The carriers mineral oil and Silwet L-77 and the botanical insecticides Neemix 4.5 and Hexacide were evaluated for their impacts on the efficacy of Beauveria bassiana (Balsamo) Vuillemin conidia against red flour beetle, Tribolium castaneum (Herbst), larvae. The dosages of liquid treatments were quantified by both conidia concentration in the spray volume and conidia deposition on the target surface. The latter approach allowed comparison with dry, unformulated conidia. The median lethal concentrations of B. bassiana in 0.05% Silwet L-77 solution or without a carrier were approximately double that for conidia in mineral oil. Carriers had highly significant effects on the efficacy of B. bassiana. The lower efficacy of conidia in aqueous Silwet L-77 may have been the result of conidia loss from the larval surface because of the siloxane's spreading properties. Neemix 4.5 (4.5% azadirachtin) delayed pupation and did not reduce the germination rate of B. bassiana conidia, but it significantly reduced T. castaneum mortality at two of four tested fungus doses. Hexacide (5% rosemary oil) caused significant mortality when applied without B. bassiana, but it did not affect pupation, the germination rate of conidia, or T. castaneum mortality when used in combination with the fungus.

This research tested the suppressive ability of Beauveria bassiana (Balsamo) Vuillemin alone and in combination with diatomaceous earth against the red flour beetle, Tribolium castaneum (Herbst). Adults did not show a dose response to B. bassiana, and the addition of diatomaceous earth (DE) did not result in a significant increase in mortality. Against larvae, however, DE at 190 mg/kg grain enhanced the efficacy of B. bassiana at all concentrations ranging from 33 to 2,700 mg of conidia per kilogram of grain. The presence of DE resulted in 17- and 16-fold decreases in the median lethal concentration of B. bassiana at 56 and 75% RH, respectively. No significant differences in larval mortality in response to B. bassiana and diatomaceous earth alone or in combination were found between 56 and 75% RH. Conidial attachment to larvae was significantly greater with 190 mg/kg DE than without it. The partial analysis of lipids taken up by DE from the larvae revealed the removal of phospholipids and long-chain fatty acids. These results support the hypothesis that diatomaceous earth enhances the efficacy of B. bassiana against larval T. castaneum, at least in part by damaging the insect cuticle, thus increasing conidial attachment and making nutrients more available to conidia for their germination.

The use of elevated temperatures (> or = 40-60 degrees C) or heat treatments for managing insects in food-processing facilities is a viable alternative to space fumigation with methyl bromide. Quantitative data are lacking on the responses of life stages of the red flour beetle, Tribolium castaneum (Herbst), an important pest of food-processing facilities worldwide, to elevated temperatures used during heat treatments. We determined time-mortality relationships for eggs, young (neonate) larvae, old larvae, pupae, and adults of T. castaneum, exposed to constant temperatures of 42, 46, 50, 54, 58, and 60 degrees C. Generally, mortality of each stage increased with an increase in temperature and exposure time. Young larvae were the most heat-tolerant stage, especially at temperatures > or = 50 degrees C. Exposure for a minimum of 7.2 h at > or = 50 degrees C was required to kill 99% of young larvae, whereas the other stages required < or = 1.8 h. Heat treatments that control young larvae should control all other stages of T. castaneum, and young larvae should be used as test insects to evaluate efficacy against T. castaneum during an actual facility heat treatment. These results provide the basis for successful use of elevated temperatures for management of T. castaneum life stages associated with food-processing facilities.

Starvation of second instar Colorado potato beetle larvae for 24h immediately after treatment with Beauveria bassiana conidia increased susceptibility to the pathogen and subsequent sporulation of cadavers but decreased time to larval death. In feeding studies, B. bassiana-treatment had no effect on subsequent larval development, and mortality occurred 5-6 days after treatment. Twenty-four hours of starvation alone retarded subsequent larval development but did not affect mortality. Mortality of B. bassiana-treated starvation stressed larvae occurred 4-5 days after treatment. Both B. bassiana treatment and 24h starvation significantly reduced total foliage consumption and daily weight gains. On the day of treatment, B. bassiana had no effect on the efficiency with which food was converted to biomass (ECI). ECI was not affected by B. bassiana or starvation alone on the day following treatment but was significantly affected by a combination of both. When larvae were exposed to a range of limited food quantities, ECI decreased with decreasing food availability but only extreme stress (starvation for 24h) increased susceptibility to B. bassiana. Topical application of Dacryodes excelsa resin (an antifeedant) to potato leaves caused a concentration dependent reduction in foliage consumption and weight gain by second instar larvae but did not affect larval mortality. When larvae were exposed to a fixed concentration of B. bassiana and a range of antifeedant concentrations there were significant linear relationships between 24h larval weight gain and mortality and 24h larval weight gain and sporulation. The interaction between starvation stress and the susceptibility to B. bassiana infection is discussed and its possible implications in pest management considered.

