This study reports the preparation of four varieties of water extract from sweet potato leaves from Taiwan, including TNG10, TNG57, TNG66 and YSP, and evaluates their antioxidative activity. The EC50 values (scavenging DPPH radicals) of TNG10, TNG57, TNG66 and YSP were 0.27 ± 0.01, 0.19 ± 0.01, 0.41 ± 0.02, and 0.31 ± 0.02 mg/ml, respectively, on a freeze-dry weight basis. The total phenolic contents of these water extracts were in the order: TNG57 > TNG10 > TNG66 > YSP. The TNG10 and TNG57 extracts exhibited better reducing power and scavenging effects of superoxide radicals than did TNG66 and YSP. At a concentration of 1 mg/ml, TNG10 and TNG57 significantly protected HaCaT cells from H2O2-induced cytotoxicity. The water extracts of YSP had more flavonoids than had those of TNG66 which may have contributed to their higher activity in many antioxidative assays. These results suggest that the water extracts of all four varieties of sweet potato leaves, and especially TNG10 and TNG57, display potent antioxidative effects.  相似文献

Agrocybe cylindracea (DC: Fr.) Mre. was available in the form of fruit bodies, mycelia and fermentation filtrate. From these three forms, hot-water extracts were prepared and their antioxidant properties were studied. Antioxidant activities of hot-water extracts from fruit bodies, mycelia and filtrate were 63.6%, 81.6% and 56.8% at 20 mg ml−1, respectively. EC50 values in reducing power were 2.72, 3.97 and 3.09 mg ml−1 whereas those in scavenging abilities of 1,1-diphenyl-2-picrylhydrazyl radicals were 0.62, 1.66 and 0.82 mg ml−1 for fruit bodies, mycelia and filtrate, respectively. At 20 mg ml−1, the scavenging abilities of hydroxyl radicals were 80.1%, 57.0% and 54.3% for fruit bodies, mycelia and filtrate, respectively. With regard to EC50 values in chelating abilities on ferrous ions, the hot-water extract from filtrate was better than that from mycelia. Total phenols were the major naturally occurring antioxidant components found in hot-water extracts and in the range of 23.74–30.16 mg g−1. From EC50 values obtained, it can be concluded that hot-water extracts from three forms of A. cylindracea were good in antioxidant properties.  相似文献

Antioxidant activities (μmol Trolox equivalent (TE)/g fresh weight) of 19 sweet potato genotypes with distinctive flesh colour (white, cream, yellow, orange and purple) were measured by oxygen radical absorbance capacity (ORAC), 2,2-diphenyl-1-picrylhydrazyl (DPPH), and 2,2′-azinobis(3-ethyl-benzothiazoline-6-sulfonic acid) (ABTS). Total phenolics were measured using the Folin–Ciocalteau method, total anthocyanins by the pH-differential method, and β-carotene by HPLC. The total antioxidant activity (hydrophilic + lipophilic ORAC) was highest (27.2 μmol TE/g fresh weight (fw)) for NC415 (purple-fleshed) and lowest (2.72 μmol TE/g fw) for Xushu 18 (white-fleshed). The hydrophilic-ORAC values were significantly correlated with the DPPH (R2 = 0.859) and ABTS (R2 = 0.761) values. However, the lipophilic-ORAC values were poorly correlated with the β-carotene contents (R2 = 0.480). The total phenolic contents (0.011–0.949 mg chlorogenic acid equivalent/g fw) were highly correlated with the hydrophilic-ORAC (R2 = 0.937) and DPPH (R2 = 0.820) values. Therefore, the total phenolic content can serve as a useful indicator for the antioxidant activities of sweet potatoes.  相似文献

