测定大口鳒Psettodes erumei线粒体控制区全序列,并与其他7种鲽形目鱼类控制区结构进行比较分析.结果表明,大口鳒线粒体控制区全长大约1601bp,5'端和3'端分别存在56bp和8bp的串联重复序列.除此之外,其控制区结构和其他种类相似,分为终止相关序列区(TAS)、中央保守区(CSB-D、CSB-E、CSB-F)、保守序列区(CSB-1、CSB-2、CSB-3),同时在CSB-D后还识别出鲽形目保守的Poly-T结构.比较了8种鲽形目鱼类控制区全序列和去除重复序列后控制区序列的碱基组成变化,结果显示前者的种间差异明显大于后者,变异系数约是后者的两倍.这种差异主要是由于重复序列的异质性如长度及其拷贝数不同、碱基组成不同所造成的.基于控制区部分序列对鲽形目12种鱼类进行系统分析,结果显示,在鲽形目内部大口鳒分类地位最原始;鲽亚目先与鳎亚目聚在一起,后与鳒亚目聚在一起,这个结果与基于形态特征的系统关系相符.  相似文献

采用长片段扩增策略,获得鲈形目石首鱼科黄鱼属小黄鱼的线粒体基因组全序列。组装分析结果表明,其长度为16470bp,H链碱基组成呈明显反G偏倚,G含量仅为16%。13个蛋白质基因起始密码子均为ATG。COI基因的终止密码子为AGA,ND1和ND3基因为TAG,COII、ND4和Cytb基因为不完全终止密码子T,其余7个基因都是完整的TAA。在小黄鱼mtDNA中发现2个特殊发夹结构,一个是轻链复制起始区(OL)的发夹环,位于tRNA簇,环区T含量仅为9%,而G含量达45%,这与哺乳动物和爪蟾该区域富含T有所不同。另一个假定发夹结构位于ATP6基因末端,这种二级结构可能与准确转录有关。该mtDNA控制区长度为799bp,5′端含有9个连续TA串联重复和2个5′-ATGTA---TACAT-3′重复,与已报道的鱼类扩展终止相关序列模式恰恰相反;中央控制区仅识别出CSB-E结构;保守序列区含有CSB-1,2,3,其中CSB-2具有高度保守性。对小黄鱼和大黄鱼的mtDNA比较进化研究,根据分子钟假说,估计二者线粒体基因组的进化分歧时间为4.8百万年前。  相似文献

黄渤海松江鲈鱼线粒体控制区结构与序列多态性分析   总被引:3,自引:0,他引:3  
测定了辽宁丹东、盘锦与河北秦皇岛35尾松江鲈鱼线粒体控制区序列977bp。共检出单倍型30个,多态位点35个,简约信息位点27个,绝大多数变异集中在192-342bp、594-783bp区段,同时识别出了线粒体控制区5’端终止序列区、中央保守区和3’端保守序列区的关键序列。3个群体总的单倍型多样性为0.990,核苷酸多样性为0.00718,丹东群体核苷酸多样性最高(0.00959)。群体间、群体内平均遗传距离低(分别为0.0067和0.0063);在NJ树上不同地理来源的个体混杂分布;Fst分析显示丹东与秦皇岛群体间遗传分化相对较大(0.14222,p0.05),其余不明显(0.02865-0.05616,p0.1),表明3个群体有可能隶属同一个种群。核苷酸不配对分布图表明松江鲈鱼可能未经历过大规模的种群扩张。  相似文献

本研究基于线粒体DNA分子标记NADH脱氢酶亚基1基因(ND1)部分序列分析了印度-西太平洋海域的胡椒鲷属及少棘胡椒鲷属共16种胡椒鲷鱼类的系统进化关系。采用最大简约法及最大似然法构建了系统进化树。进化树上,16种胡椒鲷鱼类共形成两个形态特征截然不同的类群,其中类群一的种类体表具有各种鲜艳的颜色与多变的斑纹,主要分布在西太平洋地区;类群二的种类颜色灰暗单一,少数体表具有暗色斑点与条纹,大多数分布在印度洋地区。此外,进化树上,少棘胡椒鲷属的种类都位于胡椒鲷属的类群一内部,与斑胡椒鲷,暗点胡椒鲷形成的姐妹种有较近的亲缘关系。基于遗传距离的数据,少棘胡椒鲷属与胡椒鲷属间的平均遗传距离小于胡椒鲷属内部种间的遗传距离,显示出少棘胡椒鲷属与胡椒鲷属有非常近的亲缘关系,支持少棘胡椒鲷归类于胡椒鲷属的观点。  相似文献

