The development of a direct competitive enzyme-linked immunosorbent assay (ELISA) based on polyclonal antibodies specific for semicarbazide (SEM) is described. Molecular modelling of the hapten mimics and other key components of the assay system was conducted to explain antibody properties in relation to hapten design. The small aliphatic molecule SEM was coupled to 3-carboxybenzaldehyde to produce carboxyphenyl-SEM (CPSEM), for the generation of specific antibodies. Five rabbits produced antibodies against NPSEM (used in direct and indirect ELISA formats) exhibiting a 50% binding inhibition level (IC50 values) of 0.06-2.28 μg L−1 in assay buffer for SEM. The most sensitive indirect assay based on the antibody MVK39 showed a high dynamic range providing a linear readout in the range of 0.01-0.2 μg L−1. Antibody MVK31 (IgG) allowed specific SEM detection at an IC50 = 0.14 μg L−1 in direct ELISA and was evaluated using solvent extracted SEM-spiked porcine and baby food samples. Recovery levels determined from fortified samples (0.5, 1.0, 1.5, 5, 10 and 20 μg kg−1) of porcine and baby food ranged from 82.9 to 105.3%, respectively, with a coefficient of variation less than 15.5%. Respective detection capability and threshold of the assay for porcine muscle, set on the basis of acceptance of no false negative results, was 0.3 and 0.11 μg kg−1.  相似文献

A general and broad class-specific enzyme-linked immunosorbent assay was developed for the O,O-dimethyl organophosphorus pesticides, including malathion, dimethoate, phenthoate, phosmet, methidathion, fenitrothion, methyl parathion and fenthion. Three haptens with different spacer-arms were synthesized. The haptens were conjugated to bovine serum albumin (BSA) for immunogens and to ovalbumin (OVA) for coating antigens. Rabbits were immunized with the immunogens and six polyclonal antisera were produced and screened against each of the coating antigens using competitive indirect enzyme-linked immunosorbent assay for selecting the proper antiserum. The effect of hapten heterology on immunoassay sensitivity was also studied. The antibody-antigen combination with the most selectivity for malathion was further optimized and tested for tolerance to co-solvent, pH and ionic strength changes. The IC50 values, under optimum conditions, were estimated to be 30.1 μg L−1for malathion, 28.9 μg L−1 for dimethoate, 88.3 μg L−1 for phenthoate, 159.7 μg L−1 for phosmet, 191.7 μg L−1 for methidathion, 324.0 μg L−1 for fenitrothion, 483.9 μg L−1 for methyl parathion, and 788.9 μg L−1 for fenthion. Recoveries of malathion, dimethoate, phenthoate, phosmet and methidathion from fortified Chinese cabbage samples ranged between 77.1% and 104.7%. This assay can be used in monitoring studies for the multi-residue determination of O,O-dimethyl organophosphorus pesticides.  相似文献

Furazolidone has been banned from use in food animals because of its carcinogenicity and mutagenicity, but its continued misuse is widespread in aquacultures. Therefore, there is an urgent need for a simple, reliable, and rapid method for the detection of its marker residue, 3-amino-2-oxazolidinone (AOZ), in aquatic products. In this regard, we modified a simplified indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) to address this need. A good linearity was achieved over a concentration range of 0.05-12.15 μg L−1, and the IC50 value was 0.96 μg L−1. The sample preparation was simple and effective included water bath treatments, acid hydrolysis combined with overnight derivatization of AOZ by benzaldehyde. The limit of detection and the limit of quantification were 0.15 and 0.3 μg kg−1. The recoveries of AOZ in all tissues were between 78.0-95.3% at the levels of 0.3, 1.0, and 2.0 μg kg−1. The inter-assay variability was less than 19.1%. The modified ic-ELISA was applied in quantification of AOZ elimination in carp. The results showed that AOZ was quite difficult to eliminate. Good correlations of the results obtained by ELISA and LC-MS/MS were observed in incurred carp muscle (r = 0.9923) and carp plasma (r = 0.9915) at the levels of 2.5-571.8 μg kg−1 (μg L−1). Better results were obtained by modified ic-ELISA when compared with commercial ELISA kit. Therefore, the present assay is considered a rapid, accurate, reliable, and inexpensive method for the detection of furazolidone-residues in the edible tissues of aquatic animals.  相似文献

