从实验室生物滤塔填料的生物膜上,经选择性培养基筛选,分离出4株好氧反硝化茵.好氧状态下4种菌的40h反硝化率均大于80%,其中菌种A1反硝化率可达到99.05%.跟踪菌种反硝化过程中氮元素24h变化过程,发现4株菌除A1外都有亚硝酸根积累.茵种A1为短杆菌,革兰氏阴性.生理生化特性研究与16S rDNA序列测定(GenBank接受号DQ836052.1)初步判定菌种A1为假单胞菌Pseudomonas putida.适于茵Al生长的初始pH值是7.0左右,温度30℃左右,当DO大于2.0mg/L时,DO的变化对菌种Al的反硝化效果影响很小.  相似文献

好氧反硝化生物脱氮技术自提出以来,凭借能实现同步硝化反硝化、节省基建投资及运行费用等诸多优点,受到国内外环境领域学者的广泛关注。本文首先总结了近年来好氧反硝化菌种的筛选分离情况,以及环境因子对好氧反硝化菌脱氮效能的影响,包括溶解氧(dissolved oxygen,DO)、碳氮比(C/N)、温度等。然后深入探讨了好氧反硝化生物脱氮技术的原理,好氧反硝化过程中的关键功能基因及酶,同时介绍了分子生物技术在好氧反硝化研究过程中的应用,以及好氧反硝化生物脱氮技术在实际应用方面的研究现状。最后,基于目前的研究瓶颈问题,对未来好氧反硝化生物脱氮技术的研究方向提出了科学展望。  相似文献

为研究污水厂/养殖池中好氧反硝化微生物的多样性及菌株反硝化能力,本研究采集了位于福建省厦门市和漳州市的污水处理厂、排污口、污水池、对虾养殖池的污水和污泥样品进行好氧反硝化微生物的富集、分离、鉴定和功能筛选。分别以NaNO3、NaNO2作为唯一氮源共分离纯化获得128株单菌。其中以NaNO3为唯一氮源分离得到63株,以NaNO2为唯一氮源分离得到65株。16SrRNA基因序列分析表明,128株单菌分属于γ-变形菌纲(Gammaproteobacteria,58.6%)、芽胞杆菌纲(Bacilli,6.4%)、放线菌纲(Actinobacteria,11.7%)、α-变形菌纲(Alphaproteobacteria,8.6%)、纤维菌纲(Cytophagia,2.3%)、鞘脂杆菌纲(Sphingobacteria,0.8%)和黄杆菌纲(Flavobacteria,1.6%)7个纲中的38个属。其中盐单胞菌属(Halomonas,29.7%)和芽胞杆菌属(Bacillus,12.5%)为优势菌属,并且广泛存在于各个样品中。反硝化功能初筛结果表明,35株菌能在72h内将20mg·L-1 NO-3-N/NO-2-N完全去除;复筛结果表明,21株菌能在72h内将100 mg·L-1 NO-3-N/NO-2-N完全去除,并且盐单胞菌属、卓贝尔氏菌属(Zobellella)、斯塔普氏菌属(Stappia)及节杆菌属(Arthrobactor)反硝化效果较好,其中斯塔普氏属是首次报道具有好氧反硝化功能。本研究结果表明,污水场/养殖池等环境中可培养反硝化细菌多样性丰富,同时高效反硝化菌的获得也为含氮废水的生物处理提供了良好的菌种资源。  相似文献

好氧反硝化菌是土壤、沉积物、活性污泥等介质中广泛存在的一类微生物。研究以相同种泥扩大培养的SBR(序批式活性污泥反应器)和MBR(膜生物反应器)中活性污泥为研究对象,采用平板分离法分别从运行6个月的SBR和MBR反应器中分离得到9株和11株好氧反硝化菌。经好氧反硝化性能测定发现,20株细菌中有16株总氮去除率在50%以上,且NO_2~--N累积量均在0.5 mg/L以下。经系统进化分析表明,在门分类级别中,采用同一种泥的2个反应器中好氧反硝化菌表现出极大的一致性,20株好氧反硝化菌全部属于变形菌门(Proteobacteria)。但是,进一步分析发现不同反应器中好氧反硝化菌表现出较大的属、种间的差异。其中,SBR反应器9株好氧反硝化菌分属于4个属,6个物种;而从MBR反应器获得的11株细菌分属于2个属,6个物种;仅1个物种(Pseudomonas toyotomiensis)在2个反应器均有发现。  相似文献

