Since formalin is widely used in prevention of Saprolegnia infections in salmonid fish hatcheries, there is a need for more environmentally safe treatment methods. Therefore, we screened 360 bacterial isolates against their ability to antagonize the growth of Saprolegnia parasitica hyphae in vitro, and best strains were selected according to their antagonistic properties and colonization capability on rainbow trout egg surface. Protective bacterial cultures of Pseudomonas sp. M162, Pseudomonas sp. M174 and Janthinobacterium M169 were tested for prevention of Saprolegnia sp. infections during incubation trials of rainbow trout (Oncorhynchus mykiss) eggs with UV irradiated (400 mWs cm?2) and non‐treated inlet water. UV irradiation of inlet water significantly decreased mortality during the incubation. Lowest mortalities were observed in protective culture treated groups incubated with UV‐irradiated inlet water. UV irradiation increased the dominance of the main bacterial colonizers and variation in the bacterial species diversity between the experimental units.  相似文献

Inhibitory concentrations of clove oil and ethanol against growth of Saprolegnia sp. hyphae were screened by a modification of the hemp (Cannabis sativa L.) seed MicroPlate (HeMP) method and their usability as antifungal agents during incubation of rainbow trout Oncorhynchus mykiss eggs was tested. In vitro experiment showed that in continuous static exposure, clove oil at 100 mg L?1 significantly inhibited the growth of Saprolegnia, whereas in bath exposures, clove oil at 500 mg L?1 had no significant effect at any exposure time tested (15, 60 and 240 min), but clove oil at 10 000 mg L?1 significantly inhibited growth at all exposure times. Clove oil and ethanol treatments had no visible effects on the onset or spread of the fungus during incubation of rainbow trout eggs. Clove oil at 1000 mg L?1 resulted in 95–100% mortality before the eyed stage was reached. Sublethal concentrations of clove oil and ethanol had no effects on the development rate of the embryo or growth and yolk utilization efficiency after hatching. This study suggests that clove oil and ethanol may not be options in controlling aquatic fungi infestations during incubation of rainbow trout eggs.  相似文献

Pinfish (Lagodon rhomboides Linnaeus) and pigfish (Orthopristis chrysoptera L.) are relatively new, cultured species commonly used as marine baitfish in the south‐eastern United States and currently have no defined protocols for egg incubation. Thus, experiments were conducted to determine efficient egg stocking densities during static and flow‐through incubation that yielded higher quality larvae. Eggs of each species were incubated statically at 250, 500, 1000 and 2000 eggs L?1, and after incubation, egg hatching success, larval survival to first feeding, larval morphometrics and water quality were assessed. Stocking densities above 250 eggs L?1 led to significant reductions in pinfish hatching success and water quality degradation, evident from decreased dissolved oxygen and pH levels and increased nitrogenous wastes. Increased stocking densities for pigfish also resulted in significant water quality degradation, although hatching success was unaffected up to a density of 1000 eggs L?1. A high flow‐through water exchange rate of 2000% daily resulted in significant reductions in nitrogenous wastes and greater stability in dissolved oxygen and pH levels during incubation when compared to static treatments of the same egg density. Additionally, the high exchange rate of 2000% was critical in maintaining high hatching success and larval survival to first feeding at stocking densities of 1000 eggs L?1 for pinfish and up to 4000 eggs L?1 for pigfish. No clear patterns in larval morphometrics were observed among stocking densities. Static incubation densities of 250 and 1000 eggs L?1 are recommended for pinfish and pigfish, respectively.  相似文献

Egg disinfection is considered the most important routine work in hatcheries to avoid fungal and/or bacterial infection of fish eggs. The aim of this study was to determine the effectiveness of three disinfectants: formalin, iodine and sodium chloride on the hatching success of yellow perch eggs. The disinfectants were tested in triplicate at different concentrations for 15 and 30 min bath treatments. Two experiments were conducted; formalin at five concentrations (25, 50, 100, 150 and 200 mg L?1) and 25 mg L?1 iodine were tested in the first experiment. The second experiment involved formalin at three concentrations (250, 500 and 1000 mg L?1), iodine at three concentrations (50, 100 and 250 mg L?1) and sodium chloride at three concentrations (500, 1000 and 3000 mg L?1) were used. Iodine and sodium chloride‐treated eggs hatched earlier than formalin‐treated eggs. The highest mean percentage of eyed stage, hatching rate and survival to first feeding fry was observed at 200 mg L?1 formalin for 30 min, 50 mg L?1 iodine for 15 min and 500 mg L?1 sodium chloride for 30 min. High concentrations of formalin (1000 mg L?1), iodine (250 mg L?1) and sodium chloride (1000 and 3000 mg L?1) showed toxicity to yellow perch eggs, resulting in low hatching rate and survival to first feeding fry. We recommended formalin at a concentration of 150–200 mg L?1 for 30 min as an effective, easily available and low‐cost disinfectant for routine use to improve yellow perch hatchability.  相似文献

