A total of 4,581 Salmonella strains isolated from nonhuman sources, including foodstuffs associated with foodborne Salmonella outbreaks, from January 1996 through December 2000 were serotyped at the Enteropathogens Laboratory, Instituto Adolfo Lutz, S?o Paulo, Brazil. Among the 123 different serotypes identified, Salmonella enterica subsp. enterica serotype Enteritidis (Salmonella Enteritidis) was the most prevalent (32.7%), ranking first for almost every kind of source. The next most common serotypes were Salmonella Senftenberg (10.3%), Salmonella Hadar (6.8%), Salmonella Agona (5.1%), and Salmonella Typhimurium (2.4%). Rough strains belonging to the subspecies S. enterica subsp. enterica (4.8%), S. enterica subsp. arizonae (<1%), S. enterica subsp. diarizonae (<1%), and S. enterica subsp. houtenae (<1%) were also detected. Foodstuffs (including poultry meat for consumption) contained 38.1% of the studied Salmonella strains, poultry flocks (from several farms under salmonellosis control by the owners) contained 21.7%, the environment contained 10.6%, sewage contained 9.4%, water contained 6.6%, animal feed contained 4.4%, chill water from poultry-processing operations contained 2.2%, and other sources contained 7.0%. Foodstuffs extensively contaminated with Salmonella strains were poultry meat (40%), cow meat (11%), desserts (8%), mayonnaise (6%), sausage (5%), and unpasteurized shell eggs (4%), and there were several other food sources (26%). Homemade mayonnaise was the most common vehicle for Salmonella foodborne outbreaks, and Salmonella Enteritidis was the serotype most isolated (95%) from that source. According to these data and previously published data concerning Salmonella strains isolated in S?o Paulo State, almost the same serotypes have predominated among nonhuman sources for the last decade.  相似文献

We report here on the application of a compact ultraviolet spectrometer to measurement of NO2 emissions from sugar cane field burns in S?o Paulo, Brazil. The time-resolved NO2 emission from a 10 ha plot peaked at about 240 g (NO2) s(-1), and amounted to a total yield of approximately 50 kg of N, or about 0.5 g (N) m(-2). Emission of N as NOx (i.e., NO + NO2) was estimated at 2.5 g (N) m(-2), equivalent to 30% of applied fertilizer nitrogen. The corresponding annual emission of NOx nitrogen from S?o Paulo State sugar cane burning was >45 Gg N. In contrast to mechanized harvesting, which does not require prior burning of the crop, manual harvesting with burning acts to recycle nitrogen into surface soils and ecosystems.  相似文献

Winter atmospheric measurements of gaseous lower carbonyl and carboxylic acids were carried out simultaneously (in 1999) at two distinct urban sites located in the city of S?o Paulo, Brazil. The greater metropolitan area of S?o Paulo is the largest industrialized region of Latin America and has a highly polluted atmosphere. It has an unconventional mix of vehicle types in that a variety of gasoline blends, including oxygenated ones, are used. Mixing ratios of formic and acetic acids ranged, respectively, from 0.6 to 19.4 and from 0.1 to 10.6 ppbv in one of the sites studied and from 1.4 to 18.4 and from 0.4 to 6.7 ppbv in the other site. High values of formic to acetic ratios were found, especially in the latter site (average = 4.3), suggesting that photochemical production was the predominant source of the formic and acetic acid during the afternoon. Differing from the acids, levels of carbonyls were similar at both sites. Higher average mixing ratios of acetaldehyde and formaldehyde were found in the morning (18.9 and 17.2 ppbv) and gradually decreased from midday (9.5 and 11.8 ppbv) to evening (7.2 and 10.2 ppbv). In the morning, vehicular direct emission seemed to be the main primary source of formaldehyde and acetaldehyde, whereas at midday and evening these compounds appeared to be mainly formed by photochemistry. Secondary photochemical production of organic acids and aldehydes (rather than primary emissions from vehicles) was shown to be more important in S?o Paulo's atmosphere from midday to evening, particularly on days with strong solar radiation.  相似文献

Minimally processed vegetables (MPV) may be important vehicles of Salmonella spp. and cause disease. This study aimed at detecting and enumerating Salmonella spp. in MPV marketed in the city of São Paulo, Brazil. A total of 512 samples of MPV packages collected in retail stores were tested for Salmonella spp. and total coliforms and Escherichia coli as indication of the hygienic status. Salmonella spp. was detected in four samples, two using the detection method and two using the counting method, where the results were 8.8 × 102 CFU/g and 2.4 × 102 CFU/g. The serovars were Salmonella Typhimurium (three samples) and Salmonella enterica subsp. enterica O:47:z4,z23:- (one sample). Fourteen samples (2.7%) presented counts of E. coli above the maximum limit established by the Brazilian regulation for MPV (102 CFU/g). Therefore, tightened surveillance and effective intervention strategies are necessary in order to address consumers and governments concerns on safety of MPV.  相似文献