Vegetables are the major vehicles for the entry of nitrate into the human system. Ever-increasing concern over nitrate toxicity has directed a number of countries to lay down maximum allowable threshold concentrations with regards to nitrate-N in vegetables. Fiji is an independent island nation, located in the southern Pacific Ocean, has a tropical oceanic climate and hence expected to have high nitrate-N levels in vegetables. Thus, the present study was devoted to establish a flow injection analysis (FIA) technique for nitrate-N determination in Fiji’s commonly consumed fresh and cooked root vegetables such as potato (Solanum tuberosum), dalo (Colocasia esculenta), sweet potato (Ipomoea batatas) and carrot (Daucus carota L.). Activated carbon extraction technique was applied to extract nitrate-N. FIA with colorimetric detection technique having linear dynamic range of determination 1.0–20.0 mg L−1 and detection limit of 0.042 mg L−1 (0.34 mg kg−1), using sulphanilamide and N-(1-naphthyl)ethylenediamine dihydrochloride as colour reagents, was used to determine nitrate-N contents in selected fresh and cooked root vegetables. The samples throughput was 38 h−1. The effects of various cooking (boiling, baking and frying) methods on nitrate-N contents in root vegetables have also been studied. The study shows that the nitrate content of fresh root vegetables ranges from 53.76–258.00 mg kg−1 whereas boiling reduces nitrate content by 23.30–42.62%. The frying in soya bean oil elevates nitrate contents from 204.53–299.12% but after baking nitrate contents remains almost constant with slight increasing trend from 2.80–8.43%. A comparison of the nitrate obtained by standardised method and the nitrate contents in vegetables of other countries are also presented.  相似文献

The optimised extraction conditions of red pigments (RP) from Camellia japonica obtained with an orthogonal design L9(34) were solid/liquid ratio, temperature, pH and extraction time as 1/10, 60 °C, 1.5 and 4 h, respectively. The RP were then purified by the macroporous resin method, which showed the resin LX-68 was appropriate for purifying the pigments from C. japonica. The antioxidant activities of these pigments were also investigated using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and hydroxyl radical in vitro model systems. The DPPH scavenging activity of pigment extract was comparable to that of standard butylated hydroxyanisole (BHA). The IC50 values of the RP and BHA were 4.55 and 4.17 μg ml−1, respectively. The pigments showed higher hydroxyl radical-scavenging activities than that of mannitol at the same concentration. Following activity-oriented separation, (−)-epicatechin was isolated as an active principle, which exhibited excellent DPPH free radical scavenging activities with IC50 5.08 μg ml−1.  相似文献

为比较福建产区主栽紫薯抗氧化物质成分含量及其体外抗氧化活性,对7个不同品种紫薯中总酚、总黄酮、总花色苷及花青素组分进行了分析,并测定其清除自由基活性。结果表明:福宁紫3号紫薯中所含抗氧化活性成分如总酚、总黄酮、总花色苷和花青素的含量均为最高。通过高效液相色谱法分析测定紫薯中含有的花青素单体主要为飞燕草色素、芍药色素和矢车菊色素,不同品种紫薯中花青素组成及含量表现出的一定的差异性。福宁紫3号紫薯表现出较强的自由基清除活性,0.5 mg·L-1紫薯鲜样提取液DPPH自由基、OH自由基及ABTS清除率分别达到88.4%、49.1%、90.1%。 相对于其他品种,福宁紫3号紫薯在抗氧化活性成分含量及体外抗氧化活性方面表现最佳。  相似文献

The underutilized, edible green raw (whole) fruits of amra, Spondias pinnata K. (Anacardiaceae family) from the eastern region of India were investigated for their nutraceutical and therapeutic potential. A thorough nutritional characterization of this fruit demonstrated it as a source of energy (348 kcal/100 g DM), phenolic compounds, natural antioxidants and minerals. It is also a moderate source of ascorbic acid, malic acid, calcium, phosphorus and other nutrients. The phytochemical screening revealed 5.12 ± 0.32 mg 100 g− 1 DM of alkaloids followed by saponins and tannins. All assays were carried out in three different solvent extracts of the fruit. Total phenolic, flavonoid and flavonol contents were obtained as 210 ± 1.89 mg GAE, 28.0 ± 0.91 mg CE and 9.97 ± 0.72 mg RE respectively in 100 mg mix solvent extract (MSE). Antioxidant activity of different extracts (as DPPH scavenging) ranged from 0.73 to 0.59 mg ml− 1 as IC50 value, ABTS with a trolox equivalent antioxidant concentration (TEAC) value as 0.68 to 0.83 and FRAP 5.97 to 7.93 mg TE 100 mg− 1 extract. LC- MS/MS analysis revealed the presence of gallic acid, salicylic acid, chlorogenic acid, ellagic acid, p-coumaric acid, 6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid, quercetin, catechin, myrecetin and rutin. The MSE showed the highest antimicrobial activity (against Listeria monocytogenes (MIC 1.8 mg ml− 1)) and ??-amylase inhibition capacity (as IC50: 29.3 mg ml− 1 extract). GC/MS screening showed the presence of vitamin E, furfural, phytosterol, campesterol and fatty acids. Analysis of volatile flavor showed isopropyl myristinate as a major compound followed by the other monoterpenes and sesquiterpenes. The current study explains the nutritional as well as medicinal utility of the fruit which is a rich source of minerals and antioxidants such as phenols and flavonoids.  相似文献

