Atlantic salmon Salmo salar L. pre-smolts were experimentally infected with 2 different isolates of salmonid alphavirus (SAV): a Subtype 1 isolate from Ireland and a Subtype 3 isolate from Norway. Sequential samples of tissue and blood were collected during a period of 20 wk post injection and subjected to virus isolation from kidney tissue and serum, detection of viral nucleic acid in heart tissue and serum by real-time RT-PCR, detection of specific antibodies by virus neutralisation assay, and histopathological examination. Successful reproduction of pancreas disease (PD) was obtained by intraperitoneal (i.p.) injection of both isolates. No mortality was observed post infection in either group, but typical PD histopathological lesions in heart and pancreas tissue were observed with both isolates. The prevalence and severity of lesions in the pancreas, heart, skeletal muscle and brain were similar in both groups with only subtle differences recorded. Re-isolation of virus from kidney tissue was performed at 7 and 14 d post infection (d p.i.) only and was positive for both test groups at both sampling points. Isolation of virus from sera from both groups was positive at 4 to 14 d p.i., but was negative at later sampling points when antibody production had begun. Virus may be detected only during the acute phase using both methods. Specific neutralising antibodies could be detected for both test groups from Day 21 p.i. until the end of the experiment at 140 d p.i. Peak antibody titres were seen 70 d p.i. Using real-time RT-PCR, pancreas disease virus (PDV)-specific RNA was detected frequently in serum samples up to 14 d p.i. and occasionally thereafter. In contrast, viral RNA could still be detected in the heart tissue of fish from both groups for at least 140 d p.i.  相似文献

Heart and skeletal muscle inflammation (HSMI) is a disease that affects farmed Atlantic salmon Salmo salar L. several months after the fish have been transferred to seawater. Recently, a new virus called piscine reovirus (PRV) was identified in Atlantic salmon from an outbreak of HSMI and in experimentally challenged fish. PRV is associated with the development of HSMI, and has until now only been detected in Atlantic salmon. This study investigates whether the virus is also present in wild fish populations that may serve as vectors for the virus. The virus was found in few of the analyzed samples so there is probably a more complex relationship that involves several carriers and virus -reservoirs.  相似文献

The extent and effect of disease interaction and pathogen exchange between wild and farmed fish populations is an ongoing debate and an area of research that is difficult to explore. The objective of this study was to investigate pathogen transmission between farmed and wild Atlantic salmon (Salmo salar L.) populations in Norway by means of molecular epidemiology. Piscine reovirus (PRV) was selected as the model organism as it is widely distributed in both farmed and wild Atlantic salmon in Norway, and because infection not necessarily will lead to mortality through development of disease. A matrix comprised of PRV protein coding sequences S1, S2 and S4 from wild, hatchery-reared and farmed Atlantic salmon in addition to one sea-trout (Salmo trutta L.) was examined. Phylogenetic analyses based on maximum likelihood and Bayesian inference indicate long distance transport of PRV and exchange of virus between populations. The results are discussed in the context of Atlantic salmon ecology and the structure of the Norwegian salmon industry. We conclude that the lack of a geographical pattern in the phylogenetic trees is caused by extensive exchange of PRV. In addition, the detailed topography of the trees indicates long distance transportation of PRV. Through its size, structure and infection status, the Atlantic salmon farming industry has the capacity to play a central role in both long distance transportation and transmission of pathogens. Despite extensive migration, wild salmon probably play a minor role as they are fewer in numbers, appear at lower densities and are less likely to be infected. An open question is the relationship between the PRV sequences found in marine fish and those originating from salmon.  相似文献

Many coho salmon stocks (Oncorhynchus kisutch) have been in decline during the past three decades. Canada’s most endangered salmon stock, the Thompson River coho salmon, is being studied extensively as managers attempt to reverse these population declines. Investigators are using acoustic telemetry to track the migratory behaviour and survival of the Thompson River (and other) coho salmon stocks. Coho salmon pre-smolts are relatively small compared with salmonid species that are typically studied using acoustic telemetry; therefore the identification of the appropriate sizes of fish and tags to use is critical. This study tested the effects of surgically implanting the three smallest sizes of acoustic tags currently available on the growth, survival, tag retention, swimming performance and physical condition of coho salmon pre-smolts for 300 days post-surgery. Maximum tag size to body size ratios ranged from 15–17% by fork length and 7–8% by mass for the three tag sizes (11 cm fork length for a 6 × 19 mm tag, 12.5 cm for a 7 × 19 mm tag, and 14 cm for a 9 × 21 mm tag). Based on our results, it is unlikely that coho salmon pre-smolts implanted with acoustic transmitters following these size guidelines would have poor survival in studies of freshwater migratory behaviour as a result of the surgery or the tag.  相似文献