The occurrence of aflatoxins (AF) B1, B2, G1, G2 and cyclopiazonic acid (CPA) in feeds, and AFM1 and CPA in milk was determined in dairy farms located in the northeastern region of São Paulo state, Brazil, between October 2005 and February 2006. AF and CPA determinations were performed by HPLC. AFB1 was found in 42% of feed at levels of 1.0–26.4 µg kg?1 (mean: 7.1 ± 7.2 µg kg?1). The concentrations of AFM1 in raw milk varied between 0.010 and 0.645 µg l?1 (mean: 0.104 ± 0.138 µg l?1). Only one sample was above the tolerance limit adopted in Brazil (0.50 µg l?1) for AFM1 in milk. Regarding CPA in feed, six (12%) samples showed concentrations of 12.5–153.3 µg kg?1 (mean: 57.6 ± 48.7 µg kg?1). CPA was detected in only three milk samples (6%) at levels of 6.4, 8.8 and 9.1 µg l?1. Concentrations of aflatoxins and CPA in feed and milk were relatively low, although the high frequency of both mycotoxins indicates the necessity to continuously monitor dairy farms to prevent contamination of feed ingredients.  相似文献

In the present study, 24 samples of Minas Frescal cheese and 24 samples of Minas Padrão cheese produced in the North-east region of the state of São Paulo, Brazil, were analysed for aflatoxin M1 (AFM1) by high-performance liquid chromatography (HPLC) between March and August 2008. AFM1 was detected in 13 (27.1%) samples at concentrations ranging from 0.037 to 0.313?ng?g?1. The mean concentrations of AFM1 in positive samples of Minas Frescal and Minas Padrão cheese were 0.142?±?0.118 and 0.118?±?0.054?ng?g?1, respectively. It is concluded that the incidence of AFM1 in Minas cheese may contribute to an increase in the overall ingestion of aflatoxins in the diet, hence indicating the need for the adoption of a tolerance limit for AFM1 in cheese in Brazil.  相似文献

The incidence of Listeria monocytogenes in three cheese manufacturing plants from the northeastern region of S?o Paulo, Brazil, was evaluated from October 2008 to September 2009. L. monocytogenes was found in samples from two plants, at percentages of 13.3% (n = 128) and 9.6% (n = 114). Samples of raw and pasteurized milk, water, and Minas Frescal cheese were negative for L. monocytogenes, although the pathogen was isolated from the surface of Prato cheese and in brine from one of the plants evaluated. L. monocytogenes was also isolated from different sites of the facilities, mainly in non-food contact surfaces such as drains, floors, and platforms. Serotype 4b was the most predominant in the plants studied. The results of this study indicate the need for control strategies to prevent the dispersion of L. monocytogenes in the environment of cheese manufacturing plants.  相似文献

A total of 635 raw milk samples from 45 dairy farms, from three regions of São Paulo state – Brazil, were evaluated during 15 months for aflatoxin M1 (AFM1). AFM1 was determined by high performance liquid chromatograph with fluorescence detection. AFM1 was detected (>0.003 µg kg?1) in 72.9%, 56.3% and 27.5% of the samples from Bauru, Araçatuba and Vale do Paraíba regions, respectively. The mean AFM1 contamination considering all the samples was 0.021 µg kg?1. Furthermore, the concentration of AFM1 was quite different among Bauru (0.038 µg kg?1), Araçatuba (0.017 µg kg?1) and Vale do Paraíba (<0.01 µg kg?1) regions. Only three samples (0.5%) had higher contamination than the tolerated limit in Brazil (0.50 µg kg?1) and 64 samples (10.1%) had a higher contamination than the maximum limit as set by the European Union (0.050 µg kg?1). The estimated AFM1 daily intake was 0.358 and 0.120 ng kg?1 body weight per day for children and adults, respectively.  相似文献