Elimination of egg stickiness is an important factor in artificial reproduction of pikeperch (Sander lucioperca L.). This study was conducted to evaluate the effectiveness of Alcalase enzyme to remove the adhesive layer of pikeperch eggs. The eggs were treated with Alcalase at 0.5, 1.0, 1.5, 2.0 and 5.0 mL L?1 or a milk/talc solution 2 min post insemination. Duration of exposure was 2 min in Alcalase and 60 min in milk/talc. The highest, albeit not significant, hatching rate (85.4%) was found with 1.5 mL L?1 of Alcalase, but hatching rates were similar in 0.5, 1.0 and 2.0 mL L?1. Hatching rates were significantly lower groups treated with 5.0 mL L?1 Alcalase enzyme (56.4%) compared to groups treated with milk powder and talc (61.3%). Nominally complete removal of adhesiveness was observed in 1.5 and 2 mL L?1. All Alcalase treatments led to significantly lower incubation duration compared with the traditional milk/talc treatment. The application of Alcalase successfully eliminated pikeperch egg stickiness in less time than with traditional milk/clay/talc methods.  相似文献

Egg strings and nauplii of the salmon louse Lepeophtheirus salmonis were exposed to a variety of medicinal compounds at 50 mg L?1 for 30 min in two experiment series. This medicine concentration was selected as a starting point for a screening series. Hatching of egg strings and development to copepodid larvae were monitored in one experiment, and the survival and development of nauplii were monitored in the other. Two compounds, emamectin benzoate and cypermethrin, inhibited hatching effectively. Several compounds affected nauplii, either directly or through inhibiting development to the infective stage. A total of 50 mg L?1 of azamethiphos, cypermethrin, emamectin benzoate and propoxur was lethal to >70% of the larvae. Diflubenzuron, fenoxycarb, pymetrozine, pyriprole and tebufenozide diminished the ability of nauplii developing to copepodids.  相似文献

Different laboratory synthesized metal nanoparticles viz. Copper oxide (CuO), Zinc oxide (ZnO) and silver doped titanium dioxide (Ag‐TiO2) were studied for their effect on hatching and survival of larvae and fry of Indian major carp, rohu, Labeo rohita both in direct application in tank water & coated onto tanks. Among these nanoparticles, CuO and ZnO nanoparticles exhibited highest percentage of hatching in both direct addition (78.0 ± 3.1% and 78.05 ± 4.2%, respectively) and coating onto tanks (58.6 ± 2.1% and 61.2 ± 2.7%, respectively) at 1 mg mL?1 while least percentage of hatching was recorded in Ag‐TiO2 nanoparticles irrespective of its concentration & mode of supplementation. Highest survival of L. rohita fry (50.13 ± 2.2%) was observed after 15 days post hatching in CuO coated tanks followed by ZnO coated tanks (38.6 ± 2.8%) while least was recorded in Ag‐TiO2 coated tanks (22.53 ± 3.0%). However in control tanks coated with Poly‐Urethane base with hardener and uncoated control tanks, the survival was 42.4 ± 1.2% and 41.36 ± 1.8% respectively. Further, significantly lower microbial load of water was recorded in CuO nanoparticles coated tanks (1.5 × 1010 CFU L?1) as compared to uncoated control tanks (1.1 × 1016 CFU L?1) without affecting water quality parameters. On the other hand, in Ag‐TiO2 coated tanks, significantly lower microbial load (1.0 × 106 CFU L?1) as compared to uncoated control tanks at 15 days post hatching was recorded. However, Ag‐TiO2 was toxic to L. rohita larvae & fry both in direct application and coating onto tanks. Considering the beneficial effects of CuO nanoparticle application, it has the scope of being used in a more eco‐friendly way in hatchery operations.  相似文献