Sanitation being an important component of chemical-free management of stored-products pests, the nutritional stress on insects that results from a clean environment may prove advantageous to the use of microbial controls. Dietary stress by food deprivation or suboptimal diet increased susceptibility of the red flour beetle, Tribolium castaneum (Herbst), larvae to Beauveria bassiana (Balsamo) Vuillemin. Fungus-related mortality increased linearly with the number of days of food deprivation. Rearing of larvae on a rice meal diet resulted in slower development and greater susceptibility to B. bassiana than rearing on whole wheat flour with brewer's yeast. Larvae that were fed for 24 h on flour with B. bassiana conidia consumed significantly less and weighed significantly less than those that were fed fungus-free flour. Thus, the presence of B. bassiana conidia is itself a cause of dietary stress.

The pecan weevil, Curculio caryae (Horn), is a key pest of pecan [Carya illinoinensis (Wangenh.) K. Koch]. Current control recommendations are based on chemical insecticide applications. Microbial control agents such as the entomopathogenic nematode, Steinernema carpocapsae (Weiser) and the fungus Beauveria bassiana (Balsamo) Vuillemin occur naturally in southeastern U.S. pecan orchards and have shown promise as alternative control agents for C. caryjae. Conceivably, the chemical and microbial agents occur simultaneously within pecan orchards or might be applied concurrently. The objective of this study was to determine the interactions between two chemical insecticides that are used in commercial C. caryae control (i.e., carbaryl and cypermethrin applied below field rates) and the microbial agents B. bassiana and S. carpocapsae. In laboratory experiments, pecan weevil larval or adult mortality was assessed after application of microbial or chemical treatments applied singly or in combination (microbial + chemical agent). The nature of interactions (antagonism, additivity, or synergy) in terms of weevil mortality was evaluated over 9 d (larvae) or 5 d (adults). Results for B. bassiana indicated synergistic activity with carbaryl and antagonism with cypermethrin in C. caryae larvae and adults. For S. carpocapsae, synergy was detected with both chemicals in C. caryae larvae, but only additive effects were detected in adult weevils. Our results indicate that the chemical-microbial combinations tested are compatible with the exception of B. bassiana and cypermethrin. In addition, combinations that exhibited synergistic interactions may provide enhanced C. caryae control in commercial field applications; thus, their potential merits further exploration.

Barley, Hordeum vulgare L., one of the important crops in Canada, is used in malting, feed, and food industries. Disinfestation of barley using microwaves can be an alternative to chemical methods used to kill insects. A pilot-scale industrial microwave system operating at 2.45 MHz was used in this study to determine the mortality of life stages (egg, larva, pupa, and adult) of Tribolium castaneum (Herbst) (Coleoptera: Tenebrionidae). Barley samples of 50 g each at 14, 16, and 18% moisture content (MC; wet basis) were infested with various life stages of T. castaneum and exposed to microwave energy at different power levels and exposure times, and the mortality of the insects was determined. The average temperature of the 14% MC sample exposed to 28 s at 0, 200, 300, 400, and 500 W were 27.4, 42.6, 53.7, 66.9, and 73.0 degrees C, respectively, and those exposed to 56 s at 0, 200, 300, and 400 W were 27.4, 57.3, 75.5, and 91.2 degrees C, respectively. A similar range of temperature was observed for 16 and 18% MC barley. Complete mortality of all life stages of T. castaneum can be achieved at a power level of 400 W and an exposure time of 56 s or at 500 W for 28 s. Among the life stages of T. castaneum, eggs were the most susceptible to microwave energy and adults were the least susceptible. There was no significant difference in the mortality of larvae and adults at 14, 16, and 18% MC, but the mortality was significantly different for eggs and pupae at different MCs. There was a significant increase in the mortality with an increase in power level or exposure time or both. Germination capacity of the seeds was decreased with an increase in power level or exposure time or both. Quality characteristics such as alpha-amylase, diastatic power, soluble protein, viscosity, and density of the barley malt treated at 500 W for 28 s were same as the control sample, whereas the samples treated at 400 W for 56 s were significantly lower.

