This work addresses the question of occurrence and function of photosystem II (PSII) in bundle sheath (BS) cells of leaves possessing NADP-malic enzyme-type C4 photosynthesis (Zea mays). Although no requirement for PSII activity in the BS has been established, several component proteins of PSII have been detected in BS cells of developing maize leaves exhibiting O2-insensitive photosynthesis. We used the basal fluorescence emissions of PSI (F 0I) and PSII (F 0II) as quantitative indicators of the respective relative photosystem densities. Chl fluorescence induction was measured simultaneously at 680 and 750 nm. In mature leaves, the F m(680)/F 0(680) ratio was 10.5 but less in immature leaves. We propose that the lower ratio was caused by the presence of a distinct non-variable component, F c, emitting at 680 and 750 nm. After F c was subtracted, the fluorescence of PSI (F 0I) was detected as a non-variable component at 750 nm and was undetectably low at 680 nm. Contents of Chls a and b were measured in addition to Chl fluorescence. The Chl b/(a + b) was relatively stable in developing sunflower leaves (0.25–0.26), but in maize it increased from 0.09 to 0.21 with leaf tissue age. In sunflower, the F 0I/(F 0I + F 0II) was 0.39 ± 0.01 independent of leaf age, but in maize, this parameter was 0.65 in young tissue of very low Chl content (20–50 mg m?2) falling to a stable level of 0.53 ± 0.01 at Chl contents >100 mg m?2. The values of F 0I/(F 0I + F 0II) showed that in sunflower, excitation was partitioned between PSII and PSI in a ratio of 2:1, but the same ratio was 1:1 in the C4 plant. The latter is consistent with a PSII:PSI ratio of 2:1 in maize mesophyll cells and PSI only in BS cells (2:1:1 distribution). We suggest, moreover, that redox mediation of Chl synthesis, rather than protein accumulation, regulates photosystem assembly to ensure optimum excitation balance between functional PSII and PSI. Indeed, the apparent necessity for two Chls (a and b) may reside in their targeted functions in influencing accumulation of PSI and PSII, respectively, as opposed to their spectral differences.  相似文献

The interception by crop canopies of radionuclides in rainfall can be important in determining radiation exposures to animals and man. Data were obtained on the sorption and desorption of radionuclides on the adaxial surfaces of fully expanded bean leaves by exposing them to ionic forms of caesium (Cs+), iodine (I) or sulphur (SO42−) over a six order of magnitude concentration range. The accumulation of each element was determined as a time course over a 48 h period using radioactive labels (137Cs, 125I or 35S, respectively). Time- and concentration-dependent sorption of each element to the leaf surface was analysed to determine: (a) the leaf surface-solution distribution coefficient (Kd) at equilibrium and (b) the sorption and desorption rate coefficients for each element over the range of concentrations investigated. It was expected that Cs+ would show a stronger tendency to sorb to the leaf surface than both I and SO42− because of the cation exchange properties of the cuticular membrane. The Kd for Cs+ was approximately 90× greater than that for SO42− but 5× less than that for I. This is thought to be due to either (a) the highly organophilic nature of iodide and the relatively high iodine number of cuticular waxes on plant leaf surfaces or (b) the possible oxidation of I to I0 or IO3, with consequently enhanced leaf surface sorption. Based on data obtained in this study, ranges and best estimates of sorption and desorption rate coefficients are presented for Cs+, I and SO42− for use in modelling the interception of radioactive Cs, I and S in rainfall by crops.  相似文献