This study examined the effects of water hardness and salinity on yolk sac larvae and swim‐up fry survival of Nile tilapia, Oreochromis niloticus (Chitralada strain), eggs during artificial incubation. Four experiments were conducted to evaluate the effects of hardness, salinity and the sources of saline incubation water. High water hardness treatments (500–4200 mg L?1 as CaCO3) resulted in higher yolk sac larvae and swim‐up fry survival than low water hardness treatments (50.0 and 132 mg L?1 as CaCO3); although yolk sac larvae and swim‐up fry survival did not differ among the high or low hardness treatments. Salinity of 4.0 g L?1 using seawater, and 4.0 and 8.0 g L?1 using unprocessed common salt resulted in the higher survival rate of yolk sac larvae and swim‐up fry than other salinity treatments. Yolk sac larvae and swim‐up fry survival was found to decrease with the increase in salinity and increase with the increase in water hardness. The present study demonstrated the positive effects of increased water hardness level (>132 mg L?1) on yolk sac larvae and swim‐up fry survival. The study also showed that seawater salinity of 4 g L?1 was the most appropriate salinity level for incubating Nile tilapia eggs.  相似文献

Effluent discharges from aquaculture can reduce water quality in receiving water bodies and that strategies or practices to reduce this are necessary. One possibility is to reduce, or eliminate, water renewal in grow‐out ponds. In this study, we eliminated water renewal in grow‐out ponds associated with the culture of 40 individuals m?2 of Amazon river prawn (Macrobrachium amazonicum). At the end of the culture period it was, however, necessary to drain the pond to harvest the prawns. An experiment was performed in triplicate, in which the water supply characteristics and harvest water characteristics of ponds were evaluated. To reduce these concentrations of total N and P, an aquatic macrophyte (Eichhornia crassipes, water hyacinth) treatment system (CWs) was adopted. The water characteristics in the CWs were evaluated after 1, 3, 7, 14 and 21 days. The water supply of ponds presented the average concentrations of 0.67 ± 0.32 mg L?1 and 17.4 ± 14.7 μg L?1 of total‐N and total‐P respectively. The harvest effluent of ponds had elevated concentrations of different forms of nitrogen (4.44 mg L?1 of total‐N) and phosphorous (100.9 μg L?1 of total‐P). After 1 day of the experiment we found the following reductions in key nutrients in treatment system containing E. crassipes: 90%, 78% and 45% reductions in the concentrations of particulate matter, orthophosphates and nitrates respectively. We noted that after 3 days the nitrates had been reduced by 53%. We concluded that 3 days of this treatment was sufficient for the removal of the additional nutrients that had accumulated in the Amazon river prawn ponds.  相似文献

Three in vivo experiments were conducted to measure the effectiveness of commercial Bacillus probiotics on survival and development of Litopenaeus vannamei larvae from nauplii 3–4 (N3–4) to zoea 3 (Z3) stages. Experiment I: Nine commercial Bacillus probiotics were individually added to larvae twice at N6 and Z1 at levels of 2, 20, and 100 mg L?1. Only six of nine products at 20 mg L?1 exhibited higher or significantly higher larval survival (< 0.05) than the control. Experiment II: Two superior products from the first experiment were administered to larvae with six dose frequencies at 20 mg L?1. For both products, three doses, once at each of N3–4, N6 and Z1 stages, yielded the best larval survival and development rates (< 0.05), and these were confirmed by enhanced activities of tryptase and amylase. Experiment III: The isolates of these two products, identified as Bacillus subtilis and Bacillus licheniformis, were delivered to larvae singly at concentrations of 1.0 × 108 and 1.0 × 109 CFU L?1, or at the same concentrations by mixing the two equally. At 1.0 × 109 CFU L?1, B. subtilis exerted more beneficial effects on larvae than B. licheniformis or the mixture. Therefore, the optimal dosage and dose frequency of commercial Bacillus products should be evaluated prior to large‐scale application in shrimp hatcheries to avoid futility or even adverse effects, as spore counts and non‐bacterial ingredients are key factors influencing the efficacy of Bacillus probiotics.  相似文献

