Adult epicuticular hydrocarbon profiles of male and femaleDrosophila mojavensis have been implicated as determinants of mate choice leading to premating isolation between geographically isolated populations. Hydrocarbon profiles of a Baja California and a mainland Sonora population ofDrosophila mojavensis, ayellow body mutant strain ofD. mojavensis, and a population ofD. arizonae were compared among flies that had been reared on two cactus substrates and a synthetic laboratory growth medium in order to assess the degree to which natural rearing substrates influence adult hydrocarbon composition. Twenty epicuticular hydrocarbon components, ranging from C29 to C41, were recovered by gas chromatography that represented major classes of alkanes, alkenes, and alkadienes. We found differences in relative amounts of epicuticular hydrocarbons among Baja and mainlandD. mojavensis, and theyellow body mutants. There were few differences betweenD. mojavensis andD. arizonae. The effects of rearing substrates were remarkable: most of the differences were due to the effects of lab food vs. cactus, but there were significant rearing substrate effects due to differences in the two cacti used. Eleven hydrocarbon components differed in abundance between males and females or showed significant sex × rearing substrate interactions from ANOVA. The effects of rearing substrates on epicuticular hydrocarbon composition inD. mojavensis are concordant with changes in the intensity of premating isolation between populations, implicating host ecology as a major determinant in patterns of mate choice in this species.  相似文献

Epicuticular hydrocarbon variation was investigated among the three species of the Drosophila mojavensis cluster (D. mojavensis, D. arizonae, and D. navojoa) within the large D. repleta group. Because these hydrocarbons serve as contact pheromones in adult D. mojavensis, the chemical characteristics and differences in hydrocarbon profiles in populations of these three sibling species were further investigated. Twenty-seven hydrocarbon components with chain lengths ranging from C28 to C40, including n-alkanes, methyl-branched alkanes, n-alkenes, methyl-branched alkenes, and alkadienes were observed. Hydrocarbon profiles among the three species reared on different cactus hosts were easily aligned with previously identified components in D. mojavensis. Male and female D. navojoa possessed a 31-methyldotricont-6-ene absent in both D. arizonae and D. mojavensis, while lacking the 8,24-tritricontadiene present in these two species. D. navojoa adults had far less 2-methyloctacosane than these sibling species, but the significance of this difference was obscured by the degree of variation among populations in amounts of this hydrocarbon. Mainland and Baja California populations of D. mojavensis were fixed for differences in the amounts 8,24-tritricontadiene, 9,25-pentatricontadiene, and 9,27-heptatricontadiene, consistent with all previous studies. Amounts of 18 of the 27 hydrocarbon components were greater in flies reared on Opuntia cactus. Canonical discriminant function analysis resolved all three species into distinct, nonoverlapping groups, suggesting that epicuticular hydrocarbon profiles are species-specific in the D. mojavensis cluster. Based on the amounts of interpopulation variation in hydrocarbon profiles in these three species, we hypothesize that epicuticular hydrocarbon differences may evolve early during the formation of new species.  相似文献

Drosophila mettleri is a soil-breeding, cactophilic drosophilid which lives in the Sonoran Desert. Several chemical constituents of cacti in this region have been identified as having major roles in insect-host plant relationships involvingDrosophila. For example, isoquinoline alkaloids, which are present in senita cactus, have been shown to be toxic to seven of the nine species tested. The two tolerant species areD. pachea, the normal resident, andD. mettleri. Necroses of senita cacti are often used as feeding substrates byD. mettleri adults, but this species has never been reared from senita rots. Soil, which have been soaked by juice from saguaro and cardón rots, are the typical breeding substrates of this species. The tissues of both of these cacti also contain alkaloids, chemically related to those in senita, but at much lower concentrations. Alkaloid concentration in saguaro-soaked soil was found to be 1.4–27 times the average concentration in fresh tissue. Alkaloids were extracted from saguaro tissue and used in tests of larva-to-adult viability, developmental rate, and adult longevity. Elevated concentrations of saguaro alkaloids had no significant effect on the longevity ofD. mettleri, but significantly reduced the longevity ofD. nigrospiracula andD. mojavensis, two nonsoil breeding cactophilic species. Viability and developmental rates of all three species were affected, but the effect onD. nigrospiracula was comparatively greater. It is argued that the adaptations that allowD. mettleri to utilize the saguaro soil niche also convey tolerance to alkaloids present in senita tissue. The ability to utilize senita necroses as feeding substrates represents an ecological advantage to D. mettleri, in that the density of potential feeding sites is increased as compared to species which are more specific in their host-plant relationships.  相似文献