Semicarbazide (SEM), the marker residue for the banned nitrofuran veterinary antibiotic nitrofurazone (NFZ), has been detected regularly in foods (47% of recent nitrofuran EU Rapid Alerts involve SEM). However, the validity of SEM as a definitive marker for NFZ has been undermined by SEM arising from other sources including azodicarbonamide, a plastics blowing agent and flour treatment additive. An inexpensive screening test for SEM in food matrices is needed—all SEM testing currently uses expensive LC-MS/MS instrumentation. We now report the first production of antibodies against derivatised SEM. A novel carboxyphenyl SEM derivative was used to raise a polyclonal antibody that has been incorporated into a semi-quantitative microtitre plate ELISA, validated according to the criteria set out in Commission Decision 2002/657/EC, for use with chicken muscle. The antibody is highly specific for derivatised SEM, cross-reactivity being 1.7% with NFZ and negligible with a wide range of other nitrofurans and poultry drugs. Samples are derivatised with o-nitrobenzaldehyde and simultaneously protease digested before extraction by cation exchange SPE. The ELISA has a SEM detection capability (CCβ) of 0.25 μg kg−1 when a threshold of 0.21 μg kg−1 is applied to the selection of samples for confirmation (lowest observed 0.25 μg kg−1 fortified sample, n = 20), thus satisfying the EU nitrofurans’ minimum required performance limit of 1 μg kg−1. NFZ-incurred muscles (12) containing SEM at 0.5-5.0 μg kg−1 by LC-MS/MS, all screened positive by this ELISA protocol which is also applicable to egg and chicken liver.  相似文献

Tetracyclines (TCs) are widely used for prevention and control of infectious diseases and have a great activity against variety of Gram-positive and Gram-negative bacteria. Due to the widespread use of TCs in animal husbandry, it can lead to an increase the risk of TCs remaining in human food. To protect consumers, many countries have set acceptable tolerance levels for these drugs. Therefore, it is necessary to establish a suitable analytical technique with specificity, sensitivity and simplicity.The biotin-avidin mediated ELISA method was performed to determine TC residues in honey quantitatively. By using PBS-EDTA assay buffer at pH 7.2, a honey solution of TC standard was prepared and diluted. And no additional pre-treatment of sample was required in this method. The limit of detection and limit of quantitation of the optimized method were 3.98 × 10−10 M (0.19 μg L−1) and 7.94 × 10−10 M (0.38 μg L−1), respectively, and the dynamic range was from 1.52 μg L−1 to 152 μg L−1 of TC in honey. No cross-reactivity was observed with the structurally similar compounds, and mean percent recoveries of TC spiked in honey ranged from 95% to 101%. Compared to other methods, this method was superior in terms of detection limit, dynamic range, and % recovery with simple sample-preparation.  相似文献

Polyclonal antibodies against microcystin-LR (MC-LR), a cyclic heptapeptide toxin, were generated in rabbits using MC-LR-BSA. An enzyme-linked immunosorbent assay (ELISA) was developed for the characterization of the antibodies and their potential use for analytical purposes. The concentration of MC-LR that inhibits 50% of antibody-antigen binding (IC50) was 0.5 μg L−1 for the indirect ELISA format and 0.9 μg L−1 for the direct ELISA, using MC-LR-horseradish peroxidase conjugate. The limit of detection corresponding to IC80 was found to be 0.06 μg L−1, well below the Word Health Organization level for drinking water of 1 μg L−1. The direct competitive ELISA was applied to water samples and was shown useful for screening purposes. The developed anti-microcystin antibodies were immobilized on solid supports for use in selective solid phase extraction (SPE) systems, prior to liquid chromatography (LC) quantification. An immunoaffinity cartridge (IAC), a Sepharose®-based cartridge incorporating 2 mg of antibodies allowed the selective and quantitative recovery of a mixture of 0.2 μg of MCs showing potential use in sample preparation of real matrices. When applied to water and green algae samples, average recoveries from Sepharose®-based cartridges were in the range of 86-113% for water samples and 85-92% for blue-green algae samples. Selectivity of the IAC clean-up was proven by comparison with non-specific solid phase extraction using octadecylsilica (ODS) sorbent. Results obtained using LC/UV after IAC clean-up agreed well with results obtained using liquid chromatography and mass spectrometry detection (LC/MS and LC/MS/MS) after SPE-C18 clean-up, allowing therefore to validate the resulting technique.  相似文献

