PLK1基因沉默抑制HeLa细胞凋亡   总被引:3,自引:0,他引:3  
以高表达Polo样激酶1(Polo-like kinase 1,PLK1)的宫颈癌细胞系HeLa细胞为模型,观察针对PLK1基因的短发夹状RNA(short hairpin RNA,shRNA)对其凋亡和增殖的影响.设计并合成了针对PLK1的shRNA,将其导入构建的携带增强型绿色荧光蛋白(EGFP)的RNAi(RNA interference)表达载体中.通过 RT-PCR和Western 印迹分别检测HeLa细胞PLK1基因和蛋白水平的表达,以流式细胞仪和PI-Hochest双染法测细胞凋亡,MTT法检测细胞的增殖水平.成功构建了携带EGFP的RNAi表达载体pEGFP-H1.转染shRNA后,HeLa细胞PLK1的表达降低至30%.与对照组和空载体转染组相比,shRNA转染组的HeLa细胞凋亡率明显增加,其增殖活性则明显降低.本课题构建的RNAi表达载体便于观察靶基因的转染情况,且不影响H1启动子的体内转录.PLK1的基因沉默能明显增加HeLa细胞的凋亡,抑制该细胞的增殖,有可能为未来肿瘤的治疗找到新的靶点和有效途径.  相似文献

目的:以semliki森林病毒复制子为基础,构建一类可迅速高效表达shRNA的新型RNAi载体。方法:以Semliki森林病毒衍生的复制子载体pSFV1为骨架,用CMV IE启动子替换SP6启动子并在3′-UTR下游插入SV40 polyA转录终止子,在原26S亚基因组启动子后插入带有相应改良多克隆位点的shRNA表达元件,同时加入抗新霉素选择复合体,并去掉3′-UTR的重复序列。所获载体用于沉默EGFP基因,通过体外细胞转染、病毒颗粒制备、荧光显微镜观察、RT-PCR分析等初步验证、评估其效果。结果:构建了基于Semliki森林病毒复制子的新型RNAi质粒载体pSFV-RNAi Ready。经体外实验初步证实,该载体直接转染细胞,或与辅助载体共转染,制备成具有感染能力的重组病毒颗粒后使用,均可高水平表达shRNA,沉默目的基因。其中使用病毒颗粒抑抑效率可高达90%以上。结论:该载体的成功构建,可望显著拓宽SFV载体的应用范围,丰富RNAi实施手段,并用于相关科学研究及基因药物技术开发。  相似文献

RNA干扰(RNAinterfering,RNAi)现象是动植物体内的一种序列特异性的、转录后基因沉默的过程.在哺乳动物细胞中,19~25个核苷酸长短的双链siRNA(smallinterferingRNA)可有效地抑制基因表达.利用pBS/H1SP载体,它表达的双链RNA的转录产物可以用来抑制特异性HIV-1基因的表达.在siRNA实验中,为了比较由H1启动子转录的siRNA在细胞内的作用,使用以绿色荧光蛋白(EGFP)为报告基因的载体——pEGFP-C1质粒,在荧光显微镜下,很容易地看到EGFP在细胞中表达.将HIV-1siRNA表达载体与表达相应EGFP-HIV融合基因的质粒,共转染人胚肾293细胞,结果表明,和对照质控载体相比,转染了pHIV-siRNA质粒的细胞中EGFP-HIV的表达得到显著抑制.通过该途径,筛选出能抑制HIV-1基因表达的有效siRNA.此外,还在同一载体上表达两种或三种siRNA,分别针对不同的HIV基因,获得了良好的抑制效果.  相似文献

