A facile approach is offered to synthesize well‐defined amphiphilic ABC triblock copolymers composed of poly(ethylene glycol) monomethyl ether (MPEO) as A block, poly(L ‐lysine) (PLLys) as B block, and poly(ε‐caprolactone) (PCL) as C block by a combination of ring‐opening polymerization (ROP) and click reactions. The propargyl‐terminated poly(Z‐L ‐lysine)‐block‐poly(ε‐caprolactone) (MPEO‐PzLLys‐PCL) diblock copolymers were synthesized via the ring‐opening polymerization of Nε‐carbobenzoxy‐L ‐lysine N‐carboxyanhydride (Z‐L ‐Lys NCA) in DMF at room temperature using propargyl amine as an initiator and the resulting amino‐terminated poly(Z‐L ‐lysine) then used in situ as a macroinitiator for the polymerization of ε‐caprolactone in the presence of stannous octoate as a catalyst. The triblock copolymers poly(ethylene glycol) monomethyl ether –block‐poly(Z‐L ‐lysine)‐block‐poly(ε‐caprolactone) (MPEO‐PzLLys‐PCL) were synthesized via the click reaction of the propargyl‐terminated PzLLys‐PCL and azido‐terminated poly(ethylene glycol) monomethyl ether (PEO‐N3) in the presence of CuBr and 1,1,4,7,7‐pentamethyldiethylenetriamine (PMDETA) catalyst system. After the removal of Z groups of L ‐lysine units, amphiphilic and biocompatible ABC triblock copolymers MPEO‐PLLys‐PCL were obtained. The structural characteristics of these ABC triblock copolymers and corresponding precursors were characterized by NMR, IR, and GPC. These results showed the click reaction was highly effective. Therefore, a facile approach is offered to synthesize amphiphilic and biocompatible ABC triblock copolymers consisting of polyether, polypeptide and polyester. © 2010 Wiley Periodicals, Inc. In this article, we describe the synthesis and solution properties of PEG‐b‐PTMC star block copolymers via ring‐opening polymerization (ROP) of trimethylene carbonate (TMC) monomer initiated at the hydroxyl end group of the core PEG using HCl Et2O as a monomer activator. The ROP of TMC was performed to synthesize PEG‐b‐PTMC star block copolymers with one, two, four, and eight arms. The PEG‐b‐PTMC star block copolymers with same ratio of between hydrophobic PTMC and hydrophilic PEG segments were obtained in quantitative yield and exhibited monomodal GPC curves. The amphiphilic PEG‐b‐PTMC star block copolymers formed spherical micelles with a core–shell structure in an aqueous phase. The mean hydrodynamic diameters of the micelles increased from 17 to 194 nm with increasing arm number. As arm number increased, the critical micelle concentration (CMC) of the PEG‐b‐PTMC star block copolymers increased from 3.1 × 10?3 to 21.1 × 10?3 mg/mL but the partition equilibrium constant, which is an indicator of the hydrophobicity of the micelles of the PEG‐b‐PTMC star block copolymers in aqueous media, decreased from 4.44 × 104 to 1.34 × 104. In conclusion, we confirmed that the PEG‐b‐PTMC star block copolymers form micelles and, hence, may be potential hydrophobic drug delivery vehicles. © 2008 Wiley Periodicals, Inc. Biodegradable, amphiphilic, linear (diblock and triblock) and star‐shaped (three‐armed and four‐armed) poly[(ethylene glycol)‐block‐(ε‐caprolactone)] copolymers (PEG–PCL copolymers) were synthesized by ring‐opening polymerization of ε‐caprolactone (CL) with stannous octoate as a catalyst, in the presence of monomethoxypoly(ethylene glycol) (MPEG), poly(ethylene glycol) (PEG), three‐armed poly(ethylene glycol) (3‐arm PEG) or four‐armed poly(ethylene glycol) (4‐arm PEG) as an initiator, respectively. The monomer‐to‐initiator ratio was varied to obtain copolymers with various PEG weight fractions in a range 66–86%. The molecular structure and crystallinity of the copolymers, and their aggregation behavior in the aqueous phase, were investigated by employing 1H‐NMR spectroscopy, gel permeation chromatography and differential scanning calorimetry, as well as utilizing the observational data of gel–sol transitions and aggregates in aqueous solutions. The aggregates of the PEG–PCL block copolymers were prepared by directly dissolving them in water or by employing precipitation/solvent evaporation technique. The enthalpy of fusion (ΔHm), enthalpy of crystallization (ΔHcrys) and degrees of crystallinity (χc) of