Purpose. 1. To determine properties of the estimated variance component for the subject-by-formulation interaction (2 D) in investigations of individual bioequivalence (IBE), and 2. to evaluate the prevalence of interactions in replicate-design studies published by FDA. Methods. Four-period crossover studies evaluating IBE were simulated repeatedly. Generally, the true bioequivalence of the two formulations, including 2 D= 0, was assumed, 2 D was then estimated in a linear mixed-effect model by restricted maximum likelihood (REML). The same method was applied for estimating 2 D for the data sets of FDA. Results. 1.D estimated by REML was positively biased. The bias and dispersion of the estimated Dincreased approximately linearly with the estimated within-subject standard deviation for the reference formulation (WR). Only a small proportion of the estimated D exceeded the estimated WR. 2. Distributions of the estimated D were evaluated. At WR = 0.30, a level of estimated D= 0.15 was exceeded, by random chance, with a probability of about 25%. 3. Importantly, the behaviour of the 2 D values estimated from the FDA data sets was similar to that exhibited by the simulated estimates of 2 D which were generated under the conditions of true bioequivalence. Conclusions. 1. D estimated by REML is biased; the bias increases proportionately with the estimated WR. Consequently, exceeding a fixed level of D (e.g., 0.15) does not indicate substantial interaction. 2. The data sets of FDA are compatible with the hypothesis of 2 D = 0. Consequently, they do not demonstrate the prevalence of subject-by-formulation interaction. Therefore, it could be sufficient and reasonable to evaluate bioequivalence from 2-period crossover studies.  相似文献

Summary Sympathetic efferent pathways and alpha-adrenergic receptivity were investigated in one patient with spinal cord transection (D1 level) and orthostatic hypotension. The lack of increase in catecholamine plasma levels during orthostasis and the paradoxical pressor effect of clonidine (2 g/kg orally) suggested complete interruption of efferent sympathetic pathways.The pressor response to exogenous noradrenaline was significantly higher in the patient than in 6 normal controls (0.09 vs 0.72 g·kg–1), indicating supersensitivity of vascular -adrenoceptors. The platelet 2-adrenergic receptor number, measured with [3H]yohimbine, was 507 in the patient vs 178 fmol·mg–1 protein in controls. The increase in systolic blood pressure induced by 10 mg midodrine, a specific 1-agonist, was significantly higher in the patient (=56 mm Hg) than in controls (=15 mm Hg).The results indicate that in the patient there was -adrenergic supersensitivity both of 1- and 2-adrenoceptors. This led to successfully oral treatment of the orthostatic hypotension with clonidine 150 g bd and midodrine 10 mg bd.  相似文献

Summary The present paper examines the roles of postjunctional 1- and 2-adrenoceptors for the noradrenaline (NA)-induced neurogenic contractile response to field stimulation mainly with 1–100 pulses at 2 or 20 Hz, in the tail artery of adult normotensive rats. Pharmacological tools were employed to isolate and characterize the 1- and 2-adrenoceptor-mediated components of this response. The degree to which the drugs influenced NA release or reuptake was assessed by their effects on the electrochemically determined, stimulation-induced rise in the NA concentration at the innervated outer surface of the media. This response was unaffected by ,-methylene ATP (10 M) or suramin (500 M), added to desensitize or block P2-purinoceptors, respectively prazosin (0.1 M) or SK&amp;F 104078 (6-chloro-9-[(3-methyl-2-butenyl)oxyl]-3-methyl-1H-2, 3, 4, 5-tetrohydro-3-benzazepine, 0.1 M), used to block postjunctional 1- and 2-adrenoceptors respectively, nifedipine (10 M), blocker of Ca2+ influx through L-type channels, and ryanodine (10 M), which blocks mobilization of Ca2+ from intracellular stores; it was moderately enhanced by yohimbine (0.1 M), blocker of pre- and postjunctional 2-adrenoceptors, and strongly enhanced by cocaine (3 M) or desipramine (1 M), blockers of NA reuptake. Judging from their inhibitory effects on the contractile responses to the 1- and 2-adrenoceptor agonists, phenylephrine andxylazine, prazosin (0.1 M)and SK & F 104078 (0.1 M) could be used