Concentrated muscadine pomace extract was chromatographically analyzed for its individual phenolic, flavonoid, and anthocyanidin compounds. This extract was also characterized regarding its total phenolic content (TPC), total flavonoid content (TFC), antioxidative activities in terms of scavenging DPPH free radicals, reducing ferric, and chelating Fe2+. The TPC of this product was 34.1 ± 1.8 mg of gallic acid equivalents(GAE)/g of extract, and TFC was 3.0 ± 0.3 mg of quercetin equivalents/g of extract. Some phenolic compounds including ellagic acid, gallic acid, (−)-epicatechin, (−)-epigallocatechin, catechin, myricetin, quercetin, and kaempferol were identified. Some anthocyanidins including delphinidin, cyanidin, petunidin, peonidin, and malvidin were also identified in the extract by using a combination of retention and spectral properties on a reverse-phase HPLC–PDA. In addition, 3,3′,4,4′5,5′-hexahydroxystilbene-a resveratrol analogue present in the extract was identified for the first time by LC–MS. The results from this study demonstrate that the muscadine pomace extract is rich of natural antioxidants such as phenolics, flavonoids and anthocyanins, and possesses strong antioxidant properties. Besides, the developed methods can be used for routine quality control of the muscadine products for manufacturing efficiency and consistency.  相似文献

The total phenols content (Folin–Ciocalteau assay) and antioxidant capacity (ferric reducing/antioxidant power – FRAP) of 41 plants from Córdoba (Argentina) were analyzed. Phenol content ranged from 8.2 to 100.2 mg GAE/g. FRAP ranged from 85.2 to 1862.0 μmol of Fe(II)/g. Capparis atamisguea had the lowest values of total phenols content and antioxidant capacity (8.2 mg GAE/g and 85.2 μmol of Fe(II)/g, respectively), while Ligaria cuneifolia exhibited the highest values (100.2 mg GAE/g and 1862.0 μmol of Fe(II)/g, respectively). A significant linear correlation (p < 0.05) was found (0.9125) between phenols content and antioxidant capacity. Results support the idea that these plants may be a good source of natural antioxidants for food applications. Plants from the Asteraceae family (the most representative of the Córdoba flora) were further tested for their DPPH radical scavenging activity. Some plant extracts were tested in a simple food system to investigate to their potential use in foods.  相似文献

The contents of As, Cd, Cr, Cu, Fe, Hg, Mg, Mn, Ni, Pb, and Zn in eleven fruiting bodies of Boletus tomentipes were determined. The results showed the values of the studied elements decreased in the order: Mg (208–279 mg kg−1) > Fe (106–137 mg kg−1) > Mn (29.5–46.8. mg kg−1) > Zn (18.7–23.1 mg kg−1), > Cu (11.4–15.8 mg kg−1) > Cr (3.36–4.78 mg kg−1) > Pb (1.38–3.88 mg kg−1) > Ni (1.68–3.01 mg kg−1) > Cd (0.16–0.32 mg kg−1) > As (0.10–0.24 mg kg−1) > Hg (<0.06 mg kg−1).  相似文献

