多巴胺D4受体基因启动子区多态性与精神分裂症的相关性   总被引:1,自引:0,他引:1  
目的:探讨多巴胺D4受体(Dopaminc D4 receptor,DRD4)基因启动子区的3个功能多态性与精神分裂症是否存在相关性.方法:严格按照诊断标准,选取无亲缘关系的精神分裂症患者220例,健康对照组200例提取基因组DNA,采用聚合酶链反应及等位基因特异性扩增技术检测DRD4基因启动子区-521C/T、-616C/G和-1240L/S 3个功能位点的基因型,采用HaploView4.0及SPSS11.5软件分析各位点基因型、等位基因频率及组间差异.结果:DRD4基因-1240L/S的基因型及等位基因频率分布在精神分裂症与正常对照组存在显著性差异(p<0.05).DRIM基因启动子区-521C/T和-616C/G位点的基因型及等位基因频率分布在精神分裂症组与正常对照组无统计学差异(p>0.05).结论:DRD4基因-1240L/S多态性与精神分裂症相关联,携带有-1240L/S多态性住点L等住基因的个体可能更容易患精神分裂症.  相似文献

目的:探讨多巴胺D2受体(Dopamine D2 receptors,DRD2)基因3'非翻译区Taq ⅠA、启动子区-141 Ins/Del 2个多态性位点和海洛因依赖的相关性.方法:采用聚合酶链反应-限制性片段长度多态(PCR-RFLP)技术检测320例海洛因依赖患者及300例健康对照组的TaqⅠA和-141 Ins/Del2个多态性位点的基因型.采用HaploView4.0及SPSS11.5软件分析这2个多态性位点的基因型、等位基因频率及组间差异.结果:DRD2基因Taq ⅠA位点的基因型及等位基因频率分布在海洛因依赖组与正常对照组存在显著性差异(p<0.01),海洛因依赖组TaqⅠA位点的等位基因A1频率显著高于正常对照组(x2=11.156,p=0.001,OR=1.463,95%CI=1.170~1.830);DRD2基因-141 Ins/Del位点的基因型及等位基因频率分布在海洛因依赖组与正常对照组之间无统计学差异(p<0.05).结论:DRD2基因TaqⅠA位点多态性可能与海洛因依赖有关,携带有TaqⅠA多态性位点A1等位基因的个体可能更容易对海洛因产生依赖.  相似文献

目的:探讨多巴胺D4受体基因启动子区-1240L/S,-616C/G和-521C/T三个多态性与注意缺陷多动障碍(Attention deficit hyperactivity disorder,ADHD)的关系.方法:取无亲缘关系的ADHD患者及对照组各166名,采用等位基因特异性扩增技术(allele specific amplification,ASA)及聚合酶链式反应琼脂糖凝胶电泳技术,检测ADHD患者和对照组基因型和等住基因的频率分布.结果:DRD4基因-521C/T的基因型及等位基因频率分布在ADHD组与正常对照组存在显著性差异(p<0.05),ADHD组的T等位基因的频率显著高于正常对照组(x2=9.827,p=0.002,OR=1.639,95%CI=1.202-2.234).DRD4基因启动子区2个功能多态性位点-616C/G及-1240L/S的基因型及等位基因频率在正常组与ADHD组的分布无显著性差异(p>0.05).结论:-521C/T位点与ADHD的易感性存在关联,且-521C/T等位基因是决定ADHD个体易感性的重要因素,含有T等位基因的个体罹患注意缺陷多动障碍的相对风险增高.  相似文献

目的:探讨多巴胺D1受体(Dopamine D1 receptor, DRD1)基因启动子区G-48A、外显子区T1403C两个SNP位点与注意缺陷多动障碍(attention deficit hyperactivity disorder,ADHD)的关联性.方法:选取87名ADHD患者和103名正常对照,提取基因组DNA,采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术检测G-48A和T1403C两个多态性位点的基因型,SPSS13.0软件分析各位点的等位基因及基因型频率.结果:DRD1的G-48A基因型及等位基因频率分布在ADHD和对照组之间有统计学差异(p<0.05),ADHD组中等位基因A频率显著高于正常组(p<0.05).T1403C位点基因型及等位基因频率在ADHD组与健康对照组无统计学差异.结论:DRD1基因G-48A多态性可能与ADHD的发病有关,携带有等位基因A的个体可能更容易患ADHD.T1403C多态性与ADHD的发病无明显相关性.  相似文献

