Changes of textural properties and cathepsin activity of grass carp fillets during storage in superchilling (?1.5 ± 0.2 °C) and ice (0.2 ± 0.1 °C) was investigated, and the function of cathepsin in quality deterioration of grass carp fillets was discussed. Results showed that shear force of superchilled and ice‐stored fillets decreased by 50% and 55% after 6 days of storage, respectively. The cathepsin activies in different fractions changed significantly during storage at both conditions. Cathepsin B, B+L activities in sarcoplasma, myofibrillar and heavy mitochondrial fraction significantly increased during the early 3 days postmortem, accompanied by remarkable reduction of corresponding activity in lysosome. Cathepsin D activity in sarcoplasma and myofibrillar significantly increased during 6 days of storage with corresponding decrease in lysosome and heavy mitochondria. Correlation analysis showed that changes of shear force of grass carp fillets were significantly correlated with integration of cathepsin B, D and B+L throughout storage time (r2 > 0.94). As derived from the first‐order exponential decay model, the enzyme efficiencies (ka) of cathepsin B and B+L were more than twofold higher than those of cathepsin D, suggesting the major role of cathepsin B and B+L in textural deterioration of grass carp fillets in chilling storage.  相似文献

Textural firmness is a primary determinant of consumer acceptance for evaluating freshness quality of fish fillet flesh. The objective of this study was to investigate the potential of using visible and near-infrared hyperspectral imaging (400–1000 nm) for non-destructive prediction of firmness quality of grass carp fillet as affected by frozen storage. Fillet samples were frozen at − 20 °C for 24 h and then stored at 4 °C for thawing over five days. Hyperspectral images were obtained at different thawing stages and their corresponding spectral data were extracted. Two calibration models were established between the extracted spectral data and the reference firmness values measured by the traditional mechanical method by using partial least squares regression (PLSR) and least-square support vector machine (LS-SVM) analysis. Three approaches of regression coefficients (RC) from PLSR analysis, genetic algorithm (GA) and successive projection algorithm (SPA) were utilized to recognize the most important wavelengths that possessed the greatest influence and sensitivity on the firmness prediction based upon the whole spectral range. By comparing the above-mentioned three variable selection methods, seven optimal wavelengths (450, 530, 550, 616, 720, 955 and 980 nm) were selected by GA and its corresponding simplified prediction model of GA-LS-SVM was also obtained, showing the best performance with the highest determination coefficient (R2P) of 0.941 and the lowest root mean square error estimated by prediction (RMSEP) of 1.229. The overall results of this study suggested that hyperspectral imaging technique has a potential for fast and non-destructive prediction and analysis of textural firmness of grass carp fillets as affected by frozen storage.  相似文献

BACKGROUND: Pre‐rigor fillets of Atlantic salmon were either superchilled or chilled prior to packaging in air or in modified atmosphere (MAP, 60% CO2/40% N2) with a CO2 emitter, in 5.3 L high‐density polyethylene trays with three to four layers of fillets (3.0–3.7 kg). All samples were stored at 0.1 °C for 28 days. RESULTS: Fillets stored in MAP had significantly lower bacterial growth compared to fillets stored in air, and MAP superchilled bottom fillets had lower bacterial counts compared to the corresponding chilled fillets. Samples superchilled prior to refrigerated storage in air had similar bacterial growth to ordinary chilled samples. Faster fillet softening during storage and higher liquid loss were observed in superchilled MAP samples. CONCLUSION: Combining short‐term superchilling and MAP with a CO2 emitter prolonged the shelf‐life of pre‐rigor salmon fillets, which can improve sustainability throughout the value chain. The inhibitory effect of allicin on proteolytic enzymes and textural deterioration of ice‐stored grass carp (Ctenopharyngodon idella) fillets was investigated. The results of in vitro study showed that allicin inhibited the activity of cathepsin B, L and D, calpain and collagenase in crude extract of grass carp muscle. Among endogenous enzymes, cathepsin B, L and collagenase were the most susceptible to allicin. Proteolysis of myofibrillar proteins by either crude enzyme or cathepsin B and L was almost prevented by allicin when employed at a concentration higher than 100 mm . After storage of 21 days, shear force of fillets treated with allicin at 10–100 mm was 39–51% higher than that of control. Myofibrillar proteins of fillets during storage were well protected against degradation when allicin concentration increased to 100 mm , as evidenced by SDS‐PAGE. Therefore, allicin could be a potential broad‐spectrum inhibitor to retard softening of fish fillets via mitigating myofibrillar proteolysis by endogenous enzymes especially cathepsin B and L during ice‐storage.  相似文献

