The aim of this study was to characterize the organic solvent and detergent tolerant properties of recombinant lipase isolated from thermotolerant Bacillus sp. RN2 (Lip-SBRN2). The isolation of the lipase-coding gene was achieved by the use of inverse and direct PCR. The complete DNA sequencing of the gene revealed that the lip-SBRN2 gene contains 576 nucleotides which corresponded to 192 deduced amino acids. The purified enzyme was homogeneous with the estimated molecular mass of 19 kDa as determined by SDS-PAGE and gel filtration. The Lip-SBRN2 was stable in a pH range of 9-11 and temperature range of 45-60 °C. The enzyme was a non metallo-monomeric protein and was active against pNP-caprylate (C8) and pNP-laurate (C12) and coconut oil. The Lip-SBRN2 exhibited a high level of activity in the presence of 108% benzene, 102.4% diethylether and 112% SDS. It is anticipated that the organic solvent and detergent tolerant enzyme secreted by Bacillus sp. RN2 will be applicable as catalysts for reaction in the presence of organic solvents and detergents.  相似文献

Most cases of acute gastroenteritis and foodborne disease are not ascertained by public health surveillance because the ill person does not always seek medical care and submit a stool sample for testing, and the laboratory does not always test for or identify the causative organism. We estimated the total burden of acute gastroenteritis in Miyagi Prefecture, Japan, using data from two 2-week cross-sectional, population-based telephone surveys conducted in 2006 and 2007. To estimate the number of acute gastroenteritis illnesses caused by Campylobacter, Salmonella, and Vibrio parahaemolyticus in Miyagi Prefecture, we determined the number of cases for each pathogen from active laboratory-based surveillance during 2005 to 2006 and adjusted for seeking of medical care and submission of stool specimens by using data from the population-based telephone surveys. Monte Carlo simulation was used to incorporate uncertainty. The prevalence of acute gastroenteritis in the preceding 4 weeks was 3.3% (70 of 2,126) and 3.5% (74 of 2,121) in the winter and summer months, yielding an estimated 44,200 episodes of acute gastroenteritis each year in this region. Among people with acute gastroenteritis, the physician consultation rate was 32.0%, and 10.9% of persons who sought care submitted a stool sample. The estimated numbers of Campylobacter-, Salmonella-, and V. parahaemolyticus -associated episodes of acute gastroenteritis were 1,512, 209, and 100 per 100,000 population per year, respectively, in this region. These estimates are significantly higher than the number of reported cases in surveillance in this region. Cases ascertained from active surveillance were also underrepresented in the present passive surveillance, suggesting that complementary surveillance systems, such as laboratory-based active surveillance in sentinel sites, are needed to monitor food safety in Japan.  相似文献

Deposited road particles (DRPs) collected from 13 heavily traveled roadways in an urban area of Southern Lake Biwa, Japan, were analyzed for polycyclic aromatic hydrocarbons (PAHs) in seven different particle size fractions (< or = 20 to 1000-2000 microm) and evaluated for the aryl hydrocarbon receptor (AhR) ligand activity by using a yeast bioassay. The mean compositions of individual PAHs to total PAH concentrations in different particle size fractions were 19-21% for pyrene, which was the most dominant component, 14-16% for fluoranthene, and 7-13% for benzo[g,h,i]perylene, which were the next dominant components. The total PAH distribution pattern in different particle size fractions of DRPs was different from the organic matter distribution pattern which increased with decreasing particle size of DRPs, and could be explained by the differences in their sources. Moreover, AhR ligand activities were observed in the DRP extracts of all size fractions. The activity of the DRP extracts from the smallest size fraction was approximately 5 times more potent than that of the largest size fraction in a yeast AhR ligand activity assay. The mean contribution (%) of benzo[k]-fluoranthene, chrysene, benzo[b]fluoranthene, and benzo[a]pyrene to the AhR activity of DRP extracts in all size fractions was 1.43-4.11%, 1.63-3.53%, 0.63-1.69%, and 0.31-1.42%, respectively. Although the contribution of PAHs to AhR ligand activity presented in the DRP extracts was relatively low (below 10%), it may be best to remove these DRPs before discharge to receiving water environments.  相似文献

Dyeing wastewater collected in Kyoto city, Japan, was investigated for the occurrence of aryl hydrocarbon receptor (AhR) ligands by using an AhR-responsive reporter gene assay. Concentrated extracts of wastewater samples elicited a dose-dependent increase in AhR ligand activity, and several hydrophobic HPLC fractions of the extracts were highly effective in inducing AhR ligand activity. Three potential AhR ligands were isolated from these fractions and identified to be Disperse Red 92, Disperse Yellow 64, and 3'-hydroxybenzo[b]quinophthalone by using HPLC and LC-MS/MS. Disperse Red 92, which has also been detected in the treated effluent from a sewage plant receiving the wastewater, is an anthraquinone disperse dye showing weak AhR binding affinity in the assay. Disperse Yellow 64 and 3'-hydroxybenzo[b]quinophthalone are quinoline disperse dyes capable of activating the AhR at nanomolar concentrations. In particular, Disperse Yellow 64 is a highly potent AhR ligand that was 3 times more effective in inducing AhR ligand activity than beta-naphthoflavone in the assay. Quinoline disperse dyes are suggested to be a new class of xenobiotic AhR ligands which pose a danger to aquatic biota and human health.  相似文献