We studied acclimation patterns in leaf dry mass per area (MA), nitrogen (NA) and chlorophyll (ζA) content per area, and chlorophyll to nitrogen ratio (ζ/N) along vertical light gradients in natural temperate mixed herbaceous canopy and deciduous tree canopy. In the deciduous tree canopy, all leaves are formed at approximately the same time, and the light gradient during the rest of the growing season reflects the differences in light availability during leaf development, whereas in the herbaceous canopy, leaf production continues during most of the growing season and major changes in light conditions occur after leaf maturation. MA and NA increased strongly with increasing current light availability (ID) in the tree canopy. In the herbaceous canopy, MA and NA were generally unrelated to ID. Depending on species, the correlation between chlorophyll content per leaf area (ζA) and ID was positive, negative, or non-significant. Path analyses revealed two opposite effects of ID on the amount of leaf chlorophyll. In the tree canopy, increasing ID enhanced ζA through changes in MA and NA, whereas the direct effect of light was negative in both canopies. The overall correlation network between foliage structural and chemical traits and the relationships with ID were significantly stronger in the tree canopy, suggesting limited re-acclimation potential in the mixed herbaceous canopy. Within-species acclimation patterns reflected the patterns within the main functional types. These data demonstrate that the relationships of current light availability vs. leaf dry mass per area, leaf nitrogen and chlorophyll contents, and chlorophyll to nitrogen ratio differ among multi-species herbaceous canopies and deciduous tree canopies due to contrasting canopy development.  相似文献

The slope of the initial linear range of a photosynthesis–irradiance (PI) curve, alpha (α), is frequently, but often incorrectly, used to denote the maximal quantum yield (or the “efficiency” of photosynthesis) of higher plants and macroalgae under the conditions for which the PI curve was measured. When using the increasingly popular method of pulse amplitude modulated (PAM) fluorometry, the determination of α from so-called rapid light curves (RLC) may lead to misinterpretations when comparing photosynthetic efficiencies under different environmental conditions. Furthermore, since PAM fluorometry measures the quantum yield (Y) directly, there may be no need to estimate it from the initial slopes of RLCs.We compared photosynthetic parameters derived from RLCs of Ulva sp. measured during winter and summer, and show large differences in α when electron transport rates (ETR) were plotted against incident irradiance (Ii) [α = 0.26 ± 0.00 versus 0.08 ± 0.01 during the winter (November–December) and summer (July–August), respectively], as is usually done. On the other hand, no differences in the initial slopes of the RLCs were apparent when plotting ETR versus the absorbed irradiance (Ia) (initial slope = 0.75 ± 0.01 versus 0.62 ± 0.12 during the winter and summer, respectively); this is called for since also ETR is calculated using Ia. Using the Ia based RLCs, it was also found that the values of the initial slopes equalled those of the first Y-value measurements of the RLCs (Y0) (t-test, p > 0.05, r2 = 0.85). Therefore, when using PAM fluorometry, we suggest (a) to present the x-axis of RLCs as Ia (Ii × AF × 0.5), and ETR on the y-axis as Y × Ia, and (b) that Y0 can be taken as a correct measure of the maximal quantum yield instead of estimating it from an RLC.  相似文献

An equal number of male and female rats of the King-Holtzman hybrid breed, divided into two age groups (group A1 : 21–30 days old and group A2 : 90–100 days old), were exposed to two levels of noise: N0 = 30 db and N1 = 90 db, (Ref. 0.0002μ bar), and three levels of negative air ions: IO = no measurable ion concentration, I1 = 7 × 106 ions/cm3 and I2 = 7 × 107 ions/cm3. Time and error scores of 240 rats running a modified Lashley left-right maze with an escape-from-water motive served as criteria. A randomized complete blocks design with replications (2×3×2×2×10) was selected for treatment by analysis of variance. The results indicate that (a) the males show significantly lower error score in negatively ionized air; and (b) the females swim significantly faster than males under all investigated conditions with no apparent effect of noise or ions on their performance.
Zusammenfassung Eine gleiche Anzahl m?nnlicher und weiblicher Ratten (n = 120) des King-Holtzman Stammes, unterteilt in 2 Altersgruppen (21–30 und 90–100 Tage) wurden zwei Ger?uschintensit?ten (30 db und 90 db) und 3 Konzentrationen negativer Luftionen I0: nicht messbar niedrig, I1:7×106, I2:7×107 Ionen/cm3 ausgesetzt. Die Wirkung wurde geprüft an der Schwimmzeit und der Anzahl Fehler der Tiere im Lasley-Links-Rechts-Labyrinth. Das Motiv zum Schwimmen war, aus dem Wasser zu entkommen. Die Auswertung durch Varianzanalyse erfolgte unter Nachbildung randomisierter kompletter Blockentwürfe. Die Ergebnisse zeigen, dass (a) die M?nnchen bei negativ ionisierter Luft signifikant weniger Fehler machten und (b) die Weibchen unter allen Bedingungen schneller schwammen als die M?nnchen, ohne dass das L?rmniveau oder die Luftionisation darauf einen Einfluss hatten.