Atlantic salmon, Salmo salar L. parr (age 1+), infected by the monogenean ectoparasite Gyrodactylus salaris (Malmberg, 1957), were exposed to chlorine (Cl)‐enriched water at three different concentrations: Cllow (0–5 μg Cl L?1), Clmedium (18 μg Cl L?1) and Clhigh (50 μg Cl L?1). There was a negative correlation between G. salaris infections and the hypochlorite concentrations added. The parasite infection was eliminated by day 6–8 and day 2–4 in the groups Clmedium and Clhigh, respectively, while inhibition of G. salaris population growth was observed in the Cllow group. An important note to this matter, however, is that the G. salaris specimens observed at day 6 in Clmedium and at day 2 in Clhigh were all considered dead by subjective judgement. No mortality in the salmon parr was observed during the first 8 days of the experiment, demonstrating that Cl has a stronger effect on G. salaris than on the salmonid host. The differences in sensitivity between the parasite and the Atlantic salmon indicate that hypochlorite has a potential use as a parasiticide with a therapeutic margin. The low‐dose sensitivity may imply that Cl pollution in urban areas may pose a greater risk towards biodiversity than previously assumed.  相似文献

As marine finfish aquaculture expands, there is an increasing interest in the ability to ship early life stages from breeding centres to hatcheries so that each hatchery does not have to maintain its own broodstock. Here, we conducted 24 h air‐shipping simulations with yolk‐sac larvae of California yellowtail (CYT; Seriola lalandi) and white seabass (WSB; Atractoscion nobilis) to help fill in the informational gaps for shipping marine fish larvae. We examined the effects of a pH buffer on water quality, post‐shipping larval survival and subsequent survival to first feeding at larval densities of 1000, 3000, 6000 and 9000 larvae L?1. The pH buffer, 8.3 Trizma®, was tested at varying concentrations of zero (NT = 0.00 g L?1), low (LT = 0.75 g L?1), medium (MT = 1.5 g L?1) and high (HT = 3.0 g L?1). Trials were conducted using replicate 2 L aquarium bags filled with 500 mL of seawater and held in a water bath at 19–20°C. Results showed an interspecific difference in survival at the highest shipping densities under these experimental conditions. Shipping densities up to 6750 CYT larvae L?1 or 3000 WSB larvae L?1 consistently yielded >90% survival immediately after simulated shipment and >85% survival 48 h after the simulations. Furthermore, at these densities, pH was maintained at ~8.0 when buffered at 1.5 g L?1. The highest tested densities of 9580 CYT larvae L?1 and 9940 WSB larvae L?1, yielded lower survival 69–79% and 0.0–1.3% respectively after 24 h. Final pH in the high density CYT trials were unsatisfactory (below 7.0), regardless of the buffer concentration; however pH in the WSB high density trials improved with increasing buffer concentration. On the basis of the results from these air‐shipping simulations, we recommend CYT and WSB larvae be shipped in seawater with 1.5 g L?1 Trizma® at densities not greater than 6750 larvae L?1 for CYT and 3000 larvae L?1 for WSB. We believe this represents an important step in improving long distance transport protocols for these species and provides useful guidance in air transport of other economically and ecologically important marine species. Additional research is warranted to compare these simulation results with those from actual air shipments, as we did not account for factors that may vary in flight like temperature and pressure variations, and physical agitation.  相似文献

In this study, we analysed the effect of population density and food concentration on the fecundity of a Mediterranean strain of Acartia tonsa to maximize egg production. During 4‐day feeding experiments, egg hatching success and faecal pellet production were also followed. The algae Rhinomonas reticulata was supplied at different concentrations corresponding to 250, 500, 1000, 1500, 2000 and 3000 μg C L?1 day?1 at the following adult copepod density: 40, 80 and 160 ind. L?1. Our results show a positive relationship between algal concentration and egg production under all experimental conditions confirming that the quantity of food strongly limits A. tonsa fecundity. Maximum egg production (57 eggs per female) was reached at the lowest density and at the maximum food concentration. Percentage of egg hatching success was not dependent on the quantity of food used. At the same food concentration, an increase in population density from 40 to 80 ind. L?1 induced an increase in faecal pellet production per couple which did not correspond to an increase in egg production, suggesting that higher energetic costs were shifted to swimming activity. Productivity of the A. tonsa Mediterranean strain is mainly limited by the quantity of food rather than by crowding conditions.  相似文献