用 RNA 干扰 (RNA interference , RNAi) 技术抑制哺乳动物细胞中外源报告基因的表达,以探讨该过程中 RNAi 作用的剂量和时间效应 . 应用 Lipofectamine 2000 将外源报告基因的表达载体与编码短发夹 RNA (short hairpin RNA , shRNA) 的质粒共转染 HEK293H 细胞,观察 shRNA 载体对报告基因的抑制效应 . 转染后, shRNAs 的瞬时表达可特异地抑制细胞内报告基因的表达 . 在共转染后 12 , 24 , 48 , 60 , 72 , 96 h 时检测 EGFP (enhanced green fluorescent protein , EGFP) 基因 mRNA 及蛋白质表达水平,结果显示, EGFP mRNA 及蛋白质表达在 12 h 时略有降低, 24~48 h 时表达逐渐降低, 48~72 h 时降低最明显,其后 EGFP 表达水平逐渐恢复 . 提示该过程中 RNAi 效应呈现由弱到强、又由强到弱的逐渐消逝趋势 . 共转染一系列剂量比例的 EGFP 干扰载体与靶载体的结果表明,在一定剂量范围内, RNA 干扰载体所介导的抑制效应与干扰载体剂量大小有关,当其剂量进一步加大足以抑制外源基因表达时,抑制效应则维持在一“平台期” . 此外,通过 RNAi 抑制 HeLa 细胞、 HEK293 细胞中荧光素酶基因的表达, 荧光素酶活性变化也表现出上述类似的效应 . 这些结果表明,在体外哺乳动物细胞中,基于表达载体的 RNAi 作用呈现剂量和时间依赖性效应 . 这为基于载体表达的 RNAi 技术应用研究提供了一定的理论参考及依据 .  相似文献

RNA干扰技术已经成为基因功能研究等领域的有力工具,构建带有筛选标记的siRNA载体可以在细胞中持续抑制靶基因的表达.为了利用RNAi技术开展生物学研究,在克隆载体pUC19的基础上改造构建了人类细胞小干扰RNA(small interference RNA,siRNA)表达质粒pUC19NU.该质粒具有新霉素抗性标记和真核细胞复制起点,利用连入的人U6 snRNA启动子起始siRNA的转录.以EGFP 和p53为靶基因的干扰实验证明,所构建的siRNA表达质粒可以显著抑制细胞外源性增强绿色荧光蛋白(enhanced green fluorescent protein,EGFP)及细胞内源性p53蛋白的表达,而且抑制效果具有特异性.  相似文献

RNA干扰(RNAi)是一种转录后基因沉默技术,可有效诱导序列特异性基因沉默.由RNA聚合酶Ⅱ启动子调控表达的小发卡RNA可有效介导RNAi效应,为组织特异性基因沉默提供了一条新的途径.但是,由RNA聚合酶Ⅱ启动子调控表达的小发卡RNA(shRNA)在序列上与靶基因非完全互补对RNAi效应的影响鲜有报道.本文初步探索RNA聚合酶Ⅱ启动子调控表达的shRNA碱基发生突变或缺失对RNAi效应的影响.研究表明,靶向hTERT mRNA的碱基突变shRNA显著降低RNAi效应,而靶向GFP mRNA的碱基缺失shRNA对RNAi效应没有显著影响.本研究为非完全互补shRNA对RNAi效应的进一步深入研究提供了理论与实验依据.  相似文献

为建立一条简便、有效的构建哺乳动物细胞随机RNAi文库技术路线 ,首先通过PCR将 1 9~ 2 1ntsiRNA编码序列及终止信号等元件引入U6启动子下游 ,再将该扩增产物定向克隆至H1启动子的下游。构建的双启动子载体中 ,U6和H1相对排列 ,均以其间插入的靶序列为模板 ,分别转录出其正义和反义链 ,形成功能性siRNA。以EGFP和bcl-2为靶基因的实验表明 ,该方法构建的载体可有效地干扰相应基因的表达。  相似文献

RNA干涉(TNA interference,RNAi)技术是一门新兴的基因阻断技术,可通过将外源双链RNA导入细胞介导同源靶基因mRNA序列的降解,使靶基因表达沉默,是一种转录后的基因沉默现象.目前,RNA干涉技术在植物研究中已经得到广泛应用.本文简介了植物的RNA干涉研究及RNAi表达载体的构建,综述了RNAi在棉花基因功能和功能基因组学研究、棉花品质性状的遗传改良和棉花的抗虫抗病研究中的应用进展及其在棉花研究中存在的主要问题和应用前景.  相似文献