Resume Un nombre égal de rats males et femelles (120 de chaque) de la race King-Holtzman futdivisé en deux groupes d'age (21 à 30 jours et 90 à 100 jours). Ils furent exposés à 2 intensités auditives (30 et 90 db) et à 3 concentrations d'ions négatifs: I0 non mesurables (faible), I1 : 7 × 106 et I2: 7 × 107 ions/cm3. Les effets d'un tel traitement furent examinés au moyen de deux critères: le temps pris pour sortir de l'eau et le nombre de fautes commises dans un labyrinthe droitegauche de Laslay. Pour l'analyse des variances, on a adopté une répartition due au hasard avec répétitions (2×3×2×2×10). On peut en conclure que: (a) les males font moins d'erreurs dans l'air ionisé négativement et cela de fa?on significative et (b) les femelles ont nagé plus vite que les males dans toutes les conditions de l'essai. Ni le bruit ni le degré d'ionisation ne semblent avoir d'influence sur leur comportement.

The variation of light intensity has obvious effects on leaf external morphology, internal anatomy, and physiological characteristics; it even induces changes in secondary metabolite production. The effects of different irradiance levels on biomass, gas exchange parameters, and photosynthetic pigment contents in Mahonia bodinieri (Gagnep.) Laferr. were analyzed here. Combined analyses of physiology, cytology, and HPLC were used to study the differences in leaf morphology, structure, physiological characters, and alkaloid content in response to different irradiances. The results indicated that the highest foliar biomass was observed under I 50 (50 % of full sunlight) followed by I 30 (30 % of full sunlight), the highest net photosynthetic rate, stomatal conductance, transpiration rate values were observed under I 30 followed by I 50, and lower values occurred in I 10 (10 % of full sunlight) and I 100 (full sunlight). With increased light intensity, total leaf area and the contents of chlorophyll a (Chl a), chlorophyll b (Chl b), and chlorophyll (Chl a+b) per unit leaf area were clearly reduced, whereas leaf mass per area, carotenoid content, leaf thickness, thickness of palisade and spongy parenchyma, and stomatal density were all significantly increased. Electron microscopic observation revealed that the number of grana, stroma lamellae and the number of starch grains in chloroplasts were decreased, the number of plastoglobuli was increased when irradiance levels increased. The estimated total yield of alkaloids in a single plant was higher under I 30 and I 50 than under I 10 or I 100 as a result of the higher biomass of the plants. Therefore, I 30 and I 50 were not only beneficial to increase biomass, but also suitable for the synthesis and accumulation of the major secondary metabolites (alkaloids). Our findings provide valuable data for the determination and regulation of irradiance levels during artificial cultivation of M. bodinieri.  相似文献