【目的】氮污染已成为当今水体污染的一个重要因素,为了解滇池可培养好氧反硝化细菌的多样性,获得高效好氧反硝化细菌资源,为污染水体或浅层地下水的生物修复提供材料。【方法】采用富集培养方法从滇池沉积物和水体样品中分离好氧反硝化细菌,对好氧反硝化细菌的16S r RNA基因序列进行系统发育分析,并筛选其中的高效好氧反硝化细菌。【结果】分离出260株好氧反硝化菌,经16S rRNA基因序列分析,260株菌分属于2门13科14属的59个种。假单胞菌属(Pseudomonas)为优势细菌属,其次是不动杆菌属(Acinetobacter)、气单胞菌属(Aeromonas)和代尔夫特菌属(Delftia)。筛选到12株高效好氧反硝化细菌菌株,其中8株属于假单胞菌(Pseudomonas spp.),4株为不动杆菌(Acinetobacter spp.)。定量分析发现菌株N15-6-1的反硝化效果较好。对菌株N15-6-1的脱氮条件优化结果显示,在以蔗糖为碳源,温度为30–35℃、C/N=12、静止培养时,反硝化能力较强,其在48 h内硝态氮的去除率达到98.81%,总氮的去除率达96.27%。【结论】滇池存在着较丰富的可培养好氧反硝化细菌,好氧反硝化细菌的分离丰富了好氧反硝化菌的种类,其中的高效脱氮菌株为污染水体或浅层地下水的生物修复提供了初步的候选菌株。  相似文献

从稳定运行处理竹子加工废水的生物接触氧化反应器中分离得到一株好氧反硝化菌DN7,其72 h NO3-降解率达99.4%.细胞显微镜观察显示,菌株为革兰氏阴性小杆菌,大小为0.5 μm×1.5 μm,菌落为乳白色.通过生理生化特性及16S rDNA同源性分析,初步推断该菌株为根瘤菌中的Defluvibacter lusatiensisstr.碳源、C/N、硝酸盐初始浓度、溶解氧(DO)、pH对DN7反硝化性能影响的结果表明:菌株对柠檬酸钠、葡萄糖等小分子有机物的利用较好;C/N为9时,脱氮率达99.0%;硝酸盐浓度低于138.48 mg·L-1情况下,DN7脱氮率在96%以上,且亚硝酸盐浓度均在1.0 mg·L-1以下;菌株DN7对DO不敏感,中性偏碱性环境有利于DN7反硝化反应的进行;DN7具有良好的异养硝化性能,72 h铵氮降解率达84.7%.  相似文献

一株好氧反硝化菌的分离鉴定及其除氮特性   总被引:10,自引:0,他引:10  
【目的】生物除氮中反硝化菌具有重要的作用,需氧反硝化菌研究较少,有着很好的应用潜力,本研究主要从环境样品中分离具有高效去除铵氮和亚硝酸盐氮活性的好氧反硝化菌,并对其分类及除氮特性进行研究。【方法】以高效去除铵氮、除亚硝酸盐氮和好氧反硝化能力为主要指标,从富营养化的池塘淤泥水和工厂污泥样品中进行菌株分离筛选。通过生理生化特点以及16S rRNA序列分析对活性最好的菌株进行初步鉴定。在好氧条件下,分别以NO-3-N、NH+4-N和NO-2-N作为唯一氮源,考察菌株的好氧反硝化特性、去除铵氮和亚硝酸盐氮特性,以及不同初始pH值、温度、碳源、摇床转速对该菌去除铵氮和亚硝酸盐氮特性的影响。【结果】得到的细菌中,以菌株C-4的活性最好,其16S rRNA序列与不动杆菌的同源性达99%,结合生理生化特点,初步确定菌株C-4属于不动杆菌属(Acinetobacter sp.)。以柠檬酸钠作为碳源,30℃、120 r/min振荡培养,种龄为18 h,用初始pH为8.5的200 mg/L NH +4-N培养基和初始pH为7.5的100 mg/L NO -2-N培养基进行测定,分别培养15 h与12 h,净除氮率分别达到65.8%和47.8%。【结论】从鱼塘水样中分离到一株好氧反硝化菌C-4,初步鉴定为不动杆菌属的一个种(Acinetobacter sp.),具有较高的反硝化特性和高效去除铵氮与亚硝酸盐氮的能力,在处理实际池塘污水时中,净除氮率可达73.04%以上。  相似文献