目的:探讨汉族人群中多巴胺D2受体(DRD2)基因TaqIB多态性与酒依赖的相关性.方法:采用聚合酶链式反应-限制性片断长度多态性(PCR-RFLP)技术,检测酒依赖组(80例)和对照组(95例)的DRD2基因TaqIB多态性的基因型和等位基因频率.结果:酒依赖组和时照组的DRD2基因TaqIB多态性的基因型和等位基因频率有显著性差异,等位基因B2的携带者显著降低其嗜酒的发生率(OR:1.636,P<0.05).结论:本研究提示,在汉族人群中DRD2基因TaqIB多态性与酒依赖存在相关性,TaqIB2等位基因可能是降低酒依赖发病的影响因子.  相似文献

目的:5-HT(5-hydroxytryptamine,5-HT)参与了多种中枢神经活动的生理过程,其功能异常可以影响很多行为障碍,已有研究显示,5-HT水平与多种精神疾病密切相关。5-HT受体及其转运体基因在海洛因依赖发生发展中起到了重要的作用,是海洛因依赖的主要候选基因。探讨5羟色胺2A受体(Serotonin 2A receptor,HTR2A)基因启动子区-1438A/G(rs6311)、外显子区102T/C(rs6313)与5羟色胺1B受体(Serotonin 1B receptor,HTR1B)基因外显子区861G/C(rs6296)3个单核苷酸多态性和海洛因依赖的关联性分析。方法:严格按照诊断标准,选取无亲缘关系的海洛因依赖个体616例及健康个体600例提取基因组DNA,采用PCR-RFLP方法检测rs6311、rs6313和rs6296 3个SNPs位点的基因型频率,采用SPSS16.0软件分析各位点等位基因、基因型频率在病例-对照组间差异。结果:HTR2A基因rs6311和HTR1B基因rs6296位点的等位基因及基因型频率分布在2组间存在统计学差异(P〈0.05),病例组rs6311位点的等位基因A频率显著高于对照组(X2=5.436,P=0.020,OR=1.208,CI=1.031~1.417),rs6296位点的等位基因C频率显著高于对照组(X2=12.116,P=0.000,OR=1.329,CI=1.132~1.560)。连锁不平衡检验结果显示,HTR2A基因rs6311、rs6313位点处于不连锁状态,D'〈0.5。结论:HTR2A基因rs6311和HTR1B基因rs6296多态性可能与海洛因成瘾有关,携带有rs6311 A等位基因与rs6296 C等位基因的人可能更容易对海洛因产生依赖。我们的研究为海洛因依赖易感人群筛选及药物靶向治疗提供了理论依据。  相似文献

目的:探讨微粒体甘油三酯转移蛋白(Microsomal triglyceride transfer protein,MTP)MTP基因启动子区164T/C单核苷酸多态性与(Single nucleotide polymorphism,SNP)与非酒精性脂肪肝(Non-alcoholic fatty liver disease,NAFLD)的相关性.方法:严格按照诊断标准收集NAFLD患者156例,随机选择健康体检者125例作为对照组,应用聚合酶链反应-限制性酶切片段长度多态性(Polymerase chain reaction restriction fragment length polymorphism,PCR-RFLP)技术对受试者进行基因分型.采用SPSS11.5软件分析各位点基因型、等位基因频率及组间差异.结果:病例组和对照组均检测到MTP基因-164T/C位点TT、TC和CC 3种基因型;MTP基因-164T/C位点的基因型及等位基因频率分布在病例和对照组中差别有统计学意义(P<0.05),NAFLD组等位基因C频率显著高于对照组(P<0.01).结论:MTP基因启动子区164T/C位点多态性可能与非酒精性脂肪肝有关.携带有等位基因C的个体可能更容易患非酒精性脂肪肝.  相似文献