When cultures ofPhytophthora cactorum grown on sterol-free medium were supplemented with a mixture of cholesterol and sitosterol, the sitosterol was initially taken up more rapidly than the cholesterol. Cholesterol was preferentially incorporated into the free sterol fraction and sitosterol into esters. More polar material was formed from sitosterol. Sitosterol is more active than cholesterol in promoting reproduction; this is attributed to the greater amount of polar metabolites formed from it.  相似文献

The mushroom genus Amanita has a spectrum of chemical compounds affecting survival and performance of animals. Ibotenic acid is one of such compounds found in some Amanita mushrooms. We studied the effects of ibotenic acid and its derivative, muscimol, on egg-to-pupa survival, pupation time, and pupal size in five Drosophila species (Diptera: Drosophilidae), Drosophila bizonata, Drosophila angularis, Drosophila brachynephros, Drosophila immigrans, and Drosophila melanogaster. The first three species are mycophagous and use a wide range of mushrooms for breeding, whereas D. immigrans and D. melanogaster are frugivorous. We reared fly larvae on artificial medium with 500, 250, 125, and 62.5 μg/ml of ibotenic acid and/or musimol. The three mycophagous species were not susceptible to ibotenic acid, whereas the two frugivorous species were affected. In experiments with D. melanogaster, muscimol was less toxic than ibotenic acid.  相似文献

Contribution of different gut segments to plant sterol absorption, adaptation of plant sterol absorption after partial small bowel resection, and effects of gut transplantation (necessitates extrinsic autonomic denervation and lymphatic disruption) on plant sterol biodynamics are unclear. We studied the consequences of massive proximal small bowel resection and autotransplantation of the remaining ileum on the adaptive absorption and biodynamics of plant sterols. Dietary, fecal, biliary, hepatic and plasma plant sterols, fecal elimination and absorption of cholesterol, small bowel morphology, and intestinal transit were determined before (n=5) and at 4, 8, and 14 wk after resection of the proximal 75% of the jejunoileum (n=15) and autotransplantation of the remaining ileum (n=15) or transection (n=5). Proximal gut resection significantly reduced cholesterol absorption efficiency; percentage absorption and biliary secretion of plant sterols; plasma, biliary and hepatic campesterol-to-cholesterol proportions; and sitosterol proportions in plasma and bile. Autotransplantation of the remaining ileum further significantly decreased cholesterol absorption efficiency; percentage absorption and biliary secretion of campesterol; campesterol proportions in plasma, bile and liver; and plasma proportions of sitosterol while increasing fecal excretion of neutral and acidic steroids. Plasma proportions of the two plant sterols, but absorption of just campesterol, were gradually improved with increasing cholesterol absorption and villus height after proximal gut resection; the same result was observed to a lesser degree after ileal autotransplantation. In addition, significant positive correlations were found between percentage cholesterol and campesterol absorption and the plasma plant sterol proportions in both proximal resection groups, between campesterol absorption and ileal villus height in the resection group, and between campesterol absorption and intestinal transit time in the autotransplantation group. In conclusion, plasma campesterol and sitosterol closely reflect absorption of cholesterol and plant sterols from intact and autotransplanted ileum during adaptation to proximal gut resection. A loss of proximal gut absorptive surface impairs cholesterol and campesterol absorption more than sitosterol absorption, the later being apparently less dependent on available jejunal villus surface area.  相似文献

The Cactus-Drosophila Model System of the Sonoran Desert consists of four endemic species ofDrosophila (D. mojavensis, D. nigrospiracula, D. mettleri andD. pachea) and five species of columnar cacti (agria, organpipe, saguaro, cardón and senita). Extensive collection records indicate that each cactus species has only one species ofDrosophila as the primary resident. The elimination of six of the twenty possible random combinations ofDrosophila species and cactus species can be attributed directly to phytosterols.Drosophila pachea has a strict requirement for Δ7-sterols such as 7-cholestenol and 7-campestenol. Since Δ7-sterols are found only in senita cactus,D. pachea cannot use agria, organpipe, saguaro or cardón as host plants. The lipid fractions of agria and organpipe are chemically similar and contain high concentrations of several 3β,6α-dihydroxysterols. Larval viability tests using chemical constitutents of organpipe cactus demonstrate that the sterol diols are toxic toD. nigrospiracula but not to the resident, species,D. mojavensis. Agria and organpipe are therefore unsuitable as host plants forD. nigrospiracula. These results suggest that phytosterols play a major role in determining host plant utilization by cactophilicDrosophila in the Sonoran Desert. Deceased.  相似文献