目的:以增强型绿色荧光蛋白(EGFP)作为报告基因,用流式细胞术筛选高表达EGFP的细胞,从而获得外源基因高效表达细胞株。方法:构建在EGFPC端编码区融合新霉素(neomycin)抗性基因的融合基因EGFP-Neomycin,将其插入pcDNA3.1(+)载体,构建EGFP-Neomycin融合基因表达载体pcDNAEN,转染CHO-K1细胞,G418加压筛选和倒置荧光显微镜观察证实所表达的EGFP-Neomycin融合蛋白具有新霉素抗性和激发EGFP荧光双功能;将编码组织型纤溶酶原激活剂(tPA)的cDNA插入pcDNAEN中CMV启动子下游,构建表达tPA的表达载体pcDNAEN/tPA。结果:流式细胞术分析和tPA纤维蛋白溶解活性测定表明,pcDNAEN/tPA转染CHO-K1细胞的EGFP相对荧光强度(RFT)的自然对数值与tPA表达水平呈明显的直线相关关系,相关系数为0.983;比较部分未经流式细胞仪分选的pcDNAEN/tPA转染阳性细胞克隆和RFT分布在100~1000的pcDNAEN/tPA转染阳性细胞克隆的tPA表达水平,经流式细胞术分选获得的细胞克隆的tPA平均表达水平和最高表达水平分别是未经分选获得的细胞克隆的3.9倍和4.1倍。结论:构建的EGFP-Neomycin融合基因具有双功能,建立了利用流式细胞术筛选外源基因高效表达物细胞株的方法。  相似文献

利用双链RNA(dsRNA)调控基因表达已经成为研究基因功能的有力工具。用人H1启动子构建了pBS/H1PS小干扰RNA(siRNA)表达载体,用于在哺乳动物细胞中产生特异性dsRNA转录产物。通过对293细胞中的PSMA7分子进行表达抑制,证明该siRNA载体能够有效产生针对靶基因的RNA干扰(RNAi)效应。  相似文献

A heat shock inducible and inheritable RNA interference (RNAi) system was developed in the silkworm (Bombyx mori). RNAi transgenic silkworms were generated by injecting silkworm eggs with a piggyBac transposon plasmid carrying RNAi sequence against target gene driven by the Drosophila heat shock protein 70 (HSP70) promoter and the helper plasmid expressing piggyBac transposase. The transgenic EGFP gene and the endogenous eclosion hormone (EH) gene were chosen respectively as the target genes. In the RNAi transgenic silkworms, heat shock at 42 degrees C significantly and specifically reduced the expression of EGFP or EH gene in silkworms according to the corresponding RNAi targeting sequence but not in silkworms with the irrelevant RNAi sequence demonstrating the efficiency and specificity of the RNAi effect. Heat shock in the pupal stage hampered pupal-adult eclosion and reduced egg fertility in EH RNAi transgenic silkworms but not in the wild type or EGFP RNAi transgenic silkworms. The establishment of this heat inducible and inheritable conditional RNA interference system in silkworms provided an approach for the first time to dissect the functions of target genes in silkworms at different stages.  相似文献

在真核细胞基因功能研究中, RNA 干涉 (RNAi) 已成为一种强有力的选择性沉默基因表达的实验工具. 建立一套可在哺乳动物培养细胞中高效、经济地表达 siRNA 的载体系统是 RNA 干涉研究的必要前提之一. 从 HepG2 细胞基因组 DNA 中克隆得到 H1 全长启动子 (374 bp),以之为基础构建了两套 RNA 干涉载体系统, pSL 和带有绿色荧光蛋白 (EGFP) 标签的 pESL ,并对 p53 基因进行了相应的 RNA 干涉研究. 干涉质粒瞬时转染 HepG2 细胞后,分别利用半定量 RT-PCR 和蛋白质印迹检测 p53 表达水平. 与商品化载体 pSilencerTM 3.1-H1 hygro 相比, pSL 和 pESL 对 p53 基因表达具有更高的干涉效率. 结果显示:干涉载体 pSL 和 pESL 能高效特异地下调目的基因表达,可作为哺乳动物中基因功能分析的有效工具.  相似文献