to selectively block 1- and 2-adrenoceptors respectively, while yohimbine (0.1 M) was less selective, strongly depressing 2- and slightly depressing 1-adrenoceptor-mediated responses. The 1-adrenoceptor-mediated component of the contractile response to short trains at 20 Hz was fast in onset, brief in duration and abolished by ryanodine; that mediated by 2-adrenoceptors was more delayed, prolonged and insensitive to ryanodine. Both components were dose-dependently depressed by nifedipine (0.1–10 M). The small contractile responses to single pulses, or up to 50 pulses at 2 Hz, or short train (< 4 pulses) at 20 Hz, were more markedly depressed by 0.1 M yohimbine or SK & F 104078 than by 0.1 M prazosin and, hence, mediated mainly by 2-adrenoceptors. The reverse was true of the much larger response to longer trains at 20 Hz, which thus probably was mediated mainly by 1-adrenoceptors. Cocaine or desipramine, as well as ,-methylene ATP or suramin, amplified both components of the NA induced contractile response especially that mediated via a1-adrenoceptors and caused by single pulses or short trains.The main conclusions are (i) that the small NA-induced contractile responses of this artery to single pulses, or pulses at low frequency, or in short trains at high frequency, are mediated mainly via 2-, and the larger responses to longer trains at high frequency increasingly via 1-adrenoceptors, (ii) that the 1- and 2-adrenoceptor-mediated components interact cooperatively, probably at least in part by utilizing two different pathways to increase the intracellular Ca2+, (iii) that neuronal reuptake of NA strongly restricts both components of the NA-induced contraction, especially the 1-adrenoceptor-mediated response to single pulses or short trains, and (iv) that both components of the NA-induced contraction, especially that mediated by 1-adrenoceptors, may be depressed by ATP released by field stimulation and acting via P2x-purinoceptors on smooth muscle. Based on these results a novel working hypothesis is proposed, in which it is assumed that the geometry of NA-mediated neuromuscular transmission in this vessel varies with the frequency and number of impulses in a stimulus train.Correspondence to J.-X. Summary The postsynaptic -adrenoceptors in rat aorta and in pithed rat were investigated according to their sensitivity to nine -adrenergic agonists and to the selective antagonists yohimbine (2) and prazosin (1) and the non-selective one, phentolamine. In addition, in radioligand binding studies, the affinity and selectivity of the drugs were determined on rat cerebral cortex using [3H] yohimbine and [3H] prazosin.On rat aorta, prazosin is 1,000 times more potent than yohimbine against each -adrenoceptor agonist, whether 1- or 2-selective. Rat aorta probably contains only 1-adrenoceptors.Pressor effects in pithed rats are mediated by post-junctional 1- and 2-adrenoceptors. The dose-response curve for -methylnorepinephrine in the presence of prazosin, using Hofstee's plots, revealed 1- and 2-adrenoceptors, respective proportions being 80.5 and 19.5%  相似文献

The ATP-induced increase in tritium outflow from cultured chick sympathetic neurons prelabelled with [3H]-noradrenaline was investigated.Seven days-old dissociated cell cultures of embryonic paravertebral ganglia, loaded with [3H]-noradrenaline (0.05 M), were superfused in the presence of (+)-oxaprotiline and exposed to ATP, ATP-analogues, or 1,1-dimethyl-4-piperazinium (DMPP) for 2 min. ATP (3 LM-3 mM), 2-methylthio-ATP (3–100 M), as well as DMPP (10 and 100 M) induced a significant overflow of tritium. The EC50-value of ATP was 20 M. Both the ATP-induced and the DMPP-induced tritium overflow was Ca2+-dependent and sensitive to tetrodotoxin (0.3 M) and -conotoxin (0.1 M); in addition, it was inhibited by the 2-adrenoceptor agonist 5-bromo-6-(2-imidazoline-2-ylamino)-quinoxaline (UK-14,304; 1 M). The effects of ATP and DMPP were not additive. The ATP-induced as well as the DMPP-induced overflow of tritium was diminished by the P2-purinoceptor antagonists suramin (300 M) and reactive blue 2 (3 M); in all 4 cases, the inhibition amouted to approximately 40%. The tritium overflow induced by ATP or DMPP was almost abolished by the nicotinic receptor antagonist mecamylamine (10 M) and markedly inhibited by hexamethonium (100 M). Neither ATP nor electrical stimulation caused an overflow of tritium from cultures loaded with [3H]-choline.The results suggest that ATP at molar concentrations induces noradrenaline release from cultured chick sympathetic neurons via an action on a subclass of the nicotinic cholinoceptor.  相似文献