(Z)-11-Octadecenyl acetate (Z11–18Ac) was identified as the aggregation pheromone ofDrosophila mauritiana, D. yakuba, andD. rajasekari. The amount of pheromone in the ejaculatory bulb of male flies increased with age, reaching plateau levels of ca. 240, 800, and 1100 ng/fly forD. mauritiana, D. yakuba, andD. rajasekari, respectively. Thirty to 50% of the pheromone in the ejaculatory bulb was transferred to the female during mating, with the majority transferred to the female's reproductive tract. In the subsequent 6-hr period, over half the pheromone in the female's reproductive tract was transferred to the surroundings. In a wind-tunnel olfactometer,Z11–18Ac was attractive toD. yakuba andD. mauritiana; however,D. rajasekari required food or food odors in combination withZ11–18Ac to demonstrate aggregation activity.Z11–16Ac andZ11–20Ac were not attractive forD. mauritiana, D. yakuba, andD. rajasekari. Low concentrations of dietary triacylglycerols including tristearin, triolein, and tripalmitolein, were assessed to determine their effects during egg to pupal stages on adult epicuticular hydrocarbon (EHC) variation in cactophilic Drosophila mojavensis. Triacylglycerols were added singly and in combination at concentrations of 1%, 3%, and 9% to a lipid-free culture medium. Control diets included Carolina Drosophila and lipid-free media. Egg to adult viability was reduced at triacylglycerol concentrations greater than 1%, except for tristearin. Both triolein and tripalmitolein increased EHC amounts to levels similar to those in combination and control diets. Tristearin caused significantly lower quantities of EHCs in adult flies than triolein or tripalmitolein, consistent with previous studies on reduced tristearin assimilation into adult EHCs. We rejected the hypothesis that unsaturated and saturated triacylglycerols were assimilated into unsaturated and saturated adult EHCs, respectively. Since these triacylglycerols comprise a fraction of known lipids in the columnar cacti used for breeding in nature, and EHCs serve as contact pheromones in D. mojavensis, these and other naturally occurring triacylglycerols may provide a direct causal link between host plant use and patterns of chemically mediated mate choice. Females of severalAttagenus species demonstrated calling behaviors similar to those previously reported forA. elongatulus. Attagenus rufipennis females did not call until they were 12–14 days old. OtherAttagenus species called at approx. 4 days of age. All species exhibited diurnal periods of calling activity. The cytochrome P-450 monooxygenase system has been implicated in plant utilization by at least three species ofDrosophila (D. nigrospiracula, D. mettleri, andD. mojavensis) that are endemic to the Sonoran Desert of the southwestern United States and northwestern Mexico. Basal and induced levels of total cytochrome P-450 were determined for third-instar and decapitated 2- to 5-day post eclosion adults of the three desert species. Total P-450 levels, both basal and induced for all species assayed, were significantly higher for adults than for larvae by up to 20-fold. On a per organism basis, the levels of in vitro metabolism of the cactus alkaloid, carnegine, and patterns of response to induction by cactus tissue for adult desertDrosophila approximated those of larvae. Induction by phenobarbital, however, resulted in levels of in vitro carnegine metabolism that were up to 5.6-fold higher in adults than in larvae.  相似文献

The honey bee,Apis mellifera, does not convert C28 and C29 phytosterols to cholesterol as found in most previous studies of phytophagous or omnivorous insects, but instead the workers and queens selectively transfer 24-methylenecholesterol, sitosterol and isofucosterol from their endogenous sterol pools to the brood larvae regardless of the sterol in the worker diet. Administering radiolabeled sterols by feeding and injection has made it possible to trace this selective transfer through a second generation of the honey bee. In further comparative sterol metabolism studies, the yellow fever mosquito,Aedes aegypti, was shown to be capable of dealkylating and converting a radiolabeled C29 dietary sterol ([14C] sitosterol) to cholesterol. Metabolic studies with several radiolabeled dietary sterols and an inhibitor of steroid metabolism in the yellow fever mosquito further verified this capability.  相似文献

