A series of experiments on its physical and electrical properties gave the following results.
- (1) s‐PP has sufficient flexibility compared with isotactic polypropylene (i‐PP ).
- (2) Both AC and lightning impulse breakdown strength of s‐PP in spite of no cross‐linking are superior to those of XLPE in the temperature range from 25 to 90 °C.
- (3) Degradation by copper of s‐PP is less than that of i‐PP.
- (4) s‐PP/VLDPE blend shows sufficient brittleness temperature for use.

We have demonstrated that the near-infrared and infrared absorptions in the 8000-3200 cm(-1) region of an OH group of 2-nonanol, 1-nonanol, etc., in n-heptane are excellently separated by subtraction without any serious interference down to very low concentrations at which OH groups are completely free. The separated sharp absorptions are assigned to the fundamental, combination, and overtone bands that are concerned with the OH stretching of free OH. Two components of a sharp overtone band around 7100 cm(-1), which are observed for primary and secondary alcohols, are assigned to coexisting internal rotational isomers of an OH group around the O-C bond. The frequencies of the OH stretching fundamental and overtone bands that are assigned to internal rotational positions are consistent for all the investigated alcohols, including methanol and tertiary butanol. Comparison of the separated spectrum of 2-nonanol in n-heptane with that in 1-chlorooctane or in carbon tetrachloride makes it clear that hydrocarbon is an inert solvent that does not disturb the intrinsic nature of an alcohol OH group. There actually exists a constant anharmonicity shift of 169-175 cm(-1) between the double frequency (2nu(OH)o) of the observed fundamental and the observed overtone frequency ([2nu(OH)]o) for free OH of various alcohols in n-heptane. 相似文献

The effects of the concentration dependence of the diffusion coefficient of a polymer solution (polystyrene in benzene and cyclohexane) in determining molecular weight distribution by the diffusion method are briefly discussed. The value of the ratio Dm0/DA0 in a good solvent was found to be close to 1.0 for a polydisperse polymer and less than 1.0 for monodisperse polymers. Molecular weight distribution curves of the polydisperse sample were obtained by the diffusion method in cyclohexane and benzene, respectively. The molecular weight distribution curve obtained for the polymer used in benzene solution looked as if the polymer had a narrow molecular weight distribution. The phenomena cited above were interpreted in the light of the concentration dependence of the diffusion coefficient of polymer solutions. 相似文献

The nematode Caenorhabditis elegans exhibits chemotaxis toward a wide variety of chemicals including water-soluble molecules and volatile organic compounds. We have previously discovered that C. elegans wild-type strain N2 is strongly attracted by cycloheximide which has long been known as a bitter tastant for humans and other mammals. We describe here the isolation and initial characterization of the first mutants which were defective specifically in their attraction to cycloheximide. In our screenings, we selected two mutants that were defective in chemotaxis to cycloheximide but normal in their attraction to NH4Cl and histidine. These mutants also avoided quinine hydrochloride, CuSO4 and high concentrations of NaCl similar to the wild-type strain N2. Furthermore, no observable defect was detected in their attraction to volatile odorants such as isoamyl alcohol and diacetyl. Dye-filling experiments suggested that they have no morphological defect in the sensory endings of the amphid neurons. 相似文献