Mussels of the genus Perna perna, cultivated in Ubatuba, São Paulo State, were analyzed for proximate composition and caloric value, month by month, during a year. From the nutritional point of view the proteins were outstanding (mean: 20.5% on a wet basis). The average contents of fats, ash and carbohydrates were, respectively, 3.24%, 2.24%, and 1.70%. The medium caloric value was 118 kcal 100 g−1. There were no marked variations, in the different months of the year for the proximate composition, except for the carbohydrates. This constant in the composition of the cultivated mussels indicates that there is a possibility to exploit mussels on a commercial basis.  相似文献

Detection of food-borne viruses such as noroviruses, rotaviruses and hepatitis A virus in food products differs from detection of most food-borne bacteria, as most of these viruses cannot be cultivated in cell culture to date. Therefore, detection of food-borne viruses in food products requires multiple steps: first, virus extraction; second, purification of the viral genomic material (RNA for the majority of food-borne viruses); and last, molecular detection. This review is focused on the first step, the virus extraction. All of the numerous published protocols for virus extraction from food samples are based on 3 main approaches: 1) (acid adsorption-) elution-concentration; 2) direct RNA extraction; and 3) proteinase K treatment. This review summarizes these virus extraction approaches and the results obtained from published protocols. The use of process controls is also briefly described.  相似文献

The genus Staphylococcus is recognized worldwide as a cause of bacterial infections in humans and animals. Antibiotics used in dairy cattle combined with ineffective control can increase antimicrobial resistance. The objective of this study was to characterize 95 Staphylococcus strains isolated from organic and conventional Minas Frescal cheese production regarding antibiotic resistance (phenotype and genotype), presence of sanitizer-resistant genes and biofilm-formation genes, and SCCmec typing. Most strains (25.3%) showed higher resistance to penicillin, followed by oxacillin (21.1%) and clindamycin (11.6%). Among antibiotic resistance genes, the most prevalent were blaZ (25.3%), mecA (13.7%), lsaB (6.3%), msrA (4.2%), ant4 (3.2%), and tetM (2.1%); among sanitizer-resistance genes they were qacA/B (5.3%) and qacC (6.3%); and among biofilm, bap (4.2%), icaA (29.5%), icaD (41.1%). However, there was no statistically significant difference between organic and conventional dairy products, possibly due to the lack of synthetic antibiotic use on conventional farms during the sample collection period. Methicillin-resistant Staphylococcus aureus (MRSA) had their SCCmec identified as types I and IVc, and the methicillin-resistant coagulase-negative staphylococci had nontypeable SCCmec. These results suggest that there are antibiotic-resistant strains in both organic and conventional Minas Frescal cheese production in the state of São Paulo, Brazil. This supports the idea that improved quality control is needed from the milking stage up to the final product.  相似文献

Mites can occur in large numbers in storage units, causing serious economic damage to stored products, as well as health problems such as asthma, diarrhea, acute enteritis, and allergic reactions. In Brazil, spice and medicinal dehydrated plants are widely sold in bulk, but almost nothing is known about mite infestations. Thus, the objective of this research was to evaluate the diversity and relative abundance of mites in samples of Coriandrum sativum L., Pimpinella anisum L., Petroselinum sativum Hoffm., Matricaria chamomilla L., Baccharis trimera (Less) DC, Bixa orellana L., Senna alexandrina Mill., Origanum vulgare L., Ocimum basilicum L., Melissa officinalis L., Mentha piperita L., Rosmarinus officinalis L., Peumus boldus Molina, Salvia officinalis L., Thymus vulgaris L., Laurus nobilis L., Hibiscus sabdariffa L., Myristica fragans Houtt., Capsicum annuum L., and Curcuma longa L., collected from food retailers in the metropolitan area of São Paulo, Brazil, from October 2015 to March 2016. A total of 2584 mites, distributed into 13 families, were found in the samples of these dehydrated plants. The most abundant mite species (families) were: Tyrophagus putrescentiae (Schrank) (Acaridae) (84.6%), Cheyletus malaccensis Oudemans (Cheyletidae) (5.4%), Blattisocius tarsalis (Berlese) (Blattisociidae) (4.6%), Suidasia sp. (Astigmata: Suidasiidae) (3.2%), and Typhlodromus transvaalensis (Nesbitt) (Phytoseiidae) (1.4%). Other families (Glycyfagidae, Ameroseiidae, Bdellidae, Iolinidae, Raphignatidae, Stigmaeidae, Tydeidae) together represented less than 1.0% of the mites. T. transvaalensis is recorded for the first time in stored products in Brazil. Differences among the plant species in terms of diversity, frequency and abundance of mites were observed. Although there is a regulatory standard for good food production and service practices, the results of this study demonstrate that the bulk retail market for dehydrated plants has failed to control hygiene and quality, considering the relatively high mite infestations in most of the evaluated plant species.  相似文献

