The proportion of yeast species involved in eye infections in 11 patients was examined. The presence of yeast organisms as causative agents of endophthalmitis was found in corneal smears (n=4), conjunctival swabs (4), and vitreous fluid (3). Altogether 5 strains ofCandida albicans, 2 strains ofC. krusei and one strain each ofC. guilliermondii, C. parapsilosis, C. tropicalis andCryptococcus neoformans were isolated from the clinical material. The hematogenic origin of endophthalmitis was proved in 7 cases on the basis of positive blood samples and in 2 cases by the isolation of yeasts from the tip of an intravenous catheter. Endophthalmitis-supporting risk factors such as indwelling intravenous catheters, prolonged use of broad-spectrum antibiotics and chemotherapy, surgical intervention, diabetes mellitus, and malignancy were observed in the patients.  相似文献

Young colonies of Sabouraud's glucose agar room temperature culture ofCandida species from human isolation were suspended in distilled water. The suspension was mixed with a solution of lysozyme and incubated in a 37° C water bath. Within 3–5 hours, various species ofCandida cells showed flocculation to varying degrees which occurred at varying periods of onset. Among sevenCandida species,Candida albicans andCandida stellatoidea showed the strongest flocculation, earliest onset and most solution clarity than did any other species.Candida stellatoidea was indistinguishable fromCandida albicans in its degree of flocculation, and in the clarity of solution.Candida species may be arranged in the following order according to their decreasing positivity in flocculation:
  1. Candida albicans
  2. Candida stellatoidea
  3. Candida tropicalis
  4. Candida krusei
  5. Candida pseudotropicalis
  6. Candida parapsilosis
  7. Candida guilliermondii
  8. Saccharomyces species may be placed afterCandida guilliermondii.
It seems possible to separate theCandida species into 3 groups by the rate of flocculation, and clarity of solution. Group I.Candida albicans andCandida stellatoidea. Group II.Candida tropicalis, C. krusei andCandida pseudotropicalis. Group III.Candida parapsilosis andCandida guilliermondii. Zusammenfassung An einer willkürlichen Auswahl von Stämmen aus den GeneraCandida, Torulopsis, Saccharomyces undSchizosaccharomyces konnten mit Hilfe der Immunelektrophorese außer bekannten Beziehungen der Polysaccharidantigene noch einige gemeinsame Eiweißantigene nachgewiesen werden.Übereinstimmungen bestehen mitTsuchiya in Bezug auf die enge Verwandtschaft vonC. albicans, C. tropicalis undC. stellatoidea, sowie zwischenS. cerevisiae undC. robusta.Bei Berücksichtigung aller Antigene fanden wir zusätzlich Antigengemeinschaften zwischenT. glabrata undS. rouxii, sowie weitere antigene Beziehungen beiC. krusei, C. utilis, T. famata undS. cerevisiae, die auf gemeinsamen Eiweißantigenen beruhen.Es wurde der Versuch unternommen, einige Eiweißantigene ausC. albicans zu isolieren. Nach Fällung des größten Teils der Antigene mit Universalpuffer verblieben im Rückstand 2 Eiweißantigene, mit denen Kaninchen immunisiert wurden. Von den beiden induzierten Antikörpern gegen Eiweißantigen vonC. albicans reagierte der eine nur mitC. tropicalis, der andere nur mitC. stellatoidea, jedoch nicht mit anderen Hefen.
Summary The serologic relationships of various yeast strains, arbitrarily selected from the generaCandida, Torulopsis, Saccharomyces, andSchizosaccharomyces, were investigated by means of immunoelectrophoretic technique. In addition to serologic crossings ofC. albicans, C. tropicalis, andC. stellatoidea as well as ofS. cerevisiae, andC. robusta, which had previously been reported byTsuchiya and other investigators, we have found, that protein antigens are shared by various strains, i.e.C. krusei, C. utilis, T. famata, andS. cerevisiae. Taking into account the overall picture of their antigenic patterns,T. glabrata andS. rouxii were found to stand particulary close to each other.Attempts were made to isolate some of these proteide antigens fromC. albicans. After partial precipitation of the crude homogenate by the addition ofJohnson &Lindsay's Universal Buffer followed by centrifugation, the supernatant was used to immunize rabbits. Two types of antibodies could be detected; while both reacted in the immunoelectrophoresis withC. albicans antigens, one also precipitatedC. tropicalis and the otherC. stellatoidea. None of the 10 other yeasts which have been tested showed any activity with these rabbit sera.

