斑翅蝗科四种蝗虫卵子发生阶段及种间差异的统计分析   总被引:7,自引:1,他引:6  
通过组织学观察与测量对斑翅蝗科(直翅目,蝗总科)中4种蝗虫卵子发生各阶段进行测量和分析比较,4种蝗虫分别为:大垫尖翅蝗Epacromius coerulipes(Ivan.)、黄胫小车蝗Oedaleus infernalis Sauss.、花胫绿纹蝗Aiolopus tamulus(Fabricius)和疣蝗Trilophidia annulata(Thunberg).结果显示,在蝗虫卵子发生过程中,卵母细胞体积和形态变化显著,呈现指数增长模式;细胞核体积增长在发育初期增长显著,后期变化不显著;滤泡细胞体积和形态及数量呈现周期性变化规律;进而比较种间差异,可见不同种蝗虫的卵子发生存在不同的发育进程.  相似文献

为探讨c-kit蛋白在蝗虫卵子发生过程中的表达和调控机制,应用免疫组织化学和统计分析等方法对网翅蝗科(直翅目,蝗总科)3种蝗虫卵子发生过程中8个代表性阶段c-kit蛋白表达进行观测和比较,3种蝗虫分别为:绿牧草蝗 Omocestus viridulus(Linnaeus),素色异爪蝗Euchorthippus unicolor(Ikonn.)和条纹异爪蝗Euchorthippus vittatus Zheng.结果显示蝗虫卵子发生第1~6阶段卵母细胞中有不同程度c-kit蛋白特异性表达,但随着卵黄发生的开始逐渐消失,而且3种蝗虫卵子发生过程中c-kit蛋白表达存在种间差异.以上结果提示c-kit蛋白在卵子发生中的表达暗示它参与和调控卵母细胞增殖与分化,此外c-kit蛋白表达种间差异说明在它的调控下不仅导致蝗虫卵子发生进程的迥异而且可能参与维系种间生殖隔离等机制.  相似文献

通过对吉林省四平地区6个蝗虫优势种绿牧草蝗Omocestuaviridulus(L.),黄胫小车蝗OedaleusinfernalisSauss,素色异爪蝗Euchorthippusunicolor(Ikonn.),条纹异爪蝗E.vittatusZheng,异翅负蝗AtractomorphaheteropteraB.Bienko和长翅素木蝗Shirakacrisshirakii(I.Bol.)的野外养殖,从成虫出现到死亡对6个蝗虫优势种卵巢季节动态变化进行了观察和测量研究。结果表明,(1)不同发育历期成虫体长变化差异不显著,而卵巢形态和体积变化差异显著;(2)卵巢发育具有明显的种间差异;(3)卵巢发育速度呈现先慢后快的增长模式;(4)6个蝗虫优势种卵巢发育符合5个发育级别的划分标准。  相似文献

记述采自中国山东省网翅蝗科异爪蝗属Euchorthippus Tarbinsky,1926 1新种,山东异爪蝗 Euchorthippus shandongensis sp. nov.。新种同素色异爪蝗 Euchorthippus unicolor(Ikonnikov,1913)近似,其区别特征为:头侧窝较长,长为宽的3.2倍;颜面隆起明显,具纵沟,中眼之下缩狭,往下明显宽大; 雄性前翅较长,明显超过肛上板基部;雄性下生殖板较短,侧面观长为宽的1.5倍; 阳茎基背片弓形深,两侧下端具尖突。模式标本保存于山东农业大学植保学院。  相似文献

采用常规染色体制片方法,对雏蝗属的模式种白边雏蝗Chorthippus albomarginatus(De Geer)和异爪蝗属的模式种素色异爪蝗Euchorthippus unicolor(Ikonnikov)的染色体核型进行了分析比较。结果表明,两种蝗虫的染色体数目均为2n(♂)=17=16+XO;常染色体为中部着丝粒(m,6条)和端部着丝粒(t,10条)两种类型,性染色体类型为端部着丝粒。二者的相似性显示出2属具有较近的亲缘关系,并且在进化过程中处于较近的发展阶段。但2种蝗虫的核型公式和染色体的相对长度组成则不相同,白边雏蝗K(2n,♂)=6m+11t=8L+4M+4S+XO;素色异爪蝗K(2n,♂)=6m+11t=8L+6M+2S+XO;且2种蝗虫性染色体的位次也有明显差别,白边雏蝗的性染色体为第9位,而素色异爪蝗则为第8位。由此看出,该2属的蝗虫存在的不同遗传特征。  相似文献