Atlantic salmon Salmo salar L. parr and pre-smolts from 2 Norwegian hatcheries showed reduced weight gain, abnormal behaviour and signs of circulatory disturbances. Necropsy revealed conspicuous fat deposits around the heart to be the most consistent finding. Furthermore, the ventricle/atrium ratio was altered, with the size of the ventricle significantly smaller than normal in affected fish. Histology showed poor development or absence of the outer, compact myocardium, large numbers of fat cells and melanomacrophages in the epicardium, fibrosis, and inflammation of the compactum/spongiosum interphase. Nuclei of the inner spongious myocardium showed signs of compensatory hypertrophy. The cause(s) of this malformation is(are) unknown, but a high prevalence of other malformations in fish from the same population indicates high temperature during incubation of the eggs as a possible aetiology.  相似文献

Salmonid alphaviruses (SAVs), which include the aetiological agents of salmon pancreas disease (SPD) in farmed Atlantic salmon Salmo salar L. and sleeping disease (SD) in rainbow trout Oncorhynchus mykiss (Walbaum), are significant viral pathogens of European salmonid aquaculture. SAV is horizontally transmitted and the virus can survive for extended periods in seawater. A lack of convincing evidence for vertical transmission coupled to the fact that the SPD virus (SPDV) occurs in historically infected sites irrespective of fallow period duration suggests that a substantial reservoir of infection exists in the marine environment. We used a highly sensitive real-time PCR (qPCR) assay targeting a region of the SAV nsP1 gene to screen wild marine fish species for the presence of SAV in an attempt to identify such a potential reservoir. Screened fish species were caught in the vicinity of aquaculture activity in an area with a previous history of SAV infection (Shetland Isles, Scotland). SAV RNA was detected in internal organs (kidney and heart) from the flatfish species common dab Limanda limanda, long rough dab Hippoglossoides platessoides, and plaice Pleuronectes platessa. Based on these findings, sampling was extended to an area remote from aquaculture activity (Stonehaven Bay, NE coast of Scotland) from where heart tissues obtained from common dab also tested positive. While no virus could be cultivated from these samples, qPCR detections were shown to be SAV-specific by sequencing of an alternative gene region (E2) to that targeted by the qPCR assay. Analysis of these nucleotide sequences revealed minor differences to those previously obtained from farmed salmon, and subsequent phylogenetic analysis of an E2 dataset demonstrated a subtype V-like sequence.  相似文献

Piscine reovirus (PRV) is a double stranded non-enveloped RNA virus detected in farmed and wild salmonids. This study examined the phylogenetic relationships among different PRV sequence types present in samples from salmonids in Western Canada and the US, including Alaska (US), British Columbia (Canada) and Washington State (US). Tissues testing positive for PRV were partially sequenced for segment S1, producing 71 sequences that grouped into 10 unique sequence types. Sequence analysis revealed no identifiable geographical or temporal variation among the sequence types. Identical sequence types were found in fish sampled in 2001, 2005 and 2014. In addition, PRV positive samples from fish derived from Alaska, British Columbia and Washington State share identical sequence types. Comparative analysis of the phylogenetic tree indicated that Canada/US Pacific Northwest sequences formed a subgroup with some Norwegian sequence types (group II), distinct from other Norwegian and Chilean sequences (groups I, III and IV). Representative PRV positive samples from farmed and wild fish in British Columbia and Washington State were subjected to genome sequencing using next generation sequencing methods. Individual analysis of each of the 10 partial segments indicated that the Canadian and US PRV sequence types clustered separately from available whole genome sequences of some Norwegian and Chilean sequences for all segments except the segment S4. In summary, PRV was genetically homogenous over a large geographic distance (Alaska to Washington State), and the sequence types were relatively stable over a 13 year period.  相似文献

Tissue and blood samples were collected from 152 wild boars (Sus scrofa) from the Maremma area (Grosseto district, Central Italy) between November 2002 and January 2003. The presence of pseudorabies virus (PRV) antibodies, antigen, and DNA were confirmed by an enzyme-linked immunosorbent assay (ELISA), immunohistochemistry, and polymerase chain reaction (PCR), respectively. Of 152 animals, 62 (41%) were positive for viral antigen in tonsillar tissue. Of the 80 serum samples that were suitable for testing, 41 (51%) were positive for PRV antibodies. Positive immunohistochemistry results were confirmed by PCR. A significantly higher prevalence of PRV antigen and seroprevalence was detected in older animals. No differences were detected between males and females or for animals coming from different areas sampled. Results confirm that PRV is endemic in this wild boar population with a high prevalence of infection. The results of immunohistochemistry investigations demonstrated that a large number of wild boars harbor PRV in tonsillar tissues and should be considered as an important reservoir of PRV.  相似文献

