The species-specific antigenicity of the lateral flagella of Vibrio parahaemolyticus was applied to techniques for immunological identification of the vibrio. One was a coagglutination test using staphylococcal cells sensitized with anti-lateral-flagella antibody bound to protein A located in the staphylococcal cell wall. The other technique was an enzyme-linked immunosorbent assay (ELISA) using a four-layer sandwich procedure. By means of the staphylococcal coagglutination technique, about 106 vibrio cells or 60 ng of flagellar protein could be detected and the whole procedure was performed within 1 h. The sandwich ELISA procedure was more sensitive, but the time required for the whole procedure was about 8 h. Both techniques were specific for V. parahaemolyticus, because no cross-reactivity was observed with other related vibrios, and the tests were not inhibited by heterologous bacterial cells. These results suggest that both techniques are useful for the rapid identification of V. parahaemolyticus in complex samples.  相似文献

A simplified method for the determination of forchlorfenuron in agricultural products by HPLC with UV detection was investigated. A chopped sample homogenate from agricultural products was extracted with acetone. The extract was filtrated and concentrated. The residues was loaded onto a Chem Elut column and extracted with ethyl acetate. The crude extract was purified on Oasis HLB and Bond Elut PSA mini-columns using a mixture of methanol and ethyl acetate. Forchlorfenuron was analyzed by HPLC with UV detection (263 nm). HPLC separation was performed on an ODS column with methanol-water as the mobile phase. Recoveries of forchlorfenuron from several agricultural products fortified at the level of 0.1 microg/g were in the range of 87.6-99.5%. The limit of detection (S/N=3) was 0.005 microg/g in the sample.  相似文献

The presence of lactic acid bacteria (LAB) during shochu fermentation was monitored by PCR-denaturing gradient gel electrophoresis (DGGE) and by bacteriological culturing. No LAB were detected from fermented mashes by PCR-DGGE using a universal bacterial PCR primer set. However, PCR-DGGE using a new primer specific for the 16S rDNA of Lactococcus, Streptococcus, Tetragenococcus, Enterococcus, and Vagococcus and two primers specific for the 16S rDNA of Lactobacillus, Pediococcus, Leuconostoc, and Weissella revealed that Enterococcus faecium, Lactobacillus casei, Lactobacillus fermentum, Lactobacillus nagelii, Lactobacillus plantarum, Lactococcus lactis, Leuconostoc citreum, Leuconostoc mesenteroides, and Weissella cibaria inhabited in shochu mashes. It was also found that the LAB community composition during shochu fermentation changed after the main ingredient and water were added during the fermentation process. Therefore, we confirmed that PCR-DGGE using all three primers specific for groups of LAB together was well suited to the study of the LAB diversity in shochu mashes. The results of DGGE profiles were similar to the results of bacteriological culturing. In conclusion, LAB are present during shochu fermentation but not dominant.  相似文献

A simplified HPLC determination method for maleic hydrazide in agricultural products was developed, and commercial agricultural crops were investigated. The homogenate of agricultural products was extracted with water. The crude extract was purified on an ACCUCAT Bond Elut extraction cartridge using water. Maleic hydrazide was analyzed by HPLC with UV detection (303 nm). The HPLC separation was performed on a ZORBAX SB-Aq column with acetonitrile-water-phosphoric acid(5:95:0.01) as the mobile phase. Recoveries of maleic hydrazide from 15 agricultural products fortified at 1.0 and 10 micrograms/g were in the ranges of 92.6-104.9% and 94.2-101.3%, respectively. The limit of detection was 0.5 microgram/g in samples. The proposed method was applied to the determination of 242 commercial vegetables and fruits. Maleic hydrazide was detected in 2 samples of imported onion at the levels of 4.9 and 7.2 micrograms/g.  相似文献

(1) Background: Atypical auditory perception has been reported in individuals with autism spectrum disorder (ASD). Altered auditory evoked brain responses are also associated with childhood ASD. They are likely to be associated with atypical brain maturation. (2) Methods: This study examined children aged 5–8 years old: 29 with ASD but no intellectual disability and 46 age-matched typically developed (TD) control participants. Using magnetoencephalography (MEG) data obtained while participants listened passively to sinusoidal pure tones, bilateral auditory cortical response (P1m) was examined. (3) Results: Significantly shorter P1m latency in the left hemisphere was found for children with ASD without intellectual disabilities than for children with TD. Significant correlation between P1m latency and language conceptual ability was found in children with ASD, but not in children with TD. (4) Conclusions: These findings demonstrated atypical brain maturation in the auditory processing area in children with ASD without intellectual disability. Findings also suggest that ASD has a common neural basis for pure-tone sound processing and language development. Development of brain networks involved in language concepts in early childhood ASD might differ from that in children with TD.  相似文献

Several triacylglycerols (TAG) that contained eicosapentaenoic acid (EPA) were chemically synthesized and stored at 25°C to assess the influence of TAG structure on oxidative stability and formation of oxidation products. Oxidative stability was evaluated by oxygen consumption during storage of the TAG. Autoxidation products of TAG were analyzed by high-performance liquid chromatography (HPLC) and liquid chromatography-mass spectrometry (LC-MS). Results showed that a 2:1 (mole/mole) mixture of trieicosapentaenoylglycerol (EEE) and tripalmitoylglycerol (PPP) was most susceptible to autoxidation. The oxidative stability of TAG that contained EPA and palmitic acid was negatively correlated with the moles of EPA in a single TAG molecule. When TAG with one EPA and two other fatty acids were oxidized, chainlength of constituent fatty acids hardly affected the oxidative stability of EPA-containing TAG molecules, except for stearic acid. HPLC and LC-MS analyses showed that monohydroperoxides were major oxidation products regardless of type of TAG. Bis- and tris-hydroperoxides were formed during autoxidation of EEE and dieicos-apentaenoylpalmitoylglycerol. Monohydroperoxy epidioxides were found in all autoxidized TAG. These observations suggested that TAG structure affected the oxidation of TAG with highly unsaturated fatty acids.  相似文献