Summary Three inotropic agents, ASL-7022, dobutamine and dopamine, were evaluated for their -and -adrenoceptor mediated effects in vitro in a variety of isolated organs and in radioligand binding studies. All compounds were 1-adrenoreceptor agonists in rat and guinea pig aortae, but the rank orders of potency were exactly opposite in these two tissues. Only the rank potency order of dobutamine>ASL-7022>dopamine obtained in rat aorta was consistent with the results obtained in radioligand binding studies to 1-adrenoreceptors in rat cerebral cortex and to previous results obtained in vivo in the pithed rat. The results obtained in guinea pig aorta did not parallel the radioligand binding studies in rat brain or our previous results in pithed rat, and suggests that species differences exist between postsynaptic vascular 1-adrenoreceptors in rat and guinea pig aorta, consistent with previous conclusions. ASL-7022 was found to be a potent 2-adrenoreceptor agonist in field-stimulated guinea pig ileum, and was approximately 10-fold more potent than dobutamine in this respects, which was also confirmed by radioligand binding studies to 2-adrenoreceptors in rat cerebral cortex. The 1-adrenoreceptor mediated effects of these compounds were evaluated in guinea pig atria, where the rank order of potency was dobutamine>ASL-7022>dopamine. An identical rank order of affinity was established for these compounds by displacement of 3H-dihydroalprenolol from 1-adrenoreceptors in rat cerebral cortex. The 1-adrenoreceptor mediated effects of dobutamine and ASL-7022 in guinea pig atria were completely direct in nature and not secondary to the release of endogenous catecholamines. In contrast, a major component of the 1-adrenoreceptor mediated tachycardia produced by dopamine in guinea pig atria was indirect in nature as evidenced by the marked attenuation in potency that occurred following catecholamine depletion with reserpine. All three compounds elicited 2-adrenoreceptor mediated inhibition of tone in rat uterus, with the rank order of potency being ASL-7022>dobutamine>dopamine. Again, this rank order of 2-adrenoreceptor potency was also reflected in 2-adrenoreceptor affinity as assessed by displacement of 3H-dihydroalprenolol from 2-adrenoreceptors in rat cerebellum. Based on these results, it may be concluded that for -adrenoceptors, dobutamine is a selective 2-adrenoreceptor agonist, ASL-7022 is a selective 2-adrenoreceptor agonist, and dopamine is a nonselective -adrenoceptor agonist. For -adrenoceptor mediated effect, ASL-7022 is a selective 2 agonist, while dobutamine and dopamine are nonselective -adrenoceptor agonists. It is likely that the complex inotropic and hemodynamic activities of ASL-7022, dobutamine and dopamine result from the sum of their individual effects at the -and -adrenoceptor subtypes.  相似文献

Summary For model identification and parameter estimation in the framework of linear pharmacokinetics it is most often assumed that the disposition function is a finite sum of exponential functions with time constants i and associated coefficients Ci. Least-square fitting procedures are used to estimate the coefficients Ci and the corresponding discrete locations i on the -axes.This work presents an alternative approach. It does not assume that the non-zero coefficients are located at sharply defined values of , but that they are represented by a continuous function h(), the spectrum of the disposition function. This turns the non-linear least-square problem into a linear problem, which is known to be as so-called ill-posed. Regularisation methods have been developed in recent years as suitable tools for the treatment of such ill-posed problems.Application of Tikhonov regularisation to the case of the bolus kinetics of propofol in 8 volunteers is demonstrated. In 7 of the 8 cases a spectrum with 4 to 5 peaks was found, and in one volunteer there were only 2 peaks. All spectra with more than 2 peaks showed negative values of h().The method used is described and the results are compared with those of conventional compartment analysis.  相似文献

