利用DAPI显带和rDNA-FISH技术对栽培种甘薯(‘徐薯18’)(Ipomoea batatas cv.Xushu No.18)及2种不同产地近缘野生种(Ipomoea hederacea Jacq.)进行了细胞遗传学研究。DAPI核型分析表明,‘徐薯18’核型公式为2n=6x=90=72m+18sm(18SAT),随体位于第1、3、6染色体上;美国近缘野生种核型公式为2n=2x=30=30m(4SAT),香港近缘野生种核型公式为2n=2x=30=20m+10sm(4SAT),随体均位于第6、12染色体上。rD-NA-FISH结果显示,栽培种甘薯基因组中含有3对5SrDNA位点,分别位于着丝粒区、亚着丝粒区和染色体端部;美国近缘野生种基因组中含有2对5SrDNA位点,香港近缘野生种基因组中含有1对5SrDNA位点,均位于随体部位;两种不同地域来源的近缘野生种基因组中均含有2对45SrDNA位点,分别位于第6和第12染色体上。 相似文献

应用常规方法获得了杂种猪( 野猪(♂)×家猪(♀))的核型、C带和174条带纹的G带,应用微量秋水仙素法得到了258条带纹的高分辨G带。结果表明,杂种猪的二倍体细胞染色体数目2n=38,C带具有多态性,G带和高分辨G带与家猪相比无明显差异,它们属于同一种。Abstract:Using usual method,we got karyotype of hybrid pig (wild soar(♂)×domestic pig(♀)),C-band and approximate 174 bands of G-band,and we also obtained approximate 258 bands of high resolution G-band by micro-Colchicin method.The result indicate that the diploid chromosome number is 2n=38;there is polymorphism in C-band,and compared with domestic pig in G-band and high resolution G-band there is no distinguish difference.They belong to the same seed. 相似文献

利用腹腔注射秋水仙素制备肾细胞染色体方法和DAPI(4',6'-diamidino-2-phenylindole)荧光染色的方法,对鳙鱼(Aristichthys,nobills)的染色体组型和染色质的分布进行了研究。结果表明,其二倍体数目为2n=48,核型为30M+14SM+2ST+2T。DAPI荧光染色显示间期细胞核中荧光亮度较为一致,提示异染色质在间期细胞核中分布比较均一。而DAPI荧光染色在第1和第4染色体的短臂上较为明亮,其余染色体上的明亮区都分布在着丝粒区域,表明第1和第4染色体上的异染色质主要集中在染色体的短臂上,其余染色体的异染色质主要分布在着丝粒区域。 相似文献

应用GC碱基特异性荧光染料色霉素A,辅以AT减基特异性荧光染料Hoechst33258,DAPI或喹吖因对鲤鱼,鲫鱼,大鳞副泥鳅和的有丝分裂染色体及黄鳝的有丝分裂和减数分裂染色体进行了荧光显带研究,结果发现,色霉素A3可以特异性地显示鱼类有丝分裂及减数分裂各个时期核仁组织区NORS的存在,Hoechst33258,DAPI或喹吖因则使这些区域(NORs)淡染,大鳞副泥鳞的染色体NORs 分布位置具有性别,根据实验结果,对有关鱼类染色体的荧光染色研究及其应用进行了讨论。 相似文献

目的:蕹菜(Ipomoea aquatica Forsk.)Cd(镉)积累典型品种对Cd、Pb(铅)、C(r铬)、N(i镍)等多种重金属的吸收积累及相互关系。方法:采用盆栽试验,分析4个蕹菜Cd积累典型品种在6种土壤上的两茬茎叶及根Cd、Pb、Cr、Ni含量及相关性。结果:①品种和土壤对供试蕹菜典型品种Cd含量的效应均达显著水平(P<0.05),对Pb、Cr和Ni含量的效应因重金属、收获时期及部位而不同,二者对Cd、Pb含量具一定的交互效应。②两茬茎叶Cd含量平均值均为T308>GDB>QLB>QLQ,根Cd平均含量高于茎叶Cd平均含量;除Cr外,根Pb和Ni平均含量均高于茎叶。③Cd、Pb、Cr、Ni含量呈现复杂的相关关系。茎叶Cd含量与Pb含量正相关,且第一茬相关性极显著(P<0.01);Pb含量与Ni含量相关关系明显,第一茬茎叶、根Pb含量与Ni含量的正相关达到显著(P<0.05)或极显著水平(P<0.01),但第二茬茎叶Pb含量与Ni含量却显著负相关(P<0.05);Pb含量与Cr含量的相关性仅第一茬茎叶显著(P<0.05);Cr含量与Ni含量的相关性仅第二茬茎叶达极显著水平(P<0.01)。结论:蕹菜典型品种对Cd、Pb、Cr、Ni的吸收积累存在协同和拮抗两种作用。 相似文献