Bisphenol A (BPA) is a xenoestrogen found in the environment, in consequence, for the biosensor detection of BPA we raised antibodies (polyclonal (PAbs) and monoclonal (MAbs)) against a structural analogue of BPA, 4,4 bis-(4-hydroxyphenyl) valeric acid (BVA). The kinetics of the MAb-BPA interaction were evaluated and the MAb providing the highest affinity was directly immobilized onto the sensor chip surface to evaluate a direct assay. Afterwards, the performance of the MAbs and the PAbs was compared in an inhibition assay using a BVA-coated chip.The highest sensitivity (limit of detection (LOD) of 0.4 μg L−1) was obtained with MAb 12 in the direct assay. However, the inhibition assay was the most robust and the PAbs showed the highest sensitivity (LOD of 0.5-1 μg L−1). The antibodies were specific for BVA and BPA as only minor cross-reactivities were found toward structurally related compounds or other endocrine disruptors. In the inhibition assay (with a run time of 6 min), water samples spiked with BPA at different levels (0.5-50 μg L−1) resulted in recoveries varying between 68% and 121%. The sensitivity of the inhibition assay could be improved 40 times (LOD of 0.03 μg L−1 with the Mab 12-based assay) using solid phase extraction (SPE).  相似文献

A colorimetric protein phosphatase (PP) inhibition test for the detection of microcystin-LR (MC-LR) has been developed. Three PP2As, one recombinant and two natural versions, as well as one PP1 produced by molecular engineering, were tested. First, assays were performed using the enzymes in solution to compare their sensitivity to MC-LR. The PP2A purchased from ZEU Immunotec and PP1 appeared more sensitive to the toxin than the other enzymes. With PP2A from ZEU Immunotec, the colorimetric test showed a detection limit of 0.0039 μg L−1 and an IC50 value of 0.21 μg L−1. With PP1, the assay gave a detection limit of 0.05 μg L−1 and an IC50 value of 0.56 μg L−1. Therefore, this assay allowed the detection of lower microcystin-LR (MC-LR) concentrations than the maximum level (1 μg L−1) recommended by the World Health Organisation (WHO).The main drawback of this PP-based approach in solution is poor enzyme stabilisation. To overcome this problem, enzymes were entrapped within either a photopolymer or an agarose gel. PP2A from ZEU Immunotec and PP1 were immobilised at the bottom of microwells. The agarose-based tests performed better than the photopolymer-based assay for all of the enzymes. Therefore, the agarose gel is a good candidate to replace the photopolymer, which is generally used in PP-immobilising membranes. The assays based on enzyme-entrapping agarose gels showed detection limits equal to 0.17 μg L−1 and 0.29 μg L−1 with immobilised PP2A from ZEU and PP1, respectively. In view of these performances, these tests can potentially be used for monitoring water quality.  相似文献

Competitive electrochemical enzyme-linked immunosorbent assays based on disposable screen-printed electrodes have been developed for quantitative determination of ochratoxin A (OTA). The assays were carried out using monoclonal antibodies in the direct and indirect format. OTA working range, I50 and detection limits were 0.05-2.5 and 0.1-7.5 μg L−1, 0.35 (±0.04) μg L−1 and 0.9 (±0.1) μg L−1, 60 and 100 μg L−1 in the direct and indirect assay format, respectively. The immunosensor in the direct format was selected for the determination of OTA in wheat. Samples were extracted with aqueous acetonitrile and the extract analyzed directly by the assay without clean-up. The I50 in real samples was 0.2 μg L−1 corresponding to 1.6 μg/kg in the wheat sample with a detection limit of 0.4 μg/kg (calculated as blank signal −3σ). Within- and between-assay variability were less than 5 and 10%, respectively. A good correlation (r = 0.9992) was found by comparative analysis of naturally contaminated wheat samples using this assay and an HPLC/immunoaffinity clean-up method based on the AOAC Official Method 2000.03 for the determination of OTA in barley.  相似文献