Garlic, Allium sativum L., extract administered as a therapeutic bath was shown to have antiparasitic properties towards Neobenedenia sp. (MacCallum) (Platyhelminthes: Monogenea) infecting farmed barramundi, Lates calcarifer (Bloch). The effect of garlic extract (active component allicin) immersion on Neobenedenia sp. egg development, hatching success, oncomiracidia (larvae) longevity, infection success and juvenile Neobenedenia survival was examined and compared with freshwater and formalin immersion. Garlic extract was found to significantly impede hatching success (5% ± 5%) and oncomiracidia longevity (<2 h) at allicin concentrations of 15.2 μL L?1, while eggs in the seawater control had >95% hatching success and mean oncomiracidia longevity of 37 ± 3 h. At much lower allicin concentrations (0.76 and 1.52 μL L?1), garlic extract also significantly reduced Neobenedenia infection success of L. calcarifer to 25% ± 4% and 11% ± 4%, respectively, compared with 55% ± 7% in the seawater control. Juvenile Neobenedenia attached to host fish proved to be highly resistant to allicin with 96% surviving 1‐h immersion in 10 mL L?1 (15.2 μL L?1 allicin) of garlic extract. Allicin‐containing garlic extracts show potential for development as a therapy to manage monogenean infections in intensive aquaculture with the greatest impact at the egg and larval stages.  相似文献

The effects of six 1,3;1,6‐β‐D‐glucooligo‐ and polysaccharides with different structures (ranging from 1 to 10 kDa in molecular mass and containing 10–25% of β‐1,6‐linked glucose residues) from brown algae, Saccharina cichorioides, on development of the chum salmon, Oncorhynchus keta (Walbaum), were evaluated. Exposure of chum salmon eggs to 1,3;1,6‐β‐D‐glucans with a molecular mass of more than 2 kDa increased the survival of embryos and juveniles and their resistance to Saprolegnia infection by up to 2.5‐fold, leading to a weight gain in juveniles of 40–55% compared with The control chum salmons. The 1,3;1,6‐β‐D‐glucans with molecular mass of 6–8 kDa and used at a at concentration of 0.5 mg mL?1 rendered the best stimulative effect.  相似文献

Currently, the only effective and commercially used treatment for amoebic gill disease (AGD) in farmed Tasmanian Atlantic salmon is freshwater bathing. Hydrogen peroxide (H2O2), commonly used throughout the aquaculture industry for a range of topical skin and gill infections, was trialled in vitro and in vivo to ascertain its potential as an alternative treatment against AGD. Under in vitro conditions, trophozoites of Neoparamoeba perurans were exposed to three concentrations of H2O2 in sea water (500, 1000 and 1500 mg L?1) over four durations (10, 20, 30 and 60 min) each at two temperatures (12 and 18 °C). Trophozoite viability was assessed immediately post‐exposure and after 24 h. A concentration/duration combination of 1000 mg L?1 for >10 min demonstrated potent amoebicidal activity. Subsequently, Atlantic salmon mildly affected with experimentally induced AGD were treated with H2O2 at 12 and 18 °C for 15 min at 1250 mg L?1 and their re‐infection rate was compared to freshwater‐treated fish over 21 days. Significant differences in the percentage of filaments affected with hyperplastic lesions (in association with amoebae) and plasma osmolality were noted between treatment groups immediately post‐bath. However, the results were largely equivocal in terms of disease resolution over a 3‐week period following treatment. These data suggest that H2O2 treatment in sea water successfully ameliorated a clinically light case of AGD under laboratory conditions.  相似文献

To improve the efficiency of stocking lakes with vendace (Coregonus albula) larvae, a technique for delaying egg hatching was developed. This synchronizes mass hatching with the development of suitable thermal conditions and food availability in the lake.Water temperatures were measured in Polish hatcheries and a mathematical model of the dependence of C. albula embryogenesis on incubation temperature was utilized to predict the possibilities of delaying egg hatching. Vendace hatching can be delayed by cooling the circulation water (to 1 or 2°C) during March and April. At the beginning of May, when lake conditions are optimal for stocking, the incubation temperature should be raised (at a rate of 1.5°C h?1) to that of the water supply. This ensures mass hatching of strong, normally developed larvae within 2–3 days. This technique facilitates both the delay of vendace hatching at minimal cost and the production of larvae on demand for lake stocking.  相似文献

