目的 探讨胃癌组织中Hedgehog(Hh)信号通路分子(Shh和Gli1)与基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)的表达以及临床意义.方法 采用免疫组织化学方法检测54例人胃癌组织、癌旁组织和30例正常胃黏膜组织中Shh、Gli1和MMP-9蛋白的表达.结果 胃癌组织中Shh、Gli1和MMP-9的阳性表达率分别为68.52%、61.11%和64.81%,明显高于癌旁组织(阳性表达率分别为27.78%、24.07%和22.22%)和正常组织(阳性表达率分别为36.67%、26.67%和33.33%)(P<0.05);三者的表达均与胃癌患者性别、年龄、部位无关(P>0.05);而与分化程度、浸润深度以及淋巴结转移相关(P <0.05);Shh、Gli1分别与MMP-9表达呈正相关.结论 Hedgehog( Hh)信号通路可能通过上调MMP-9的表达促进胃癌的侵袭转移.联合检测胃癌组织中Shh、Gli1和MMP-9蛋白水平,可作为胃癌预后的客观参考指标.  相似文献

目的:研究C型凝集素结构域家族1成员B(CLEC1B)在肝细胞癌(HCC)组织中的表达并检测其对肝癌细胞恶性生物学行为的影响。方法:实时荧光定量PCR(qRT-PCR)检测15例HCC患者新鲜癌组织及癌旁正常组织中CLEC1B基因表达差异,免疫组化检测85例HCC组织及癌旁正常组织中CLEC1B蛋白表达,并分析CLEC1B表达与临床病理特征之间的相关性;建立CLEC1B过表达的HepG2稳转细胞株,qRT-PCR及Western blot进行验证;采用克隆形成实验检测HepG2细胞增殖能力变化;Western blot检测Hedgehog信号通路相关因子(Shh、Gli1、Ptch1)蛋白表达情况。结果:HCC组织中CLEC1B表达量较癌旁正常组织明显降低(P<0.05),CLEC1B在HCC中的表达量与肿瘤大小及远处转移有关,且CLEC1B表达越高肿瘤体积相对越小,远处转移概率越低;过表达HepG2细胞CLEC1B表达量,细胞增殖能力明显下调(P<0.05),且细胞中的Shh、Gli1、Ptch1蛋白表达量明显降低(P<0.05)。结论:CLEC1B在肝细胞癌中表现...  相似文献

Shh(Sonichedgehog)作为最重要的Hh同源基因产物,它的异常激活与胃癌的发生、发展关系密切成为近年研究热点。经典的Shh信号通路是由Shh配体、Ptch和Smo组成的受体复合物以及下游转录因子Gli蛋白(Glil、G1i2、Gli3)组成。研究发现相比正常组织,胃癌组织中Shh的表达都有不同程度的升高,且升高程度与胃癌的发展转移呈正相关,本文就Shh信号通路与胃癌的研究进展做一综述。  相似文献

摘要:Hedgehog(Hh)信号通路在脊椎动物的各种生理过程中都发挥着重要作用,包括细胞的生长和分化等。Sonic hedgehog(Shh)信号通路是 Hh信号通路中研究最深入的成员之一,主要在真核生物中发挥生物学功能。国内外多项研 究表明,人类各种肿瘤的发生发展都涉及Shh信号通路,多种因素引起的Shh异常活化会导致人体系统多种恶性肿瘤的 发生。神经胶质瘤相关癌基因同源蛋白(glioma-associatedoncogenehomologue,Gli)是 Shh信号通路的关键成员之一, 参与调控肿瘤的发生和进展。该文总结了Shh信号通路中 Gli蛋白激活的分子机制及其在肿瘤发生发展中的作用,并 讨论了 Gli蛋白与肿瘤干细胞的相互作用,以及 Gli蛋白靶向治疗相关肿瘤的新思路。  相似文献