The four stereoisomers of 8-methyl-2-decyl propanoate were tested in the United States and Mexico for attractiveness toDiabrotica virgifera virgifera LeConte, the western corn rootworm,D. v. zeae Krysan and Smith, the Mexican corn rootworm, andD. porracea Harold. Males ofD. v. virgifera andD. v. zeae responded strongly to the (2R,8R)-isomer and secondarily to (2S,8R), whileD. ponacea responded exclusively to the (2S,8R)-isomer. Matsunone, (2E,4E)-4.6,10,12-tetramethyl-2,4-tridecadien-7-one, is the primary sex pheromone ofMatsucoccus resinosae, M. thunbergianae, andM. matsumurae. The synthetic compound was tested in the laboratory and in field tests with all three species, and significantly more males responded to synthetic matsuone than to controls. The attraction of synthetic matsuone to males in field tests was not significantly different from that of virgin females at the same matsuone release rate. An analog of matsuone previously shown to be biologically active was also tested in the laboratory withM. resinosae andM. thunbergianae, and field tested withM. resinosae. The honey bee,Apis mellifera, is one of only a few species of phytophagous insects known to be unable to convert C-24 alkyl phytosterols to cholesterol. Regardless of the dietary sterols available to worker bees, the major tissue sterol of brood reared by the workers is always 24-methylenecholesterol, followed by sitosterol and isofucosterol. Normally, little or no cholesterol is present in honey bee sterols. The maintenance of high levels of certain sterols is accomplished through a selective transfer of sterols from the endogenous sterol pools of the workers to the developing larvae through the brood food material secreted from the hypopharyngeal and mandibular glands and/or the honey stomach of the workers. The selective uptake and transfer of radiolabeled C27, C28 and C29 sterols have been studied to correlate these aspects of sterol utilization with the discovery of an unusual molting hormone (ecdysteroid) in honey bee pupae as the major ecdysteroid of this stage of development. The phylogenetic implications of this selective transfer phenomenon in the honey bee and comparison with sterol metabolism in certain other hymenopteran species emphasize the diversity of steroid biochemistry in insects.  相似文献

Behavioral analysis of male attraction to stereoisomers of 8-methyl-2-decyl propanoate was conducted withDiabrotica virgifera virgifera andD. barberi using a flight tunnel.D. barberi males were attracted to the 2R,8R isomer only, and the response was inhibited by the addition of 2S,8R.D. v. virgifera males were also attracted to the 2R,8R isomer and were neither attracted nor inhibited by any of the other isomers. Males attracted to the lures displayed a series of behaviors characterized by a slow hovering flight upwind towards the odor source. While significant numbers of males of both species landed on the pheromone lure, none attempted copulation, indicating that other cues are necessary to induce this behavior.  相似文献

The effect of tomatine on larval growth ofHeliothis zea andSpodoptera exigua was assessed by rearing larvae on diets with different concentrations of the chemical added. When reared from neonates, linear dose-response relationships were obtained for both species, withS. exigua being three times more sensitive to tomatine thanH. zea. Tomatine toxicity was completely alleviated inH. zea by the addition of equimolar cholesterol into the diet; however, inS. exigua some toxicity was maintained. Larvae ofS. exigua that were started on control diet were insensitive to tomatine after five days; larvae started on diet with an EC50 of tomatine and then switched to control diet after five days failed to recover from toxicosis. Larval growth ofH. zea, on the other hand, was affected both at the neonate and third-instar stage, but normal growth resumed when the larvae were transferred to control diet. Tomatine had little or no affect on food consumption, assimilation, or dietary utilization of the food by third-instar larvae ofS. exigua, except at a concentration 10 times the EC50. In contrast, the efficiency of food utilization ofH. zea larvae decreased with increasing tomatine concentrations. Assimilation of the food tended to increase, although not significantly, as tomatine levels increased. Food consumption ofH. zea larvae also increased when the tomatine concentration was greater than an EC50. The addition of equimolar cholesterol to diets with an EC50 of tomatine restored weight gain and nutritional indices values to control values. These results are related to the utility of using tomatine in host-plant resistance programs.  相似文献

