The concepts of biocitizenship and biosociality, in many ways developed as a reaction to the former critique of genetification and fears of a return of eugenics, have gained a stronghold in much of the current debates on the social effects of modern-day genetics. In contrast to claims of a return to eugenics, the literature on biocitizenship highlights the new choice-enhancing possibilities involved in present-day biomedicine, underlining the break with past forms of biopower. In this analysis, hope becomes a life-inducing and vitalizing force, opening new avenues of civic participation and engagement. Most critics of this analysis have attacked the claims to novelty attributed to these concepts, arguing that more traditional forms of biopower remain as important as ever. In contrast, we argue that the biocitizenship literature underestimates the radical nature of this break with the past, ending up with a too narrow and one-sided interpretation of the ramifications of the new discourse of hope. On the basis of two different case stories, the “Portraits of Hope” campaign from California, USA and the “Mehmet Case” from Norway, we indicate an alternative “darker” reading of the new discourse of hope, arguing that its driving force is not so much future possibilities as present despair.  相似文献

The issues of nativity and identity of Linnaean Physalis minima are long-debated while the use of the name P. maxima Mill. and its report from India are recent. The available evidence indicates that the name “P. minima L.” is misapplied to two different elements, viz., P. angulata L. and P. lagascae Roem. & Schult. The name Physalis minima L. may be rejected as nomen confusum, for which the paper provides the primary information. As on today, it is submerged under the synonymy of P. angulata L. The correct name for the widely known P. minima is P. lagascae. The name “P. maxima Mill.” applied to the escape and naturalized weed in the Indian subcontinent and elsewhere is to be substituted by P. pruinosa L., a name misapplied to P. grisea (Waterf.) M. The origins of field guides and other plant identification manuals have been poorly understood until now because little attention has been paid to 18th century botanical identification guides. Identification manuals came to have the format we continue to use today when botanical instructors in post-Revolutionary France combined identification keys (step-wise analyses focusing on distinctions between plants) with the “natural method” (clustering of similar plants, allowing for identification by gestalt) and alphabetical indexes. Botanical works featuring multiple but linked techniques to enable plant identification became very popular in France by the first decade of the 19th century. British botanists, however, continued to use Linnaeus’s sexual system almost exclusively for another two decades. Their reluctance to use other methods or systems of classification can be attributed to a culture suspicious of innovation, anti-French sentiment and the association of all things Linnaean with English national pride, fostered in particular by the President of the Linnean Society of London, Sir James Edward Smith. The British aversion to using multiple plant identification technologies in one text also helps explain why it took so long for English botanists to adopt the natural method, even after several Englishmen had tried to introduce it to their country. Historians of ornithology emphasize that the popularity of ornithological guides in the 19th and 20th centuries stems from their illustrations, illustrations made possible by printing technologies that improved illustration quality and reduced costs. Though illustrations are the most obvious features of late 19th century and 20th century guides, the organizational principles that make them functional as identification devices come from techniques developed in botanical works in the 18th century.  相似文献

There has been a great deal of interest in the regulation of muscle contraction. Prior biochemical studies have demonstrated that the binding of regulated actin to S-1-ATP is unchanged at low Ca2+, even though the ATPase activity of regulated actomyosin is inhibited under these conditions. Prior structural studies using X-ray diffraction techniques have suggested that the tropomyosin-troponin complex may move and inhibit the actomyosin interaction at low Ca2+ (i.e., steric blocking). In physiologic fiber experiments, “weak” binding crossbridges have been found to bind to the actin filament at low Ca2+, especially at low ionic strength, and other experiments have suggested that Pi release is not directly regulated by calcium. In biochemical studies in the absence of ATP, inhibition of the binding of strong binding states have been reported in both equilibrium and transient kinetic studies. The current work suggests that all of these observations can be explained in terms of a six-state model in which regulation affects one particular actomyo sin state that contains both strongly bound ADP and Pi. This further implies that regulation affects both a kinetic transition as well as a weak binding constant.  相似文献

Impurities of free aromatic amino acids (Phe and Tyr) and the elastin protein were found in the heparin commercial drug (Hep) by spectral luminescent and spectrophotometric methods. The fluorescence quenching of the Trp, Tyr, and Phe amino acids by the Hep drug was studied, and the Stern-Folmer constants (K) that reflected stability of the Hep complexes with amino acids were determined. The stability of AA-Hep complexes increased in the following sequence: Trp < Tyr < Phe (K = 19 ± 2 < 39 ± 3 < 710 ± 70 M?1, respectively). These values probably determined the dominant contribution of the phenylalanine impurity in the heparin drug. The contamination of animal elastin whose structure differed from that of the human elastin is thought to be a reason for allergic reactions and even anaphylactic shock during medical treatment with this drug.  相似文献

