目的:以原代培养的胚胎大鼠大脑皮质神经细胞进行缺氧缺糖再给氧处理,观察党参总皂苷对神经细胞凋亡的抑制作用。方法:胚胎大鼠大脑皮质神经细胞原代培养,含连二亚硫酸钠的无糖Earle's液进行缺氧缺糖处理造模,MTT法测定细胞存活率,Hoechst33342和PI原位双染法荧光显微镜观察细胞形态学变化和坏死、凋亡细胞百分率,APO-BRDU法流式细胞术检测凋亡细胞百分率,Fluo-3法流式细胞术检测细胞内Ca2+浓度,评价药效。结果:细胞存活率:与模型组犤(19.69±5.80)%犦比较党参总皂苷0.52mg/L组39.21±12.21)%,0.052mg/L组犤(犦犤(37.91±4.67)%犦能显著提高细胞存活率(P<0.001);细胞坏死率:与模型组犤(44.99±12.81)%犦比较党参总皂苷0.52mg/L组犤(15.66±4.67)%犦,0.052mg/L组犤(23.98±4.04)%犦和0.0052mg/L组犤(20.50±5.19)%犦能显著降低细胞坏死率P<0.001);原位双染法细胞凋亡(率:与模型组犤(17.44±4.82)%犦比较党参总皂苷0.52mg/L组犤(10.58±2.04)%犦,0.052mg/L组犤(11.61±2.35)%犦能显著降低细胞凋亡率(P<0.05);流式细胞术检测细胞凋亡率:与模型组犤(8.55±3.84)%犦比较党参总皂苷0.52mg/L组犤(3.85±1.92)%犦,0.0052mg/L组犤(3.45±2.22)%犦能显著降低细胞凋亡率P<0.05);(细胞内Ca2+平均荧光值,与模型组(47.  相似文献

骨缺损修复材料可降解速固化骨水泥结构特征及力学性能   总被引:4,自引:2,他引:4  
目的分析异体脱钙骨基质(DBM)骨粒复合磷酸钙骨水泥(CPC)及氰基丙烯酸正丁酯粘合剂(NBCA)后材料的扫描电镜结构特征及生物力学性能。方法将DBM骨粒与CPC以1:1体积比搅拌均匀,加入适量NBCA,制成DCN骨水泥;扫描电镜观察复合材料结构特征,力学试验机测定材料生物力学性能。结果扫描电镜发现,CPC与DBM均匀复合,CPC被覆DBM表面呈规则有序海绵状,其中存在较多大小不规则、相互连通的自然裂隙;材料的抗压及拉伸极限强度较CPC明显提高。结论DCN骨水泥具有相互联通的自然裂隙及潜在的骨粒通道,并可提供较好的机械支持和固定作用的骨修复材料。  相似文献

目的探讨异体脱钙骨基质颗粒复合骨水泥修复严重粉碎性骨折的效果。方法制造兔右股骨粉碎性骨折的动物模型,a组用异体脱钙骨基质颗粒复合骨水泥并表面贴附自体骨渣进行修复;b组用异体脱钙骨基质颗粒复合骨水泥进行修复;c组单纯植入骨水泥进行修复作为对照组。分别于术后不同时间行X射线照相、99mTc-MDP骨扫描和组织学观察。结果a组和b组4周开始形成骨痂,12周骨痂最丰富,24周复合材料与宿主骨边缘模糊,边缘部分的异体脱钙骨基质颗粒被新生骨替代,并与宿主骨融合。a组较b组骨痂形成快。99mTc-MDP骨扫描计数a组大于b组,b组大于c组(P<0.01)。结论异体脱钙骨基质颗粒复合骨水泥易塑形,并具有生物学活性,能修复粉碎性骨折,表面贴附自体骨渣进行修复效果更为理想。  相似文献

AIM:To analyze constructive and biomechanical properties of different quality ratio material impregnated decalcified bone matrix(DBM) with bone cement(BC).METHODS:The DBM particles and the materials impregnated 0mg/g,300mg/g and 400mg/g mass ratio DBM particles with BC were made according to the methods of Urist et al.The compound material constructions were observed by scanning electron microscope and the biomechanical properties were measured by Instron mechanics test-ing-machine.RESULTS:The DBM particles with irregular gaps existing within interspace were connected with BC by the multipoint mode in the compound material.The ultimate compressive strength were 0mg/g DBM in (59.3&;#177;2.2)MPa,300mg/g in(27.1&;#177;1.8)MPa,400mg/g in (19.3&;#177;1.6)MPa.The ultimate bending strength were 0mg/g in (54.3&;#177;3.7)MPa,300mg/g in (18.5&;#177;1.1)MPa,400mg/g in (13.3&;#177;1.4)MPa.CONCLUSION:The materials of DBM impregnated with BC had perfect plastic property with much more irregular gaps existing within interspace.The materials could provide abundant biomechanical support.  相似文献

目的观察活性煅烧骨(TBC)复合骨水泥(BC)修复骨缺损后的血管化及结构变化。方法将活性TBC和BC复合后植入兔尺骨缺损部位,不同时间行印度墨汁动脉血管灌注及材料断面扫描电镜观察,观察血管生成情况及结构变化。结果墨汁灌注2周时见TBC周围血管形成密集,并向材料中长入,4、8和12周时新生血管逐渐增多,并深入材料中央。电镜观察4周时TBC周围及内部孔隙被胶原纤维束充填,部分胶原矿化。8~12周新生骨组织附于TBC颗粒及BC表面,原孔隙接近消失,TBC颗粒少量降解。结论活性TBC复合BC具有骨诱导作用,新生血管及新生骨组织生成良好,是理想的骨缺损修复材料。  相似文献

目的通过磷酸钙骨水泥(CPC)复合同种异体脱钙骨基质颗粒(DBM)及骨形态发生蛋白(bBMP)修复皮质骨缺损,探讨其对骨缺损的修复情况。方法在兔前肢桡骨制作长1cm的骨缺损,填充CPC与复合bBMP异体同种的DBM的复合物,应用大体标本观察、X线摄片、组织学观测、印度墨汁灌注等方法观察其对骨缺损的修复情况,对侧以单纯的CPC做对照。结果第2周开始复合材料组有血管长入,第4周X线片上显示可见、骨痂形成,12周后,复合材料与两端的骨质愈合良好,局部板层骨致密,哈夫氏系统排列整齐。结论磷酸钙骨水泥复合bBMP及同种异体脱钙骨颗粒修复、皮质骨缺损有较好的效果。  相似文献

目的分析不同质量比的牛煅烧骨颗粒复合骨水泥结构特征及生物力学性能.方法把提取过bBMP的牛骨颗粒制成煅烧骨,以不同的质量比与骨水泥混合制成复合材料,测定其生物力学性能与电镜下结构.结果复合材料呈均匀混合分布并多点状结合,其中存在较多的裂隙;含牛煅烧骨颗粒质量比为500mg/g的复合材料的生物力学性能最为适宜.结论牛煅烧骨颗粒复合骨水泥后具有新骨长入的通道,含煅烧骨质量比为500mg/g的复合材料生物力学性能良好,能作为支架材料修复骨缺损.  相似文献