The yeastCandida albicans coaggregates with a variety of streptococcal species, an interaction that may promote oral colonization by yeast cells.C. albicans andCandida tropicalis are the yeasts most frequently isolated from the human oral cavity and our data demonstrate that both these species bind toStreptococcus gordonii NCTC 7869 while two otherCandida species (Candida krusei andCandida kefyr) do not. Adherence ofC. albicans was greatest when the yeast had been grown at 30° C to mid-exponential growth phase. For 21 strains ofC. albicans there was a positive correlation between the ability to adhere toS. gordonii and adherence to experimental salivary pellicle. Whole saliva either stimulated or slightly inhibited adherence ofC. albicans toS. gordonii depending on the streptococcal growth conditions. The results suggest that the major salivary adhesins and coaggregation adhesins ofC. albicans are co-expressed.  相似文献

Distribution ofCandida species was investigated by examining 245 samples from skin lesions and nails. The isolates were identified using standard laboratory methods including germ tube test, micromorphology of colonies on rice agar, the commercial kit, saccharide assimilation and fermentation tests. Eight species ofCandida were identified:C. albicans accounted for 56.4% of the isolates,C. parapsilosis 29.1,C. tropicalis 7.8,C. pulcherrima 2.9,C. guilliermondii 1.5,C. krusei andC. zeylanoides for 0.9% each, andC. robusta for 0.5%. The factors significantly associated with colonization were prolonged antibiotic therapy, parenteral nutrition, low birth body mass of infants, intubation, duration of stay in hospital, indwelling intravenous catheter, malignancies, diabetes, surgery, and obesity.  相似文献

A cell wall lytic enzyme (gamete wall-autolysin) and a polyclonal antiserum raised against one of the major cell wall glycopeptides ofChlamydomonas reinhardtii were used to study their cross-reactivities with the cell walls of variety of members of the Volvocales. Lytic enzyme was able to digest completely the cell walls of five species ofChlamydomonas (C. reinhardtii group), six species ofGonium and two species ofAstrephomene. The colonial structures ofGonium andAstrephomene were broken into individual cells by exposure to the enzyme and protoplasts were then formed. These organisms also showed a strong cross-reactivity with anti-cell wall glycopeptide by an indirect-immunofluorescence test. The cell walls ofChlamydomonas angulosa, Dysmorphococcus globosus, Pandorina morum, Eudorina elegans, Volvulina steinii, Pleodorina california andVolvox carteri all showed a strong cross-reactivity to the antibody, although they were insensitive to the lytic enzyme. Many other species ofChlamydomonas, Carteria crucifera, Chlorogonium elongatum, Polytoma uvella, Haematococcus lacustris, Lobomonas piriformis, Phacotus lenticularis, Pteromonas angulosa, Stephanosphera pluvialis, andPyrobotrys casinoensis had cell walls which were resistant to the enzyme and showed no or weak cross-reactivity with the antibody. Based on the results, a possible evolutionary sequence from a unicellular relative ofC. reinhardtii to the multicellular algae is discussed.  相似文献

A study of ultrathin sections ofMicrococcus denitrificans andM.halodenitrificans has shown similar cell structures. The cell wall consists of several layers corresponding to those of the cell wall of gram-negative bacteria. The thickness of the cell wall is 250 – 350 Å; that of the cytoplasmic membrane 70 Å. The cytoplasm in both species contains ribosomes and inclusions of polymetaphosphate. Comparison with ultrathin sections ofThiobacillus novellus shows too much difference to consider the former two species to be identical with the latter one. The species most frequently found in the group of neonates wasC. parapsilosis, its incidence reaching 48%. Other yeasts isolated from blood wereC. albicans (41%),C. tropicalis (7%),C. krusei andC. pseudotropicalis (2% each). Six yeast species (C. albicans 44%,C. parapsilosis 28%,C. tropicalis 11%,C. krusei 8%,C. guilliermondii 6%,C. lusitaniae 3%) were detected in cancer patients. The presence of an intravenous catheter as a possible risk in the development of candidaemia was identified in all neonates and in 69% of cancer patients.C. krusei candidaemia was associated with prophylactic fluconazole therapy.  相似文献

