银鲳亲鱼不同组织的氨基酸及其随性腺发育的变化   总被引:3,自引:1,他引:2  
测定了海捕银鲳亲鱼肌肉,肝脏和性腺组织的氨基酸,并分析了不同性腺发育阶段的氨基酸变化。结果表明,雄性V期亲鱼精巢中的牛磺酸含量最高(27.8mg/g),显著高于肌肉和肝脏组织,而雄性V期亲鱼氨基酸总量在肌肉中最高(631.4 mg/g),其次是精巢(544.8 mg/g)和肝脏(468.0 mg/g)组织。雌性亲鱼肝脏中的牛磺酸含量(13.7 mg/g)显著高于卵巢和肌肉组织。在卵巢从III期发育到V期过程中,雌性亲鱼肌肉和肝脏中的氨基酸总量在V期下降显著,而卵巢氨基酸总量维持稳定。在总氨基酸的组成上,肌肉中含量高的氨基酸为Glu,Lys, Asp,Leu和Arg;肝脏中含量高的为Glu,Lys, Asp,Val和Leu;精巢中含量高为Glu,Asp, Lys, Arg和Val;卵巢含量高中为Glu,Lys,Leu,Val和Asp。性别和性腺发育阶段对亲鱼肌肉总氨基酸组成无影响。游离氨基酸含量在III 、IV和 V期卵巢中的含量分别为17.7 mg/g ,41.5 mg/g和29.6 mg/g,且其组成随性腺发育阶段的不同而有显著差异。  相似文献

研究了EPA和DHA水平对牙鲆生长的影响,饲料中含0.5%EPA和1.0%~1.5%DHA能保证牙鲆幼鱼最适生长,鱼体水分最低,肝体指数最小,脂肪含量有较大幅度提高,肝脏极性脂中EPA和DHA达到最大积累;在肝脏和肌肉的非极性脂部分,各组间的脂肪酸组成没有显著变化,而极性脂部分能体现出饲料中n-3 HUFA含量对鱼体脂肪酸组成的影响,极性脂中的EPA和DHA含量远高于非极性脂;在肌肉和肝脏的极性脂和非极性脂中都含有较高的16:0和18:1n-9; 18:1n-9/n-3HUFA可以作为必需脂肪酸满足程度的一个判据,18:1n-9值的升高往往是缺乏必需脂肪酸的表现,在生长最佳时18:1n-9/n-3HUFA比值下降,为0.62和0.74.  相似文献

为研究脂肪酸对牙鲆繁育性能的影响, 采用生物化学方法, 对野生和养殖牙鲆亲鱼肌肉、肝脏及卵中脂肪酸组成分别进行测定。结果表明: (1) 牙鲆肌肉、肝脏和卵中脂肪含量大小关系为肝脏>卵>肌肉。野生亲鱼肌肉和卵中脂肪含量显著低于养殖亲鱼(P<0.05), 肝脏脂肪含量与养殖亲鱼无显著差异; (2) 牙鲆亲鱼3种组织中均检测出21种脂肪酸。野生亲鱼肌肉中饱和脂肪酸(SFA)与养殖亲鱼无显著差异, 单不饱和脂肪酸(MUFA)显著低于养殖亲鱼(P<0.05)。肝脏和卵中SFA显著高于养殖亲鱼(P<0.05), MUFA与养殖亲鱼无显著差异; (3) 野生亲鱼肌肉、肝脏和卵中高不饱和脂肪酸(PUFA)的含量, 尤其是肝脏和卵中C20:5n-3(EPA)、C22:6n-3(DHA)的含量均显著低于养殖亲鱼(P<0.05), 但肌肉和卵中的C20:4n-6(ARA)含量明显高于养殖亲鱼(P<0.05); (4) 野生牙鲆亲鱼肌肉、肝脏和卵中n-3/n-6 PUFA及EPA/ARA显著低于养殖亲鱼(P<0.05), 肝脏中DHA/EPA显著高于养殖亲鱼(P<0.05), 但野生亲鱼肌肉和卵中的DHA/EPA与养殖亲鱼无显著差异。比较结果说明, DHA、EPA和ARA等PUFA是与牙鲆繁殖性能密切相关的重要脂肪酸。在牙鲆亲鱼养殖过程中, 除了提供牙鲆亲鱼足够的脂肪酸营养外, 也应注意各种脂肪酸, 尤其是PUFA中各种脂肪酸之间的添加比例, 从而保证亲鱼的繁殖性能及卵和仔鱼的质量。  相似文献