A 2‐year study on epidemic progress of apple scab was conducted at Randwijk, the Netherlands, in 1998 and 1999. The summer epidemic caused by conidia was studied instead of the well‐described spring season epidemic originating from ascospores. The aim was to investigate relationships between disease measurements, i.e. disease incidence and severity measures of apple scab, and their implications for the development of predictive models and threshold levels. The study characterized good relationships between the measurements on cultivar Jonagold using regression analyses in three disease control regimes (untreated, organic and integrated). For fruit quality prediction, the relationship between fruit incidence (If) and leaf incidence (Il) in the organic control regime was given by If = 1.966 + 0.402 × (Il) (R2 = 0.92). As a result of low level of disease in the integrated control regime, shoot incidence (Is), with higher values than leaf incidence, was better suited for prediction. The relationship was given by If = ?0.162 + 0.028 × (Is) (R2 = 0.91). For the integrated control regime, disease threshold levels were constructed for timing of the final fungicide application. If an apple grower wants to keep fruit infection under 1% incidence (harvest scab threshold), the timing of the final fungicide application (action threshold) should correspond to 4% shoot scab incidence at the beginning of August. The results are compared with similar studies and their biological interpretation is discussed.  相似文献

Arthrobacter sialophilus neuraminidase catalyzes the hydration of 5-acetamido-2,6-anhydro-3, 5-dideoxy-d-glycero-d-galacto-non-2-enonic acid (2,3-dehydro-AcNeu) with Km and kcat values of 8.9 × 10?4m and 6.40 × 10?4 s?1, respectively. The methyl ester of 2,3-dehydro-AcNeu as well as 2,3-dehydro-4-epi-AcNeu are also hydrated by the enzyme. The product resulting from the enzymatic hydration of 2,3-dehydro-AcNeu is N-acetylneuraminic acid. A series of derivatives of 2,3-dehydro-AcNeu (KI 1.60 × 10?6m) including 2,3-dehydro-4-epi-AcNeu (2.10 × 10?4m) and 2,3-dehydro-4-keto-AcNeu (KI = 6.10 × 10?5 m) were each competitive inhibitors of the enzyme. The methyl esters of these ketal derivatives were also competitive enzyme inhibitors. Dissociation constants for these ketals were determined independently by fluorescence enzyme titrations which gave values similar to those found kinetically. These six relatives of 2,3-dehydro-AcNeu were also competitive inhibitors for the influenza viral neuraminidases. For the viral neuraminidases, the dissociation constant for 2,3-dehydro-AcNeu and its methyl ester were 2.40 × 10?6 and 1.17 × 10?3m, respectively. The interpretation placed upon the KI values determined for these ketals against the Arthrobacter versus influenza neuraminidases is that the bacterial enzyme has a more flexible glycone binding site.  相似文献

The aim of this work was to determine the concentrations and properties of free radicals in piperacillin, ampicillin, and crystalline penicillin after gamma irradiation. The radicals were studied by electron paramagnetic resonance (EPR) spectroscopy using an X-band spectrometer (9.3 GHz). Gamma irradiation was performed at a dose of 25 kGy. One- and two-exponential functions were fitted to the experimental data, in order to assess the influence of the antibiotics’ storage time on the measured EPR lines. After gamma irradiation, complex EPR lines were recorded confirming the presence of a large number of free radicals formed during the irradiation. For all tested antibiotics, concentrations of free radicals and parameters of EPR spectra changed with storage time. The results obtained demonstrate that concentration of free radicals and other spectroscopic parameters can be used to select the optimal parameters of radiation sterilization of β-lactam antibiotics. The most important parameters are the constants τ (τ 1(A),(I) and τ 2(A),(I)) and K (K 0(A),(I), K 1(A),(I), K 2(A),(I)) of the exponential functions that describe free radicals decay during samples storage.  相似文献

A mixture of ten steroids was separated by microemulsion and micellar (SDS and glycodeoxyholate) electrokinetic chromatography systems. Separations were done on a 50 cm (to the detector) × 50 μm I.D. fused-silica capillary. Complete separation of all the test compounds in the micellar mode was obtained with glycodeoxycholate (50 mM) in 25 mM borate buffer, pH 6.5, as the micelle-forming agent. The best results, however, were obtained using microemulsion electrokinetic chromatography in which higher aliphatic alcohols were used as the microemulsion-forming modifiers. The system consisted of n-hexanol (0.81%), SDS (3.31%) and n-butanol (6.61%) in 20 mM phosphate buffer, pH 10.0 (89.28%, w/w). In the microemulsion mode, linear calibration for steroid standards was obtained in the concentration range 3 × 10−4 − 3 × 10−5 mol l−1 with a detection limit of 1 pmol. The method was validated and applied to an 11β-hydroxysteroid dehydrogenase assay in tissues.  相似文献