目的研究Hedgehog信号通路核心蛋白Sonic hedgehog(Shh)和Patched(Ptch)在肝门部胆管癌中的表达及与肿瘤侵袭性和根治性切除术后肿瘤复发的关系,探讨其相关分子机制。方法采用免疫组织化学技术检测44例肝门部胆管癌、7例胆管炎性病变组织中Shh和Ptch的表达,分析Shh、Ptch与肝门部胆管癌临床病理特征及根治术后复发的关系。应用Kaplan-Meier法对肿瘤根治术后至肿瘤复发时间跨度进行单因素分析,并经Log-rank检验;选择可能对肝门部胆管癌患者预后产生影响的临床病理指标进行量化,依次引入COX风险比例模型进行多因素回归分析。结果肝门部胆管癌Shh和Ptch阳性表达强度明显高于胆管炎性病变组织;Shh和Ptch表达与肝门部胆管癌侵袭性呈正相关;Shh和Ptch表达与肝门部胆管癌根治术后早期复发呈正相关;单因素生存分析显示Shh和Ptch高表达组肝门部胆管癌患者根治术后复发时间更短(P<0.01);多因素回归分析显示Shh和Ptch表达是影响肝门部胆管癌患者根治术后复发的独立因素。结论 Shh和Ptch表达与肝门部胆管癌根治术后短期复发有关,Shh和Ptch可以作为预测胆管癌根治术后复发的独立影响因子。  相似文献

目的:研究Hh信号通路抑制剂Cyclopamine作用Hela细胞后Gli1、FoxM1、上皮间质转化(EMT)过程相关标记物表达的改变及对其增殖、侵袭迁移能力的影响,探究Gli1、FoxM1、EMT过程的可能作用关系,以期为宫颈癌的分子靶向治疗提供多个靶点。方法:采用实时定量RT-PCR、Western Blot技术检测Hela、Siha细胞系Hh信号通路中Shh、Ptch1、Smo的表达情况,选择Hh信号通路表达较高的Hela细胞系作为后续实验细胞,利用四甲基偶氮唑蓝比色法(MTT法)检测不同浓度Cyclopamine作用Hela细胞后其增殖能力的改变;采用Transwell小室试验检测Hela细胞侵袭、迁移能力的改变,并采用实时定量RT-PCR、Western Blot技术检测Gli1、FoxM1、EMT过程标记物的表达情况。结果:(1)Hela细胞中Hh信号通路成分(Shh、Ptch1、Smo)在mRNA水平显著高于Siha细胞(P<0.01),且相应蛋白的表达强度也高于Siha细胞;(2)Cyclopamine对Hela细胞增殖的抑制作用具有时间、剂量依赖性;(3)Cyclopamine作用Hela细胞48h后,细胞侵袭试验显示,实验组侵袭细胞数较对照组明显降低(P<0.01)。细胞迁移试验显示,实验组细胞迁移细胞数较对照组明显降低(P<0.01);(4)Cyclopamine作用Hela细胞48h后,实验组Gli1、FoxM1、Vimentin在mRNA水平的表达显著低于对照组(P<0.05),且相应蛋白的表达强度也低于对照组;E-cadherin在实验组中mRNA层面的表达显著高于对照组(P<0.05),且相应蛋白的表达强度也高于对照组。结论:抑制Hh信号通路可显著降低Hela细胞系增殖、侵袭迁移能力,FoxM1很可能是Hh信号通路末端转录因子Gli1的下游靶基因,且其对宫颈癌细胞系增殖、侵袭迁移的作用很可能是通过EMT实现的。  相似文献

目的 探讨Hedgehog(Hh)信号通路中效应蛋白在人大肠癌、正常大肠组织中的表达及与病理特征的相关性.方法 采用免疫组化方法检测60份大肠癌和20份正常大肠组织中Shh、Ptchl和Glil蛋白的表达,并与临床病理因素进行相关性分析.结果 Shh、Ptchl和Glil蛋白在大肠癌组织中均为高表达,Shh、Ptchl蛋白与Gli1蛋白的表达呈正相关.结论 综合检测Shh、Ptchl和Gli1 蛋白对大肠癌的诊断、治疗及预后有较明显的指导意义.  相似文献

Objective: To evaluatel the value of D-dimers in patients with acute aortic dissection (AAD). Methods: This study consisted of 16 patients with AAD and 27 non-AAD patients. Serum D-dimets were measured by Sta-Liatest D-DI immunoturbidimetric assay. Results: D-dimer level was higher (P < 0.001) in patients with AAD(7.91 ± 5.52 μg/ml) than that in non- AAD group(1.57±1.24 μg/ml). D-dimer was positive (>0.4 μg/ml) in all patients with AAD and in 10 control group patients (37%). Among patients with acute AAD, D-dimers tended to be higher in Stanford A than in Stanford B (8.67 ± 4.31 μg/ml vs. 3.24±1.27 μg/ml, P <0.01). D-dimer values tended to be higher in more extended disease(3.84 ± 1.65 μg/ml, 8.57 ± 3.58 μg/ml and 11.87 ± 5.69 μg/ml in thoracic aorta, thoracic and abdominal aorta, thoracic and abdominal aorta and iliacal arteries, respectively, P < 0.05 for both 8.57 ± 3.58 and 11.87 ± 5.69 vs. 3.84 ± 1.65 ). Including the control group into the analysis, we found a sensitivity of 100%, a negative predictive value of 100%, and a specificity of 66% and a positive predictive value of 64% for D-dimer in diagnosis of AAD in our patients with suspected AAD. Conclusion: D-dimer was elevated in patients with AAD. A negative D-dimer test result could be useful in excluding AAD.  相似文献