PEG blocks in copolymers and the copolymer aggregates in aqueous solutions were influenced by their PEG weight fractions and molecular architecture. Well‐defined asymmetric amphiphilic ABA′ block copolymers composed of poly(ethylene oxide) monomethylene ether (MPEO) with different molecular weights as A or A′ block and poly(styrene) (PS) as B block were synthesized by the combination of atom transfer radical polymerization (ATRP) and click reactions. First, bromine‐terminated diblock copolymer poly(ethylene oxide) monomethylene ether‐block‐poly(styrene) (MPEO‐PS‐Br) was prepared by ATRP of styrene initiated with macroinitiator MPEO‐Br, which was prepared from the esterification of MPEO and 2‐bromoisobutyryl bromide. Then, the azido‐terminated diblock copolymers MPEO‐PS‐N3 were prepared through the bromine substitution reaction with sodium azide. Propargyl‐terminated MPEO with a different molecular weight was prepared under the basic condition from propargyl alcohol and p‐toluenesulfonyl‐terminated MPEO, which was prepared through the esterification of MPEO and p‐toluenesulfochloride using pyridine as solvent. Asymmetric amphiphilic ABA′ block copolymers, with a wide range of number–average molecular weights from 1.92 × 104 to 2.47 × 104 and a narrow polydispersity from 1.03 to 1.05, were synthesized via a click reaction of the azido‐terminated diblock copolymers and the propargyl‐terminated MPEO in the presence of CuBr and 1,1,4,7,7‐pentamethyldiethylenetriamine (PMDETA) catalyst system. Dually responsive amphiphilic diblock copolymers consisting of hydrophilic poly(N‐isopropyl acrylamide) [poly(NIPAAm)] and hydrophobic poly(9‐anthracene methyl methacrylate) were synthesized by reversible addition fragmentation chain‐transfer (RAFT) polymerization with 3‐(benzyl sulfanyl thiocarbonyl sulfanyl) propionic acid as a chain‐transfer agent. In the first step, the poly(NIPAAm) chain was grown to make a macro‐RAFT agent, and in the second step, the chain was extended by hydrophobic 9‐anthryl methyl methacrylate to yield amphiphilic poly(N‐isopropyl acrylamide‐b‐9‐anthracene methyl methacrylate) block copolymers. The formation of copolymers with three different hydrophobic block lengths and a fixed hydrophilic block was confirmed from their molecular weights. The self‐assembly of these copolymers was studied through the determination of the lower critical solution temperature and critical micelle concentration of the copolymers in aqueous solution. The self‐assembled block copolymers displayed vesicular morphology in the case of the small hydrophobic chain, but the morphology gradually turned into a micellar type when the hydrophobic chain length was increased. The variations in the length and chemical composition of the blocks allowed the tuning of the block copolymer responsiveness toward both the pH and temperature. The resulting self‐assembled structures underwent thermally induced and pH‐induced morphological transitions from vesicles to micelles and vice versa in aqueous solution. These dually responsive amphiphilic diblock copolymers have potential applications in the encapsulation of both hydrophobic and hydrophilic drug molecules, as evidenced from the dye encapsulation studies. © 2018 Wiley Periodicals, Inc. A series of amphiphilic poly(N‐vinyl carbazole) (PVK) block copolymers containing tris(8‐hydroxyquinoline) aluminum (Alq3) have been synthesized via reversible addition‐fragmentation chain transfer (RAFT) radical polymerization with trithiocarbonate terminated polyethylene glycol (PEG) as a RAFT agent. The chemical structures of the block copolymers have been identified by 1H nuclear magnetic resonance and Fourier transform infrared spectrometer. The analysis of gel permeation chromatography indicates that the polydispersity indices of the block copolymers are lower than 1.30. The results of thermogravimetric analysis show that the decomposition temperatures of the copolymers are higher than 330 °C. The optical properties of the copolymers have been investigated and the obviously enhanced emission from Alq3 has been found due to resonance energy transfer from PVK to Alq3. © 2016 Wiley Periodicals, Inc. Previously, star polystyrenes (PSs) have been