A simple and cost effective sample pre-treatment method, dispersive liquid–liquid microextraction (DLLME), has been developed for the extraction of six fluoroquinolones (FQs) from chicken liver samples. Clean DLLME extracts were analyzed for fluoroquinolones using liquid chromatography with diode array detection (LC-DAD). Parameters such as type and volume of disperser solvent, type and volume of extraction solvent, concentration and composition of phosphoric acid in the disperser solvent and pH were optimized. Linearity in the concentration range of 30–500 μg kg−1 was obtained with regression coefficients ranging from 0.9945 to 0.9974. Intra-day repeatability expressed as % RSD was between 4 and 7%. The recoveries determined in spiked blank chicken livers at three concentration levels (i.e. 50, 100 and 300 μg kg−1) ranged from 83 to 102%. LODs were between 5 and 19 μg kg−1 while LOQs ranged between 23 and 62 μg kg−1. All of the eight chicken liver samples obtained from the local supermarkets were found to contain at least one type of fluoroquinolone with enrofloxacin being the most commonly detected. Only one sample had four fluoroquinolone antibiotics (ciprofloxacin, difloxacin, enrofloxacin, norfloxacin). Norfloxacin which is unlicensed for use in South Africa was also detected in three of the eight chicken liver samples analyzed. The concentration levels of all FQs antibiotics in eight samples ranged from 8.8 to 35.3 μg kg−1, values which are lower than the South African stipulated maximum residue limits (MRL).  相似文献

A highly selective sequential injection (SI) method for the automated determination of weak-acid-dissociable cyanides is reported. The analytical procedure is based on the on-line reaction of the analyte with ninhydrin in carbonate medium to form a coloured product (λmax = 510 nm). Cyanides are removed from sample matrix by acidification through a gas-diffusion step incorporated in the SI manifold. The effect of instrumental and chemical variables was studied. By adopting an on-line standard addition protocol, the sensitivity of the proposed method was enhanced drastically, without affecting the determination range. The assay was validated in terms of linearity (up to 200 μg L−1), limit of detection (cL = 2.5 μg L−1), limit of quantitation (cQ = 7.5 μg L−1), precision (sr < 2.5% at 100 μg L−1) and selectivity. High tolerance against critical species such as sulfides and thiocyanates was achieved. The applicability of the method was demonstrated by analyzing tap and mineral water samples at levels below the limits established by international E.U. and U.S. organizations. The percent recoveries were satisfactory in all cases, ranging between 94.2 and 103.6%.  相似文献

A gas chromatography–mass spectrometric (GC–MS) method has been established for the determination of hydrazine in drinking water and surface water. This method is based on the derivatization of hydrazine with ortho-phthalaldehyde (OPA) in water. The following optimum reaction conditions were established: reagent dosage, 40 mg mL−1 of OPA; pH 2; reaction for 20 min at 70 °C. The organic derivative was extracted with methylene chloride and then measured by GC–MS. Under the established condition, the detection and the quantification limits were 0.002 μg L−1 and 0.007 μg L−1 by using 5.0-mL of surface water or drinking water, respectively. The calibration curve showed good linearity with r2 = 0.9991 (for working range of 0.05–100 μg L−1) and the accuracy was in a range of 95–106%, and the precision of the assay was less than 13% in water. Hydrazine was detected in a concentration range of 0.05–0.14 μg L−1 in 2 samples of 10 raw drinking water samples and in a concentration range of 0.09–0.55 μg L−1 in 4 samples of 10 treated drinking water samples.  相似文献

A fast and highly sensitive assay for pepsinogen I (PG I) and pepsinogen II (PG II) by using time-resolved fluoroimmunoassay (TRFIA) detection technique has been developed for the determination of serum PG I and PG II against gastrointestinal diseases. On the noncompetitive assay, one monoclonal antibody (McAb) coated on wells was directed against a specific antigenic site on the PG I or PG II. The McAb, called as labelling McAb, was prepared with the europium-chelate of N-(p-isothiocyanatobenzyl)-diethylenetriamine-N,N,N,N-tetraacetic acid and directed against a different antigenic site on the PG I or PG II molecule. After bound/free separation by washing, the fluorescence counts of bound Eu3+-McAb were measured. The levels of PG in sera from patients or healthy volunteers were determined by PG I and PG II TRFIA using the autoDELFIA1235 system. The measurement ranges of PG I-TRFIA were 3.5-328.0 μg L−1 and those of PG II-TRFIA were 2.0-55.0 μg L−1. The within-run and between-run CVs of the PG I-TRFIA were 1.9% and 4.7%, respectively, and those of PG II-TRFIA were 2.1% and 3.8%, respectively. The recovery rates of PG I-TRFIA and PG II-TRFIA were 102.7% and 104.6%, respectively. The detection limitations of PG I and PG II were 0.05 μg L−1 and 0.02 μg L−1, respectively. The dilution experiments showed the percentage of expected value of PG I-TRFIA was 93.2-102.3% and of PG II-TRFIA was 97.3-110.6%. The cross-reacting rate between PG I and PG II was negligible. The linear correlation of radioimmunoassay (RIA) and TRFIA measurements resulted in a correlation coefficient as 0.926 of PG I and as 0.959 of PG II. The europium-labelling McAbs were stable for at least one year at −20 °C, and the results of the TRFIA with same reagents were reproducible over one year as well. The means of 1600 healthy volunteers were 162.4 ± 52.1 μg L−1 for serum PG I, 11.7 ± 6.8 μg L−1 for serum PG II, and 13.8 ± 7.4 for the PG I/PG II ratio. The normal ranges of Serum PG I levels for healthy volunteers were 58.2-266.6 μg L−1, and those of serum PG II levels were less than 25.3 μg L−1. The availability of a highly sensitive, reliable, and convenient PG-TRFIA method for quantifying PG will allow investigations into the possible diagnostic value of this analysis in various clinical conditions, including gastric carcinoma, duodenal ulcer, gastric ulcer and gastritis. The sensitivity and reproducibility of the assay were satisfactory for clinical applications.  相似文献