以野生日本鬼鲉人工催产获得的受精卵为实验材料,定量检测了日本鬼鲉胚胎及卵黄囊仔鱼发育过程中脂肪组成及脂肪酸含量.结果表明:日本鬼鲉胚胎及卵黄囊仔鱼的总脂肪含量为13.85%~ 11.66%,极性脂肪占总脂肪含量为75.39%~ 72.20%.总脂肪及极性脂肪含量在胚胎发育阶段无显著变化,在卵黄囊仔鱼阶段随发育而显著下降.中性脂肪含量在胚胎发育阶段有显著变化,在卵黄囊仔鱼阶段其含量相对稳定.野生日本鬼鲉胚胎及卵黄囊仔鱼总脂肪的主要脂肪酸为DHA(22∶6n-3),16∶0,ARA(20∶4n-6),EPA(20∶5n-3),18∶0和18∶1n-9.总脂肪及极性脂肪的DHA,ARA,EPA含量(mg/gDW)均随胚胎和卵黄囊仔鱼的发育而显著下降,且DHA和ARA含量均在胚胎囊胚期至尾芽期大幅降低.中性脂EPA和DHA含量随发育呈先升后降,其峰值分别出现在初孵仔鱼和2日龄(2DPH)卵黄囊仔鱼.中性脂ARA含量随发育逐步升高,峰值出现在3日龄(3DPH)卵黄囊仔鱼.在胚胎发育前期,总脂肪DHA和ARA相对EPA被选择性消耗,饱和脂肪酸(SFAs)和多不饱和脂肪酸(PUFAs)相对单不饱和脂肪酸(MUFAs)被机体选择性消耗,N-6PUFA相对N-3 PUFA被选择性消耗;在胚胎发育后期及卵黄囊仔鱼阶段,总脂肪EPA相对DHA和ARA被选择性消耗.在胚胎和卵黄囊仔鱼发育过程中,SFAs中16∶0相对18∶0被选择性消耗.胚胎发育后期阶段和卵黄囊仔鱼阶段,总脂肪MUFAs中16∶1相对18∶1被选择性消耗.实验表明日本鬼鲉胚胎及卵黄囊仔鱼发育阶段极性脂肪中DHA、ARA和EPA可以向中性脂肪中转移,胚胎和卵黄囊仔鱼对不同类别脂肪中的重要脂肪酸的消耗具有选择性,且其选择性与发育阶段相关.  相似文献

为获知棘头梅童鱼亲鱼性腺发育后期脂类及脂肪酸蓄积特点,采用内标定量法,对性腺发育至Ⅳ期的雌性棘头梅童鱼亲鱼肌肉、肝脏和卵巢组织中的脂肪及脂肪酸含量进行了测定,并比较分析了3种组织中的脂肪酸组成。结果表明:雌性棘头梅童鱼亲鱼卵巢、肝脏和肌肉中脂肪的质量分数分别为(4.84±0.05)%、(2.66±0.05)%和(0.56±0.05)%,不同组织间具有显著性差异(P<0.05)。雌性棘头梅童鱼亲鱼的卵巢、肌肉和肝脏中分别检出29种脂肪酸,含量最高的饱和脂肪酸依次为C16∶0、C18∶0和C14∶0;含量最高的单不饱和脂肪酸依次为C16∶1、C18∶1n9c和C20∶1;含量最高的多不饱和脂肪酸依次为C22∶6n3(DHA)、C20∶5n3(EPA)和C20∶4n6(ARA)。从相对含量来看,肌肉中饱和脂肪酸的质量分数为(58.22±0.24)%,显著高于肝脏(52.71±0.24)%和卵巢(36.42±0.24)%(P<0.05);卵巢中单不饱和脂肪酸的质量分数为(23.78±0.09)%,显著高于肌肉(16.15±0.07)%和肝脏(10.79±0.05)%(P<0.05);卵巢中多不饱和脂肪酸质量分数为(39.79±0.19)%,显著高于肝脏(36.49±0.21) %和肌肉(25.63±0.19)%(P<0.05)。肌肉、肝脏和卵巢中EPA和DHA的总量依次升高,质量分数分别为(19.34±0.15)%、(26.59±0.16)%和(27.78±0.13)%。卵巢、肝脏和肌肉组织中的DHA/EPA分别为1.95、2.28和1.89,n-3系和n-6系多不饱和脂肪酸的比值分别为3.29、3.81和6.25。  相似文献