四平地区六种优势蝗虫精巢发育的动态变化   总被引:1,自引:1,他引:0  
在吉林省四平地区对6种优势蝗虫进行野外罩养,从成虫出现到死亡期间,对蝗虫精巢发育动态变化进行解剖学测量、统计分析和组织学观察。结果表明,蝗虫成虫刚出现时精巢发育水平较高,精巢长度显著增长但宽度增长差异不显著,随着虫体发育精巢体积达到峰值后出现下降趋势。不同种蝗虫精巢发育具有明显的种间差异,成虫出现时其精巢发育水平依次为长翅素木蝗>黄胫小车蝗>绿牧草蝗>素色异爪蝗>条纹异爪蝗>异翅负蝗,并且各蝗虫类群精巢发育的体积动态变化各不相同,体现出各蝗虫类群精巢生理功能的强弱。组织学观察显示,蝗虫发育不同时期精巢内以不同类型的精母细胞为主,精巢体积增长是精母细胞增长和成熟精子积累的结果,精巢体积下降为交配活动导致的大量精子外排以及其生理机能减退所至。  相似文献

异爪蝗属(Euchorthippus)是雏蝗属(Chorthippus) 的一个近缘属,两者在外部形态及地理分布上都十分接近。在异爪蝗属内已知的某些种,原先是被放在雏蝗属内的。到1925年,Tarbinsky以其跗节爪的左、右不等长为主要特征而区别于雏蝗属,并建立了以草原异爪蝗Eu.pulvinatus为属模的异爪蝗属。后来便以此特征来描述该属的新种。1964年以来,郑哲民陆续发表了五个新种,即按此特征来描述的。到目前为止,我国已报导的异爪蝗共有9个种(表1),分布范围基本同雏蝗属。  相似文献

A method is described which permits the preparation of descrete classes of oocytes of different sizes from all stages of oogenesis in Xenopus laevis. This technique is used in the determination of the content of microtubule protein in oocytes during the course of oogenesis. These experiments show that microtubule protein is present in oocytes of all sizes assayed and that the amount is simply related to the volume of the oocyte. In the largest oocytes microtubule protein constitutes 1% of the soluble protein and this amount does not change on maturation and fertilization. These results show that the changes occurring in the oocyte on maturation which allow the cytoplasm to support microtubule polymerization occur as a result of a modification of the pre-existing microtubule protein, not from protein synthesis de novo. These experiments also indicate that the synthesis of microtubule protein either form 'masked' mRNA or from newly synthesized mRNA plays an insignificant role in microtubule protein synthesis at maturation, ovulation and immediately post-fertilization.  相似文献

Cdc2 kinase is a catalytic subunit of maturation-promoting factor (MPF), a central factor for inducing the meiotic maturation of oocyte. To understand the role of Cdc2 kinase on the oocyte maturation in crustacean, a complete cDNA sequence of Cdc2 kinase was cloned from Chinese mitten crab Eriocheir sinensis and its spatial-temporal expression profiles were analyzed during oogenesis at RNA and protein levels. The crab Cdc2 cDNA (1364 bp) encodes for a 299 amino acids protein with calculated molecular weight of 34.7 kDa. The Cdc2 mRNAs level showed no significant change in the ovary during oogenesis, whereas higher protein level was found at previtellogenesis, late vitellogenesis and germinal vesicle breakdown (GVBD) stages. Two forms (35 kDa and 34 kDa) of Cdc2 proteins were simultaneously identified in ovary at all stages. Immunocytochemistry analysis revealed that Cdc2 proteins locate exclusively in ooplasm of previtellogenic oocyte, and then relocate into germinal vesicle at vitellogenesis stage and accumulate on meiotic spindle at oocyte maturation. These findings suggest that Cdc2 kinase has essential roles in inducing GVBD and generating meiotic apparatus during the crab oocyte maturation.  相似文献