本研究测定双线舌鳎Cynoglossus bilineatus (Lacepède,1802)、长钩须鳎Parapla gusia bilineata (Bloch,1787)和短钩须鳎Paraplagusia blochii( Bleeker,1851)3种舌鳎亚科鱼类的线粒体(mtDNA)控制区全序列,并与舌鳎亚科其它4种及鲽形目其他科的5种鱼类mtDNA控制区结构和序列进行了比较分析.结果显示,3种舌鳎鱼类的线粒体控制区全长从655~1 155 bp,长度差异明显;舌鳎亚科鱼类控制区结构与其他鲽形目鱼类相似性很低,保守区系列中只能识别出CSB-A和TAS.碱基组成比较发现这3种舌鳎鱼类的AT含量比鲽形目其它鱼类要高.分析认为上述事件可能都是由该类群鱼类控制区的快速进化导致的.所有3种舌鳎的控制区在5'端都存在串联重复序列,每个重复单元中都存在类似于TAS的相关序列,并且重复序列可以形成非常稳定的二级结构.这2种特征为形成线粒体重复序列的非正常延伸机制(Illegitimate elongation model)提供了新的证据,同时,推测了这些舌鳎鱼类重复序列可能的延伸过程.  相似文献

利用PCR技本扩增了6个野生地理群体大泷六线鱼(Hexagrammos otakii)线粒体DNA控制区的部分序列,共获得352bp碱基序列,31尾个体定义了22种单倍型.中性检验Fu's Fs值为-0.14881(P<0.01).分子变异分析(AMOVA)结果显示:遗传分化指数Fst=0.7398(P<0.01),73.98%的变异来自群体间,26.02%的变异来自群体内.根据不同个体间的遗传距离构建了NJ和MP系统树,两种方法获得了相似的进化树.其中青岛群体与旅顺群体的一部分个体单独成支,其它群体聚为1支.研究结果表明,大泷六线鱼群体遗传多样性较为丰富,群体间发生了较大的遗传分化,证明线粒体DNA控制区基因可用于大泷六线鱼群体内及群体间遗传多样性的分析.  相似文献

利用常规 PCR 未纯化产物直接测序的方法,获得紫红笛鲷(Lutjanus argentimaculatus)全长为16543 bp 线粒体 DNA 全序列, GenBank 登录号为 JN182927.结合 GenBank 中的已有数据比较分析表明:紫红笛鲷与笛鲷属(Lutjanus)线粒体基因组成基本一致,符合脊椎动物的特征,各基因间进化速率差异显著;13个蛋白编码基因拼接序列贝叶斯建树的后验率显著高于单基因或全序列,更适用于笛鲷属的贝叶斯法系统分化分析和年代推断;13个蛋白编码基因及其拼接序列进行的系统进化分析均支持紫红笛鲷与勒氏笛鲷(Lutjanus russellii)及海鸡母笛鲷(Lutjanus rivulatus)亲缘关系较近;基于13个蛋白编码基因拼接序列的贝叶斯松散分子钟推算出笛鲷属鱼类两个主要支系大约在61.5Ma 前古新世的达宁阶与蒙蒂阶交界时期发生分化.  相似文献

为了解杂色龙虾(Panulirus versicolor)线粒体基因组多态位点及遗传变异,对龙虾类的分类研究奠定基础,作者通过常规PCR步移扩增法获得杂色龙虾线的线粒体基因组全序列,分析了其线粒体基因组的基本特征以及基因排列情况,并结合中国龙虾(Panulirus stimposni)、波纹龙虾(Panulirus homarus)线粒体基因组全序列,综合分析了龙虾属线粒体编码基因多态位点及基因变异状况。结果表明:杂色龙虾线粒体基因组全序列为15 700 bp,由13个蛋白质编码基因、22个t RNA基因、2个r RNA基因和D-loop区组成,保持了泛甲壳动物线粒体基因组的原始排列。杂色龙虾线粒体DNA的碱基组成具有AT偏好性,A+T含量为67%,4种碱基的含量分别为(A=34.6%,T=32.1%,C=20.5%,G=12.8%)。预测了杂色龙虾18个t RNA以及2个r RNA的二级结构。龙虾属线粒体基因组中的nad 2、nad 5基因以及D-loop区的多态位点比例较高,适合作为分子标记用于龙虾类系统进化关系、种内多态性以及遗传分化等方面的研究。  相似文献