The susceptibility of organisms of the speciesCandida to sulfamethoxazole were tested in-vitro using the disc sensitivity method.Under specified conditions a zone of growth inhibition surrounding sulfamethoxazole tablet sensi disc was consistently produced in cultures ofCandida albicans, Candida stellatoidea andSaccharomyces cervisiae.Diameters of growth inhibition zone were measured and found to be greatest in cultures ofCandida stellatoidea andSaccharomyces, but growth inhibition inCandida albicans cultures was observed to a lesser degree.The correlation between in vivo pathogenicity of organisms of the speciesCandida, and degree of growth inhibition by sulfamethoxazole sensi disc presents an interesting relationship.  相似文献

In the period from November 1998 to June 2001 13 cases of nosocomial meningitis were reported.Candida albicans was isolated from 54% of the patients (7);C. parapsilosis from 23% (3);C. tropicalis from 15% (2) andC. krusei from 8% (1).C. albicans was isolated from the cerebrospinal fluid (CSF) of five children with the following diagnoses: nonspecified tumor of the central nervous system, Hodgkin's disease, meningitis, suspect neuroinfection, and sepsis. Examination of CSF allowed us to detect 2 strains ofC. albicans from adult patients, one after neurosurgery because of a brain tumor and one with a vascular disease of the brain.C. parapsilosis was found in CSF from two premature children and one child with epilepsy. Two isolates ofC. tropicalis were obtained from both blood and the CSF of a child from the neonatal intensive care unit and from a child from pediatric oncology with multiple malignant neoplasms. Only one strain ofC. krusei was found in the oral cavity and CSF of a patient after neurosurgery performed after head trauma.  相似文献

Each of seven pathogenic species of Candida exhibits a unique pattern of light and dark recovery responses to ultraviolet irradiation. C. guilliermondii, C. parapsilosis and C. pseudotropicalis photoreactivate whereas C. albicans, C. krusei, C. stellatoidea and C. tropicalis do not. Within eachof these groups, individual species are distinguishable by whether or not they express differential dark recovery during postirradiation growth at 25 C or 37 C on oxidative vs fermentative carbon sources, on inorganic vs amino acid nitrogen sources or in the presence rather than absence of ergosterol. Equivalent recovery patterns are obtained for species of Candida and the ascosporogenous species which are their corresponding perfect forms. These observations indicate strongly that the postirradation recovery is a reliable, species-specific characteristic of yeasts.These studies were added in part by a contract (AT11-1)-1772 with the U.S. Atomic Energy Commission.  相似文献

The yeasts of patients with oral cancer has been studied before and during Xr-therapy. Gram and PAS smears revealed an increase of yeast-like structures, during treatment, from 56% to 66% of the cases. Before radiotherapy oral yeasts were isolated from 56% of the patients with cancer represented by Candida albicans (30%); C. tropicalis (12%); C. glabrata and C. krusei (4%), besides six other different species (2%). During radiotherapy yeasts were isolated in 72% of the cases, as follow: C. albicans (36%); C. tropicalis (16%); Rhodotorula rubra (6%); C. kefyr; C. krusei and Pichia farinosa (4%), besides other nine species (2%). C. albicans serotype A represented 93% of the isolated samples, before treatment and 88,8% during Xr-therapy.  相似文献

The proportion ofCandida and non-Candida species in the clinical material from patients. with respiratory-tract diseases was determined.C. albicans was isolated in 102 cases. An additional 89 strains of yeasts, isolated in association with respiratory diseases, belonged to 10 non-albicans Candida spp. andCryptococcus spp. The prevailing species, which occurred in 47 cases, wasC. parapsilosis. C. tropicalis, C. glabrata, andC. guilliermondii were isolated in 12, 10, and 9 cases, respectively. C. stellatoidea differs from both C. albicans and C. tropicalis in its i) much greater growth differential on minimal and amino acid enriched media and ii) unique inability to grow on minimal medium containing glycerol as carbon source at 37C. The relative responses to amino acid enrichment occur on media containing either fermentative or oxidative carbon sources, at 25C or 37C. Under any given conditions of carbon source and temperature, different assortments of individual amino acids are stimulatory for each of the three species. All assortments include one or more members of the glutamic acid family. However, sulfur amino acids stimulate only C. stellatoidea on all three carbon sources. On minimal-glycerol medium, wild type strains of C. stellatoidea grow prototrophically at 25C but are auxotrophic for amino acids at 37C; the particular auxotrophies expressed vary from strain to strain. Slow growing, mycelial mutants, prototrophic on glycerol at 37C arise spontaneously in wild type strains at frequencies indicating nuclear gene mutation. Such mutants can be induced by both transition and frame shift mutagens. The implications of these observations for the taxonomic relationships between the three Candida species and for identification of C. stellatoidea in particular are discussed.  相似文献