为了解不同季节不同鱼类不同组织中的不同脂肪酸含量,科学地指导鱼类膳食消费,本实验研究了冬夏两季,采集自上海市场常见的5种经济鱼类:大黄鱼(海洋肉食性),银鲳(海洋杂食性),日本鳗鲡(淡水肉食性),莫桑比克罗非鱼(淡水杂食性),草鱼(淡水草食性),分别检测鱼背部肌肉、腹部肌肉、尾部肌肉、肝脏和腹腔脂肪组织的脂肪含量和脂肪酸绝对含量。结果显示,5种鱼肌肉脂肪酸谱存在显著差异,并与各自的生活环境及食性均有关系;在鱼的腹腔脂肪或肝脏中,饱和脂肪酸(SFAs)和单不饱和脂肪酸(MUFAs)含量较高,且与组织脂肪含量密切相关;而多不饱和脂肪酸(PUFAs)、n-3系多不饱和脂肪酸(n-3 PUFAs)和n-6系多不饱和脂肪酸(n-6 PUFAs)含量较低,且与组织脂肪含量关系不大;大黄鱼和银鲳各肌肉组织中的n-3 PUFAs、二十碳五烯酸(EPA)和二十二碳六烯酸(DHA)含量以及n-3/n-6值高于莫桑比克罗非鱼和草鱼,并与组织脂肪含量呈正相关;冬季草鱼腹部肌肉、莫桑比克罗非鱼尾部肌肉以及日本鳗鲡和银鲳的肝脏中的n-3 PUFAs含量较夏季高。研究表明,脂肪酸组成与物种、食性、水域环境以及季节温度和组织部位均有关系。从补充n-3 PUFAs摄入的角度分析,日本鳗鲡、大黄鱼和银鲳营养价值高于莫桑比克罗非鱼和草鱼,冬季鱼类的营养价值高于夏季。  相似文献

为掌握养殖银鲳的性腺发育规律及其与性类固醇激素水平的关系,本研究采用组织切片技术和酶联免疫吸附法(Elisa),观察养殖条件下银鲳的精巢与卵巢发育特征和血清中性类固醇激素的周年变化,并分析性腺成熟指数(GSI)、肝重指数(HSI)和肥满度(CF)与性腺发育的关系。结果显示,养殖银鲳雌鱼1龄即可发育成熟,成熟卵巢呈一对""形的囊状结构。11月份有近一半个体发育至III期,33.3%已发育至IV期,12月—翌年1月越冬期间约2/3退化至II期,3月又迅速发育至IV期,4月份达到V期并产卵,5月份产卵结束卵巢进入恢复期IV期。银鲳精巢属小叶型,III期精巢出现小叶腔,IV期精巢出现精子,精巢发育速率快于卵巢,即"雄性早熟",7月龄约20%个体可达到性成熟,越冬时退化至III期,3月重新发育,精子分批成熟排精。雌鱼GSI值为0.19%~12.89%,HSI为0.97%~2.30%,CF为2.30~3.08 g/cm~3,雄鱼的GSI为0.08%~2.62%,HSI值为0.73%~1.83%,CF为2.11~2.80 g/cm~3,GSI值在V期时达到最大,HSI和CF值则于IV期达到峰值。雌鱼血清中雌二醇(E2)周年表达水平为23.27~59.13 pg/m L,雄鱼为15.90~36.20 pg/m L,雌鱼的睾酮(T)为14.57~68.67 nmol/L,雄鱼的T为18.62~66.49 nmol/L,E2水平与HSI值呈现显著正相关关系,T表达水平与GSI值呈现显著正相关关系。研究表明,银鲳血清中性类固醇激素含量与精巢和卵巢的发育密切相关,可作为了解养殖银鲳性腺发育的重要指标。  相似文献