RNA interference (RNAi) provides a powerful new means to inhibit viral infection specifically. However, the selection of siRNA-resistant viruses is a major concern in the use of RNAi as antiviral therapeutics. In this study, we conducted a lentiviral vector with a H1-short hairpin RNA (shRNA) expression cassette to deliver small interfering RNAs (siRNAs) into mammalian cells. Using this vector that also expresses enhanced green fluorescence protein (EGFP) as surrogate marker, stable shRNA-expressing cell lines were successfully established and the inhibition efficiencies of rationally designed siRNAs targeting to conserved regions of influenza A virus genome were assessed. The results showed that a siRNA targeting influenza M2 gene (siM2) potently inhibited viral replication. The siM2 was not only effective for H1N1 virus but also for highly pathogenic avian influenza virus H5N1. In addition to its M2 inhibition, the siM2 also inhibited NP mRNA accumulation and protein expression. A long term inhibition effect of the siM2 was demonstrated and the emergence of siRNA-resistant mutants in influenza quasispecies was not observed. Taken together, our study suggested that M2 gene might be an optimal RNAi target for antiviral therapy. These findings provide useful information for the development of RNAi-based prophylaxis and therapy for human influenza virus infection.  相似文献

BACKGROUND: Current methods to establish stably transfected cell lines by nonviral techniques involve coselection for a drug selection marker. However, this approach suffers from several drawbacks. We developed a fluorescence-activated cell sorting (FACS)-based protocol for the selection and isolation of stable hematopoietic electrotransfectants without the need for selective growth conditions. METHODS: Leukemic K562 cells were electroporated with the enhanced green fluorescent protein (EGFP) reporter gene and FACsorted to obtain stably EGFP-expressing cells. Stable EGFP(+) clones were established by single-cell sorting. RESULTS: Efficiency of stable EGFP gene expression increased steadily in function of number of consecutive FACsorts. Stable transfectants (>99% EGFP(+)) were obtained after four FACsorts. Furthermore, several single-cell derived clones with variable levels of stable EGFP expression were isolated and cultured without the use of selective growth media. CONCLUSIONS: EGFP is an effective selection marker for the generation and isolation of stably transfected hematopoietic cell clones without the need for selection in toxic media that could create a potentially undesirable stress environment for stably transfected cells.  相似文献

RNA-mediated interference (RNAi) is a powerful technique that is now being used in mammalian cells to specifically silence a gene. Some recent studies have used this technique to achieve variable extent of depletion of a nuclear enzyme poly(ADP-ribose) polymerase-1 (PARP-1). These studies reported either transient silencing of PARP-1 using double-stranded RNA or stable silencing of PARP-1 with a DNA vector which was introduced by a viral delivery system. In contrast, here we report that a simple RNAi approach which utilizes a pBS-U6-based DNA vector containing strategically selected PARP-1 targeting sequence, introduced in the cells by conventional CaPO(4) protocol, can be used to achieve stable and specific silencing of PARP-1 in different types of cells. We also provide a detailed strategy for selection and cloning of PARP-1-targeting sequences for the DNA vector, and demonstrate that this technique does not affect expression of its closest functional homolog PARP-2.  相似文献