There is a widespread assumption, even among those who reject the Synthetic Theory of Evolution, that the form of “modern”Homo sapiens is somehow superior to that of archaicHomo sapiens (Tattersall 2000). Those who accept the general outlook of evolutionary biology also tend to assume that “modern” form emerged because it was selected for, which also implies that it was better than that which preceded it. However, after years of using craniofacial measurements to compare human populations, I finally came to realize that, with only a few exceptions, the dimensions measured have no relation to differences in adaptation (Brace 1989, 1996, 2000; Brace et al., 1993). Elsewhere the conclusion has been supported that what is shown by craniometrics is selectively neutral on the average (Relethford 2002). With the documentation that approximately 95% of the genome is not functional, molecular genetics has proved to be useful in documenting the length of time of separation of related human populations by investigating the differences that have accumulated in the neutral parts of the genome. Not surprisingly, the picture revealed by the study of genetic differences is very similar to the one revealed by the study of craniometric differences (Brace et al., 2001). For this reason, the logic behind the “neutral theory” in molecular genetics is very similar to that applied to what happens to morphological characteristics when selection ceases (Brace 1963; Kimura 1968). The difference is that random changes in the neutral part of the genome have no other consequences. However, random changes in the genes that produce specific aspects of morphology will be visible even when selection is no longer controlling the particular trait in question. From an assessment of what random changes in genes controlling morphological traits are likely to do, it follows that the most likely change will probably be a reduction in the trait in question, i.e. the Probable Mutation Effect will produce structural reduction. When survival in the temperate zone during the last glaciation dependend on “obligatory cooking”, one of the unintended consequences was a reduction in the selective pressures maintaining a Middle Pleistocene-sized dentition. The result was a gradual reduction in tooth size and a conversion, of a Neanderthal-sized face into one of “modern” dimensions. The manufacture and use of string for snares and nets similarly reduced the selective pressures maintaining post-cranial levels of robustness and muscularity. The reduction in the latter resulted in the emergence of moderm post-cranial robustness out of what had been a Neanderthal level,in situ wherever the technology can be documented and without any need for invasions and replacements.  相似文献

Mitochondrial Complex II (succinate:ubiquinone oxidoreductase) is purified in a partially inactivated state, which can be activated by removal of tightly bound oxaloacetate (E.B. Kearney, et al., Biochem. Biophys. Res. Commun. 49 1115–1121). We crystallized Complex II in the presence of oxaloacetate or with the endogenous inhibitor bound. The structure showed a ligand essentially identical to the “malate-like intermediate” found in Shewanella Flavocytochrome c crystallized with fumarate (P. Taylor, et al., Nat. Struct. Biol. 6 1108–1112) Crystallization of Complex II in the presence of excess fumarate also gave the malate-like intermediate or a mixture of that and fumarate at the active site. In order to more conveniently monitor the occupation state of the dicarboxylate site, we are developing a library of UV/Vis spectral effects induced by binding different ligands to the site. Treatment with fumarate results in rapid development of the fumarate difference spectrum and then a very slow conversion into a species spectrally similar to the OAA-liganded complex. Complex II is known to be capable of oxidizing malate to the enol form of oxaloacetate (Y.O. Belikova, et al., Biochim. Biophys. Acta 936 1–9). The observations above suggest it may also be capable of interconverting fumarate and malate. It may be useful for understanding the mechanism and regulation of the enzyme to identify the malate-like intermediate and its pathway of formation from oxaloacetate or fumarate.  相似文献

Four rheumatic diseases—ankylosing spondylitis, the arthritis accompanying ulcerative colitis or regional enteritis, psoriatic arthropathy, and Reiter''s syndrome—formerly considered to be forms of rheumatoid arthritis, are now distinguished from that disorder and should be recognized by the physician as entities. These arthritides may be distinguished from each other by a number of clinical and radiographic characteristics, principally (1) the roentgenographic appearance of the spine when spondylitis is present, (2) the location of periosteal new bone formation, (3) the location of arthritis in the joints of the limbs, and (4) the presence of characteristic skin lesions.  相似文献

El Chichón is an active volcano located in the Chiapas Volcanic Arc characterized by high temperatures, low pH, and large heavy metal concentrations. High temperatures are known to affect microorganisms, so the bacterial community structure was investigated in crater-lake sediment at a ‘low’ 50?°C and a ‘high’ 92?°C by sequencing the 16S rRNA bacterial gene. Most metal concentrations, and bacterial richness, and diversity indexes were higher at 50?°C than at 92?°C. Fifteen phyla were found in the sediment at 50?°C dominated by Actinobacteria (33.1%), Proteobacteria (29.1%) and Acidobacteria (20.1%). Nine phyla dominated by Firmicutes (52.7%, mostly Alicyclobacillus and Sulfobacillus) and Proteobacteria (44.8%, mostly Bradyrhizobium, Methylobacterium, Sediminibacterium) were detected in the sediment at 92?°C. The predictive functional profiling indicated metabolic pathways related to amino acid metabolism, membrane transport, replication, and repair as the most important. It was found that although a large number of bacterial groups were well adapted to 92?°C, the higher temperature reduced strongly, the bacterial diversity and species richness in the El Chichón volcano crater-lake system, and altered the bacterial community structure and their functionality.  相似文献