目的:探讨强啡肽原(prodynorphin,PDYN)基因3'非翻译区的3个单核苷酸多态性(Single Nucleotide Polymorphism,SNP)位点与海洛因依赖的相关性.方法:严格按照诊断标准,选取无亲缘关系的海洛因依赖患者212例,健康对照200例提取基因组DNA,采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术检测PDYN基因3'非翻译区rs1022563、rs2235749、rs9100803个多态性位点的基因型,采用HaploView及SPSS11.5软件分析各位点基因型、等位基因频率及组间差异.结果:PDYN基因rs2235749的基因型及等位基因频率分布在海洛因依赖组与正常对照组存在显著性差异(p<0.01),在海洛因依赖组中等位基因A的频率显著高于正常对照组(p<0.01).连锁不平衡结果显示,这3个SNPs位点不连锁,D'=0.215.结论:PDYN基因3'非翻译区rs2235749与海洛因相关联,携带有等位基因A的个体可能对海洛因更容易产生依赖.  相似文献

目的:探讨云南地区人群中DRD2(TaqIA、-141C)、DRD4和DAT基因多态性与酒精依赖的相关性.方法:采用聚合酶链式反应-限制性片断长度多态性(PCR-RFLP)分析技术以及聚合酶链式反应-可变数目串联重复序列多态性(PCR-VNTR)分析技术检测酒依赖组(80例)和对照组(70例)在3个候选基因中的基因型和等位基因频率.结果:上述三个基因多态性的基因型和等位基因频率在酒依赖组和对照组中差异均无统计学意义(P>0.05),对DRD2基因中的TaqIA和-141C进行单倍型分析时发现,Del/Al是一种保护单倍型,能抑制酒依赖综合征的发生.结论:在云南地区人群中,携带有Del/Al单倍型基因的人不易形成酒依赖.  相似文献

目的:探讨5-羟色胺转运体基因(solute carrier family 6 member 4,SLC6A4)基因4个单核苷酸多态性(single nucleotide polymorphism,SNP)位点与海洛因依赖之间的关系。方法:严格按照诊断标准,选取无亲缘关系的海洛因依赖个体397例(病例组)及健康对照个体402例(对照组)提取基因组DNA,采用SNaPshot SNP分型技术对SLC6A4基因4个SNP位点(rs1042173,rs3813034,rs6354,rs7224199)进行基因分型,比较病例-对照组间各位点等位基因、基因型频率的差异。结果:病例组和对照组SLC6A4基因rs1042173和rs3813034位点的基因型和等位基因频率比较存在显著性差异(P0.05),rs1042173的C等位基因(P=0.031,OR=1.317,95%CI=1.026-1.691)及rs3813034的C等位基因(P=0.013,OR=1.375,95%CI=1.069-1.768)是海洛因依赖的危险因素。病例组TCC单倍型(rs7224199、rs3813034和rs1042173)的比例较对照组显著增高(P0.05)。结论:SLC6A4基因rs1042173和rs3813034多态性可能与海洛因成瘾有关,携带有rs1042173的C等位基因和rs3813034的C等位基因的个体及携带TCC单倍型的个体可能更容易对海洛因产生依赖。  相似文献

On the origin of the Hirudinea and the demise of the Oligochaeta   总被引:10,自引:0,他引:10  
The phylogenetic relationships of the Clitellata were investigated with a data set of published and new complete 18S rRNA gene sequences of 51 species representing 41 families. Sequences were aligned on the basis of a secondary structure model and analysed with maximum parsimony and maximum likelihood. In contrast to the latter method, parsimony did not recover the monophyly of Clitellata. However, a close scrutiny of the data suggested a spurious attraction between some polychaetes and clitellates. As a rule, molecular trees are closely aligned with morphology-based phylogenies. Acanthobdellida and Euhirudinea were reconciled in their traditional Hirudinea clade and were included in the Oligochaeta with the Branchiobdellida via the Lumbriculidae as a possible link between the two assemblages. While the 18S gene yielded a meaningful historical signal for determining relationships within clitellates, the exact position of Hirudinea and Branchiobdellida within oligochaetes remained unresolved. The lack of phylogenetic signal is interpreted as evidence for a rapid radiation of these taxa. The placement of Clitellata within the Polychaeta remained unresolved. The biological reality of polytomies within annelids is suggested and supports the hypothesis of an extremely ancient radiation of polychaetes and emergence of clitellates.  相似文献