The objective of this work was to analyse the role of desmin in texture softening and water‐holding capacity (WHC) of ice‐stored grass carp fillet. Results indicated that shear force of the fillet decreased sharply within 3 days, while drip loss increased during the whole ice storage. Water mobility and distribution were measured by low‐field 1H NMR T2 relaxation that revealed a positive correlation (P < 0.01) between WHC and the mobility of the immobilised water (T21 relaxation time). Meanwhile, negative correlation could be established between intact desmin and drip loss (< 0.01). Intact desmin was extremely correlated with shear force (< 0.01) and positively correlated with calpains activity (< 0.05). This study demonstrated that the degradation of desmin was involved in the mobility of the myofibrillar water and softening of grass carp fillet during ice storage. Furthermore, calpains autolysis seemed to result in desmin degradation.  相似文献

Effects of chitosan coating combined with essential oils from clove, cinnamon and lemon grass on quality and antioxidant enzyme activities of grass carp fillets stored at 4 ± 0.5 °C were evaluated. The quality parameters (including pH, total volatile basic nitrogen (TVB‐N), K value, thiobarbituric acid (TBA) value, shear force and total viable count (TVC)) and antioxidant enzyme activities were analysed periodically. The results indicated that composite chitosan coatings presented better preservation effects than chitosan coating alone. In addition, chitosan–clove essential oil coating had the best quality enhancement effects among treatments by inhibiting deterioration of physicochemical quality and microbial growth and maintaining antioxidant enzyme activities of fillets during refrigerated storage. The negative correlation was observed between the changes in TBA values and antioxidant enzyme activities in fillets. Based on these findings, chitosan–essential oil coatings effectively mitigated oxidative stress and extended shelf life of refrigerated grass carp fillets.  相似文献

The effect of chilled, frozen and freeze‐chilled storage on quality of grass carp fillets and soups was evaluated by sensory score, total aerobic counts and biochemical quality. Fish fillets were stored at 4 °C for 6 days (T1), ?40 °C for 12 h and then at ?20 °C for 5 days (T2), ?40 °C for 12 h and then at ?20 °C for 5 days, followed by at 4 °C for 4 days (T3). T1 showed higher sensory score, salt‐soluble protein content, better colour and texture qualities than T2 and T3 within 3 days. All fillets kept good quality based on the acceptable limit of sensory score, total volatile basic nitrogen and total aerobic count during storage time. According to the transportation and retail time, chilled storage is appropriate when it is within 3 days. If it extends for 5 days, freeze‐chilling treatment keeps better quality, but later chilled fillets should be retailed within 4 days.  相似文献

The aim of the present study was to investigate the underlying mechanisms of softening texture in chilled grass carp filletsunderpinning collagen solubility, gelatinolytic proteinase activity and physicochemical parameters. Acid-soluble collagen (ASC) and heat soluble collagen (HSC) increased markedly, while a significant decline was detected in total collagen and insoluble collagen (ISC) during the first 3 days of storage, coinciding with a loss of shear force and water-holding capacity (WHC) (< 0.05). Moreover, the activity of gelatinolytic proteinases was gradually activated and reached the peak at day 3 (P < 0.05). Pearson coefficient analysis showed that gelatinolytic proteinase activity revealed a significant correlation with collagen solubility. Total collagen, ASC, ISC and HSC were significantly correlated with shear force and WHC. Our study clarified that the increase of collagen solubility by gelatinolytic proteinases played an important role for texture softening in the early stage of chilled grass carp fillets.  相似文献

Quality changes in Atlantic mackerel fillets during superchilled, chilled and frozen storage focusing on lipid quality, colour and changes in low molecular weight metabolites relevant for quality and safety were studied. Low formation of oxidation products was observed in chilled (up to 7 days), superchilled (up to 14 days) and frozen storage (up to one year). Only slight formation of TBARS was measured in the samples which correlated with the increasing fillet yellowness. The profile of low molecular weight compounds, analysed by high-resolution NMR, showed clear grouping of the different samples according to both treatment and storage time. The increase in K-value was highest in the chilled samples, reaching a K-value of 93 ± 3% on day 5, exceeding the proposed limit of human consumption (80%) while superchilling reached a K-value of ca 90 % after 14 days. Frozen samples showed acceptable quality in the whole storage period.  相似文献