为了探明野生和人工养殖褐牙鲆亲鱼的繁育性能和卵巢发育之间的关系,采用生物化学方法,将处于Ⅲ~Ⅳ期和Ⅴ期的野生和养殖褐牙鲆亲鱼中肌肉、肝脏及卵巢氨基酸含量分别进行测定,结果表明,(1)褐牙鲆肌肉、肝脏、卵巢3种组织器官中总氨基酸含量大小关系为肌肉>卵巢>肝脏;(2) 发育同期的野生和养殖亲鱼肌肉中大部分氨基酸含量差异不显著(P>0.05),并且随着性腺的成熟,野生和养殖亲鱼肌肉中大部分氨基酸较为稳定,其中含量变化显著的必需氨基酸为蛋氨酸、组氨酸(P<0.05);(3) 发育同期的养殖亲鱼肝脏中大部分氨基酸含量高于野生亲鱼,随着性腺发育,野生和养殖亲鱼肝脏中氨基酸含量呈明显上升趋势;(4) 在Ⅴ期卵巢氨基酸组成中,除蛋氨酸、组氨酸含量存在显著差异外,野生和养殖亲鱼卵巢中氨基酸总量及必需氨基酸总量均不存在显著差异(P>0.05),同时,随着性腺发育,野生和养殖亲鱼卵巢中氨基酸含量呈明显上升趋势,差异显著的必需氨基酸有精氨酸、蛋氨酸、组氨酸(P<0.05)。  相似文献

本研究测定和评估了青岛地区5种底栖型海水鱼[(黄鮟鱇(Lophius litulon)、鲬(Platycephalus indicus)、长绵鳚(Zoarces viviparu)、角木叶鲽(Pleuronichthys cornutus)和带纹条鳎(Zebrias zebrinus)]的形体指标、肌肉和肝脏的粗成分,以及肌肉、肝脏、脑、眼、皮肤、肠道等组织的脂肪酸组成,以全面评估底栖型代表鱼类的脂肪和脂肪酸组成特征。实验鱼每种鱼9尾,3尾一组混样作为一个重复。结果显示,在5种鱼中,肌肉脂肪含量整体偏低,黄鮟鱇和带纹条鳎肌肉脂肪含量尤其低(0.3%~0.4%),但黄鮟鱇、鲬和角木叶鲽具有较高的肝脏脂肪含量(19%~29%),而带纹条鳎和长绵鳚肝脏脂肪含量偏低(4%~5%)。脂肪酸组成方面,黄鮟鱇肌肉中n-3系列长链多不饱和脂肪酸(long chain-polyunsaturated fatty acid, LC-PUFA)含量显著高于其他鱼,尤其以DHA最为明显。黄鮟鱇背肌中还具有最高含量的18:1n-9。长绵鳚背肌脂肪酸组成最明显的特征是较低的16:0含量和较高的20:4n-6及EPA含量。肝脏脂肪酸中,鲬和角木叶鲽具有显著高的18:1n-9等单不饱和脂肪酸含量,但其DHA含量较低。在黄鮟鱇、带纹条鳎和长绵鳚的大多组织中均有较高的DHA、20:4n-6和22:5n-3,但EPA含量较低。研究表明,即使同为底栖型鱼类,不同种类间脂肪和脂肪酸组成差异也非常显著。黄鮟鱇具有显著高的n-3 LC-PUFA含量,脂肪酸营养价值较高。  相似文献

为研究中华鲟对脂肪酸的选择利用,采用国标方法,对野生与人工养殖中华鲟成熟鱼卵和肌肉的脂肪酸组成进行了测定分析。试验结果显示:野生中华鲟卵细胞中C18:1和C22:6(DHA)脂肪酸的含量高于养殖的中华鲟成熟卵细胞;在相同的养殖环境和投饲策略下,野生中华鲟幼鱼合成或富集n-3多不饱和脂肪酸的能力高于养殖的中华鲟幼鱼。研究表明,中华鲟亲鱼在发育过程中对营养素的累积具有差异性,多不饱和脂肪酸,尤其是DHA、EPA和ARA脂肪源,对中华鲟亲鱼性腺发育和幼鱼生长发育具有重要意义。  相似文献