A gas chromatography–tandem mass spectrometric (GC–MS/MS) method has been established for the determination of cyanide in surface water. This method is based on the derivatization of cyanide with 2-(dimethylamino)ethanethiol in surface water. The following optimum reaction conditions were established: reagent dosage, 0.7 g L−1 of 2-(dimethylamino)ethanethiol; pH 6; reaction carried out for 20 min at 60 °C. The organic derivative was extracted with 3 mL of ethyl acetate, and then measured by using GC–MS/MS. Under the established conditions, the detection and quantification limits were 0.02 μg L−1 and 0.07 μg L−1 in 10-mL of surface water, respectively. The calibration curve had a linear relationship relationship with y = 0.7140x + 0.1997 and r2 = 0.9963 (for a working range of 0.07–10 μg L−1) and the accuracy was in a range of 98–102%; the precision of the assay was less than 7% in surface water. The common ions Cl, F, Br, NO3, SO42−, PO43−, K+, Na+, NH4+, Ca2+, Mg2+, Ba2+, Mn4+, Mn2+, Fe3+, Fe2+ and sea water did not interfere in cyanide detection, even when present in 1000-fold excess over the species. Cyanide was detected in a concentration range of 0.07–0.11 μg L−1 in 6 of 10 surface water samples.  相似文献

Li2O-ZrO2-BaO-SiO2 glass fibers were produced and their surfaces were coated with zinc oxide. The fibers’ surface morphology was examined by scanning electron microscopy and the zinc oxide layer was characterized by mapping the Kα and Lα lines of zinc by energy dispersive X-ray spectroscopy. The results indicated that a homogeneous and porous layer of ZnO was formed on the fibers’ surface. This layer was subjected to a simultaneous determination of trihalomethanes using headspace-solid phase microextraction-gas chromatography. The study was conducted after evaluating the ideal time of incubation (15 min), extraction (15 min) and desorption (10 min), as well as the effect of the addition of salt (15%, m/v) on the analytical response. A good linear dynamic range was observed individually for trihalomethanes aqueous solutions containing 20 μg L−1 and 500 μg L−1 of trichloromethane, 15 μg L−1 and 250 μg L−1 of dichlorobromomethane and dibromochloromethane and 10 μg L−1 and 100 μg L−1 of tribromomethane, with all the compounds showing correlation coefficients higher than 0.9900.  相似文献