Objective: To set up a simple and reliable rat model of combined liver-kidney transplantation. Methods: SD rats served as both donors and recipients. 4℃ sodium lactate Ringer's was infused from portal veins to donated livers,and from abdominal aorta to donated kidneys, respectively. Anastomosis of the portal vein and the inferior vena cava (IVC) inferior to the right kidney between the graft and the recipient was performed by a double cuff method, then the superior hepatic vena cava with suture. A patch of donated renal artery was anastomosed to the recipient abdominal aorta. The urethra and bile duct were reconstructed with a simple inside bracket. Results: Among 65 cases of combined liver-kidney transplantation, the success rate in the late 40 cases was 77.5%. The function of the grafted liver and kidney remained normal. Conclusion: This rat model of combined liver-kidney transplantation can be established in common laboratory conditions with high success rate and meet the needs of renal transplantation experiment.  相似文献

Shock wave lithotripsy (SWL) is a treatment of choice for upper urinary stones. However, this procedure is inappropriate for obese patients because the focus is often unable to reach the target owing to the limited focal distance in shock wave source. Although treating such patients in a blast path may increase the application length of shock wave source, it's difficult to find this path on the lithotripter monitor. Excess production of reactive oxygen species(ROS)of mitochondrion mediated by hyperglycemia is the common pathogenesis of angiopathic complications of diabetes.TCM holds that the damp from the dysfunction of spleen.kidney and liver is the causative factor of complications of diabetes.This is similar to the mechanism of Ros resulting in angiopathic complications of diabetes.When the angiopathic complications of type II diabetes mellitus(T2DM)are difierentiated as caused by turbid damp in TCM can be explained as ROS.Since the obstruction of pathogenic damp in channels and collaterals is said to be the main pathogenesis,the treating principle should be dissolving the damp to remove the obstruction.  相似文献

Objective To observe blood pressure change with age in salt-sensitive teenagers whose salt sensitivity were determined by repeated testing.Methods Salt sensitivity was determined through intravenous infusion of normal saline combined with volume-depletion by oral diuretic furosemide in 55 teenagers. After five years, salt sensitivity was re-examined and subject blood pressure was followed up. Blood pressure changes in salt-sensitive teenagers were compared to that of non-salt sensitive teenagers over five years.Results After 5 years, the repetition rate of salt sensitivity determined by intravenous saline loading is 92.7%. In teenagers with salt sensitivity on the baseline, both the systolic blood pressure increments and increment rates were much higher than non-salt sensitive teenagers (12.7±12.1 mmHg vs. 2.8±5.2 mmHg, P< 0.01; 12.2%± 12.0% vs. 2.5% ±4.4%, P< 0.001,respectively). There was a similar trend for diastolic blood pressure (8.4 ± 6.4 mmHg vs. 3.7 ± 6.4 mmHg, P = 0.052; 13.2% ±10.6 % vs. 6.8%± 10.1%, P = 0.053, respectively).Conclusions Salt sensitivity determined by intravenous saline loading showed good reproducibility. Blood pressure increments with age were much higher in salt-sensitive teenagers than non-salt sensitive teenagers, especially in terms of systolic blood pressure.  相似文献

Objective: To observe the therapeutic effects in acupunture treatment of primary dysmenorrhea combined with spinal Tui Na, and study its mechanism. Methods: Thirty cases of the treatment group were treated by acupuncture combined with spinal Tui Na, and thirty cases in the control group were treated by routine acupuncture. Results: The total effective rate was 93.3% in the treatment group, and 73.3% in the control group, with a significant difference between the two groups (P<0.05). Conclusions: Acupuncture combined with spinal Tui Na has good prospects for treatment of primary dysmenorrhea.  相似文献