Total lipids, lipid classes, fatty acids and vitamin E contents were measured in ovary, liver, dark muscle, white muscle and mesenteric perigonadal fat in four maturational stages of Atlantic female northern buefin tuna (Thunnus thynnus thynnus L.) during migration to Mediterranean spawning grounds through the Strait of Gibraltar. Ovary lipid content increased significantly by 37.6-fold from inmature to spawning stage, accumulating large quantities of steryl/wax esters, whereas the ratio of mesenteric perigonadal fat (primarily triacylglycerols) weight to ovary weight decreased linearly throughout maturation. In contrast, liver, dark red and white muscles lipid content did not show a major depletion during maturation compared to that of perigonadal fat. The fatty acid composition of ovary from bluefin tuna showed that docosahexaenoic acid (DHA; 22:6n-3) was the most abundant fatty acid and DHA: EPA: AA and DHA: EPA ratios were 19.3/4.3/1 and 4.5/1, respectively. Neutral lipids predominated over polar lipids (2-3:1) in ovary and triacylglycerol and steryl/wax ester were major lipid classes. In general, fatty acid composition of most tissues examined were similar to each other but, in contrast, the fatty acid profile of perigonadal fat was rich in monoenes such as 18:1n-9, 20:1n-9 and 22:1n-11, and DHA decreased significantly at spawning stage. The total lipid balance suggested for the disappearance of lipid from perigonadal fat depots and, to a lesser degree, from muscle and liver depots, reflected lipid utilization for gonadal development on the one hand and non-gonadal metabolism in the other. The former included lipid transferred to the gonad and lipid catabolized to provide metabolic energy for the biosynthesis of gonadal constituents and the later was likely to be lipid catabolism to provide energy for swimming during spawning migration. 采用生化方法测定了瓯江彩鲤(Cyprinus carpio var.color)卵巢不同发育时期的成熟系数及卵巢脂类、脂肪酸含量与组成,旨在了解瓯江彩鲤卵巢中脂类、脂肪酸的变化与卵巢发育及卵细胞成熟之间的关系。结果显示:卵巢中总脂含量在产卵前最高,占干重17.62%,产卵后显著下降到12.84%(P<0.05),到快速生长期又出现稍微下降,之后在越冬期开始呈上升趋势。4个时期中,C16∶0(软脂酸)、C18∶1n-9c(油酸)、C18∶2n-6c(亚油酸)均是卵巢中的主导脂肪酸,其含量显著高于其它脂肪酸(P<0.05);产卵前,卵巢中脂肪酸含量高,但种类较少,产卵后,卵巢中脂肪酸含量低,但种类较多;随卵巢的发育,不饱和脂肪酸的比例增加,从快速生长期到产卵前,C20∶4n-6(ARA)与C22∶6n-3(DHA)的含量逐渐上升,最高分别达11.63 mg/g、17.80 mg/g。结果表明:不同时期卵巢中脂类含量与成熟系数呈正相关,卵巢成熟需积累较多脂类物质。不饱和脂肪酸尤其是DHA、ARA对卵巢的成熟具有重要作用。  相似文献

The fatty acid accumulation in broodstock influences the fecundity of spawners as well as egg quality. A comprehensive understanding of the lipid and fatty acid changes in broodstock during the reproductive period can increase our knowledge of the nutritional requirements in artificial breeding programs. The study assessed the lipid and fatty acid compositions of muscle, liver and ovary of American shad at different stages of reproductive development under reared conditions. The results indicated that there were significant differences in lipid content among ovary, liver and muscle. Mean total lipid levels in ovary, liver and muscle were 15.92%–10.87%, 13.62%–6.89%, 10.39%–6.11% respectively. During the development season, the total lipid content in muscle in stages V and VI were significantly lower than those in stages II and III. The total lipid content in liver in stages IV, V and VI was significantly lower than in stages II and III, whereas the total lipid content in the ovary increased significantly as the gonad developed and reached its highest value at stage V. From II to VI stage, the liver content of docosahexaenoic acid (DHA) decreased and the ovarian content of DHA increased, whereas the content of eicosapentaenoic acid (EPA) remained relatively constant. The content of C16:0 and C18:1n‐9 decreased in muscle and liver, but significantly increased in ovary. It is therefore suggested that the lipids were preferentially transferred from muscle and liver to ovary beginning in stage IV as the gonad matured. At stage IV, the lipid transfer accelerated, and saturated and unsaturated fatty acids, such as C16:0, C18:1n‐9, DHA, EPA and arachidonic acid, were selectively transferred to and conserved in ovaries.  相似文献