Forty strains ofCandida and one ofTorulopsis were isolated from patients with cutaneous candidiasis. The isolates comprised 29 strains ofC. albicans, 7 strains ofC. tropicalis, 2 strains ofC. guilliermondii, and one each ofC. parakrusei, C. lipolytica, andT. famata were identified by the ordinary method. Besides the common pathogenC. albicans, a few other species ofCandida may be etiologic organisms of cutaneous candidiasis. These strains were re-examined by combined use of sucrose agar slants and slide agglutination tests with IgG monofactorial antibodies as a rapid identification method, especially for determining serotypes ofC. albicans. The new method was useful and reliable for rapid identification ofC. albicans and related species. All strains ofC. albicans isolated from skin lesions proved to be standard serotypes ofC. albicans.

Neunundzwanzig Stämme vonC. albicans, 7 vonC. tropicalis, 2 vonC. guilliermondii, und je einer vonC. parakrusei, C. lipolytica undT. famata wurden mit dem ordinären Methode identifiziert. Außer dem gewohnlichen Erreger,C. albicans, konnten auch ein Paar andere Spezies vonCandida als den Erreger betrachtet werden. Sechsunddreißig Stämme vonC. albicans undC. tropicalis wurden mit der von uns verbesserten kombinierten serologischen und biologischen Methode untersucht, besonders um den Serotypus vonC. albicans festzusetzen. Die neue Methode war gut und zuverlässig als die rapide Identification vonC. albicans und verwandten Spezies. Alle aus der Hautläsion isoliertenC. albicans waren der in Japan allgemeine Serotypus vonC. albicans.

Summary Protoplasts ofAmmi visnaga initiated cell wall formation within 2 days in culture; after 13 days the new cells were enclosed by a cell wall similar to the walls on the original cultured cells. Budding occurred in protoplasts with little or no detectable cell wall. No evidence was obtained for direct participation of any organelle in cell wall formation. The cytoplasm of regenerating cells contained numerous organelles and appeared typical of actively growing plant cells; they were easily distinguished from degenerate cells and protoplasts. While coated vesicles were common, spiny vesicles occurred in only a few cells. Sustained cell division yielded multicellular aggregates. Multinucleate protoplasts, formed by spontaneous fusion, did not divide; some of them contained annulate lamellae with few pore complexes.Supported by the National Research Council of Canada, Grant A6304.  相似文献

A unicellular cyanophycean culture contaminant had features of both Geitleribactron Kom. and Cyanophanon Geitl. The cells were elongated, sheathless, mostly similar in diameter throughout their length, and attached polarly in rosettes or groups and produced only a single elongated exocyte (=exospore). Young cells were moderately elongated and resembled Geitleribactron. As cells aged, they greatly elongated and then resembled Cyanophanon. Some cells formed Y-shaped bifurcations, features of C. mirabile Geitl. and C. minus Geitl., but they lacked the basal sheath (pseudovagina) of C. mirabile. During exocyte formation, a thick and localized L-II wall layer protuberance extended the exocyte away from the parent cell. This terminal wall thickening then appeared to move to one side from subsequent and unequal cell wall growth. Cells sovnetimes bent abruptly, occasionally opposite a thickening in the L-II wall layer. Further studies in culture of putative Geitleribactron and Cyanophanon isolates are necessary to ascertain the breadth of their structural diversity and the identity of the present taxon.  相似文献

Freeze-fracturing of outer wall layers ofCladosporium conidia revealed two types of ultrastructure, coinciding with taxonomic characteristics. The outer conidial layers were essentially smooth in the human pathogenic species,C. bantianum, C. carrionii, andC. trichoides. In contrast, mosaic arrays of rodlets on conidia were observed with freeze-fracturing in the saprobic species,C. cladosporioides, C. coralloides, C. herbarum, C. sphaerospermum, andC. variabile. Conidia ofC. elatum were an exception among the saprobic species as they had smooth surfaces. The present study supports the suggestion that the human pathogenicCladosporium species should be transferred to another genus.  相似文献

CHROMagar has been reported to be useful for the rapid and accurate identification of Candida species. We tested 135 isolates of Candida species isolated from oropharyngeal candidiasis in HIV patients and found that it was useful in the presumptive identification of Candida albicans and Candida krusei. Occasional strains of C. tropicalis produced colonies with a greenish tinge making it difficult to differentiate from C. albicans.  相似文献