Summary In the isolated rat vas deferens stimulated at 0.2 Hz, a series of 2, 3-, and 5-substituted adenine nucleotides all inhibited the twitch responses, their actions being potentiated by the nucleoside transport inhibitors, HNBTGR, NBMPR and dipyridamole.The metabolism of these nucleotides was examined utilising HPLC analysis of the bathing medium after exposure to 30 M nucleoside or nucleotide for 5 min. 5-AMP, 5-ADP, 5-ATP, and NAD+ were all partially hydrolysed to adenosine, the relative extent of this being 5-AMP>5-ADP=5-ATPNAD+. However, the other nucleotides examined were not detectably converted to adenosine or to adenosine deamination products.These results indicate that the 2-, 3- and 5-substituted nucleotides studied act at a P1-purinoceptor in rat vas deferens to inhibit neurotransmission and, with the exception of 5-AMP, 5-ADP, 5-ATP and NAD+, all appear to act directly at this receptor. However, the 5-adenine nucleotides (AMP, ADP and ATP) and NAD+ all appear to act at least partially indirectly subsequent to their hydrolysis to adenosine.Abbreviations. Summary Presynaptic 2-autoreceptors in rat and rabbit brain cortex were compared by means of antagonists and agonists. Brain cortex slices were preincubated with [3H]-noradrenaline and then superfused and stimulated by 3 (rat) or 4 (rabbit) pulses at a frequency of 100 Hz.The 2-adrenoceptor agonist bromoxidine (UK 14 304) reduced the electrically evoked overflow of tritium with EC50 values of 4.5 nmol/l in the rat and 0.7 nmol/l in the rabbit. The antagonists phentolamine, 2-[2H-(1-methyl-1,3-dihydroisoindole)methyl]-4,5-dihydroimidazole (BRL 44408), rauwolscine, 1,2-dimethyl-2,3,9,13b-tetrahydro-1H-dibenzo(c,f)imidazo(1,5-a)azepine (BRL 41992), 2-(2,6-dimethoxyphenoxyethyl)aminomethyl-1,4-benzodioxane (WB 4101), 6-chloro-9-[(3-methyl-2-butenyl)oxy]-3-methyl-1H-2,3,4, 5-tetrahydro-3-benzazepine (SKF 104078), imiloxan, prazosin and corynanthine did not per se increase the evoked overflow of tritium but shifted the concentration-inhibition curve of bromoxidine to the right in a manner compatible with competitive antagonism. Up to 4 concentrations of each antagonist were used to determine its dissociation constant KD. The KD values correlated only weakly between the rat and the rabbit. Dissociation constants KA of bromoxidine were calculated from equieffective concentrations in unpretreated brain slices and slices in which part of the 2-adrenoceptors had been irreversibly blocked by phenoxybenzamine. The KA value was 123 nmol/l in the rat and 7.2 nmol/l in the rabbit.The results confirm the species difference between rat and rabbit brain presynaptic 2-autoreceptors. Comparison with data from the literature indicates that the rat brain autoreceptors can be equated with the 2D subtype as defined by radioligand binding, whereas the rabbit brain autoreceptors conform to the 2A subtype. For example, the antagonist affinities for the rat autoreceptors correlate with their binding affinities for the gene product of 2-RG20, the putative rat 2D-adrenoceptor gene (r = 0.97; P<0.01), but not with their binding affinities for the gene product of 2-C10, the putative human 2A-adrenoceptor gene. Conversely, the rabbit autoreceptors correlate with the 2-C10 (r = 0.98; P<0.001) but not with the 2-RG20 gene product. Since presynaptic 2-autoreceptors are also 2D in rat submaxillary gland and perhaps vas deferens and 2A in rabbit pulmonary artery, the possibility arises that the majority of 2-autoreceptors generally are 2D in the rat and 2A in the rabbit. Summary The effect of prostaglandin (PG) E2, F2, the thromboxane A2 mimetic U46619 (9,11-dideoxy-9,11-methanoepoxy-prostaglandin F2) and the prostacyclin mimetic iloprost was investigated in feline and rat pial arteries in-situ using perivascular microapplication and measurement of vascular diameter. U 46619 induced a concentration-dependent vasoconstriction with a maximal response of 24% at 10–6 mol · l–1 and an EC50 of 4.9 × 10–8 mol · l–1. This effect was inhibited by the thromboxane receptor blocking drugs AH23848B (([1(z), 2ß,5]-(±)-7-[5-[(1,1-(biphenyl)-4-yl]methoxy]-2-(4-morpholinyl)-3-oxocyclo-pentyl]-4-heptenoic acid)) and EP092 ((±)-5-endo-(6-carboxyhex-2z-enyl)-6-exo[1-[N-(phenylthiocarbamoyl)-hydrazano]-ethyl]-bicyclo[2,2,1]-heptane)), indicating the involvement of thromboxane-prostanoid-receptors. PGF2 produced vasodilatation in cats with an increase in vessel diameter of 30% at 10–3 mol · l–1, but constricted rat pial arteries concentration-dependently with a maximal response of 23% at 10–4 mol · l–1. PGE2 and iloprost induced concentration-dependent (10–9 - 10–5 mol · l–1) dilatations with apparent maximal responses of 39% and 34% at 10–5 mol · l–1, respectively. The corresponding EC50 values were 2.45 × 10–7 mol · l–1 (PGE2) and 3.5 × 10–7 mol · l–1 (iloprost). These data demonstrate the presence of prostanoid receptors mediating constriction and dilatation under in-situ conditions. Since the norepinephrine- but not the potassium-free artificial cerebrospinal fluid-induced constriction in cats was blocked by the thromboxane-prostanoid-receptor antagonists AH 23848 B and EPO 92, it is likely that the effect of norepinephrine is, at least partly, mediated by the release of an endogenous prostanoid acting at constriction-inducing thromboxane-prostanoid-receptors. The sites of action at which ATP elicits contraction of the rat vas deferens were studied by means of the P2-purinoceptor antagonists pyridoxalphosphate-6-azophenyl-2,4-disulfonic acid (PPADS), suramin and reactive blue 2.Increasing concentrations of PPADS (up to 1 mM), suramin (up to 1 mM) and reactive blue 2 (up to 320 M) reduced and eventually abolished contractions elicited by the P2X-purinoceptor-selective agonist ,-methylene ATP 3 M with IC50 values of 2.1, 10.1 and 27.0 M, respectively. In contrast, PPADS and suramin caused only a partial inhibition of contractions elicited by ATP 1 mM, maximal reduction by about 40%, IC50 values 1.3 and 5.0 M, respectively; reactive blue 2 did not change ATP-induced contractions. In tissues exposed to PPADS 320 M throughout, increasing concentrations of reactive blue 2 or suramin decreased contractions elicited by ATP 1 MM, IC50 values 2.6 and 14.5 M, respectively. In tissues exposed to suramin 320 M throughout, increasing concentrations of PPADS decreased contractions elicited by ATP 1 mM, IC50 37.9 M, whereas reactive blue 2 slightly enhanced these contractions. In tissues exposed to reactive blue 2 100 M throughout, increasing concentrations of PPADS reduced contractions elicited by ATP 1 MM, IC50 26.6 M, whereas suramin caused no change. Pre-exposure to ,-methylene ATP 1 M to desensitize P2X-purinoceptors reduced the response to ATP 1 mM by 91% in otherwise untreated tissues, but did not reduce the response to ATP 1 mM in tissues exposed throughout to PPADS 320 M, suramin 320 M or reactive blue 2 100 M. Neither PPADS nor suramin nor reactive blue 2 altered contractions elicited by KCl 35 mM. The P1-purinoceptor antagonist 8-(p-sulfophenyl) theophylline 100 M did not change contractions elicited by ,-methylene ATP 3 M or ATP 1 mM.It is concluded that ATP 1 mM elicits contraction of the rat vas deferens through three sites: P2X-purinoceptors which are blocked by PPADS, suramin and reactive blue 2; P2Y-purinoceptors blocked by reactive blue 2 and suramin but resistant to PPADS; and non-P2X-non-P2Y-purinoceptors blocked by PPADS but resistant to inhibition by suramin and reactive blue 2. ICV cromakalim, a K+ channel opener, produced antinociception. This effect was completely antagonized by ICV glibenclamide, a selective adenosine triphosphate-sensitive K+ channel (KATP channel) blocker. Furthermore, direct opening of central KATP channels by ICV cromakalim increased the spinal noradrenaline (NA) turnover. On the other hand, the antinociception induced by ICV morphine ( opioid agonist), but not ICV U-50,488H ( opioid agonist) was markedly potentiated by cromakalim. These findings suggest that the opening of central KATP channels may elicit the antinociceptive effect and activate the descending NAergic pathway, and central KATP channels play an important role as a modulator of the antinociception induced by agonists but not agonists.  相似文献