Fruit weight, antioxidant capacity, total anthocyanins, total phenolics, ascorbic acid, soluble solid content (SSC), reducing sugar and acidity of a number of selected cornelian cherry (Cornusmas L.) genotypes of varied pigmentation were investigated. Two methods, namely β-carotene bleaching and ferric reducing antioxidant power (FRAP) were used to determine total antioxidant capacity, while Folin–Ciocalteu reagent was used to determine total phenols. Fruit weight, SSC and ascorbic acid content of genotypes were 2.09–9.17; 12.53–21.17% and 29–112 mg/100 g, respectively. Antioxidant activity and total phenolic content varied among genotypes and 44-18 genotype had the highest antioxidant capacity using both methods. This genotype also had the highest total phenolic (74.8 mg GAE/g DW) and total anthocyanin (115 mg cyanidin-3-glucoside equivalents /100 g FW) content. There are linear relationships between antioxidant capacities and total phenols. The present study demonstrates the potential of certain cornelian cherry genotypes, notably 44-18, for improvement of nutritional value through germplasm enhancement programmes.  相似文献

Macedonian Vranec wines were analysed by HPLC coupled with DAD and MS detections and by spectrophotometric methods. ESI-IT MS and MS–MS methods with alternating ionisation polarity were used for identification of the phenolic compounds. Both, nonflavonoids (stilbens, hydroxybenzoic and hydroxycinnamic acids and derivatives) and flavonoids (flavonols, flavan-3-ols and anthocyanins) were detected in the samples. Vranec wines were produced under different fermentation conditions: maceration time of 3, 6 and 10 days, two doses of SO2 (30 and 70 mg l−1) and two yeasts for fermentation, in order to examine their effects on the extraction of phenolic compounds from grapes into the wine.  相似文献

Dichloromethane, ethyl acetate, acetone, methanol, and water extracts prepared from the fruits and leaves of Laurocerasus officinalis Roem. (LO) (Rosaceae) were screened for their cholinesterase inhibitory activity against acetylcholinesterase (AChE) and butyrylcholinesterase (BChE), the key enzymes in pathogenesis of Alzheimer's disease (AD), using ELISA microplate reader at 50, 100, and 200 ??g mL−1. As AD is associated with oxidative stress, the antioxidant activity of the extracts was also tested by radical-forming methods against 2,2-diphenyl-1-picrylhydrazyl (DPPH), N,N-dimethyl-p-phenylendiamine (DMPD), and superoxide radicals as well as iron-related methods; iron-chelating capacity and ferric-reducing antioxidant power (FRAP) assays. Total phenol and flavonoid quantification was achieved using Folin-Ciocalteau and AlCl3 reagents, respectively. The highest AChE (44.01 ± 1.75%) and BChE (19.91 ± 0.37%) inhibition was caused by the LO-leaf-methanol extract 200 ??g mL−1, while it showed the best radical-scavenging activity against DPPH at 2000 ??g mL−1. Only, the dichloromethane and water extracts of the fruits and the leaf water extract had an iron-chelating capacity, while the leaf methanol extract displayed the highest FRAP. The leaf methanol extract (113.45 ± 0.71 mg g−1 extract) was found to be the richest in total phenols, while the leaf acetone extract (139.90 ± 4.64 mg g−1 extract) had the most abundant amount of total flavonoids.  相似文献

Reverse osmosis process for the concentration of black currant juice was carried using AFC-99 tubular membrane at 30 °C and 45 bar. The contents of selected flavonols and anthocyanins were analyzed after centrifugation; enzyme treatment by Panzym Super E and by Rohapect berry followed by centrifugation; and ultrafiltration black currant juices and juice concentrates. The total soluble solid (TSS) content of the juices increased from the initial 17.6–17.9 °Brix to 24–24.8 °Brix in the case of the centrifuged juice in the concentration process. Similarly, it increased from 14.5–15.5 °Brix to 23.1–23.4 °Brix for the Panzym Super E treated juice, and from 16.1–16.9 °Brix to 22.5–23.1 °Brix for the Rohapect berry treated black currant juices. The ultrafiltered juice had the lowest initial TSS content between 14.1 and 14.9 °Brix and it increased to 22.1–23.1 °Brix. The average permeate fluxes during the concentration process were 7.3 L m−2 h−1 for the centrifuged juice, 11.9 L m−2 h−1 for the Panzym Super E treated juice, 9.2 and 13.1 L m−2 h−1 for the Rohapect berry treated and ultrafiltered juice, respectively. Analysis indicated that the enzymatic treatment resulted in the increase of anthocyanin and flavonol content of the juices. The centrifugation process decreased the amount of anthocyanins and flavonols to some extent. The juice clarified by ultrafiltration had significantly lower concentrations of anthocyanins and flavonols, while the juices treated by Panzym Super E had the highest levels of these flavonoids. This study recommends enzymatic pre-treatment by Panzym Super E, since it improves the permeate flux in reverse osmosis during the concentration process, and results in a juice concentrates highest in anthocyanins and flavonols.  相似文献