The success of interpolation techniques relies heavily on the density and regularity of field reference data points. For instance temperature interpolations in the Arctic are hampered by few and scattered meteorological stations. The major objective of this study is to analyze the spatial relationship between plants, defined in terms of an index of thermophily (It) and temperature distribution. The study area is located in Kongsfjorden, northwest Spitsbergen (Svalbard). A systematic recording of floristic data covering the study area was made within quadrates of 1 km × 1 km (93 units). For each of them, the It was calculated. It provides a synthetic measure by which plants are taken as temperature indicators at a long time scale. Temperature values were recorded by means of 39 temperature loggers during the summer 2000. The model for spatial interpolation of temperature was developed using multiple regression of remote sensed data (Landsat TM) and topographical features derived from a digital elevation model (DEM). Continuous temperature layers were calculated at a spatial resolution of 50 m × 50 m, and aggregated to a resolution of 1 km × 1 km in order to correspond with the observed botanical units. Different maps were produced showing spatial distribution of the modelled temperature and It. Correlations between the It and temperature values derived from the modelled temperature layers were systematically explored. Correlation between the It and temperatures works well as standard deviation of residues is 0.7 °C only. Highest correlations (r) of It and the spatial distribution of temperature were obtained for: (a) maximum average temperature for August, excluding all areas higher than 100 m above sea level (0.75), (b) average daily maximum temperature for July–October (0.67), (c) average temperature for July and August (0.64, 0.65), and (d) when temperature range is >8 °C (0.55). Areas with low correlations between It and temperature were mainly attributed to the fact that these measurements represent (a) different time scales and (b) different spatial scales. However, results from this study have shown that calculating It provide a mean for restoring selected temperature parameters and thus can contribute to fill in and extend the network of field data points for temperature interpolation purposes.  相似文献

Uterine fibroid (UFs) affect 77 % of women by menopause and account for $9.4 billion in healthcare costs each year. Although UFs are heritable, genetic risk is poorly understood. The first genome-wide association study (GWAS) of UFs was recently performed in a Japanese population, with reported genome-wide significance for single nucleotide polymorphisms (SNPs) across three chromosomal regions. We tested these SNPs for association with UFs in US cohorts. Women were enrolled in the Right from the Start (RFTS) cohort and the BioVU DNA repository. UF status in both cohorts was determined by pelvic imaging. We tested 65 candidate and haplotype-tagging SNPs for association with UFs presence using logistic regression in RFTS and the top three GWAS-associated SNPs in BioVU. We also combined association results from both cohorts using meta-analysis. 1,086 European American (EA) cases and 1,549 controls were examined. Two SNP associations replicated [blocked early in transport 1 homolog (BET1L) rs2280543, RFTS–BioVU meta-odds ratio (OR) = 0.67 95 % confidence interval (CI) 0.38–0.96, Q = 0.70, I = 0, p = 6.9 × 10?3; trinucleotide repeat containing 6B (TNRC6B) rs12484776, RFTS–BioVU meta-OR = 1.21, 95 % CI 1.07–1.35, Q = 0.24, I = 28.37, p = 8.7 × 10?3). Meta-analyses combining evidence from RFTS, BioVU, and prior GWAS showed little heterogeneity in effect sizes across studies, with meta-p values between 7.45 × 10?8 and 3.89 × 10?9, which were stronger than prior GWAS and supported associations observed for all previously identified loci. These data suggest common variants increase risk for UF in both EA and Japanese populations. However, further research is needed to assess the role of these genes across other racial groups.  相似文献

