Influence of sorption to dissolved humic substances on transformation reactions of hydrophobic organic compounds in water. I. Chlorination of PAHs

The effect of sorption to dissolved humic acids (HAs) on the chlorination of PAHs in aqueous solution was studied. The addition of HA accelerated the chlorination of fluoranthene and naphthalene in hypochlorite solutions at pH 5, the stronger effect being observed for fluoranthene that is sorbed to a higher extent than naphthalene. Sorption coefficients (K(DOC)) of the analytes were determined by solid-phase microextraction (SPME). The observed rate constant for fluoranthene chlorination is, for example, larger by a factor of 5 in the presence of 10 mg L(-1) of an aquatic HA as compared to HA-free solution (k' = 0.02 h(-1) at 60 mg L(-1) active chlorine, pH 5, without HA). While Cl2 is the dominant reactive species in pure aqueous solution for both PAHs, the reaction of fluoranthene seems to involve an additional pathway of chlorination by HOCl in the presence of HA. It was found that not only did HA not protect PAHs from the electrophilic attack of the chlorinating species, but the sorption of PAHs on the hydrophobic domains of the HA favored instead the extent of the chlorination reaction.     

Coherence between emotional experience and physiology: Does body awareness training have an impact?

Two fundamental issues in emotion theory and research concern: (a) the role of emotion in promoting response coherence across different emotion systems; and (b) the role of awareness of bodily sensations in the experience of emotion. The present study poses a question bridging the two domains; namely, whether training in Vipassana meditation or dance, both of which promote attention to certain kinds of bodily sensations, is associated with greater coherence between the subjective and physiological aspects of emotion. We used lag correlations to examine second-by-second coherence between subjective emotional experience and heart period within individuals across four different films. Participants were either: (a) experienced Vipassana meditators (attention to visceral sensations), (b) experienced dancers (attention to somatic sensations), and (c) controls with no meditation or dance experience. Results indicated a linear relationship in coherence, with meditators having highest levels, dancers having intermediary levels, and controls having lowest levels. We conclude that the coherence between subjective and cardiac aspects of emotion is greater in those who have specialized training that promotes greater body awareness. BTK and PI3K Inhibitors Reveal Synergistic Inhibitory Anti-Tumoral Effects in Canine Diffuse Large B-Cell Lymphoma Cells

Bruton’s tyrosine kinase (BTK) and phosphoinositide 3-kinase (PI3K) in the B-cell receptor (BCR) signaling pathway are considered potential therapeutic targets for the treatment of B-cell lymphomas, among which, diffuse large B-cell lymphoma (DLBCL) is the most common type. Herein, we comparatively evaluated the single and combined application of the BTK inhibitor ibrutinib and the selective PI3Kγ inhibitor AS-605240 in the canine DLBCL cell line CLBL-1. For further comparison, key findings were additionally analyzed in canine B-cell leukemia GL-1 and human DLBCL cell line SU-DHL-4. While ibrutinib alone induced significant anti-proliferative effects on all cell lines in a dose-dependent manner, AS-605240 only induced anti-proliferative effects at high concentrations. Interestingly, ibrutinib and AS-605240 acted synergistically, reducing cell proliferation and increasing apoptosis/necrosis in all cell lines and inducing morphological changes in CLBL-1. Moreover, the combined application of ibrutinib and AS-605240 reduced relative phosphorylation and, in some instances, the levels of the BTK, AKT, GSK3β, and ERK proteins. Comparative variant analysis of RNA-seq data among canine B- and T-lymphoid cell lines and primary B-cell lymphoma samples revealed potentially high-impact somatic variants in the genes that encode PI3K, which may explain why AS-605240 does not singly inhibit the proliferation of cell lines. The combination of ibrutinib and AS-605240 represents a promising approach that warrants further in vivo evaluation in dogs, potentially bearing significant value for the treatment of human DLBCL.