TMB-8 has been characterized as an inhibitor of the release of Ca+ from intracellular pools. We have studied the modification of the pressor responses to selective l-adrenoceptor agonists (methoxamine and phenylephrine), and to selective 2-adrenoceptor agonists (B-HT 920 and B-HT 933) in pithed rats, produced by TMB-8. We have compared this modification with that produced by the calcium antagonist nifedipine. Nifedipine (100 g/kg, 300 g/kg, and 1000 g/kg) inhibited in a dose-dependent manner the pressor responses to the 1- and 2-adrenoceptor agonists, the dose-response curves to the 2-adrenoceptor agonists being shifted further to the right. TMB-8 at a dose of 3000 g/kg did not modify the pressor effects of the l-adrenoceptor agonists, and neither did it reinforce the inhibition of such responses produced by nifedipine. By contrast, TMB-8 pretreatment (0.03 g/kg, 0.3 g/kg, 3 g/kg, 30 g/kg, 300 g/kg and 3000 g/kg) inhibited the responses to both 2-adrenoceptor agonists, the inhibition being more pronounced with B-HT 920. A similar effect was obtained with 0.03 g/kg TMB-8 and 0.3 g/kg TMB-8, particularly in the case of B-HT 920. It was stronger with higher doses, but similar for all doses over 3 g/kg. The inhibition of the pressor responses mediated by the stimulation of 2-adrenoceptors by TMB-8 was less in rats treated with the Ca2+ entry promoter BAY K 8644 (300 g/kg), and could also be reduced by the continuous infusion of CaCl2 (0.25 g/min). These results suggest that in pithed rats TMB-8 may also behave as an inhibitor of the Ca+ influx into vascular smooth muscle.  相似文献

Summary Long-term -adrenergic stimulation has been shown to desensitize the -adrenoceptor/adenylyl cyclase signalling pathway at both the receptor and the G-protein level. To further elucidate the cellular mechanism of G-protein regulation we investigated the influence of prolonged infusion of isoprenaline (2.4 mg/kg·d) on myocardial mRNA levels of different G-protein -subunits in rats. For comparison rats were treated with triiodothyronine (T3; 0.5 mg/kg·d) which induces cardiac hypertrophy like isoprenaline but has different effects on the adenylyl cyclase system. Isoprenaline- and T3-treated animals developed an increase in heart/body weight ratio of 41±3% and 27±4%, respectively (P<0.05). Isoprenaline increased myocardial total RNA concentration by 39±6% (P<0.05). Hybridization with 32P-labeled rat cDNAs demonstrated an expression rank order of Gs-mRNA>Gi-2-mRNA>Gi–3-mRNA and no detectable expression of Gi–1-mRNA in rat myocardium. mRNA levels of Gs Gi–2 and Gi–3 were 36.9±1.28, 10.7±1.07 and 3.7±0.19 pg/g total RNA, respectively. Isoprenaline increased Gi–2 and Gi–3-mRNA concentrations per g total RNA by 49±18% and 27±710, respectively (P<0.05). This effect was abolished by simultaneously administered propranolol (9.9 mg/kg·d), indicating a,-adrenoceptor-mediated mechanism. In contrast, T3-induced cardiac hypertrophy was not accompanied by changes in Gi-mRNA expression. Gsa-mRNA levels were unaffected by either treatment.In conclusion, long-term stimulation with isoprenaline in vivo induces a -adrenoceptor-mediated increase in myocardial Gi–2 and Gi–3-mRNA without affecting Gs-mRNA. Purpose. We studied the feasibility of using the Kohlrausch-Williams-Watts stretched exponential function (KWW equation) to describe protein aggregation in lyophilized formulations during storage. Parameters representing mean aggregation time (a) and stretched exponential constant (a) were calculated according to the KWW equation by assuming that the time required for protein molecules to aggregate () varies because of the fact that protein aggregation occurs at a rate that depends on the degree of protein deformation resulting from stresses created during freeze-drying. The temperature dependence of the parameters near the glass transition temperature was examined to discuss the possibility of predicting protein aggregation by accelerated testing. Methods. Protein aggregation in lyophilized bovine serum -globulin (BGG) formulations containing dextran or methylcellulose, at temperatures ranging from 10 to 80°C, was followed by size-exclusion chromatography. Results. Non-exponential BGG aggregation in lyophilized formulations could be described by the KWW equation. The a and a parameters changed abruptly around the NMR relaxation-based critical mobility temperature for formulations containing dextran and methylcellulose. In the glassy state, in contrast, the a parameter of these formulations exhibited continuous temperature dependence. The parameter , as calculated from a and a, reflected differences in values between the two excipients. Conclusions. The results indicate that the parameter a is reflective of physical changes wihtin lyophilized formulations. Within the temperature range, during which no abrupt changes in a were observed, knowledge regarding the aand a parameters allows the rate of protein aggregation to be predicted. The parameter was found to be useful in comparing the protein aggregation behavior of formulations having different a and a values.  相似文献