prepared by atom transfer radical polymerization (ATRP) of N‐[2‐(2‐bromoisobutyryloxy)ethyl]maleimide (BiBEMI) with a large excess of styrene (St) in one pot. But linear PSs were also present during the formation of the star polymers. In the work reported here, we found that control of the formation of star polymers using a one‐pot approach can be improved by using a two‐step process. The polymerization was conducted first at a low temperature to form multifunctional cores by copolymerization of BiBEMI and St. Second, on increasing the temperature, homopolymerization of St occurred to grow PS arms. Then a series of amphiphilic star polystyrene‐block‐poly(acrylic acid)s, (S14Ax)16, were prepared by ATRP of tert‐butyl acrylate with the star PSs as macroinitiators, followed by selective acidolysis of the poly(tert‐butyl acrylate) blocks. Their micellization was studied using dynamic light scattering, which suggested that (S14A112)16 amphiphilic star block copolymers could form unimolecular micelles in a basic aqueous solution. Then pyrene molecules were encapsulated using the (S14A112)16 amphiphilic star copolymers and the loading capacity was investigated with UV and fluorescence spectroscopy. © 2013 Society of Chemical Industry  相似文献

Functionalized poly(ethylene glycol) (PEG) containing four chloride end functional groups (PEG‐Cl4) was synthesized through reaction between cyanuric chloride and PEG‐(OH)2. Chloride end functional groups of PEG‐Cl4 were able to initiate the ring opening polymerization of 2‐ethyl‐2‐oxazoline and star copolymers containing a PEG core, and poly(2‐ethyl‐2‐oxazoline) (POX) arms were obtained. Polymerization was quenched using diethanolamine, and star copolymers containing hydroxyl end functional groups (PEG‐POX‐OH) were obtained. ε‐Caprolactone was then polymerized using the hydroxyl end functional groups of star copolymers and amphiphilic linear‐dendritic copolymers containing PEG and POX, and poly(caprolactone) (PCL) blocks were synthesized. Linear‐dendritic copolymers were able to load the organic and inorganic guest molecules. BACKGROUND: The aim of this work was to develop polyhydroxyalkanoates (PHAs) for blood contact applications, and to study their self‐assembly behavior in aqueous solution when the PHAs are incorporated with hydrophilic segments. To do this, poly(ester‐urethane) (PU) multiblock copolymers were prepared from hydroxyl‐terminated poly(ethylene glycol) (PEG) and hydroxylated poly[(R)‐3‐hydroxyalkanoate] (PHA‐diol) using 1,6‐hexamethylene diisocyanate as a coupling reagent. The PEG segment functions as a soft, hydrophilic and crystalline portion and the poly[(R)‐3‐hydroxybutyrate] segment behaves as a hard, hydrophobic and crystalline portion. In another series of PU multiblock copolymers, crystalline PEG and completely amorphous poly[((R)‐3‐hydroxybutyrate)‐co‐(4‐hydroxybutyrate)] behaved as hydrophobic and hydrophilic segments, respectively. RESULTS: The formation of a PU series of block copolymers was confirmed by NMR, gel permeation chromatography and infrared analyses. The thermal properties showed enhanced thermal stability with semi‐crystalline morphology via incorporation of PEG. Interestingly, the changes of the hydrophilic/hydrophobic ratio led to different formations in oil‐in‐water emulsion and surface patterning behavior when cast into films. Blood compatibility was also increased with increasing PEG content compared with PHA‐only polymers. CONCLUSION: For the first time, PHA‐based PU block copolymers have been investigated in terms of their blood compatibility and aggregation behavior in aqueous solution. The aim of this study is to develop azido‐carrying biodegradable polymers and their postfunctionalization with alkynyl compounds via click chemistry and to investigate their potential use in drug delivery. Azido polymers were prepared by ring‐opening polymerization of cyclic carbonate monomer, 2,2‐bis(azidomethyl)trimethylene carbonate (ATC) with lactide using stannous octoate as catalyst. Several alkynyl compounds were selected to investigate the feasibility and reaction condition of click chemistry. With microwave‐assisting, the reaction time of click chemistry was shortened to 5 min. By