Acute or chronic sublethal exposure of Colorado potato beetle larvae to the CryIIIA delta-endotoxin of Bacillus thuringiensis Berliner did not significantly (P > 0.05) alter their subsequent susceptibility to Beauveria bassiana (Balsamo) Vuillemin. During the period of exposure to B. bassiana there was continued mortality from previous exposure to delta-endotoxin, and B. bassiana also caused significant mortality. Acute and chronic exposure to delta-endotoxin significantly prolonged larval development. The weights of prepupae and adults were significantly reduced by exposure to delta-endotoxin, with the greatest effect being from chronic exposure. Separation of the manifestations of stress in time (feeding vs soil stages) and space (toxin damage to the insect gut vs fungal penetration of the cuticle and activity in the hemocoel) may have precluded alteration of insect susceptibility to infection by B. bassiana. Endemic populations of B. bassiana are not expected to influence the development of resistance in the Colorado potato beetle to the delta-endotoxin of B. thuringiensis.

The pecan weevil, Curculio caryae (Horn), is a key pest of pecan. Endemic levels of the entomopathogenic fungus Beauveria bassiana (Balsamo) Vuillemin can occur in pecan orchards and contribute to natural control of C. caryae. Commercial formulations of the fungus can also be applied for suppression of C. caryae. We hypothesized that a clover cover crop enhances B. bassiana efficacy and persistence (e.g., by protecting the fungus against abiotic environmental stresses). The hypothesis was tested by conducting field trials in a pecan orchard in Byron, GA, in 2009 and 2010. The study included four treatments arranged in a factorial with two levels of fungus (endemic fungus only, and application of a commercial B. bassiana product), and two levels of clover (white clover, Trifolium repens L., and no clover). Fungal persistence was measured by determining the number of CFUs per gram of soil over time (during 42 d postapplication of B. bassiana in 2009 and 29 d in 2010). Efficacy was measured by capturing naturally emerging C. caryae and subsequently determining mortality and mycosis (over 24 d in 2009 and 17 d in 2010). In 2009, greater prevalence of B. bassiana conidia was detected in plots receiving fungal applications compared with no fungus applications, and no clear effect of clover was observed in plots receiving B. bassiana applications in either year. In 2010, B. bassiana prevalence in the endemic fungus plus clover treatment was higher than fungus without clover, and was similar to plots receiving additional B. bassiana applications. Given that we observed enhanced persistence of endemic B. bassiana in 2010 but not 2009, the impact of clover appears to be a cumulative effect. Mortality of C. caryae (averaged over the sampling periods) ranged between 68-74% in plots receiving B. bassiana applications and 51-56% in plots with endemic fungus only. C. caryae mortality and mycosis data also provided evidence that endemic B. bassiana efficacy was enhanced by clover relative to plots without clover (with no clear clover effect on plots receiving fungus applications). Thus, we conclude that natural control of C. caryae can increase when clover is grown in pecan orchards with endemic populations of B. bassiana.

Injection of zymosan or dead yeast cells enhanced the inhibitory activity against exocellular Beauveria bassiana proteases in the cell - free haemolymph of Galleria mellonella larvae . Pre - injected larvae exhibited no decreased mortality after subsequent injection with living B. bassiana blastospores but survived for a prolonged time before death . Increased levels of protease inhibitors in the haemolymph were also observed after injection of B. bassiana proteases . In contrast , no enhanced inhibitory activity against B. bassiana proteases was detected in infected larvae when mycosis was initiated with conidia which enabled the fungus to invade host larvae through the integument in a natural manner . B. bassiana proteases were not completely inhibited by the addition of cell - free haemolymph . Protease inhibitors obtained after heat and trichloroacetic acid precipitation of cell - free haemolymph were added to the protein medium of B. bassiana to study the effect on its growth in vitro. Enriched fractions from pre - injected larvae delayed fungal growth in comparison with fractions from untreated larvae , suggesting that delayed mortality of immunized G. mellonella larvae infected with B. bassiana is due to enhanced levels of protease inhibitors . A non - virulent form of the same strain exhibited reduced capacity to release proteases in vitro. The results strongly suggest that the capacity of insects to release inhibitors against fungal proteases influences their susceptibility against entomopathogenic fungi .