The effects of freezing and freezing-thawing cycles during cold storage have been studied in meagre (Argyrosomus regius) fillets. Fillets were subjected to three conservation protocols: fresh, freezing at –20°C, and repeated freezing-thawing cycles. Fresh fillets were stored (4°C, 15 days), and the same protocol was followed for freezing and freezing-thawing after the freezing period. Freezing and freezing-thawing fillets were softer and presented lower water holding capacity than fresh, mostly attributable to collagen solubilization, and partial myofibrillar protein degradation. Cold storage (4°C) during 15 days caused softening in fresh and frozen fillets attributable to myofibrillar protein hydrolysis. Freezing-thawing cycles increased proteolysis, this leading to unacceptable softening even from early stages of further cold storage, and this was also revealed by SDS-PAGE.  相似文献

This study investigated the effect of chitosan coating (1 and 2%) on the quality of grass carp fillets stored at 4°C for 20 days. During storage, the physicochemical properties (pH, thiobarbituric acid value, total volatile basic nitrogen value, trimethylamine nitrogen value, K-value, water loss, and instrumental texture), microbiology (total viable count and psychrotrophic count), and sensory properties were evaluated. The results showed that chitosan coating could effectively inhibit bacterial growth, improve physicochemical and sensory qualities, and it reduced the deterioration of the quality of grass carp fillets. Compared with fillets without chitosan coating, the shelf life of fillets with 1 and 2% chitosan coating was extended by ~3 and 6–7 days, respectively.  相似文献

对鲜切草鱼片在4℃冷藏过程中的鲜度变化进行研究,并采用选择性培养基对鱼片中的微生物生长规律进行分析.实验表明4℃冷藏的鲜切草鱼片在冷藏3d内处于一级鲜度范围,冷藏8d后鱼肉严重腐败,其在4℃冷藏的贮藏期限为5~6d;在冷藏期间各种腐败微生物数量均呈现明显的上升趋势,初步确定腐败希瓦氏菌、假单胞菌和乳酸菌为鲜切鱼片中生长的优势菌群.  相似文献

采用4、10(无光照)、10(5 000 lx)、25℃条件,同时贮藏草鱼鱼片(除去红肉)和三文鱼鱼片,研究光照和温度对草鱼和三文鱼贮藏品质的影响。结果表明,草鱼和三文鱼贮藏过程中pH值、挥发性盐基氮(total volatile basic nitrogen,TVB-N)值、硫代巴比妥酸(thiobarbituric acid,TBA)值和色差变化差异显著(P0.05),且菌落总数(total viable count,TVC)、pH值、TVB-N值、TBA值、色差、ATP分解产物(adenosine triphosphate-related decomposition products)K值均随着贮藏时间的延长而增加。光照(5 000 lx)对于草鱼、三文鱼贮藏过程中的pH值变化、TBA值变化影响显著(P0.05),能使鱼肉中pH值增加量高于避光保存,促进光氧化,提高TBA值增加速率,而对于TVC、TVB-N值、K值、色差变化影响不显著(P0.05)。贮藏温度的差异(4、10℃)对两种鱼肉品质变化都有显著影响(P0.05)。  相似文献

Quality changes of aerobically packed cod fillets stored under superchilling and abusive temperature conditions were characterized by the growth of specific spoilage organisms (SSO) and the production of microbial metabolites measured by an electronic nose along with traditional sensory and chemical analysis (total volatile basic nitrogen [TVB‐N], pH). A new process based on quick contact freezing and cold air blasting was used to achieve superchilling of fillets before chilled (0.5 °C) or superchilled (‐1.5 °C) storage. Photobacterium phosphoreum dominated under temperature abusive conditions coinciding with high levels of TVB‐N and increased electronic nose responses indicating increased levels of alcohols and aldehydes at sensory rejection. Dominating growth of Pseudomonas spp. in 1 batch was associated with the origin, the catching method, and the cooling conditions during processing. The superchilling process followed by superchilled storage (‐1.5 °C) extended the sensory shelf life of the fillets for at least 3 d compared with traditional process, resulting in a total shelf life of 15 d. High content of TVB‐N was observed in superchilled fillets at sensory rejection. P. phosphoreum counts were lower under superchilling conditions (6.0 to 6.8 log colony‐forming units [CFU]/g), compared with the traditionally processed chilled fillets (7.2 log CFU/g). However, H2S‐producing bacteria appeared to grow steadily under superchilling conditions reaching counts as high as 7.6 log CFU/g at sensory rejection.  相似文献