The principal antioxidant components and content of cinnamon (Cinnamomum cassia), turmeric (Curcuma longa) and golden thread (Coptidis rhizoma) extracts were determined using high performance liquid chromatography (HPLC) with UV detection. In general, C. cassia, C. longa and C. rhizoma extracts from domestic Taiwan were rich in cinnamaldehyde, curcumin, and berberin, respectively. The contents of cinnamaldehyde, curcumin, and berberin in the acetone extracts were 1911, 2029, and 840 mg l−1, respectively. The Folin–Ciocalteu method was used to measure the total phenolic concentrations of extracts, which had the content of 9.6 (C. cassia), 2.6 (C. longa), and 4.3 (C. rhizoma) mM l−1. In addition, DPPH radical-scavenging, ferric-reducing antioxidant power (FRAP), and ferric thiocyanate (FTC) assays were employed to measure antioxidant activities. The C. cassia fresh extracts had higher antioxidant activities which were 84–90% (DPPH), 17–33 μmol l−1 (FRAP), and 53–82% (FTC). The activities of C. longa fresh extracts were 22–44% (DPPH), 7–11 μmol l−1 (FRAP), and 53–81% (FTC) while C. rhizoma were 53–64% (DPPH), 18–26 μmol l−1 (FRAP), and 59–82% (FTC).  相似文献

Berries and red fruits are rich dietary sources of polyphenols with reported health benefits. More than 50 different flavonols (glycosides of quercetin, myricetin, kaempferol, isorhamnetin, syringetin and laricitrin) have been detected and quantified with HPLC–MSn in fruits of blueberry, bilberry, cranberry, lingonberry, eastern shadbush, Japanese wineberry, black mulberry, chokeberry, red, black and white currants, jostaberry, red and white gooseberry, hardy kiwifruit, goji berry, rowan, dog rose, Chinese and midland hawthorn, wild and cultivated species of blackberry, raspberry, strawberry and elderberry. The phenolic constituents and contents varied considerably among the analyzed berry species. Elderberry contained the highest amount of total flavonols (450–568 mg kg−1 FW), followed by berry species, containing more than 200 mg kg−1 FW of total: chokeberry (267 mg kg−1), eastern shadbush (261 mg kg−1), wild grown blackberry (260 mg kg−1), rowanberry (232 mg kg−1), american cranberry (213 mg kg−1) and blackcurrants (204 mg kg−1). Strawberry (10.5 mg kg−1) and white currants (4.5 mg kg−1) contained the lowest amount of total flavonols. Quercetins represent the highest percentage (46–100%) among flavonols in most analyzed berries. In wild strawberry and gooseberry the prevailing flavonols belong to the group of isorhamnetins (50–62%) and kaempferols, which represent the major part of flavonols in currants (49–66%). Myricetin glycosides could only be detected in chokeberry, rowanberry and species from the Grossulariaceae, and Adoxaceae family and Vaccinium genus. Wild strawberry and blackberry contained from 3- to 5-fold higher total flavonols than the cultivated one.  相似文献