The changes in distribution and density of mitochondria and the level of mitochondrial RNA during Drosophila oogenesis were studied simultaneously in the 3 cell types ie follicle cells, nurse cells and oocyte, making up the egg chamber. Up to stage 6, mitochondrial density (mitochondrial and cellular areas ratio) was elevated and increased similarly in both follicle and nurse cells. Thereafter the mitochondrial density of follicle cells continued to increase and that of the nurse cells declined markedly while the nurse cell mitochondria assembled in dense groups and decreased in size. This can be related to a transfer of nurse cell cytoplasm, including mitochondria, to the oocyte. In the oocyte from stage 4 to stage 7 we observed a significant decrease of the mitochondrial density due to the absence of mitochondrial biogenesis. Then the cytoplasm transfer caused mitochondrial density to increase up to the level found in the nurse cells at the end of oogenesis. The mature oocyte contains enough mitochondria to supply 15,000 somatic cells. Our results strongly suggest that the variations in size, distribution and density of mitochondria relate to the particular energetic requirements of the different cell types during the first half of oogenesis. Later they relate to the developmental requirements of the nurse cells and the oocyte, in particular the storage of mitochondria in the oocyte. The level of mitochondrial RNA was studied through in situ hybridization. Throughout oogenesis the follicle and nurse cell RNA evolved similarly. Up to stage 9, there was no change in RNA densities in these cells, suggesting a correlation with the cell volume and/or the nuclear DNA content. Thereafter the cellular RNA concentration declined rapidly. In the oocyte the RNA concentration evolved differently especially from stage 10 to the end, the RNA density being stabilized. This can be related to the injection of nurse cell mitochondria, followed by their assignment to reserve status. Our results suggest that the mt RNA density is under extramitochondrial control mechanisms.  相似文献

Reproductive division in termites is the most significant biological process that leads to the formation of caste‐specific differences in tasks and status. However, little is known about the mechanism of reproductive division that underlies caste differentiation. In the present study, ovarian development and stage‐specific apoptotic patterns are investigated during oogenesis in reproductive, worker and soldier termites Reticulitermes aculabialis Tsai & Hwang. The results show that the mean lengths of the ovaries of reproductives are two‐fold longer compared with those of workers and six‐fold longer compared with soldiers. By contrast to the reproductives, the process of oogenesis in the workers includes only the oogonium differentiation stage (stage I) and oocyte growth stage (stage II), and oogenesis in the soldiers stops at stage I. Vitelogenic oocytes (stage III) are absent from workers and soldiers. During stage II in the reproductives and workers, the layer of follicle cells has a thickness of 7.56 ± 0.52 and 2.81 ± 0.34 µm, respectively. In addition, there are significant differences in the number and size of the germ cells at the same stage in the various castes. The existence of two apoptotic patterns during oogenesis is demonstrated by the terminal deoxynucleotidyl transferase‐mediated dUTP nick end labelling (TUNEL) assay. First, the majority of the cells showing apoptosis occur at stage I of oogenesis in reproductives, workers and soldiers. Second, DNA fragmentation is demonstrated by TUNEL staining of the follicle cell layers and oocytes at stage II in reproductives. Finally, the proliferation activity of follicle cells in the reproductives is observed by 5‐bromo‐2′‐deoxy‐uridine labelling. The level of oogenesis may explain the significant discrepancies in the reproductive capacity among the reproductives, soldiers and workers. These large discrepancies are controlled by apoptosis during early oogenesis.  相似文献

Stages of oocyte development in Cynops pyrrogaster are defined, and changes of annulate lamellae in their fine structure, number, sizes and locations during oogenesis are described. The results show that two different types of annulate lamellae occur during oogenesis. One type differentiates in or at the periphery of vesicle-rich cytoplasm at the early stages of vitellogenesis and increases in number and size. The maximum number of about 40 stacks per median section of oocyte is reached at the stage of complete differentiation of the animal and the vegetal hemispheres. In these growing oocytes, all the stacks show elongate appearances and tetragonal arrangements of annuli as common characteristics. A second type of stacks of annulate lamellae is added anew in full-grown oocytes, increasing the number of stacks per median section of the oocyte to about 90. The new stacks occur in close contact with electron-dense bodies in the cytoplasm and have a massive appearance and hexagonal array of annuli. It is suggested that they appear coincidentally with the onset of oocyte maturation. The possible significance of the observed results is discussed.  相似文献