European sea bass juveniles (14.4±0.1 g mean weight) were fed diets containing different levels of fish oil then of n-3 highly unsaturated fatty acids (n-3 HUFA) for 12 weeks. The fish performance as well as fatty acid (FA) composition of neutral and polar lipids from whole body after 7 and 12 weeks feeding were studied. The requirements of juvenile sea bass for n-3 highly unsaturated fatty acids (n-3 HUFA) were studied by feeding fish diets containing six different levels of n-3 HUFA ranging from 0.2% to 1.9% of the diet, with approximately the same DHA/EPA ratio (1.5:1).

The growth rate at the end of the trial showed significant differences. Fish fed low dietary n-3 HUFA (0.2% DM of the diet) showed significantly lower growth than the diet 3 (0.7%), then no further improvement (P>0.05) of growth performance was seen by elevating the n-3 HUFA level in the diet up to 1.9% (diet 6). No difference in feed efficiency, protein efficiency ratio or protein retention was observed among treatments, nor in protein and total lipid content. However, the n-3 HUFA levels in diets highly influenced fish fatty acid composition in neutral lipid, while polar lipid composition was less affected. Comparison of polar lipid content after 7 or 12 weeks indicated that DHA remained stable at the requirement level, while arachidonic acid decreased with time. Results of this experiment suggest that the requirement for growth of n-3 HUFA of juvenile sea bass of 14 g weight is at least 0.7% of the dry diet.  相似文献

Two hundred and twenty rainbow trouts (IBW: 700 g) were randomly allotted to four tanks, with a male/female ratio of 0.56. Fish were fed for 168 d with four experimental diets containing herring oil, cod liver oil and coconut oil with the following inclusion rates: diet A: 12-1-0% respectively; diet B: 6-1-6%; diet C: 0-1-12%; diet D: 0-0-13%. Irrespective of the dietary treatment, weight gains of broodstocks were high (> 3 g/d) and FCR below 2. No significant difference was observed concerning the total amount of eggs spawn, egg average weight (82.5 mg/egg) and lipid content (5.4 mg/egg). However, the fatty acid profile of eggs was significantly affected by the dietary treatments. The content of unsaturated fatty acids, particularly the n-3 fatty acid series (EPA and DHA) significantly decreased with increasing levels of coconut oil in the diet.  相似文献

刀鲚(Coilia nasus)是一种江海生殖洄游性鱼类,长江口是刀鲚重要的洄游通道。本研究首次关注洄游至长江口卵巢发育比较成熟的繁殖群体的营养状况,并比较分析了其不同组织总脂和脂肪酸组成,为探明长江口水域刀鲚繁殖群体的营养状况及繁殖性能提供参考信息。结果显示,肌肉、肝脏和卵巢中的水分含量依次降低,总脂含量依次增加。饱和脂肪酸(SFA)总量在肝脏中最高,肌肉中最低,3种组织间差异显著(P<0.05);单不饱和脂肪酸(MUFA)中C18:1n9c的含量最高,其在肌肉、肝脏和卵巢中的含量依次为(40.88±0.19)%、(35.06±1.84)%和(42.85±2.14)%,肝脏中含量显著低于其他2个组织(P<0.05);多不饱和脂肪酸(PUFA)、n3-PUFA及二十二碳六烯酸(DHA)含量在肌肉、肝脏和卵巢中依次升高,卵巢中的含量显著高于肝脏和肌肉(P<0.05),而后二者间的差异不显著(P>0.05)。肌肉、肝脏和卵巢中不饱和脂肪酸(UFA)/SFA的比值依次为2.35、1.67和4.49,n3-PUFA/n6-PUFA的比值依次为4.94、3.87和5.13,2组比值均在卵巢中最高,其次为肌肉,肝脏中最低。综上所述,本研究发现,卵巢发育至IV期的刀鲚成体的脂类和多不饱和脂肪酸营养存在向卵巢中选择性积累的特点,这是对该时期刀鲚繁殖活动的一种适应,该结果为进一步探明卵巢不同发育期刀鲚成体的营养需求差异及必需脂肪酸的合成转化途径提供了基础资料。  相似文献