The enzymological, physical, and immunological properties of soluble and bound forms of intracellular acid carboxypeptidase isolated from fresh mycelia ofAspergillus saitoi are reported. In the broken mycelia, about 60% of the total activity was found in the 2,000×g precipitate, with most of the remainder in the 100,000×g supernantant. The highly purified enzymes, Ia and Ib, from the 100,000×g supernatant were found to be homogeneous by such criteria as disc gel electrophoresis at pH 9.4 The bound enzyme, II, was solubilized from the 2,000×g precipitate by self-digestion at pH 6.4 and was highly purified by chromotography. The two forms of intracellular enzymes, the soluble enzymes (Ia and Ib) from the 100,00×g supernatant and the solubilized enzyme (II) from the 2,000×g precipitate, were closely related to, but not completely identical with, the extracellular acid carboxypeptidase.  相似文献

Pelargonium × hortorum ‘Grand Prix’ which is susceptible to leaf yellowing and ‘Bergpalais’ which is not susceptible to leaf yellowing were chosen for the experiments. Ethylene production and action as well as the associated morphological response of Pelargonium shoots grown in the presence of a precursor of ethylene biosynthesis 1-aminocyclopropane-1-carboxylic acid (ACC), ethylene inhibitors: α-aminooxyacetic acid (AOA) and silver nitrate (AgNO3) and different cytokinins: (meta-topolin) (mT) or 6-benzylaminopurine (BAP) were studied. It was found that ‘Grand Prix’ was more sensitive to ethylene than ‘Bergpalais’ and it showed the leaf yellowing in response to 0.1 mg l?1 ACC. Moreover, it was noted that ACC added separately or together with cytokinin influenced Pelargonium morphogenesis. Depending on the concentration of ACC (0.1–2.0 mg l?1), it either stimulated or inhibited shoot and root formation as well as the growth of shoots and leaf blades. ACC-induced leaf yellowing in ‘Grand Prix’ was effectively inhibited by mT. In contrast, BAP did not enhance shoot quality. Simultaneously, the presence of mT in the medium resulted in up to a twofold increase in the ethylene production by ‘Grand Prix’ shoots throughout the culture period compared with the shoots growing on the BAP-medium. The inhibitor of ethylene action (AgNO3) added with cytokinin prevented the yellowing of Pelargonium shoots, but simultaneously influenced the formation of mature shoots with limited long-term multiplication potential. The shoots of P. × hortorum ‘Grand Prix’ treated with AgNO3 and mT emitted two- and sevenfold more ethylene after 11th and 21st day of culture compared with those treated with AgNO3 and BAP. It is suggested that mT inhibits the early senescence of Pelargonium in vitro by decreasing its sensitivity to ethylene.  相似文献

Livestock are frequently provided nutrient-depleted diets, which can negatively impact animal health and productivity. In our previous trial, we found that iodine (I) supplementation (not selenium (Se)) could increase cashmere production. In order to explore the role of I and Se in cashmere growth, we investigated the effects of dietary I and Se supplementation in Liaoning cashmere goats. Serum thyroid hormone status and the mRNA expression levels of skin monodeiodinase (MDII, MDIII) were measured during the cashmere fiber growth period. Forty-eight 2.5-year-old Liaoning cashmere goats (38.6?±?2.65 kg BW) were divided into six equal groups, and their diets were supplemented with I (0, 2, or 4 mg/kg DM) and Se (0 or 1 mg/kg DM) in a 2?×?3 factorial treatment design. The six treatment groups were: I0Se0, I2Se0, I4Se0, I0Se1, I2Se1, and I4Se1. Concentrations of I and Se in the basal diet (group I0Se0) were 0.67 and 0.09 mg/kg DM, respectively. The trial started in September of 2009 and lasted 70 days. For every measured parameter, supplemental Se had no significant effect on thyroid hormones, but improved the mRNA expression levels of skin MDIII (P?<?0.01). However, supplemental I increased levels of thyroid hormones (thyroxine and triiodothyronine) and improved the mRNA expression levels of skin MDII (P?<?0.05). These results show that the addition of I to cashmere goat feedstock may be an effective means of increasing cashmere production through thyroid hormones regulating the mRNA expression of skin MDII.  相似文献