using poly(ethylene glycol) (PEG) as macroinitiator, amphiphilic block copolymer mPEG‐b‐P(LA‐co‐ATC) was obtained and it could self‐assemble into micelles by solvent replacement method. The pendant groups were used for conjugating anticancer drugs gemcitabine and paclitaxel and fluorescent dye Rhodamine B. 3‐(4,5‐Dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide was used to assay the cytotoxicity of the conjugate micelles against SKOV‐3 and HeLa cell lines. © 2012 Wiley Periodicals, Inc. A new amphiphilic Y‐shaped copolymer, comprised of hydrophobic Poly(trimethylene carbonate) (PTMC) and hydrophilic Poly(N,N‐dimethylamino‐2‐ethyl methacrylate) (PDMAEMA), was designed and synthesized by a combination of atom transfer radical polymerization (ATRP) and ring‐opening polymerization (ROP) using a new heterofunctional initiator, Br‐Init‐(OH)2, bearing one initiation site for ATRP and two for ROP. At first, a new trifunctional core molecule bearing hydroxyl group and bromine moieties, Br‐Init‐(OH)2, was synthesized via protection followed by esterification reaction of 5‐ethyl‐5‐hydroxymethyl‐2,2‐dimethyl‐1,3‐dioxane with 2‐bromoisobutyryl bromide and deprotection. In the presence of trifunctional core molecule, Br‐Init‐(OH)2, target Y‐shaped miktoarm star copolymers, (PTMC)2‐ b‐PDMAEMA, were successfully synthesized by sequence conducting the ROP of TMC and ATRP of DMAEMA. The Y‐shaped copolymers were characterized by 1H NMR and GPC measurements. Subsequently, the self‐assembly behavior of these copolymers was investigated by dynamic light scattering method and transmission electron microscopy, which indicated that these amphiphilic Y‐shaped copolymers can self‐assemble into micelles and possess distinct pH‐dependent size in aqueous milieu. Amphiphilic thermally sensitive poly(N‐isopropylacrylamide)‐block‐poly(tetramethylene carbonate) block copolymers were synthesized by ring‐opening polymerization of tetramethylene carbonate with hydroxyl‐terminated poly(N‐isopropylacrylamide) (PNiPAAm) as macro‐initiator in the presence of stannous octoate as catalyst. The synthesis involved PNiPAAm bearing a single terminal hydroxyl group prepared by telomerization using 2‐hydroxyethanethiol as a chain‐transfer agent. The copolymers were characterized using 1H NMR and Fourier transform infrared spectroscopy and gel permeation chromatography. Their solutions show reversible changes in optical properties: transparent below the lower critical solution temperature (LCST) and opaque above the LCST. The LCST depends on the polymer composition and the media. Owing to their amphiphilic characteristics, the block copolymers form micelles in the aqueous phase with critical micelle concentrations (CMCs) in the range 1.11–22.9 mg L?1. Increasing the hydrophobic segment length or decreasing the hydrophilic segment length in the amphiphilic diblock copolymers produces lower CMCs. A core‐shell structure of the micelles is evident from 1H NMR analyses of the micelles in D2O. Transmission electron microscopic analyses of micelle morphology show a spherical structure of both blank and drug‐loaded micelles. The blank and drug‐loaded micelles have an average size of less than 130 nm. Observations show high drug‐entrapment efficiency and drug‐loading content for the drug‐loaded micelles. Two series of poly(ε‐caprolactone)‐b‐poly(ethylene glycol)‐b‐poly(ε‐caprolactone) triblock copolymers were prepared by the ring opening polymerization of ε‐caprolactone in the presence of poly(ethylene glycol) and dibutylmagnesium in 1,4‐dioxane solution at 70°C. The triblock structure and molecular weight of the copolymers were analyzed and confirmed by 1H NMR, 13C NMR, FTIR, and gel permeation chromatography. The crystallization and thermal properties of the copolymers were investigated by wide‐angle X‐ray diffraction (WAXD) and differential scanning calorimetry (DSC). The results illustrated that the crystallization and melting behaviors of the copolymers were depended on the copolymer composition and the relative length of each block in copolymers. Crystallization exothermal peaks (Tc) and melting endothermic peaks (Tm) of PEG block were significantly influenced by the relative length of PCL