The zooplankton composition is studied in the thermokarst, glacial and meteorite lakes, channels, former riverbeds, and hollows in the basin of Anadyr’. We found 174 taxa: 78, Rotatoria, 55, Cladocera, and 41, Copepoda. The most diverse is the lake fauna: 51 taxa of Rotatoria, 48, Cladocera, and 37, Copepoda. The thermokarst Lake Maiorskoe hosts 68 taxa: 31, Rotatoria, 14, Cladocera, and 23, Copepoda, wheras the cold ultraoligotrophic Lake El’gygytgyn features only one species of Cyclop of the group scutifer Cyclops neymanae Strel., and Rotatoria and Cladocera are present as allochtonous forms. The Copepoda illustrate the relations of the Anadyr’ fauna with those of Europe, North America, and Japan.  相似文献

In different medical systems, notions of person, as patient and as healer, vary widely. An analysis of the talk of a practitioner of internal medicine in the U.S.A. reveals an understanding of patients as syndromes. The work of medicine is physiological integrity, distinct from personal and interpersonal integrity. It is suggested that the world view of this practitioner is consistent with the institutions of medicine in which he practices, and with historical movements in U.S. society and Western civilization more broadly.  相似文献

In the last decades, the concept of human races was considered scientifically unfounded as it was not confirmed by genetic evidence. None of the racial classifications, which strongly differ in the number of races and their composition, reflects actual genetic similarity and genealogy of human populations inferred from variability of classical markers and DNA regions. Moreover, intercontinental (interracial) variability was shown to be far lower than that within populations: the former constitutes 7 to 10% of the total genetic variation and the latter about 85% of it. It is believed that the low level of differentiation of regional population groups contradicts their race status and suggests a recent origin of humans from one ancestral population. The results of studies of various genetic systems are in agreement with the latter conclusion rejecting the hypothesis of regional continuity. According to this hypothesis, the populations of continents regarded as large races have developed during long evolution from local types of archaic humans, in particular, Neanderthals. Phenotypic similarity of different, sometimes unrelated, populations united into one race is explained by strong selection since race-diagnostic traits characterize body surface and thus are directly subjected to the influence of environmental (primarily climatic) factors. It has been recently established that variability of the most important of these traits, body and hair pigmentation, is largely controlled by one locus (MC1R), which accounts for its high evolutionary lability. Other traits used for race identification are also likely to be labile and controlled by major genes. However, the fact that the currently existing race classifications are groundless does not mean that such classifications are impossible in principle. Commonly used argumentation (races do not exist because populations are not genetically separated) does not hold water. A polytypic species is characterized by genetic continuity of allopatric populations rather than the presence of narrow genetic boundaries between them. Borderlines between races are usually conventional and arbitrary. As to intergroup variation in humans, it is indeed low but comparable with that in a number of other species. There are no obstacles to the development of genetic systematics of human races.  相似文献

An historical background is provided for the term, “aniline dye,” which is still widely used as a synonym for “synthetic dye.” The discovery of aniline and the role of Hofmann in clarifying it are described. The subsequent discovery of mauveine (mauve) by a student of Hofmann's, William Perkin, and his difficulties in transforming an academic synthesis into a commercial product also are discussed. The key role of Scottish dyers, the Pullars of Perth and Thomas Keith in London, in this technology transfer is described. The subsequent ascendancy of the German dyestuff industry over British manufacturers is noted.  相似文献

In 1936, Frank Macfarlane Burnet published a paper entitled “Induced lysogenicity and the mutation of bacteriophage within lysogenic bacteria,” in which he demonstrated that the introduction of a specific bacteriophage into a bacterial strain consistently and repeatedly imparted a specific property – namely the resistance to a different phage – to the bacterial strain that was originally susceptible to lysis by that second phage. Burnet’s explanation for this change was that the first phage was causing a mutation in the bacterium which rendered it and its successive generations of offspring resistant to lysogenicity. At the time, this idea was a novel one that needed compelling evidence to be accepted. While it is difficult for us today to conceive of mutations and genes outside the context of DNA as the physico-chemical basis of genes, in the mid 1930s, when this paper was published, DNA’s role as the carrier of hereditary information had not yet been discovered and genes and mutations were yet to acquire physical and chemical forms. Also, during that time genes were considered to exist only in organisms capable of sexual modes of replication and the status of bacteria and viruses as organisms capable of containing genes and manifesting mutations was still in question. Burnet’s paper counts among those pieces of work that helped dispel the notion that genes, inheritance and mutations were tied to an organism’s sexual status. In this paper, I analyze the implications of Burnet’s paper for the understanding of various concepts – such as “mutation,” and “gene,” – at the time it was published, and how those understandings shaped the development of the meanings of these terms and our modern conceptions thereof.  相似文献