Polyamine analogues have been studied as potential inhibitors or substrates of barley leaf polyamine oxidase. NH2(CH2)3NH(CH2)10NH2 was particularly effective as an inhibitor of spermine oxidation at pH 4·5 (Ki = 5 × 10?6 M). Methylglyoxal-bis(guanylhydrazone) inhibited spermine oxidation only slightly (Ki = 10?4 M). Activity with the polyamine analogues as substrates was generally 10% or less of the activity with spermine. The Km for oxygen was 3 × 10?4 M. The Km for spermine oxidation was independent of oxygen concentration. Using the N-methyl-2-benzothiazolone hydrazine reagent, 1-(3-aminopropyl)pyrroline was shown to be formed stoichiometrically by the enzyme on oxidation of spermine. The enzyme will not function as a dehydrogenase in the presence of oxygen with either potassium ferricyanide or dichlorophenolindophenol as electron acceptors. Activity in the leaves increased with age, up to 4 weeks. In the leaves of 11-week-old plants activity was lower than in leaves of 1-week-old plants. The enzyme was mainly associated with an easily-sedimented particulate fraction, and relatively small proportions were found in the cell wall or soluble fractions.  相似文献

Offspring born from normal litter size (10 to 15 piglets) but classified as having lower than average birth weight (average of the sow herd used: 1.46 ± 0.2 kg; mean ± s.d.) carry at birth negative phenotypic traits normally associated with intrauterine growth restriction, such as brain-sparing and impaired myofiber hyperplasia. The objective of the study was to assess long-term effects of intrauterine crowding by comparing postnatal performance, carcass characteristics and pork quality of offspring born from litters with higher (>1.7 kg) or lower (<1.3 kg) than average litter birth weight. From a population of multiparous Swiss Large White sows (parity 2 to 6), 16 litters with high (H = 1.75 kg) or low (L = 1.26 kg) average litter birth weight were selected. At farrowing, two female pigs and two castrated pigs were chosen from each litter: from the H-litters those with the intermediate (HI = 1.79 kg) and lowest (HL = 1.40 kg) birth weight, and from L-litters those with the highest (LH = 1.49 kg) and intermediate (LI = 1.26 kg) birth weight. Average birth weight of the selected HI and LI piglets differed (P < 0.05), whereas birth weight of the HL- and LH-piglets were similar (P > 0.05). These pigs were fattened in group pen and slaughtered at 165 days of age. Pre-weaning performance of the litters and growth performance, carcass and meat quality traits of the selected pigs were assessed. Number of stillborn and pig mortality were greater (P < 0.05) in L- than in H-litters. Consequently, fewer (P < 0.05) piglets were weaned and average litter weaning weight decreased by 38% (P < 0.05). The selected pigs of the L-litters displayed catch-up growth during the starter and grower–finisher periods, leading to similar (P > 0.05) slaughter weight at 165 days of age. However, HL-gilts were more feed efficient and had leaner carcasses than HI-, LH- and LI-pigs (birth weight class × gender interaction P < 0.05). Meat quality traits were mostly similar between groups. The marked between-litter birth weight variation observed in normal size litters had therefore no evident negative impact on growth potential and quality of pigs from the lower birth weight group.  相似文献