IR and 1H NMR studies in CDCl3 and CCl4 of a series of tertiary aminoxylidides with the amino group in the 2 to 6 position of the acyl chain are described. Lidocaine, diethylaminoaceto-2,6-xylidide, forms an intramolecular five-membered ring hydrogen-bonded monomer at all concentrations in both solvents. -Diethyl-amino-propiono-2,6-xylidide forms an intramolecular six-membered ring hydrogen-bonded monomer in CDCl3 and CCl4 but a trans intermolecularly associated species is the major form present at high concentrations in CCl4. The longer-chain homologues are mixtures of nonassociated trans and cis monomers at low concentrations but associated trans forms predominate at high concentrations. Evidence for the presence of a hydrogen-bonded seven-membered ring intramolecular monomer in CDCl3 for -diethylaminobutyro-2,6-xylidide is presented. The relationship between the molecular conformation and the partition coefficient is discussed.  相似文献

Summary Evidence for a general role of phospholipase D in signal transduction is accumulating. In the present study, the activity of the enzyme was investigated in heart tissue under basal conditions and after addition of phorbol esters or aluminum fluoride (AlF inf4 sup– ; 10 mM NaF plus 10 M AlCl3). Atria of rats and chickens were incubated with [3H]-myristic acid in order to label preferentially phosphatidylcholine. Under basal conditions, the tissues generated choline and phosphatidic acid (PtdOH), the primary catalytic products of phospholipase D. When 0.5 or 2.0% ethanol was present, [3H]-phosphatidyl-ethanol (PETH) was rapidly formed at the expense of [3H]-PtdOH. This transphosphatidylation reaction is specific for phospholipase D activity. The basal formation of PETH was not inhibited by a Ca2+-free, EGTA-containing medium. - The phorbol ester 4-phorbol-12,13-dibutyrate (PDB), which is known to activate protein kinase C, enhanced the net formation of choline, whereas the inactive 4-phorbol-13-acetate (PAc) was ineffective. PDB (0.2 M), in contrast to PAc, also increased the formation of [3H]-PtdOH and, in the presence of ethanol, of [3H]-PETH. The PDB-evoked formation of PETH occurred again at the expense of PtdOH. Treshold and maximum effective concentrations of PDB were 10 nM and 0.2–0.6 M, respectively. The effects of PDB on either choline efflux and generation of PETH showed the same Cat+-dependency, i.e., both effects were blocked by a Ca2+-free, EGTA-containing medium, but not by a Ca2+-free medium without EGTA. In protein kinase C-deficient tissue which was prepared by pretreatment with 0.61 M PDB for 27 h, PDB failed to enhance the formation of PtdOH and PETH. - A1F4–, a known activator of G-proteins, increased not only the tissue content of inositol phosphates, but also markedly enhanced choline efflux and formation of [3H]-PtdOH and PETH. In conclusion, in mammalian and avian atria a high phospholipase D activity was found even under basal conditions. In the present study we have prepared crude, methanolic extracts of bovine lung and bovine brain and, using radioligand binding assays in conjunction with a number of simple chromatographic techniques, provided evidence for the presence of a non-catecholamine clonidine-displacing substance (CDS). The level of CDS in lung extracts (9 units/g wet weight n = 11) is approximately 3 times that in the brain extracts. Furthermore, the effect of the crude, methanolic extracts are selective for non-adrenoceptor, imidazoline (labelled by [3H]-idazoxan) and 2-adrenoceptor binding sites (labelled by [3H]-clonidine); both extracts are 5–10-fold more potent displacers of ligand binding to 2-adrenoceptors compared with binding to opiate receptors (labelled by [3H]-etorphine) and practically inactive against 1-adrenoceptor and muscarinic binding sites (labelled by [3H]prazosin and [3H]-quinuclidinyl benzilate, respectively). With the exception of the non-adrenoceptor, imidazoline binding assay, which used rat kidney membranes labelled by [3H]-idazoxan in the presence of the 2-adrenoceptor antagonist RS-15385-197, all radioreceptor assays involved bovine cerebral cortex membranes. Although the extracts contain catecholamines (brain only), histamine (lung only) and monovalent cations (both), which have the potential to interfere with the radioligand binding assays, their concentrations were too low to account for the effects observed. Preliminary attempts at purification of the extracts revealed that CDS activities from the two tissues are similar, i.e., practically insoluble in organic solvents at room temperature, not affected by either Sep-Pak C18 column or anion exchange resins but retained (along with the monovalent cations) by cation exchange resin. However, following chromatographic separation on a Biogel P2 column, the CDS-containing eluates are cation-free and exhibit qualitatively similar elution profiles. Future experiments will involve further purification of clonidine-displacing substance to characterize its interaction with 2-adrenoceptor binding sites in greater detail and establish whether it has biological activity consistent with the properties implied by its effects in radioligand binding assays.  相似文献

Presynaptic 2-autoreceptors in mouse atria were characterized in terms of the 2A, 2B, 2C and 2D subtypes. Segments of the atria were preincubated with 3H-noradrenaline and then superfused and stimulated electrically. The affinity of up to 16 antagonists for the autoreceptors was assessed as (1) pEC30% values, i.e. concentrations that increased previously autoinhibited release of 3H-noradrenaline (120 pulses, 3 Hz) by 30%, and (2) pKd values against the release-inhibiting effect of 5-bromo-6-(2-imidazolin-2-ylamino)-quinoxaline (UK 14,304) under conditions of no or little autoinhibition (2 trains of 20 pulses, 50 Hz, train interval 120 s).The pKd values correlated well with the pEC30% values (r = 0.98; P<0.001; slope of regression line 0.93), indicating that UK 14,304 and released noradrenaline modulated the release of noradrenaline through pharmacologically identical receptors. Comparison with antagonist affinities for (1) prototypic native 2 radioligand binding sites, (2) radioligand binding sites in COS cells transfected with 2 subtype genes, and (3) previously classified presynaptic 2-autoreceptors — all taken from the literature — indicated that the mouse atrial autoreceptors corresponded to the 2D subtype. For example, the pKd values at mouse atrial autoreceptors correlated closely with pKd values at native 2D binding sites in the bovine pineal gland (r = 0.96; P<0.001); with pKd values at 2D binding sites in COS cells transfected with the rat 2D gene (r= 0.85; P<0.01); and with pKd values at guinea-pig cerebral and atrial and mouse cerebral 2D-autoreceptors (r=0.96–0.98; P<0.001). The antagonist pKd values at mouse atrial autoreceptors correlated less with pKd values at 2A, 2B, and 2C sites.It is concluded that the presynaptic 2-autoreceptors in mouse atria are 2D This identification supports the hypothesis that at least the majority of 2-autoreceptors belong to the 2A/D pair of orthologous 2-adrenoceptors.  相似文献

Summary Dehydro-digitoxosides are metabolites of digitalis glycosides. In order to study their possible biological activity their affinity to (Na++K+)-activated ATPase was determined and compared with unchanged glycosides. Based on the dissociation constants of glycoside-enzyme-complexes, the affinity of the dehydro-digitoxosides ranged in the same order of magnitude as that of the native glycosides. Comparing mono-, bis-, and tris-digitoxosides of digitoxigenin (dt-1, dt-2, dt-3) and of digoxin (dg-1, dg-2, dg-3) with the corresponding dehydrodigitoxosides (3-dehydro-dt-1, 9-dehydro-dt-2, 15-dehydro-dt-3, 3-dehydro-dg-1 and 9-dehydro-dg-2, respectively) the dehydro-digitoxosides had lower affinities to the enzyme. The highest dissociation constants (K D)were found for 3-dehydro-dt-1 and 3-dehydro-dg-1. The half maximal inhibition of (Na++K+)-ATPase activity (I50) corresponded to affinity measurements in all but two cases: dehydro-dt-3 and dehydro-dt-2 showed very low I50 values.  相似文献