The objective of the current study was to analyze and develop some important automation steps in a superchilling process line. In order to control the product quality there is a need for online measurements of ice fraction and distribution in inhomogeneous products. Moreover, automatic handling of such superchilled products is currently commercially unavailable. The current study presents a new method for monitoring and handling superchilled product of varying form and consistency. Observation of the shift in the water absorption peak, measured by near-infrared spectroscopy (NIR) transflection mode, was used to determine the ice level in superchilled salmon, scanning approximately 1.5 cm into the fillets. The salmon fillets were stored on ice at 0 °C for 5–7 days before superchilling at −24 °C to target ice contents of 10%, 15% and 30%. Online NIR measurements of ice fraction showed promising results, with a low prediction error of 2.5%. The storage study confirmed former quality results with a microbiological shelf life of 15–17 days with only minor differences in values for drip loss and water-holding capacity between superchilled and chilled samples.  相似文献

To understand the quality changes in Songpu mirror carp (Cyprinus carpio) fillets during chilled storage, sensory score, total aerobic counts (TAC), total volatile basic nitrogen (TVB‐N) and electrical conductivity (EC) of the fillets stored at 270, 273, 276, 282 and 288 K were investigated. Furthermore, both Arrhenius model and log‐logistic model were established to predict the quality changes. The high regression coefficients (R2 > 0.90) represented the acceptability of both Arrhenius model and log‐logistic model. Relative errors between predicted values and observed values were all within ±5% for models based on EC, within ± 25% for models based on TAC and TVB‐N (except the TVB‐N value on 9th and 15th day). The relative errors of sensory score were all within ±35% in the first 9 days. It is concluded that both Arrhenius model and log‐logistic model based on EC, TAC or TVB‐N can be applied in modelling quality changes in Songpu mirror carp fillets during storage within 270–288 K.  相似文献

This study aimed to determine the phenolic content and antioxidant effect of grape pomace extract on silver carp fillets during refrigerated storage. Total phenolic content of grape pomace extract was quantified by colorimetric methods. Silver carp samples were treated with 0, 2, and 4% of grape pomace extract (g extract/100 g flesh) and stored 15 days in a refrigerator (4°C). Changes in pH, peroxide value, thiobarbituric acid, and heme iron at 0, 3, 6, 9, 12, and 15 days of storage were investigated. Results showed that the values of pH, peroxide value, and thiobarbituric acid increased and iron levels decreased in all treatments during storage (p < 0.05). The phenolic contents were generally much more influenced by the quality parameters treated compared to the control. The addition of grape pomace extract delayed lipid oxidation in silver carp fillet considerably during refrigerated storage. These results suggested that grape pomace extract has the potential to be used as a natural antioxidant.  相似文献

Cinnamon essential oil was used in gelatin coatings to maintain the quality of refrigerated rainbow trout fillets over a period of 20 days. Fish fillets were coated with a solution of 4% gelatin incorporated with cinnamon essential oil and then stored in refrigerator (4 ± 1 °C). Coating’s preservative effect was assessed periodically (every 5 days) by microbial analyses (aerobic plate count and psychrotrophic count), chemical determinations (total volatile basic nitrogen, thiobarbituric acid, free fatty acid) and sensory characteristics. When gelatin coating with cinnamon applied to the preservation of trout fillets, a reduction in the growth of total bacteria was observe after 15 days of storage. Fish fillets with gelatin coating containing 1%, 1.5% and 2% v/v cinnamon oil produced significantly lower (P < 0.05) total volatile basic nitrogen then gelatin‐coated fillets and control until 15 days of storage period (35, 22.86, 30.33, 57.76 and 53.26 mg per 100 g of fillet, respectively). The obtained results indicate that gelatin in the form of coating enriched with cinnamon oil is suitable for the preservation of rainbow trout fresh fillets and to efficiently maintain the quality attributes to an acceptable level during storage.  相似文献

The computer vision and a stereoscopy technique were used to characterize and detect changes in the capacity of the salmon fillet surface to recover its original form after a constant weight was applied. A curvature index (associated to fillet firmness) calculated by means of 3D information data obtained with the stereoscopy technique from the fat stripe on the fillets was estimated over 6 months and was suggested as a characteristic of the recovery property of the fresh salmon fillet surface. Simultaneously, instrumental firmness using a LFRA1500 texture was measured. The values for the fresh salmon fillet measurement curvature index during a period of 6 months were established in the range of 0.05 and 0.10 (dimensionless), and this value was used in the experiments to represent fresh salmon fillets. Changes in the curvature index (associated with similar changes in firmness) for salmon fillet surface treated with enzymes (pepsin and papain) were registered on the second day during storage at 2 °C. In general, stereoscopy is a technique that can be used on salmon fillets to detect changes in the recovery properties of the salmon fillet surface and to determine when salmon fillets can be classified as a fillet that has not been processed with enzymes.  相似文献