In order to determine the ferrous and ferric ion capacities: 3
for an MgO-saturated MgO + CaO + Al2O3 slag, two experiments were carried out at 1873 K: (1) the distribution of iron between Fe x O dilute slags of the system and Pt + Fe alloys under controlled atmosphere, and (2) the equilibrium among molten iron or iron alloys, magnesiowustite, and molten slags. Although the activity of iron and the partial pressure of oxygen in each experiment are remarkably different, the values of the ferrous and ferric ion capacities agree well with each other. The influence of the MgO:CaO:Al2O3 ratio on the values of and was found to be limited within the experimental composition range. Using and , the relationship between total iron content, (pct Fe T ), and partial pressure of oxygen, , under iron saturation was calculated. The change in log with respect to the bulk slag composition is less than 0.2 within the range of (pct Fe T ) < 5.  相似文献

To theoretically explore amorphous materials with a sufficiently low dielectric loss, which are essential for next-generation communication devices, the applicability of a nonequilibrium molecular dynamics simulation employing an external alternating electric field was examined using alkaline silicate glass models. In this method, the dielectric loss is directly evaluated as the phase shift of the dipole moment from the applied electric field. This method enabled us to evaluate the dielectric loss in a wide frequency range from 1 GHz to 10 THz. It was observed that the dielectric loss reaches its maximum at a few THz. The simulation method was found to qualitatively reproduce the effects of alkaline content and alkaline type on the dielectric loss. Furthermore, it reasonably reproduced the effect of mixed alkalines on the dielectric loss, which was observed in our experiments on sodium and/or potassium silicate glasses. Alkaline mixing was thus found to reduce the dielectric loss.  相似文献

Phase relations in the composition range from UO2+x to U3O8?z were studied by electrical-conductivity measurements and X-ray diffraction in the ranges 1025°C ? T ? 1140°C and 10?7atm ? PO2 ? 10?3atm. The plot of log σ versus log PO2 showed straight lines with distinct slopes, which corresponded to four regions (UO2+x, U4O9?y, U4O9+y and U3O8?z). The existence of the hyperstoichiometric U4O9+y phase was suggested in the temperature range from 1025 to 1126°C. The peritectoid temperature (U4O9±y = UO2+x + U3O8?z) was estimated to be present between 1126 and 1131°C. The partial free enthalpies and entropies for the two-phase equilibrium (U4O9+y + U3O8?z, and U4O9?y + UO2+x) were calculated and compared with previous results. From the dependence of the electrical conductivity on the oxygen partial pressure the nonstoichiometric defect structures of UO2+x and U4O9±y were interpreted as consisting of doubly charged oxygen interstitials (Oi'') and doubly charged oxygen vacancies (VO''). At room temperature, the homogeneity range of the U4O9 phase was investigated with a Debye-Scherrer camera.  相似文献

This article evaluates the performance of flexible manufacturing systems with finite local buffers and fixed or dynamic routing rules, and addresses the optimal design or system configuration problem of maximizing the system throughput. The costs include machine cost, part (or pallet) cost, and local buffers cost. First, the system throughputs and their behaviors are considered with both queueing network analysis and simulation, and it is shown for a fixed routing model that the system throughput in the case of finite local buffers is greater than in the case of infinite local buffers. For a fixed versus dynamic routing rule, it is also found that the throughput in the former case can be close to the one in the latter case by changing the setting parameters. Next, the design problems of maximizing the system throughput are considered numerically for fixed and dynamic routing cases. Then, it is seen that better combination of design variables is a class of the monotonicity in local buffers, service rates, and routing probabilities.  相似文献

Well-defined La2Sn2O7 with a phase-pure pyrochlore structure was produced by hydrothermal synthesis at temperatures as low as 200°C. Production of phase-pure La2Sn2O7 requires a pH above 10, and higher pH accelerates the crystallization process. The synthesis produced spherical particles of average particle size ∼0.59 μm (±0.12) and surface area ∼14.1 m2/g. SEM and TEM observation for morphologic evolution and kinetic analysis during crystallization indicated that La2Sn2O7 formation probably proceeds via a two-step reaction. First a transient dissolution–precipitation occurs. Then the primary crystallites aggregate because of their colloidal instability, and heterocoagulation with the lanthanum hydrous oxide precursor particles also occurs. The sluggish reaction rate at the later stage of reaction is characterized by an in situ transformation, where the soluble tin species is diffused through the porous La2Sn2O7 aggregates to react with entrapped lanthanum precursors.  相似文献

The modeling of particle aggregation under a simple shear flow and the extension of the model to a stirred vessel is described. The model quantitatively demonstrates the change of the number of aggregates with time for each shear rate. This number increased with higher shear rate and, conversely, the aggregate size became small when raising the shear rate. This was because aggregates were broken by the stronger shear force. The number of aggregates for different impellers was determined. The shear rate was back‐calculated from the experimentally obtained aggregate size and the model equation. This shear rate was different from that estimated from the Metzner‐Otto method, consequently, some revisions of the Metzner‐Otto equation might be necessary for its application to particle aggregation.  相似文献