Field experiments were conducted in steel bins containing 13,600 kg of hard red winter wheat, Triiticum aestivum L. One bin was treated with ozone and the second bin served as a control. Stored grain insects were placed in bins for 1-, 2-, 3-, and 4-d exposure periods in sampling tubes to test ozone concentrations of 0, 25, 50, and 70 parts per million by volume (ppmv). Ozone treatments on eggs and larvae of Plodia interpunctella (Hübner) were not effective, but pupae were more susceptible. Sitophilus oryzae (L.) adults were the most susceptible species with 100% mortality reached after 2 d in all ozone treatments. However, some progeny were produced at all concentrations and exposure periods. Tribolium castaneum (Herbst) adults had 100% mortality only after 4 d at 50 or 70 ppmv. No T. castaneum progeny were produced after 2-4 d at 70 ppmv. For Rhyzopertha dominica (F.), Cryptolestes ferrugineus (Stephens), and Oryzaephilus surinamensis (L.), 100% mortality was never achieved and progeny were produced at all ozone concentrations. Laboratory experiments, testing the effectiveness of ozone in controlling psocids, were conducted in two polyvinyl chloride cylinders each containing 55 kg of hard red winter wheat. Ozone treatment at a concentration of 70 ppmv was highly effective against adult female Liposcelis bostrychophila Badonnel and Liposcelis paeta Pearman after only 1 d of exposure. However, it was not effective against eggs of both species at all exposure periods. Ozonation has potential for the control of some stored grain insect pests on wheat.

Interactions between the immature stages of Diadegma semiclausum, an endolarval parasitoid of Plutella xylostella, and the fungal entomopathogen Beauveria bassiana were investigated in the laboratory. Detrimental effects of B. bassiana on D. semiclausum cocoon production and adult parasitoid emergence increased with increasing pathogen concentration and some parasitoid larvae became infected by B. bassiana within hosts. The negative impact of B. bassiana on D. semiclausum cocoon production decreased as temporal separation between parasitism and pathogen exposure increased. Adult parasitoid emergence was significantly compromised by the highest rates of B. bassiana tested even when exposure of host larvae to the pathogen was delayed until one day before predicted parasitoid cocoon formation. Parasitoid pupae were infected by the pathogen in all B. bassiana treatments which did not preclude their development.

Beauveria bassiana, an entomopathogenic fungus, was evaluated for its potential against second and third instar larvae of Culex quinquefasciatus, Anopheles stephensi and Aedes aegypti. Conidiospores of this fungus were effective in causing infection leading to mortality of different larval instars. Larvae of Cx. quinquefasciatus were more susceptible to infection than An. stephensi and the second instar larvae of these two species were more susceptible than third instar larvae. Larvae of Ae. aegypti were resistant to infection by B. bassiana.

Imidacloprid and the entomopathogenic fungus Beauveria bassiana (Balsamo) Vuillemin are both used to control the whitefly Bemisia argentifolii Bellows & Perring. We tested whether the two control strategies acted additively, synergistically, or antagonistically when combined for whitefly control. We found antagonism in that B. bassiana inhibited the effectiveness of imidacloprid. When B. bassiana was combined with imidacloprid, insect response was either less than or similar to (depending on B. bassiana rates) that when imidacloprid was used alone. Adding imidacloprid to B. bassiana treatments always increased mortality, but the increase was less than additive. Beauveria bassiana spore germination and colony formation were not inhibited by imidacloprid in vitro, and B. bassiana did not adsorb or degrade imidacloprid in a tank mix. We hypothesize that B. bassiana caused a behavioral response that reduced insect feeding and uptake of imidacloprid.

The influence of sanitation on responses of life stages of the red flour beetle, Tribolium castaneum (Herbst) (Coleoptera: Tenebrionidae), was investigated in a pilot flour mill subjected to three, 24-h heat treatments by using forced-air gas heaters fueled by propane. Two sanitation levels, dusting of wheat flour and 2-cm-deep flour, were created in 25 plastic bioassay boxes, each holding eggs, young larvae, old larvae, pupae, and adults of T. castaneum plus two temperature sensors. Data loggers (48) were placed on the five mill floors to record air temperatures. The time required to reach 50 degrees C, time above 50 degrees C, and the maximum temperature among mill floors and in bioassay boxes were measured. The maximum temperature in bioassay boxes and in the mill was lower on the first floor than on other floors. This trend was apparent in time required to reach 50 degrees C and time above 50 degrees C, especially in compartments with 2-cm-deep flour. The mean +/- SE mortality of T. castaneum life stages on the first floor was 55.5 +/- 12.9-98.6 +/- 0.8%; it was 93.2 +/- 6.7-100 +/- 0.0% on other floors. Adults were the least susceptible stage. Mortality of T. castaneum stages in compartments with 2-cm-deep flour was generally lower than those with flour dust. Costs for the three heat treatments ranged from US$27,438 to $28,838. An effective heat treatment can be conducted within 24 h, provided temperatures on mill floors reach 50 degrees C in 8-12 h and are held above 50 degrees C for at least 10-14 h, with maximum temperatures held between 50 and 60 degrees C.