Osteoprotegerin (OPG) is an osteoblast-derived secreted member of the tumour necrosis factor receptor superfamily that inhibits osteoclastogenesis. Mice that are OPG-deficient have severe bone loss, including growth plate cartilage destruction. Using OPG-deficient mice as a useful animal model, we attempted to clarify differentiation and ultrastructural features of osteoclasts located on destructed growth plate cartilage and trabecular bone matrix. In the humerus and femur of OPG homozygous (-/-) mice, adjacent to the growth plate cartilage, bone trabeculae without a calcified cartilage core were characteristically formed at the metaphyseal side of the medullary cavity, which resulted in an irregular chondrocyte distribution and arrangement in growth plate cartilage. During growth plate cartilage destruction, osteoclasts positive for tartrate-resistant acid phosphatase showed unusual localization on both type-II collagen-positive cartilage and type-I collagen-positive trabecular bone matrix at the ossification centre of the epiphyseal/metaphyseal border. Although multinucleated osteoclasts were distributed within open lacunar canals in the growth plate, those on uncalcified cartilage matrix lacked a ruffled border. Facing the calcified cartilage matrix within lacunar canals, osteoclasts showed irregularly formed ruffled borders. After growth plate destruction, a thin bone layer was deposited on the remaining cartilage surfaces by invading osteoblasts. Osteoclasts formed prominent ruffled border structures on bone matrix, deposited on the remaining growth plate cartilage. These results suggest that, in OPG (-/-) mice, terminal osteoclast differentiation requires the presence of newly produced bone matrix, as the coupled phenomenon of bone formation and resorption, as well as osteoblast-derived cytokines. 相似文献

Using 3-day-old newborn rats, we examined the differentiation processes of osteoclasts associated with the destruction of the femoral growth plate cartilage and primary trabecular bone. In the growth plate cartilage, thin mineralized areas were detected solely in the longitudinal septal cartilage matrix in the hypertrophic zone, but the transverse septal cartilage matrix between adjacent chondrocytic lacunae within a row of chondrocytes remained unmineralized. The longitudinal septal cartilage between adjacent rows of chondrocytes appeared to persist, forming the walls of opened lacunar canals. Consistent with the removal of the transverse septal cartilage matrix, the longitudinal canals of opened chondrocytic lacunae were deeply invaded by capillary vessels, mononuclear cells and multinucleated pre-osteoclasts lacking a ruffled border. CD34-positive endothelial cells of capillary vessels deeply penetrated into the transverse septal cartilage matrix facing the medullary cavity and the opened chondrocytic lacunae. ED1-positive monocytes/macrophages were distributed at the chondro-osseous junction, but they were distant from the erosive front of the transverse septa. Tartrate-resistant acid phosphatase-positive multinucleated pre-osteoclasts lacking a ruffled border and differentiated osteoclasts with a ruffled border were localized mainly at two locations: the chondro-osseous junction and the growth front of primary bone trabeculae. Osteoclasts were located on the type-I collagen-positive bone trabeculae close to the growth plate, but they appeared to be distant from the type-II collagen-positive cartilage matrix. Even within opened chondrocytic lacunae, when osteoclasts were distant from the cartilage and bone matrix, they lacked polarized cytoplasmic organization and a ruffled border. The osteoclasts located in the remaining septal cartilage also exhibited neither a ruffled border nor a clear zone. Osteoclasts with a prominent ruffled border and clear zone were located in bone matrix covering the remaining septal cartilage. These results suggest that osteoclasts require hydroxyapatite crystals and bone matrix constituents for ruffled border formation and are not involved in resorption of the unmineralized transverse and mineralized longitudinal septal cartilage without covering bone matrix at the chondro-osseous junction. 相似文献

A novel optical sensor based on spectroscopic measurement of the plasmon absorption of a colloidal gold submonolayer immobilized upon a planar waveguide has been developed for label-free detection of biomolecular interactions at the sensor's surface. We fabricated the sensor by locally modifying a 50-microm-thick glass plate with hemoglobin (Hb) and then self-assembling gold colloids from the aqueous solution onto the Hb-modified area of the glass plate. Polychromatic light from a xenon-arc lamp was launched into the thin-film glass plate by use of a broadband fiber-coupling method. With the use of a CCD detector to monitor the light beam emitted from an end face of the glass plate, the plasmon absorption spectrum of the colloidal gold submonolayer was determined to depend on the polarization states of the guided light and to change with the refractive index of the medium in contact with the colloids. In addition to simplicity of fabrication and the ease of use, the sensor yields a larger absorbance sensitivity than the normal transmission measurement. 相似文献