blocks, due to the hindrance of the lateral PCL blocks. With increasing of the length of PCL blocks, the diffraction and the melting peak of PEG block disappeared gradually in the WAXD patterns and DSC curves, respectively. Star‐shaped copolymers poly(ε‐caprolactone)‐bolck‐poly(ε‐benzyloxycarbonyl‐l ‐lysine) (SPPCL‐b‐PZLLs) with porphyrin core were synthesized by a sequential ring‐opening polymerization (ROP) of CL and Nε‐Benzyloxycarbonyl‐l ‐lysine N‐Carboxyanhydride. After the deprotection of benzyloxycarbonyl groups in polylysine blocks, the star‐shaped amphiphilic copolymers SPPCL‐b‐PLLs were obtained. These amphiphilic copolymers can self‐assemble into micelles or aggregates in aqueous solution. Investigation shows that the morphology of micelles/aggregates varied according to the change of pH values of media, indicating the pH‐responsive property of SPPCL‐b‐PLL copolymers. Furthermore, associated with conjugated porphyrin cores, the SPPCL‐b‐PLL copolymers micelles showed a certain degree of Photodynamic Therapy (PDT) effects on tumor cells, suggesting its potential application as carrier for hydrophobic drug with additional therapeutic ability of inherent porphyrin segments. © 2013 Wiley Periodicals, Inc. Poly(A)‐block‐poly(B), poly(A)‐block‐poly(B)‐block‐poly(A) and B(A)2 block copolymers were prepared through coordinated anionic ring‐opening polymerization of ε‐caprolactone (CL) and lactic acid (LA) using hydroxy‐terminated polydimethylsiloxane (PDMS) as initiator. A wide range of well‐defined combinations of PDMS‐block‐PCL and PDMS‐block‐PLA diblock copolymers, PCL‐block‐PDMS‐block‐PCL and PLA‐block‐PDMS‐block‐PLA triblock copolymers and star‐PDMS(PCL)2 copolymers were thus obtained. The number‐average molar masses and the structure of the synthesized block copolymers were identified using various analytical techniques. The thermal properties of these copolymers were established using differential scanning calorimetry. Considering PDMS‐block‐PCL copolymers, the results demonstrate the complex effect of polymer architecture and PCL block length on the ability of the PDMS block to crystallize or not. In the case of diblock copolymers, crystallization of PCL blocks originated from stacking of adjacent chains inducing the extension of the PDMS block that can easily crystallize. In the case of star copolymers, the same tendency as in triblock copolymers is observed, showing a limited crystallization of PDMS when the length of the PCL block increases. In the case of PDMS‐block‐PLA copolymers, melting and crystallization transitions of the PLA block are never observed. Poly(methyl methacrylate)‐block‐polyurethane‐block‐poly(methyl methacrylate) tri‐block copolymers have been synthesized successfully through atom transfer radical polymerization of methyl methacrylate using telechelic bromo‐terminated polyurethane/CuBr/N,N,N,N″,N″‐pentamethyldiethylenetriamine initiating system. As the time increases, the number‐average molecular weight increases linearly from 6400 to 37,000. This shows that the poly methyl methacrylate blocks were attached to polyurethane block. As the polymerization time increases, both conversion and molecular weight increased and the molecular weight increases linearly with increasing conversion. These results indicate that the formation of the tri‐block copolymers was through atom transfer radical polymerization mechanism. Proton nuclear magnetic resonance spectral results of the triblock copolymers show that the molar ratio between polyurethane and poly (methyl methacrylate) blocks is in the range of 1 : 16.3 to 1 : 449.4. A facile synthetic pathway to a multi‐arm star graft polymer has been developed via a grafting‐onto strategy using a combination of a reversible addition–fragmentation chain transfer (RAFT) arm‐first technique and aldehyde–aminooxy click reaction. A star backbone bearing aldehyde groups was prepared by the RAFT copolymerization of acrolein (Ac), an existing commercial aldehyde‐bearing monomer, with styrene (St), followed by crosslinking of the resultant poly(St‐co‐Ac) macro‐RAFT agent using divinylbenzene. The aldehyde groups on the star backbone were then used as clickable sites to attach poly(ethylene glycol) (PEG) side chains via the click reaction between the aldehyde groups and aminooxy‐terminated PEG, leading to a structurally well‐defined star graft copolymer with arms consisting of poly(St‐co‐Ac) as backbone and PEG as side chains. Crystalline morphology and self‐assembly in water of the obtained star graft copolymer were also investigated. Opportunities are open for the star graft copolymer to form either multimolecular micelles or unimolecular micelles via control of the number of grafted PEG side chains. © 2013 Society of Chemical Industry  相似文献