A chemoenzymatic synthesis was developed for new highly specific fluorogenic substrates for cysteine proteases of the papain family, Abz-Phe-Ala-pNA (I) and Glp-Phe-Ala-Amc (II) (Abz, pNA, Glp, and Amc are o-aminobenzoyl, p-nitroanilide, pyroglutamyl, and 4-amino-7-methylcoumaride, respectively). Substrate (I) was obtained in an aqueous-organic medium using native chymotrypsin. Substrate (II) was synthesized in DMF-MeCN by the treatment with chymotrypsin and subtilisin Carlsberg immobilized on polyvinyl alcohol cryogel. Hydrolysis of substrate (I) with papain, ficin, and bromelain was accompanied by a 15-fold increase in fluorescence intensity, and that of substrate (II), by a change in the fluorescence spectrum. Unambiguity of enzymatic hydrolysis of the substrates after the Ala residue was shown. The specific activity of the substrate hydrolysis with papain, bromelain, and ficin and was determined. Papain showed the greatest activity for both substrates. The activity of all proteases under study was essentially higher for substrate (II), than for substrate (I). The lowest detectable papain concentrations were 2.4 × 10?10 M for (I) and 1.2 × 10?11 M for (II). A high selectivity of cysteine proteases for Glp-Phe-Ala-Amc was established.  相似文献

The effects of nitrogen (N) nutrition on growth, N uptake and leaf osmotic potential of rice plants (Oryza sativa L. ev. IR 36) during simulated water stress were determined. Twenty-one-day-old seedlings in high (28.6 × 10 ?4M) and low (7.14 × 10 4M) N levels were exposed to decreased nutrient solution water potentials by addition of polyethylene glycol 6000. The roots were separated from the solution by a semi-permeable membrane. Nutrient solution water potential was ?0.6 × 105 Pa and was lowered stepwise to ?1 × 105, ?2 × 105, ?4 × 105 and ?6 × 105 Pa at 2-day intervals. Plant height, leaf area and shoot dry weight of high and low nitrogen plants were reduced by lower osmotic potentials of the root medium. Osmotic stress caused greater shoot growth reduction in high N than in low N plants. Stressed and unstressed plants in 7.14 × 104M N had more root dry matter than the corresponding plants in 28.6 × 104M N. Dawn leaf water potential of stressed plants was 1 × 105 to 5.5 × 105 Pa lower than nutrient solution water potential. Nitrogen-deficient water-stressed plants, however, maintained higher dawn leaf water potential than high nitrogen water-stressed plants. It is suggested that this was due to higher root-to-shoot ratios of N deficient plants. The osmotic potentials of leaves at full turgor for control plants were about 1.3 × 105 Pa higher in 7.14 × 10?4M than in 28.6 × 10?4M N and osmotic adjustment of 2.6 × 105 and 4.3 × 105 Pa was obtained in low and high N plants, respectively. The nitrogen status of plants, therefore, affected the ability of the rice plant to adjust osmotically during water stress. Plant water stress decreased transpiration and total N content in shoots of both N treatments. Reduced shoot growth as a result of water stress caused the decrease in amount of water transpired. Transpiration and N uptake were significantly correlated. Our results show that nitrogen content is reduced in water-stressed plants by the integrated effects of plant water stress per se on accumulation of dry matter and transpiring leaf area as well as the often cited changes in soil physical properties of a drying root medium.  相似文献

Stomatal regulation of transpiration was studied in hedgerow coffee (Coffea arabica L.) at different stages of canopy development encompassing a range of leaf area indices (L) from 0·7 to 6·7. Stomatal (gs) and crown (gc) conductance attained maximum values early during the day and then declined as both leaf-to-bulk air water vapour mole fraction difference (Va) and photosynthetically active photon flux density (I) continued to increase. Covariation of environmental variables during the day, particularly V, I, and wind speed (u), obscured stomatal responses to individual variables. This also caused diurnal hysteresis in the relationship between gc and individual variables. Normalization of gs and gc by I removed the hysteresis and revealed a strong stomatal response to humidity. At the crown scale, transpiration (E) increased linearly with net radiation (Rn) and seemed to increase with increasing wind speed. Increasing wind speed imposed higher leaf interior to leaf surface water vapour mole fraction differences (Vs) at given levels of Va. However, strong relationships between declining gc and E and increasing wind speed were obtained when gc and E were normalized by I and Rn, respectively, without invoking additional potential interactions involving temperature or CO2 concentration at the leaf surface. Apparent stomatal responses to wind were thus at least partially a reflection of the stomatal response to humidity.  相似文献