Atlantic salmon (Salmo salar L.) mariculture has been associated with epidemics of infectious diseases that threaten not only local production, but also wild fish coming into close proximity to marine pens and fish escaping from them. Heart and skeletal muscle inflammation (HSMI) is a frequently fatal disease of farmed Atlantic salmon. First recognized in one farm in Norway in 1999[1], HSMI was subsequently implicated in outbreaks in other farms in Norway and the United Kingdom[2]. Although pathology and disease transmission studies indicated an infectious basis, efforts to identify an agent were unsuccessful. Here we provide evidence that HSMI is associated with infection with piscine reovirus (PRV). PRV is a novel reovirus identified by unbiased high throughput DNA sequencing and a bioinformatics program focused on nucleotide frequency as well as sequence alignment and motif analyses. Formal implication of PRV in HSMI will require isolation in cell culture and fulfillment of Koch''s postulates, or prevention or modification of disease through use of specific drugs or vaccines. Nonetheless, as our data indicate that a causal relationship is plausible, measures must be taken to control PRV not only because it threatens domestic salmon production but also due to the potential for transmission to wild salmon populations.  相似文献

Recent advances in micro-electronics make the study of the migration of even small marine animals (>12 cm) over many 1000s of kilometres a serious possibility. Important assumptions in long-term studies are that rates of tag loss caused by mortality or tag shedding are low, and that the tagging procedure does not have an unacceptable negative effect on the animal. This paper reports results from a study to examine the retention of relatively large (24 × 8 mm) surgically-implanted dummy acoustic tags over a 7-month period in steelhead pre-smolts (O. mykiss), and the effects of implantation on growth and survival. Although there was some influence on growth to week 12, survival was high for animals > 13 cm FL. In the following 16-week period, growth of surgically implanted pre-smolts was the same as the control population and there was little tag loss from mortality or shedding. Currently available acoustic tags can be implanted in salmonid fish ≥12 cm FL, although combined losses from mortality and tag shedding were 33–40% for animals in the 12 and 13 cm FL size classes. By 14 cm FL, combined rates of tag loss (mortality plus tag shedding) for surgically implanted tags dropped to <15% and growth following surgery was close to that of the controls. Our results suggest that studies of ocean migration and survival over periods of many months are now feasible even for animals as small as salmon smolts. Surgically implanted salmon smolts are therefore good candidates for freshwater and coastal ocean-tracking studies on relatively long time scales (months). On such time scales, even relatively small salmon smolts may move thousands of kilometers in the ocean.  相似文献

Infectious salmon anaemia (ISA) is a viral disease which targets the vascular and endocardial endothelial cells. An in vitro preparation of the spontaneously beating working heart of Salmo salar L. (i.e. able to generate physiological values of output pressure, cardiac output, ventricle work and power) was used to study cardiac performance under basal (i.e. in the absence of stimuli) and loading (i.e. Frank-Starling response) conditions in both control and ISAV-affected fish. In contrast to control fish, the heart preparations of the infected counterparts showed an impairment of the Frank-Starling response, particularly evident in fish infected with a higher virus dose. The Frank-Starling response was progressively impaired with the progression of the viral disease from the time of the virus administration until the 20th day. The potential involvement of nitric oxide (NO) in cardiac dysfunction was investigated by using the authentic nitric oxide synthase (NOS) substrate L-arginine and the NOS inhibitors L-NMMA and L-NIL. In contrast to control fish, infected hearts were particularly sensitive to the inducible nitric oxide synthase (iNOS) inhibitor L-NIL and insensitive to L-arginine. While pretreatment with NOS inhibitors reduced the Frank-Starling response in control hearts, it restored this response in infected counterparts. Taken together, the results indicate that cardiac dysfunction and the NO-transduction-pathway can be mechanistically linked in infected salmon.  相似文献

Atlantic salmon (Salmo salar) pre-smolts vaccinated with Norvax®Protect (NP) or injected with saline, were demonstrated to have a significantly (P≤0·0001) higher probability of being IPNV-infected when sampled during 12 weeks post challenge (PC) with IPNV (11·7 times and 32·5 times higher, respectively), than fish vaccinated with Norvax®Protect-IPN (NP-IPN), a vaccine identical to NP except for the presence of recombinant VP2. Furthermore, following experimental challenge with IPNV, an IPNV-specific secondary humoral immune response was detected in the group vaccinated with NP-IPN. Statistical analysis revealed that NP-IPN-vaccinated fish had a significantly (P≤0·0001) higher probability of producing IPNV-specific antibodies during 12 weeks PC with IPNV, compared to NP-vaccinated or saline-injected fish (5·3 times and 7·6 times higher, respectively). The results support efficacy data from field trials in Norway, where NP-IPN has proven successful in the prevention of IPN in Atlantic salmon post-smolts. However, the immunological mechanisms behind the increased IPNV clearance remain unknown.  相似文献