肌肉生长抑制素(MSTN)基因对肌肉生长起负调控作用,在动物育种上具有潜在的应用前景.报道了眼斑拟石首鱼(Sciaenops ocellatus)的MSTN基因的序列结构特征及其组织特异性表达.MSTN基因具有3个外显子和2个内含子,编码376个氨基酸.在内含子Ⅰ和3'-UTR区内,分别发现了(TC)4CC(TC)4CT(TC)4和(AC)7微卫星序列.推断的眼斑拟石首鱼的MSTN氨基酸序列具很强的保守性,与石鼓鱼、金鲷、条纹石鮨、白鲈、金眼石鮨之间的相似性在90%以上.在结构上,眼斑拟石首鱼的MSTN包含N端信号序列(1~22氨基酸残基),TGF-β前肽域(41~256氨基酸残基),RXXR蛋白水解位点(264~267位的RARR)和TGF-β区域(282~376氨基酸残基).在检测的10种组织中,MSTN只在眼斑拟石首鱼的肌肉、脑和眼组织中表达,其中在肌肉表达最强.  相似文献

Determining the nursery habitat of fishes that have moved from estuarine nursery habitats is difficult. The elemental fingerprints of otoliths of three species of sparids were determined to investigate their utility as a natural tag of the nursery habitat. Juvenile Pagrus auratus (snapper), Rhabdosargus sarba (tarwhine) and Acanthopagrus australis (bream) were collected from two sites in each of 15, six and three estuaries, respectively, and their otoliths analysed by solution-based inductively coupled plasma-mass spectrometry. Significant differences in otolith chemistry were found for all three species of juveniles collected from different estuaries. The same patterns among estuaries were not seen for all species, although it was not possible to sample the same sites within an estuary for all species. For bream, significant differences in otolith chemistry were found among all three estuaries, whereas for tarwhine the six estuaries were separated into three groups. For snapper, a number of estuaries could be separated, but there was some overlap for other estuaries. All three species were collected from the same site within one estuary and their otoliths analysed. Significant differences were found among species, but the implication of this finding remains unclear as the three species show differences in microhabitat use and may also differ in age. Because the elemental fingerprints of juveniles vary among estuaries or groups of estuaries, the nursery or recruitment estuary of adult fish could now be determined by analysing the juvenile region of adult otoliths. Thus, connectivity between estuaries and open coastal populations could be determined. Such information will have major implications for fisheries management because it will provide information on the distance that fish have moved from their recruitment estuary and the number of estuaries that contribute to each adult population.  相似文献

Snapper (Chrysophrys auratus) is an important coastal fish species in New Zealand for a variety of reasons, but the large amount of research conducted on snapper has not been reviewed. Here, we review life history information and potential threats for snapper in New Zealand. We present information on snapper life history, defining stages (eggs and larvae, juvenile and adult), and assess potential threats and knowledge gaps. Overall we identify six key points: 1. post-settlement snapper are highly associated with certain estuarine habitats that are under threat from land-based stressors. This may serve as a bottleneck for snapper populations; 2. the largest knowledge gaps relate to the eggs and larvae. Additional knowledge may help to anticipate the effects of climate change, which will likely have the greatest influence on these early life stages; 3. ocean acidification, from land-based sources and from climate change, may be an important threat to larval snapper; 4. a greater understanding of population connectivity would improve certainty around the sustainability of fishery exploitation; 5. the collateral effects of fishing are likely to be relevant to fishery productivity, ecosystem integrity and enduser value; 6. our understanding of the interrelationships between snapper and other ecosystem components is still deficient.  相似文献

Feeding was examined in 772 snapper, Chrysophrys auratus (Bloch and Schneider), from the western Bay of Plenty, New Zealand. Occurrence, points (bulk), and numbers methods of food analysis were used in the study. Diet was extremely varied, and, by all methods of food analysis, crustaceans, polychaetes, echinoderms, molluscs, and teleosts formed the main snapper food groups, with crustaceans being the most important. Size of snapper, depth, sampling area, and time of day affected the occurrence, volume, and numbers of food items, while sex did not.