Meadowsweet was extracted in water at a range of temperatures (60–100 °C), and the total phenols, tannins, quercetin, salicylic acid content and colour were analysed. The extraction of total phenols followed pseudo first-order kinetics, the rate constant (k) increased from 0.09 ± 0.02 min−1 to 0.44 ± 0.09 min−1, as the temperature increased from 60 to 100 °C. An increase in temperature from 60 to 100 °C increased the concentration of total phenols extracted from 39 ± 2 to 63 ± 3 mg g−1 gallic acid equivalents, although it did not significantly affect the proportion of tannin and non-tannin fractions. The extraction of quercetin and salicyclic acid from meadowsweet also followed pseudo first-order kinetics, the rate constant of both compounds increasing with an increase in temperature up until 90 °C. Therefore, the aqueous extraction of meadowsweet at temperatures at or above 90 °C for 15 min yields extracts high in phenols, which may be added to beverages.  相似文献

Biopolymers mixed systems have been widely studied in the past two decades, resulting in advances in knowledge of the key parameters involved in macromolecules–macromolecules interactions. Sound understanding and control of these parameters should enable food scientists to design coating or film with desired functional properties. In this study the phase behaviour of high methoxyl pectin and soy flour at pH 4.6 was investigated. The aim was to investigate the effect of different concentrations of pectin (P) and soy flour (SF) solution and different P/SF ratios on mixture phase behaviour. Pectin suspensions at 16 mg ml−1 and 8 mg ml−1 concentrations and soy flour suspensions at 13 mg ml−1 and 6.5 mg ml−1 concentrations were prepared and mixed together in different ratios (10/90, 25/75, 50/50, 75/25, 90/10). Turbidity and rheological measurements were performed to obtain information on pectin–soy flour interaction in electrostatic compatibility conditions. The experimental data show that soy flour solution is a non-stable system in which complexes may be formed during aging. It becomes insoluble and settles to the bottom of the suspension. By blending charged soy flour, at high concentration, with anionic pectin an associative interaction between biopolymers occurs which stabilizes the protein in solution. The best pectin and soy flour suspension for forming film with a continuous network was at concentrations of 16 mg ml−1 and 13 mg ml−1.  相似文献

Anthocyanins in the flowers of pomegranate (Punica granatum L.) were extracted and separated by reverse-phase high-performance liquid chromatography and Sephadex LH-20 chromatography. Two anthocyanins were identified to be pelargonidin 3,5-diglucoside and pelargonidin 3-glucoside by spectral methods, ultraviolet–visible spectroscopy, 1H nuclear magnetic resonance spectroscopy, mass spectroscopy and comparisons with the literature. These two anthocyanins were identified from pomegranate flowers for the first time. Furthermore, the antioxidant activities of purified anthocyanins were screened for their antioxidative potential using 2,2′-diphenyl-1-picryl hydrazyl (DPPH) and 3-ethyl-benzo-thiazoline-6-sulphonate (ABTS) systems. The purified anthocyanins showed strong radical scavenging activities. Pelargonidin 3-glucoside showed higher antioxidant activity (50% inhibition values of 19.56 μg/ml for DPPH and Trolox equivalents antioxidant activity of 2.39 μg/μg for ABTS) than pelargonidin 3,5-diglucoside.  相似文献

Antioxidant activity of the ethanolic extracts of the fruits of Fritillaria pontica Wahlenb. (Liliaceae), Euonymus latifolius (L.) Mill. ssp. latifolius (Celastraceae), and Vicia sativa L. ssp. nigra (L.) Ehrh. var. nigra L. (Fabaceae), the aerial parts of Turritis laxa (Sibth & Sm.) Hayek (Brassicaceae), Vicia cracca L. (Fabaceae), and Oxyria digyna (L.) Hill. (Polygonaceae) was screened by 2,2-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and ferric-reducing antioxidant power (FRAP) assays at 0.5, 1.0, and 2.0 mg ml−1 concentrations. Total phenolic contents of the extracts were determined using Folin-Ciocalteau reagent. T. laxa was also tested for its anti-acetylcholinesterase activity. The extracts were analyzed by LC–DAD–MS for their flavonoid content and the ethanolic extract of T. laxa has been found to contain rutin in appreciable amounts (7.63 ± 0.2%). Rutin and hyperoside were detected qualitatively in F. pontica, where vitexin was identified in O. digyna. It was also the most active in the antioxidant tests.  相似文献