Bismuth and Sb were evaluated as internal standards (IS) to minimize matrix effects on the direct and simultaneous determination of As, Cu, and Pb in cachaça by graphite furnace atomic absorption spectrometry using W-coated platform plus Pd-Mg(NO3)2 as modifier. For 20 μL injected sample, calibration within the 0.5-10 μg L−1 As, 100-1000 μg L−1 Cu and 0.5-30 μg L−1 Pb intervals were established using the ratios As absorbance to Sb absorbance, Cu absorbance to Bi absorbance and Pb absorbance to Bi absorbance versus analytes concentration, respectively. Typical linear correlations of 0.998, 0.999 and 0.999 were, respectively, obtained. The proposed method was applied for direct determination of As, Cu and Pb in 10 commercial cachaça samples and results were in agreement with those obtained by inductively coupled plasma mass spectrometry at 95% confidence level. The found characteristic masses were 30 pg As, 274 pg Cu and 39 pg Pb. The useful lifetime of the graphite tube was around 760 firings. Recoveries of As, Cu and Pb added to cachaça samples varied, respectively, from 98% to 109%, 97% to 108% and 98% to 104% with internal standards and from 48% to 54%, 53% to 92% and 62% to 97% without internal standards. The limits of detection were 0.13 μg L−1 As, 22 μg L−1 Cu and 0.05 μg L−1 Pb. The relative standard deviations (n = 12) for a spiked sample containing 20 μg L−1 As, Pb and 500 μg L−1 Cu were 1.6%, 1.0%, and 1.8% with IS and 4.3%, 5.2%, and 5.5% without IS.  相似文献

An ELISA was developed for routine examination for extensive monitoring and screening programs for the residues of salbutamol in swine serum, animal feed, meat, and meat-related products destined for human consumption in Taiwan. Objectives of the study were to investigate the use of a new immunoassay for the detection of salbutamol residues in swine meat and animal feed samples, and to compare with a commercial kit in field test screens. A fast, simple and reliable sample preparation method for the determination of salbutamol was established. Field trials with 222 swine meat and 120 animal feed samples that were taken from local meat markets, auction markets and feed mills. The application and the results of two ELISA kits (a homemade and a commercial kit) for the screening of salbutamol were presented. Adopting 2 μg kg−1 salbutamol as a cut-off value for swine meat, the commercial β-agonist ELISA had a sensitivity of 85.3% and a specificity of 95.2% versus GC-MS at a cut-off of 2 μg kg−1. The homemade salbutamol ELISA had a sensitivity of 100% and a specificity of 90.9% and gave no false-negative rate results. Furthermore, adopting 20 μg kg−1 salbutamol as a cut-off value for animal feed, both the commercial and homemade ELISA showed 100% sensitivity and 100% specificity of the assays. In conclusion, a sensitive, specific salbutamol polyclonal antibody-based ELISA has been developed that could serve as a rapid screening assay, and the detection of positive samples at the place of sampling can result in more effective control of the illegal use of β-agonists.  相似文献

In this study, the steroid hormone levels in river and tap water samples were determined by using a novel dispersive liquid-liquid microextraction method based on the solidification of a floating organic drop (DLLME-SFO). Several parameters were optimized, including the type and volume of the extraction and dispersive solvents, extraction time, and salt effect. DLLME-SFO is a fast, cheap, and easy-to-use method for detecting trace levels of samples. Most importantly, this method uses less-toxic solvent. The correlation coefficient of the calibration curve was higher than 0.9991. The linear range was from 5 to 1000 μg L−1. The spiked environmental water samples were analyzed using DLLME-SFO. The relative recoveries ranged from 87% to 116% for river water (which was spiked with 4 μg L−1 for E1, 3 μg L−1 for E2, 4 μg L−1 for EE2 and 9 μg L−1 for E3) and 89% to 102% for tap water (which was spiked with 6 μg L−1 for E1, 5 μg L−1 for E2, 6 μg L−1 for EE2 and 10 μg L−1 for E3). The detection limits of the method ranged from 0.8 to 2.7 μg L−1 for spiked river water and 1.4 to 3.1 μg L−1 for spiked tap water. A HPLC method using a coulometric electrode array detector (CEAD) to analyse 4-ethylcatechol in wine was established. The procedure does not require any sample preparation or analyte derivatisation and performs chromatographic separation in a short time. The assay method is linear up to 1520 μg L−1 and precise (R.S.D. < 3%), with limits of detection and quantitation of 1.34 μg L−1 and 2.2 μg L−1, respectively. Recoveries in spiked wine samples ranged from 95% to 104% with a median value of 102% and matrix effects were not observed. The method was applied to the evaluation of the concentration of 4-EC in 250 commercial Italian wines. The red wines analysed had median, 75° percentile and maximum values of 37 μg L−1, 89 μg L−1 and 1610 μg L−1, respectively. For Sangiovese-based wines the mean ratios of 4-EP and 4-EG to 4-EC were 3.7:1 and 0.7:1, respectively. The feasibility of a cheaper fluorimetric approach to 4-EC quantification was investigated.  相似文献
