Although genetic and protein manipulations have been the cornerstone for the study and understanding of biological processes for many decades, complimentary nanoscale observations have only more recently been achieved in the live-imaging mode. It is at the nano measurement level that events such as protein-protein interactions, enzymatic conversions, and single-molecule stochastic behavior take place. Therefore, nanoscale observations allow us to reinterpret knowledge from large-scale or bulk techniques and gain new insight into molecular events that has cellular, tissue, and organismal phenotypic manifestations. This review identifies pertinent questions relating to the sensing and directional component of cancer cell chemotaxis and discusses the platforms that provide insight into the molecular events related to cell motility. The study of cell motility at the molecular imaging level often necessitates the use of devices such as microinjection, microfluidics, in vivo/intravital and in vitro chemotaxis assays, as well as fluorescence methods like uncaging and FRET. The micro- and nanofabricated devices that facilitate these techniques and their incorporation to specialized microscopes such as the multiphoton, AFM, and TIR-FM, for high-resolution imaging comprise the nanoplatforms used to explore the mechanisms of carcinogenesis. In real-time observations, within a milieu of physiological protein concentrations, true states of dynamic and kinetic fluxes can be monitored.  相似文献

Neuroimaging of primary headache syndromes, such as cluster headache and migraine, has begun to provide a glimpse of the neuroanatomical and physiological basis of the conditions. Although these headache types have been widely described as vascular, there is now considerable imaging and clinical evidence to suggest that they are primarily driven from the brain. The shared anatomical and physiological substrate for both of these clinical problems is the neural innervation of the cranial circulation. Functional imaging with positron emission tomography (PET) has shed light on the genesis of both syndromes, documenting activation in the midbrain and pons in migraine, and in the hypothalamic grey in cluster headache. These areas are involved not simply as a response to first division nociceptive pain impulses but specifically in each syndrome, probably in some permissive or dysfunctional role. In a recent PET study in cluster headache, as well as brain activation, tracer pooled in the region of the major basal arteries. This is likely to be due to vasodilatation of these vessels during the acute pain-attack and represents the first convincing activation of neural vasodilator mechanisms in humans. The author takes the view that the known physiology and pathophysiology of the systems involved dictate that these disorders should be collectively regarded as neurovascular headaches to place emphasis on the interaction between nerves and vessels, which is the underlying characteristic of these syndromes. Understanding this neurovascular relationship facilitates an understanding of the pain mechanisms, while characterising the CNS dysfunction will ultimately allow us to dissect out the basic pathogenesis of these disorders.  相似文献

As a part of Ministry of New and Renewable Energy (MNRE), Government of India programme for promoting small hydropower (shp) development, efficiency testing and performance evaluation of Small Hydro Power plants is mandatory for receiving financial incentives by plant developer. The task of efficiency testing and performance evaluation of shp plants was assigned by MNRE to Alternate hydro Energy Centre (AHEC) Indian Institute of Technology Roorkee in the year 2003 along with financial support to acquire the required instrumentation and develop expertise. Over 200 small hydro plants (SHPs) of different capacity (1–25 MW) as well as types viz dam toe based, run of river and canal fall based have been tested for efficiency and performance evaluation during the period 2005–2018 in different states of India as per IEC-60041, IEC-62006, ISO -1438-1 and ISO-748 to the extend feasible as per available resources and time. During efficiency testing of hydro plants, it has been found that discharge measurement is very challenging activity as the most of the plant owners did not provide the required space and provisions for mounting/fixing the instruments for discharge measurement. After commissioning of the plants it becomes extremely difficult to select the ideal/suitable locations and providing the provisions for mounting different measuring instruments. Various problems observed during discharge measurement are discussed in this paper and may be helpful for testing in future.  相似文献

N-连接糖基化修饰是蛋白质上常见的翻译后修饰,发生在特征氨基酸序列N-X-T/S/C(X≠P)中的天冬酰胺上。N-连接糖具有五糖核心结构,包括高甘露糖型、杂合型和复杂型等3种主要类型。N-糖基化在修饰位点上具有微观和宏观不均一性,其功能与修饰位点和N-连接糖结构对应。对蛋白质N-糖基化位点和结构特异性的精准鉴定和定量,是基于质谱的N-糖蛋白质组学发现N-糖蛋白相关疾病诊断和预后标志物、药物靶点,以及研发高效靶向大分子药物的主要任务。随着高效富集材料、液相色谱分离技术、串联质谱和检测技术以及生物信息学数据库搜索方法的逐步发展和成熟,目前N-糖蛋白质组学已能够对体系中的N-糖基化进行高通量位点和结构特异性鉴定和定量。本文主要介绍N-糖蛋白质组学的样本制备、高效液相色谱-质谱联用分析、生物信息学数据处理等基本步骤,以及由这些步骤组成的完整分析流程的代表性应用研究。  相似文献

This paper describes the design, construction, and characterization of a variable-temperature wafer apparatus for use in the detection of electrically active defects which produce deep levels in the band gap of silicon. In its present form, the wafer chuck can heat and cool wafers as large as 51 mm in diameter over the temperature range from -196 degrees to 350 degrees C. Heating rates as high as 7 degrees C/s have been achieved. Sensitivity for electrical measurements is sufficient to allow current measurements as low as 0.2 pA or capacitance changes (1 MHz) as small as 5 fF. The use of this apparatus is illustrated by wafer mapping the gold defect density in diodes fabricated across a silicon wafer.  相似文献