Shiga toxin-producing Escherichia coli (STEC), is the most important recently emerged group of foodborne pathogens. Ruminants, especially cattle, have been implicated as a principal reservoir of STEC, undercooked ground beef and raw milk being the major vehicles of foodborne outbreaks. Enteropathogenic E. coli (EPEC) strains are defined as eae-harboring diarrheagenic E. coli that possess the ability to form A/E lesions on intestinal cells and that do not possess Shiga toxin genes. In order to determine the occurrence, serotypes and virulence markers of STEC and EPEC strains, 546 fecal samples from 264 diarrheic calves and 282 healthy calves in beef farms in S?o Paulo, Brazil, were screened by PCR. STEC and EPEC were isolated in 10% and 2.7% of the 546 animals, respectively. Although IMS test was used, the STEC serotype O157:H7 was not detected. The most frequent serotypes among STEC strains were O7:H10, O22:H16, O111:H(-), O119:H(-) and O174:H21, whereas O26:H11, O123:H11 and O177:H11 were the most prevalent among EPEC strains. In this study, serotypes not previously reported were found among STEC strains: O7:H7, O7:H10, O48:H7, O111:H19, O123:H2, O132:H51, O173:H(-), and O175:H49. The eae gene was detected in 25% of the STEC and 100% of EPEC strains. The intimin type theta/gamma2 was the most frequent among STEC, whereas the intimin beta1 was the most frequent intimin type among EPEC strains. To our knowledge, this is the first report of the occurrence of the new intimin muB in one strain of animal origin. This new intimin was detected in one atypical EPEC strain of serotype O123:H? isolated from diarrheic cattle. The enterohemolysin (ehxA) was detected in 51% of the STEC and 80% of the EPEC strains, whereas STEC autoagglutinating adhesin (saa) virulence gene was detected only in those STEC strains negative for eae gene. All 15 bovine EPEC strains isolated in this study were negative for both eaf and bfp genes. Our data shows that in Brazil cattle are not only a reservoir of STEC and atypical EPEC, but also a potential source of infection in humans, since the important STEC serotypes previously described and associated with severe diseases in humans, such as O111:H(-), O113:H21, O118:H16, and O174:H21 were isolated.  相似文献

The aim of this study was to assess the hygienic quality of raw milk used in the manufacture of S?o Jorge, a Protected Denomination of Origin Portuguese semihard cheese, as well as to ascertain the sanitary conditions prevailing during its processing. Viable counts of Enterobacteriaceae and Micrococcaceae were accordingly obtained, pertaining to 21 independent batches (including samples of raw milk, curd, and cheeses after 1, 3, and 4 months of ripening), from 7 dairy farms. Standard plate counts (log CFU per milliliter or per gram) ranged from 6.1 to 8.6 in raw milk, whereas they ranged from 7.0 to 8.0 in 4-month-old cheeses. Viable counts of Enterobacteriaceae ranged between 5.9 and 7.0 in raw milk and between 0.0 and 1.3 in 4-month-old cheeses. Species identified within this family encompassed Klebsiella oxytoca, Klebsiella cloacae, Klebsiella pneumoniae, Enterobacter sakazakii, and Escherichia coli; Klebsiella ornithinolytica, Klebsiella terrigena, and Serratia odorifera were detected only in raw milk. No Salmonella whatsoever could be detected in any of the samples. Viable counts of Micrococcaceae ranged between 4.7 and 5.9 and between 1.3 and 3.3 in raw milk and 4-month-old cheeses, respectively. Species identified within this family encompassed Staphylococcus sciuri, Staphylococcus epidermidis, Staphylococcus saprophyticus (which was found mainly in ripened cheeses), and Staphylococcus aureus (which was not detected in 4-month-old cheeses). Accompanying physicochemical analyses included determination of moisture, salt, and pH. Statistical analyses revealed a negative correlation between salt content and viable numbers of Enterobacteriaceae in cheese, whereas in the case of Micrococcaceae, a more negative correlation was found between viable numbers and moisture content than between viable numbers and pH. The results of our study indicate, in general, poor milk handling conditions in all farms, given that the indicators total mesophile and Enterobacteriaceae counts were high, between 100- and 1,000-fold those enforced by international standards pertaining to the matrices in question. However, by the time of regular consumption (i.e., after 4 months of ripening), S?o Jorge cheeses exhibit low levels of contamination by Enterobacteriaceae and S. aureus, as well as absence of Salmonella.  相似文献