在邻苯二甲酸(2-乙基己基)酯(DEHP)不同污染程度的水稻土上盆栽不同品种通菜,应用GC/MS联机检测技术研究了通菜-土壤系统中DEHP的环境行为与归宿.结果表明,通菜中DEHP的含量为0.335~12.710mg·kg^-1,与土壤的污染程度呈正相关.不同品种通菜之间对DEHP的吸收累积效应存在显著差异,与其叶子大小存在一定的正相关关系.种植不同品种通菜后土壤中DEHP的残留量(1.424~19.834mg·kg^-1)存在显著差异.通菜对土壤中DEHP的BCFs都小于1.0,与土壤的污染程度呈负相关.不同通菜品种的BCFs之间存在显著差异,中等叶子通菜品种的BCFs相对较大。 相似文献

Reductants are often used to reduce Cr(VI) in chemical treatments, yet the effects of the reductants on Cr(VI) phytoremediation are not fully understood. This study investigates the effects of different reductants on Cr(VI) phytoremediation by Ipomoea aquatica in simulated solution with 3 mg L?1 of Cr(VI), pH0 of 6, and an incubation time of 5 days. Results indicate that the applications of S2O32?, Fe0, and Fe2+ at low doses notably increased root Cr concentrations, which were obviously higher than that those in the control (Cr6+ alone). However, high reductant concentrations decreased bioaccumulation of Cr in the roots and shoots of the plant.Statistical results indicate that Cr concentrations were significantly and negatively correlated with Fe concentrations in the roots and shoots of the plant (p < 0.05). This suggest that Fe accumulation inhibited Cr accumulation in the plant. A Cr(VI) concentration of 3 mg L?1 caused short, brown lateral roots with tip necrosis, leaf chlorosis, and noticeable shoot wilting. The leaf necrosis and shoot wilting is caused by oxidative damage of lateral roots by Cr(VI) rather than by the reactive oxygen species generated by the oxidative stress. Addition of the reductants effectively reduced these plant injuries. 相似文献

Thioredoxin reductase 1 (Trr1) from Saccharomyces cerevisiae is a member of the family of pyridine nucleotide‐disulfide oxidoreductases capable of reducing the redox‐active disulfide bond of the cytosolic thioredoxin 1 (Trx1) and thioredoxin 2 (Trx2). NADPH, Trr1 and Trx1 (or Trx2) comprise the thioredoxin system, which is involved in several biological processes, including the reduction of disulfide bonds and response to oxidative stress. Recombinant Trr1 was expressed in Escherichia coli as a His6‐tagged fusion protein and purified by nickel‐affinity chromatography. The protein was crystallized using the hanging‐drop vapour‐diffusion method in the presence of PEG 3000 as precipitant after treatment with hydrogen peroxide. X‐ray diffraction data were collected to a maximum resolution of 2.4 Å using a synchrotron‐radiation source. The crystal belongs to the centred monoclinic space group C2, with unit‐cell parameters a = 127.97, b = 135.41, c = 75.81 Å, β = 89.95°. The crystal structure was solved by molecular‐replacement methods and structure refinement is in progress. 相似文献

Reaction of 5,6-dihydro-5,6-epoxy-1,10-phenanthroline (L) with Cu(ClO4)2·6H2O in methanol in 3:1 M ratio at room temperature yields light green [CuL3](ClO4)2·H2O (1). The X-ray crystal structure of the hemi acetonitrile solvate [CuL3](ClO4)2·0.5CH3CN has been determined which shows Jahn-Teller distortion in the CuN6 core present in the cation [CuL3]2+. Complex 1 gives an axial EPR spectrum in acetonitrile-toluene glass with g|| = 2.262 (A|| = 169 × 10−4 cm−1) and g⊥ = 2.069. The Cu(II/I) potential in 1 in CH2Cl2 at a glassy carbon electrode is 0.32 V versus NHE. This potential does not change with the addition of extra L in the medium implicating generation of a six-coordinate copper(I) species [CuL3]+ in solution. B3LYP/LanL2DZ calculations show that the six Cu-N bond distances in [CuL3]+ are 2.33, 2.25, 2.32, 2.25, 2.28 and 2.25 Å while the ideal Cu(I)-N bond length in a symmetric Cu(I)N6 moiety is estimated as 2.25 Å. Reaction of L with Cu(CH3CN)4ClO4 in dehydrated methanol at room temperature even in 4:1 M proportion yields [CuL2]ClO4 (2). Its 1H NMR spectrum indicates that the metal in [CuL2]+ is tetrahedral. The Cu(II/I) potential in 2 is found to be 0.68 V versus NHE in CH2Cl2 at a glassy carbon electrode. In presence of excess L, 2 yields the cyclic voltammogram of 1. From 1H NMR titration, the free energy of binding of L to [CuL2]+ to produce [CuL3]+ in CD2Cl2 at 298 K is estimated as −11.7 (±0.2) kJ mol−1. 相似文献