The susceptibility of different geographical strains of Phlebotomus papatasi to a cytoplasmic polyhedrosis virus (CPV) was determined experimentally by feeding polyhedra to larvae. Of the Indian P. papatasi, 15.6% became infected, whereas Egyptian P. papatasi were mostly refractory. Infection rates were not augmented in colony flies from the Jordan Valley, 23.8% of which were naturally infected with CPV. The infectivity of Serratia marcescens and Beauvaria bassiana to P. papatasi were determined experimentally. A suspension of B. bassiana spores or S. marcescens bacteria, ingested by P. papatasi in sucrose solution, did not significantly augment mortality rates or reduce the number of eggs oviposited. However, B. bassiana spores smeared on a filter paper constituting 1 or 5% of the surface area available to flies induced 100% mortality of P. papatasi on days 5 and 4, respectively. Mortality in Lutzomyia longipalpis reached 100% on day 4. There were markedly lower mortality rates in the control groups and more eggs were produced by these females (P. papatasi: control = 48.5; experimental = 0.9-1.6 eggs/female; L. longipalpis; control = 17.1; experimental = 0 eggs/female). From wild-caught Colombian Lutzomyia spp., a nonfluorescent pseudomonas, an Entomophthorales fungus, and a Trypanosomatid protozoon (probably Leptomonas) were isolated in culture media. Gregarines (Ascogregarina saraviae) and nematodes (Tylenchida and Spirurida) were also recorded. In laboratory-reared flies, an ectoparasitic fungus was associated with high mortality rates of first instar Lutzomyia spp. larvae. Opportunistic ectoparasitic aggregates of bacteria, yeast, and fungi on the tarsi of colonized L. longipalpis and P. papatasi hindered their mobility and were associated with reduced colony vigor. Aspergillus flavus, B. bassiana, and S. marcescens were isolated from laboratory-bred P. papatasi adults.

The spectrum and abundance of entomopathogenic fungi in agricultural soil receiving different pesticide applications were evaluated . Seven small field plots within a barley crop were selected . Each plot had received a different pesticide treatment at slightly higher than the field rate each year for the previous 12-19 years . The field plots received either benomyl (fungicide) , triadimefon (fungicide) , aldicarb (insecticide) , chlorfenvinphos (insecticide) , glyphosate (herbicide) , all five of these pesticides or no pesticides at all (control) . Soil sampled from each plot was baited with Galleria mellonella larvae at either 18 or 26 C . Five species of entomopathogenic fungi infected these larvae . Beauveria bassiana was the dominant species , and the only species for which infection levels were high enough to be analyzed statistically . Significantly fewer G. mellonella larvae became infected with B. bassiana in soil treated with benomyl than in other treatments . This deleterious effect was confirmed in in vitro experiments where benomyl inhibited fungal growth significantly . Chlorfenvinphos also reduced the proportion of G. mellonella becoming infected with B. bassiana slightly (although not statistically significantly) . This effect was much greater in in vitro experiments in which chlorfenvinphos inhibited fungal growth significantly . B. bassiana- induced mortality was significantly greater in G. mellonella larvae placed in soil treated with triadimefon than in other treatments . In vitro studies , however , demonstrated that triadimefon inhibited rather than stimulated growth . A similar anomaly between field and laboratory observations was seen in the case of aldicarb . There was no significant difference in the number of G. mellonella larvae found infected with B. bassiana in soil treated with aldicarb than in the control soil . However , in the in vitro studies , aldicarb stimulated fungal growth at all concentrations except 10 times the field rate . In general , significantly fewer larvae became infected with B. bassiana at 26 C than at 18 C in all treatments . Pesticides may have a direct impact on the natural occurrence , infectivity and population dynamics of entomopathogenic fungi . They also affect other macro - and microorganisms in the soil which may interact with the ento mopathogenic fungi . In combination with climate and soil variables , these effects are difficult to separate . Although any suppression of these fungi may be detrimental to their capacity as natural enemies , this study has demonstrated that pesticides used under field conditions are unlikely either to kill all the entomopathogenic fungi present in the treated area or to limit their recolonization . It seems likely , therefore , that pesticides have the potential to be used in conjunction with mycoinsecticides in integrated pest management systems .