【目的】研究反硝化除磷菌特性。【方法】通过微生物筛选和生物学特性研究方法,从对虾养殖池塘中筛选出多株可在有氧条件下同时具有反硝化除磷功能的菌种。【结果】菌株LY-1可在18 h内将初始量为10 mg/L的亚硝酸盐氮降低至0.04 mg/L,PO43?-P降低至0.05 mg/L。在DO浓度为5.0?5.9 mg/L时,该菌反硝化除磷率近100%。试验选取具有反硝化除磷功能的枯草芽孢杆菌为阳性对照菌,大肠杆菌为阴性对照菌,比较研究了菌株LY-1在不同pH、温度、盐度、PO43?-P浓度、亚硝酸盐浓度时反硝化除磷的强弱,在pH为5?9范围时,该菌亚硝酸盐氮去除率近99%,PO43?-P去除率86%;温度为30°C时,该菌反硝化除磷率近100%;盐度为5‰?15‰、PO43?-P浓度为10 mg/L、亚硝酸盐氮浓度为20 mg/L时,该菌亚硝酸盐氮和PO43?-P去除率均可达99%。【结论】菌株LY-1反硝化除磷性能显著高于对照菌(P<0.05)。通过菌株LY-1形态学观察、生理生化及16S rRNA基因序列分析,初步鉴定为蜡样芽孢杆菌(Bacillus cereus)。  相似文献

基于响应面法对一株好氧反硝化菌脱氮效能优化   总被引:2,自引:1,他引:1  
【目的】水体富营养化是当今我国水环境面临的重大水域环境问题,氮素超标排放是主要的引发因素之一。好氧反硝化菌构建同步硝化反硝化工艺比传统脱氮工艺优势更大。获得高效的好氧反硝化菌株并通过生长因子优化使脱氮效率达到最高。【方法】经过序批式生物反应器(Sequencing batch reactor,SBR)的定向驯化,筛选获得高效好氧反硝化菌株,采用响应面法优化好氧反硝化过程影响总氮去除效率的关键因子(碳氮、溶解氧、pH、温度)。【结果】从运行稳定的SBR反应器中定向筛选高效好氧反硝化菌株Pseudomonas T13,采用响应面法对碳氮比、pH和溶解氧关键因子综合优化获得在18 h内最高硝酸盐去除率95%,总氮去除率90%。该菌株的高效反硝化效果的适宜温度范围为25?30 °C;最适pH为中性偏碱;适宜的COD/NO3?-N为4:1以上;最佳溶解氧浓度在2.5 mg/L。【结论】从长期稳定运行的SBR反应器中筛选获得一株高效好氧反硝化菌Pseudomonas T13,硝酸盐还原酶比例占脱氮酶基因的30%以上,通过运行条件优化获得硝氮去除率达到90%以上,对强化废水脱氮工艺具有良好应用价值。  相似文献

Reddish filaments in two fragments of unusual iron oxide bearing stalactites, "the Rusticles" from Lechuguilla Cave, New Mexico, are found only within the central canals of the Rusticles. The curved, helical, and/or vibrioidal filaments vary from 1 to 6 w m in outer diameter and 10 to > 50 w m in length. SEM and TEM show the filaments have 0.5- w m diameter central tubes, with goethite crystals radiating outwardly along their lengths. The diameter of the central tubes is consistent with the diameter of many ironoxidizing filamentous bacteria. Although most iron oxide depositing bacteria do not deposit well-crystallized radiating goethite, we propose thick hydrous iron oxide was slowly crystallized from amorphous material to goethite, in place, over a relatively long period of time. From the gross morphology and the particular setting, we suggest this represents an occurrence of fossilized, acidophilic iron-oxidizing bacteria.  相似文献

Nutritional studies on four different strains of Chloroflexus, a new genus of filamentous, photosynthetic bacteria are described. This organism appears to be related to several different procaryotic groups, and in particular to the green sulfur bacteria and blue-green algae. Unlike these autotrophs, however, Chloroflexus is nutritionally diverse, being able to grow aerobically as a light-independent heterotroph, and anaerobically as a photoautotroph or photoheterotroph. Numerous organic carbon sources including hexoses, amino acids, short chain fatty acids, organic acids, and some alcohols are utilized under various growth conditions. These results suggest that this organism may be among the most nutritionally versatile organisms known.  相似文献

A series of bis-indolone-N-oxides, 1a–f, was prepared from bis(ethynyl)benzenes and o-halonitroaryls and studied for their in vitro antiplasmodial activities against Plasmodium falciparum and representative strains of bacteria and candida as well as for their cytotoxicity against a human tumor cell line (MCF7). They did not cause any haemolysis (300 μg mL−1). Of the synthesized bis-indolones, compound 1a had the most potent antiplasmodial activity (IC50 = 0.763 μmol L−1 on the FcB1 strain) with a selectivity index (CC50 MCF7/IC50 FcB1) of 35.6. No potency against the tested microbial strains was observed.  相似文献