Understanding the factors limiting copepod productivity in dense cultures is a prerequisite for the partial or entire replacement of Artemia and rotifers as live feed for finfish larvae. In dense cultures, high encounter rates between individuals may increase stress, cannibalism incidents and potentially trigger resting egg production. We conducted an experiment to evaluate the potential egg production and egg quality of Acartia tonsa stocked at densities ranging from 10 to >5000 ind. L?1. Egg Production (EP), Delayed Hatching Eggs production (DHE), hatching success (HS), egg mortality and water quality were used as end points. In the present system, A. tonsa was raised at >5000 ind. L?1 without affecting the mortality, confirming that attaining this high density in culture is possible. However, egg harvest reached an optimum of 12 000 egg L?1 day?1 at ~2500 ind. L?1 indicating that increasing stocking density above this level is not of practical interest. Calculations showed that the loss in egg harvest at stocking densities <2500 ind. L?1 is of 1.3% for every additional 100 adult copepods L?1. The increasing adult density did not affected the proportion of DHE produced (~10% of harvest) but decreased significantly the HS, though not to a point that would be problematic in a commercial production. Understanding the biology of copepods when stocked at high density is important to improve copepod culture systems and increase egg harvest yields. Technical solutions such as the continuous separation of eggs from adults in the water column, recirculation and the continuous provision of food are seen as potential solutions.  相似文献

Observations were made in an experimental stream tank (total area 14.7 m2) on juvenile Atlantic salmon, Salmo salar L (parr), relating experimental observations to field observations, including the reported diurnal fasting behaviour of juvenile salmon at water temperatures <10 °C. Densities in the tank ranged from five to twenty parr, at water temperatures ranging from 4.6 °C to 15.8 °C. The wide channel of the stream tank, with mean water velocity of 18.8 cm·s?1, was the preferred section, where territorial behaviour was observed. Biomass was regulated in the wide channel by territorial mosaics or by dominance hierarchies. Dominance hierarchies were reflected in coloration of the fish. Dominant salmon were generally in the wide channel. Densities of salmon parr (of mean fork length 10.2 cm) in the channel ranged from 0.84 m?2 to 1.73 m?2, with an average biomass of 14.2 g·m?2. Growth was least at the 5.9 °C temperature treatment. In experiments at temperatures below 10 °C, feeding, dominance hierarchy and territorial behaviour were observed in daylight hours, contrary to the published literature. Interactions with other species may affect behaviour. Some observations were made on a closely related species, brown trout (Salmo trutta L.), a commonly cohabiting species in many systems. Trout displaced salmon from their preferred locations in the tank and were more aggressive than the salmon, reducing agonistic behaviour by the salmon. The commonest agonistic act shown by salmon was ‘charge’ and that by the trout was ‘approach’. Some field observations affecting behaviour and production are discussed.  相似文献

Heart deformities are a concern in aquaculture and are linked to egg incubation temperature. Diploid and triploid Atlantic salmon, Salmo salar L., were incubated at 6, 8 and 10 °C and analysed for aplasia of the septum transversum (= 150 ploidy?1 incubation temperature?1). Heart morphology (size and shape) was assessed in fish incubated at 6 °C and in fish with and without aplasia of the septum transversum (= 9 group?1) incubated at 10 °C. Egg mortality was significantly higher in triploids than in diploids at all incubation temperatures, and increased egg incubation temperatures increased mortality in both ploidy. Triploids grew quicker than diploids after egg incubation at 10 °C, but not at 6 °C. Aplasia of the septum transversum occurred only in triploid fish after incubation at 6 °C and 8 °C (0.7% and 3.3%, respectively) and was significantly greater (≤ 0.05) in triploids after incubation at 10 °C compared with diploids (30% and 18%, respectively). Aplasia of the septum transversum significantly increased heart mass and resulted in a long flat ventricle compared with fish displaying a septum transversum.