Data on the ontogeny of the posterior haptor of monogeneans were obtained from more than 150 publications and summarised. These data were plotted into diagrams showing evolutionary capacity levels based on the theory of a progressive evolution of marginal hooks, anchors and other attachment components of the posterior haptor in the Monogenea (Malmberg, 1986). 5 + 5 unhinged marginal hooks are assumed to be the most primitive monogenean haptoral condition. Thus the diagrams were founded on a 5 + 5 unhinged marginal hook evolutionary capacity level, and the evolutionary capacity levels of anchors and other haptoral attachement components were arranged according to haptoral ontogenetical sequences. In the final plotting diagram data on hosts, type of spermatozoa, oncomiracidial ciliation, sensilla pattern and protonephridial systems were also included. In this way a number of correlations were revealed. Thus, for example, the number of 5 + 5 marginal hooks correlates with the most primitive monogenean type of spermatozoon and with few sensillae, many ciliated cells and a simple protonephridial system in the oncomiracidium. On the basis of the reviewed data it is concluded that the ancient monogeneans with 5 + 5 unhinged marginal hooks were divided into two main lines, one retaining unhinged marginal hooks and the other evolving hinged marginal hooks. Both main lines have recent representatives at different marginal hook evolutionary capacity levels, i.e. monogeneans retaining a haptor with only marginal hooks. For the main line with hinged marginal hooks the name Articulon-choinea n. subclass is proposed. Members with 8 + 8 hinged marginal hooks only are here called Proanchorea n. superord. Monogeneans with unhinged marginal hooks only are here called Ananchorea n. superord. and three new families are erected for its recent members: Anonchohapteridae n. fam., Acolpentronidae n. fam. and Anacanthoridae n. fam. (with 7 + 7, 8 + 8 and 9 + 9 unhinged marginal hooks, respectively). Except for the families of Articulonchoinea (e.g. Acanthocotylidae, Gyrodactylidae, Tetraonchoididae) Bychowsky's (1957) division of the Monogenea into the Oligonchoinea and Polyonchoinea fits the proposed scheme, i.e. monogeneans with unhinged marginal hooks form one old group, the Oligonchoinea, which have 5 + 5 unhinged marginal hooks, and the other group form the Polyonchoinea, which (with the exception of the Hexabothriidae) has a greater number (7 + 7, 8 + 8 or 9 + 9) of unhinged marginal hooks. It is proposed that both these names, Oligonchoinea (sensu mihi) and Polyonchoinea (sensu mihi), will be retained on one side and Articulonchoinea placed on the other side, which reflects the early monogenean evolution. Except for the members of Ananchorea [Polyonchoinea], all members of the Oligonchoinea and Polyonchoinea have anchors, which imply that they are further evolved, i.e. have passed the 5 + 5 marginal hook evolutionary capacity level (Malmberg, 1986). There are two main types of anchors in the Monogenea: haptoral anchors, with anlages appearing in the haptor, and peduncular anchors, with anlages in the peduncle. There are two types of haptoral anchors: peripheral haptoral anchors, ontogenetically the oldest, and central haptoral anchors. Peduncular anchors, in turn, are ontogenetically younger than peripheral haptoral anchors. There may be two pairs of peduncular anchors: medial peduncular anchors, ontogentically the oldest, and lateral peduncular anchors. Only peduncular (not haptoral) anchors have anchor bars. Monogeneans with haptoral anchors are here called Mediohaptanchorea n. superord. and Laterohaptanchorea n. superord. or haptanchoreans. All oligonchoineans and the oldest polyonchoineans are haptanchoreans. Certain members of Calceostomatidae [Polyonchoinea] are the only monogeneans with both (peripheral) haptoral and peduncular anchors (one pair). These monogeneans are here called Mixanchorea n. superord. Polyonchoineans with peduncular anchors and unhinged marginal hooks are here called the Pedunculanchorea n. superord. The most primitive pedunculanchoreans have only one pair of peduncular anchors with an anchor bar, while the most advanced have both medial and lateral peduncular anchors; each pair having an anchor bar. Certain families of the Articulonchoinea, the Anchorea n. superord., also have peduncular anchors (parallel evolution): only one family, the Sundanonchidae n. fam., has both medial and lateral peduncular anchors, each anchor pair with an anchor bar. Evolutionary lines from different monogenean evolutionary capacity levels are discussed and a new system of classification for the Monogenea is proposed.In agreeing to publish this article, I recognise that its contents are controversial and contrary to generally accepted views on monogenean systematics and evolution.