The RNA interference pathway functions as an antiviral defense in invertebrates. In order to generate a phenotypic marker which "senses" the status of the RNAi pathway in Aedes aegypti, transgenic strains were developed to express EGFP and DsRED marker genes in the eye, as well as double-stranded RNA homologous to a portion of the EGFP gene. Transgenic "sensor" mosquitoes exhibited robust eye-specific DsRED expression with little EGFP, indicating RNAi-based silencing. Cloning and high-throughput sequencing of small RNAs confirmed that the inverted-repeat transgene was successfully processed into short-interfering RNAs by the mosquito RNAi pathway. When the A. aegypti homologues of the genes DCR-2 or AGO-2 were knocked down, a clear increase in EGFP fluorescence was observed in the mosquito eyes. Knockdown of DCR-2 was also associated with an increase in EGFP mRNA levels, as determined by Northern blot and real-time PCR. Knockdown of AGO-3, a gene involved in the germline-specific piRNA pathway, did not restore EGFP expression at either the mRNA or protein level. This transgenic sensor strain can now be used to identify other components of the mosquito RNAi pathway and has the potential to be used in the identification of arboviral suppressors of RNAi.  相似文献

RNA interference (RNAi) mediated by small interfering RNA (siRNA) has become a popular tool of examining the function of various genes. However, many studies have failed to identify any inhibitory effect of the siRNAs on the expression of the target gene, even though the siRNA being tested had been designed sequence-specifically. In order to determine if this failure is due to the incorrect choice of observation time rather than that of the target site of the gene of interest, this study examined the RNAi efficiency of a vector-driven siRNA targeting two different reporter proteins, EGFP and d2EGFP, whose targeted sequences were identical but the half-lives within the cells differed remarkably from each other (>24h versus 2h), during the time course after transfection. The EGFP expression levels in both cells were reduced in time-dependent manner but the reduction patterns were quite different from each other. The RNAi efficiency varied among the different observation time points and the time required for the maximum RNAi efficiency was proportional to the half-life of the target protein. Stable knocked down cell lines for EGFP expression were then established and the reduced EGFP expression levels in these cell lines were retained for a long period. These results suggest that the choice of an adequate observation time or the establishment of stable knocked down cells by antibiotic selection might be required for making an accurate evaluation of the RNAi effect on the target protein possessing a long half-life.  相似文献

BACKGROUND: RNA interference (RNAi) has become a powerful tool in silencing target genes in various organisms. In mammals, RNAi can be induced by using short interfering RNA (siRNA). The efficacy of inducing RNAi in mammalian cells by using siRNA depends very much on the selection of the target sequences. METHODS: We developed an siRNA target sequence selection system by first constructing parallel-type siRNA expression vector libraries carrying siRNA expression fragments originating from fragmentized target genes, and then using a group selection system. For a model system, we constructed parallel-type siRNA expression vector libraries against DsRed and GFP reporter genes. RESULTS: We carried out the first screening of groups containing more than 100 random siRNA expression plasmids in total for each target gene, and successfully obtained target sequences with very strong efficacy. Furthermore, we also obtained some clones that express dsRNAs of various lengths that might induce cytotoxicity. CONCLUSIONS: This system should allow us to perform screening for powerful target sequences, by including all possible target sequences for any gene, even without knowing the whole sequence of the target gene in advance. At the same time, target sequences that should be avoided due to cytotoxicity can be identified.  相似文献

本研究对金针菇Flammulina velutipes的一个RNAi转化子菌株1382R3进行了高通量测序,以本实验室先前获得的野生型W23基因组数据为参考,分析了该转化子的基因插入位点以及拷贝数。转化子菌株1382R3是通过农杆菌介导将fv-hmg1-RNAi载体转化至金针菇菌株并通过PCR检测筛选标记而得到。通过BLAST将转化子测序的reads对外源载体和基因组定位,找到具有基因组序列(GS)和外源载体序列(ES)两种序列的临界reads,并据此使用PERL语言程序成功在转化子1382R3菌株中找到两个插入位点。对两个插入位置的序列分析表明:在插入位点1,T-DNA片段部分插入;在插入位点2,T-DNA全部插入到基因组。两个插入位点都对基因组内源基因的表达造成了一定的干扰。此方法拓宽了高通量测序技术的应用范围,将其运用到遗传转化插入位置和拷贝数的研究中,有利于食用菌的功能基因组及基因工程研究。  相似文献