A collection of Pseudomonas strains was isolated in different regions of Tunisia in the period 2016–2017 from the fruits and leaves of Citrus sinensis cv. ‘Valencia Late’ and Citrus limon cv. ‘Eureka’ plants with symptoms of blast and black pit disease. A phylogenetic analysis of the housekeeping gene rpoD was used for strain identification at the species level. The results demonstrated the affiliation of these strains with the genus Pseudomonas and revealed the presence of 11 strains representing two putative new species in two monophyletic branches. These strains were analyzed morphologically and genotypically by multilocus sequence analyses of the rpoD, gyrB and 16S rRNA (rrs) gene sequences, and their phenotypic characteristics by API 20NE and Biolog GEN III. Plant pathogenic properties were confirmed on fruits and detached leaves of C. limon cv. ‘Eureka’. Fatty acids and WC MALDI-TOF MS major protein profiles were determined. The genomes of both representatives were sequenced. The average nucleotide index and genome-to-genome distance from KC12T and E10BT are below the cut-off established for a described species. These results support the conclusion that the strains KC12T, KC17, KC20, KC22, KC24A, KC25 and KC26 represent a novel species of Pseudomonas, for which the name of Pseudomonas kairouanensis is proposed. The type strain is KC12T (=CECT9766 and CFBP 8662). The strains E10BT, E10AB, E10CB1 and Iy3BA represent another novel species of Pseudomonas for which the name of Pseudomonas nabeulensis is proposed; the type strain is E10BT (=CECT9765 and CFBP 8661).  相似文献

测定绿脓杆菌冷休克率方法的研究   总被引:1,自引:0,他引:1  
本研究基于某些革兰氏阴性杆菌当处于不同生长阶段时对冷敏感性表现不同的现象,建立了绿脓杆菌冷休克率的测定技术,可用于测定群体绿脓杆菌的生长情况。应用该技术,实验群体绿脓杆菌生长各期的肉汤培养物,观察冷休克率变化规律,对照群体细菌生长曲线可见两者有一定程度吻合,同时也有差别,后者主要在于细菌冷休克率峰值出现早于细菌活菌数峰值,而当细菌达到稳定期时,其数量居高不下而后才逐渐降低,此时,冷休克率的降低则迅速而明显,可清楚地表明细菌生长状态,较早地预示群体细菌生长势头。对实验动物及少量烧伤病人创面标本检测的结果表明冷休克率测定有助于检出生长的优势菌群,此点将在机会致病菌的微生物病原性确定方面有一定的应用前景。  相似文献

About 1,000 houses built on excavated nonweathered mudstone sediments, originally deposited in the Neogene, have been damaged by microbially induced heaving of foundations. The maximal height of the heaving was 48 cm. The presence of sulfate-reducing, sulfur-oxidizing, and acidophilic iron-oxidizing bacteria in the mudstone indicated that the joint activity of these three types of bacteria could account for the heaving. A hypothesis is presented in which, first, the temperature of the newly exposed mudstone sediments increased above 25 °C, which stimulated the sulfate-reducing bacteria in the mudstone to actively reduce sulfate to hydrogen sulfide. The mudstone sediments under the houses gradually dried, and became permeable to air. Consequently, sulfur-oxidizing bacteria oxidized the hydrogen sulfide to sulfuric acid and the environmental pH decreased to approximately 3. Next, the acidophilic iron-oxidizing bacteria actively oxidized the sulfur in pyrite to produce much more acid. The resulting sulfuric acid reacted with calcium carbonate and with ferric and potassium ions to produce gypsum and jarosite, respectively. A combination of the increased volume of gypsum and jarosite crystals and the production of CO 2 as a by-product of their formation made the mudstone sediments bulky. The end result was widespread heaving.  相似文献

新生儿重症监护病房常见病原菌耐药率比较分析   总被引:2,自引:0,他引:2  
目的探讨新生儿重症监护病房与其他病房分离病原菌的耐药率。方法采用VITEK全自动微生物分析仪进行药物敏感性分析,对深圳市儿童医院1999~2004年分离的2组常见病原菌的耐药率进行统计分析,耐药率的显著性比较用χ2检验。结果新生儿重症监护病房常见病原菌为表皮葡萄球菌、凝固酶阴性葡萄球菌和肺炎克雷伯菌,而其他病房以金黄色葡萄球菌、表皮葡萄球菌、凝固酶阴性葡萄球菌、大肠埃希菌和铜绿假单胞菌为主。新生儿重症监护病房病原菌对左氧沙星、环丙沙星、头孢吡肟、头孢他啶和哌拉西林三唑巴坦的耐药率分别为376%、368%、712%、712%和505%,显著高于儿科其他病房分离菌株的耐药率(P<0001)。结论新生儿重症监护病房分离菌的耐药率高于其他病房病原菌的耐药率。  相似文献