The metabolism of various dietary sterols and the effects of an azasteroid on sitosterol metabolism in the free-living nematodeCaenorhabditis elegans was investigated. The major unesterified sterols ofC. elegans in media supplemented with sitosterol, cholesterol or desmosterol included 7-dehydrocholesterol (66.5%, 40.5%, 31.2%, respectively), cholesterol (6.7%, 52.3%, 26.9%), lathosterol (4.4%, 3.6%, 1.7%) and 4α-methylcholest-8(14)-en-3β-ol (4.2%, 2.1%, 3.8%). Esterified sterols, representing less than 20% of the total sterols, were somewhat similar except for a significantly higher relative content of 4α-methylcholest-8(14)-en-3β-ol (23.3%, 23.4%, 10.6%). ThusC. elegans not only removes the substituent at C24 of dietary sitosterol but possesses the unusual ability to produce significant quantities of 4α-methylsterols. WhenC. elegans was propagated in medium supplemented with sitosterol plus 5 μg/ml of 25-azacoprostane hydrochloride, the azasteroid strongly interfered with reproduction and motility ofC. elegans and strongly inhibited the Δ24-sterol reductase enzyme system; excluding sitosterol, the major free sterols of azacoprostane-treatedC. elegans were cholesta-5, 7, 24-trien-3β-ol (47.9%), desmosterol (9.4%), fucosterol (2.1%) and cholesta-7,24-dien-3β-ol (2.0%). These 4 sterols are likely intermediates in the metabolism of sitosterol inC. elegans.  相似文献

Heliothis zea (corn earworm), an insect that fails to synthesize sterols de novo, was reared on an artificial diet treated with 18 different sterol supplements. Larvea did not develop on a sterol-less medium. Δ5-Sterols with a hydrogen atom, a methylene group, an E-or Z-ethylidene group, or an α- or β-ethyl group (cholesterol, ostreasterol, isofucosterol, fucosterol, sitosterol, and clionasterol, respectively) at position C-24, and Δ5-sterols doubly substituted in the side chain at C-24 with an α-ethyl group and at C-22 with a double bond (stigmasterol) supported normal larval growth to late-sixth instar (prepupal: maturity). The major sterol isolated from each of these sterol treatments was cholesterol, suggesting that H. zea operates a typical 24-dealkylation pathway. The sterol requirement of H. zea could not be met satisfactorily by derivatives of 3β-cholestanol with a 9β, 19-cyclopropyl group, gem dimethyl group at C-4, a Δ5,7-bond or Δ8-bond, or by side-chain modified sterols that possessed a Δ25(27)-24β-ethyl group, Δ23(24)-24-methyl group, or 24-ethyl group, or Δ24(25)-24-methyl or 24-ethyl group. The major sterol recovered from the larvae (albeit developmentally arrested larvae) treated with a nonutilizable sterol was the test compound. Sterol absorption was related to the degree of sterol utilization. The most effective sterols absorbed by the insect ranged from 27 to 66 μg per insect, whereas the least effective sterols absorbed by the insect ranged from 0.6 to 6 μg per insect. Competition experiments using different proportions of cholesterol and 24-dihydrolanosterol (from 9:1 to 1:9 mixtures) indicated that abnormal development of H. zea may be induced on less than a 1 to 1 mixture of utilizable (cholesterol) to nonutilizable (24-dihydrolanosterol) sterols. The results demonstrate new structural requirements for sterol utilization and metabolism by insects, particularly with respect to the position of double bonds in the side chain and functionalization in the nucleus. The novel sterol specificities observed in this study appear to be associated with the dual role of sterols as membrane inserts (nonmetabolic) and as precursors to the ecdysteroids (metabolic).  相似文献

The ripe fruit ofMorinda citrifolia, host plant forDrosophila sechellia is highly toxic for three closely related species (D. melanogaster, D. simulans, andD. mauritiana). Green and rotten fruits are not toxic for all species tested. Short fatty acids were found to be present in large quantities in the extract of the ripe fruit. The most abundant (octanoic acid) was tested pure for its toxicity in a dose-dependent manner;D. sechellia is five to six times more resistant thanD. melanogaster to octanoic acid. Octanoic acid alone seems to be sufficient to explain the toxic effect of the pulp. It is less abundant in the rotten fruit and absent in the green fruit.  相似文献