Seasonal feeding patterns on most species/categories of food varied locally within each sampling area, and there were no uniform seasonal feeding patterns for any food/category in the four sampling areas examined.  相似文献

Anecdotal data sources may constitute an important component of the information available about an exploited species, as record keeping may not have occurred until after exploitation began. Here, we aimed to fill any gaps in the exploitative history of the sparid snapper (Pagrus auratus), using social and historical research methods. Social research consisted of interviews with recreational fishers, focusing on the most and largest snapper they had caught. In addition, the diary‐logs of two recreational fishers were analysed. Historical research consisted of investigation of old books, photos, archives and unpublished sources unconventional to fishery science. Interviews with fishers demonstrated no or weak trends in snapper abundance or size, and were likely impeded by a lack of ability to detect change in a fish stock that may still be considered abundant. The fishers’ perception of change, however, largely reflected recent experiences (last c. 10 years), when biomass is understood to have increased, and mostly did not consider experiences before the 1980s. Alternatively, diary‐logs of fisher catch rates produced a pattern that matched formal stock assessments of snapper biomass, suggesting declines in abundance up until the 1990s and an increase in biomass after that time. Historical research, although more qualitative, had the ability to investigate periods where formal records were not kept and described a fishery vastly different from the current one. Snapper were easily caught, in great abundance and in unusual locations. Localised depletion of snapper was first noticed in the early 20th century, despite spectacular catches of snapper occurring after that time. Snapper behaviour was also likely different, with visual sightings of snapper by onlookers a common occurrence. Although predictions from stock assessment models are consistent with that of the anecdotes listed here (i.e., high biomass in the past), these anecdotes are valuable as they explain lost biomass in a perspective meaningful to all. This perspective may be valuable for managers trying to consider the non‐financial value of a shared fishery but, if unrecognised, represents a shifting baseline.  相似文献

The potential of the first line of the active oxygen-scavenging system, partial cDNA encoding Cu/Zn superoxide dismutase (SOD) was isolated in three aquatic mollusc species: Ruditapes decussatus (marine clam), Dreissena polymorpha (continental water mussel) and Bathymodiolus azoricus (hydrothermal vent mussel). These SOD cDNA fragments were amplified by PCR with degenerate oligonucleotide primers derived from the amino acid sequence conserved in the Cu/Zn-SOD from several other organisms. A partial cDNA of CuZn-SOD was obtained for R. decussates (510 bp), D. polymorpha (510 bp) and B. azoricus (195 bp). The deduced amino acid sequence showed high similarity among the three mollusc species (57-63%) and among other species (50-65%). The residues involved in coordinating copper (His-47, 49, 64, 121) and zinc (His-64, 72, 81 and Asp-84) were well conserved among the three Cu/Zn-SOD sequences.  相似文献

Many fish species rely on estuarine nursery habitats as they transition to adult life stages. Quantifying nursery value, however, requires identification of the life stages (often small and short-lived) that utilise nursery habitats, and survey methods that provide comparable estimates across habitats. We focused our surveys on post-settlement snapper (Chrysophrys auratus) using video camera deployments across habitats within a northeastern New Zealand harbour. Post-settlement snapper abundance was higher among structured habitat types relative to bare sediments, with the type of structured habitat not influential. The exception appeared to be for reef habitat (although sampling was limited). Reef sites were structurally complex, but largely inhabited by older life stages (snapper and other fish species). Overall, nursery value for snapper appears to be connected to structure, rather than structure type. This result emphasises the importance of a broadened scope to coastal fishery management, one that incorporates not just fish extraction, but also habitat maintenance or restoration.  相似文献

A comparison was made between the electrophoretic patterns of isozymes of esterases and glucosephosphate isomerases of adult snapper Chryosophrys auratus, suspected snapper larvae, jack mackerel Trachurus novaezelandiae larvae, and blue mackerel Scomber australasicus larvae. By occurrence and bulk, crustaceans, polychaetes, echinoderms, molluscs, and teleosts formed the main snapper food groups, with crustaceans being the most important. Size of snapper, bottom type, depth, time of day and geographical regions affected the occurrence and importance of food items, while sex of fish probably did not.  相似文献