Polyethylene‐b‐poly (ethylene glycol) (PE‐b‐PEG) was successfully synthesized by a coupling reaction of hydroxyl‐terminated polyethylene (PE‐OH) and isocyanate‐terminated poly (ethylene glycol) (PEG‐NCO). PE‐OH was prepared by coordination chain transfer polymerization (CCTP) using 2,6‐bis[1‐(2,6‐diisopropylphenyl)imino ethyl] pyridine iron (II) dichloride /dry ethylaluminoxane (DEAO) /diethyl zinc (ZnEt2) as catalyst and subsequent in situ oxidation with oxygen. The active centers of this catalyst system were counted, indicating that the active centers were more stable using DEAO as cocatalyst than using dry methylaluminoxane (DMAO) as cocatalyst. PEG‐NCO was synthesized through the condensation reaction of monomethylpoly(ethylene glycol) (PEG) with isophoronediisocyanate (IPDI). Subsequently, the thermal characterization, morphological characterization and the application of these diblock copolymers was investigated. The results indicated that the diblock copolymers were effective compatilizers for polyethylene/poly(ethylene glycol) blends. Meanwhile, they were excellent surface modification agents for polyethylene membrane and glass sheet, it can efficiently turn a hydrophobic surface into a hydrophilic surface, or vice versa. © 2015 Wiley Periodicals, Inc. Two new alkyne‐terminated xanthate reversible addition‐fragmentation chain‐transfer (RAFT) agents: (S)‐2‐(Propynyl propionate)‐(O‐ethyl xanthate) (X3) and (S)‐2‐(Propynyl isobutyrate)‐(O‐ethyl xanthate) (X4) were synthesized and characterized and used for the controlled radical polymerization of N‐vinylpyrrolidone (NVP). X3 showed better chain transfer ability in the polymerization at 60°C. Molecular weight of the resulted polymer increased linearly with the increase in monomer loading. Kinetics study with X3 showed the pseudo‐first order kinetics up to 67% monomer conversion. Molecular weight (Mn) of the resulting polymer increased linearly with the increase in the monomer conversion up to around 67%. With the increase in the monomer conversion, polydispersity of the corresponding poly(NVP)s initially decreased from 1.34 to 1.32 and then increased gradually to 1.58. Chain‐end analysis of the resulting polymer by 1H‐NMR and FTIR showed clearly that polymerization started with radical forming out of xanthate RAFT agent. Living nature of the polymerization was also confirmed from the successful homo‐chain extension experiment and the hetero‐chain extension experiment involving synthesis of poly(NVP)‐b‐polystyrene amphiphilic diblock copolymer. Formed alkyne‐terminated poly(NVP) also allowed easy conjugation to azide‐terminated polystyrene by click chemistry to prepare well‐defined poly(NVP)‐b‐polystyrene block copolymers. Magnetic star‐shaped amphiphilic copolymers (S‐Fe3O4‐PLA‐b‐MPEG) consisting of Fe3O4 as the core, poly(L ,D ‐lactide) (PLA) as the inner layer, and monomethyl polyethylene glycol (MPEG) as the out shell were synthesized. The syntheses included ring‐opening polymerization of L ,D ‐lactide initiated by hydroxyl modified Fe3O4 (Fe3O4‐(OH) n), followed by the esterification of the PLA with MPEG. The structure of the star block copolymers were characterized by Fourier Transform infrared spectroscopy, thermogravimetric analysis, X‐ray diffraction, transmission electron microscopy, nanoparticle size analyzer, and vibrating sample magnetometer. The nanoparticles in aqueous solution were made from the amphiphilic star copolymer. The average size of the nanoparticles was adjustable and increased with the increase of the PLA segments in the copolymer. The cytotoxicity grade of the nanoparticles was zero determined by the analysis of cytotoxicity.
