Ochratoxin A (OTA) is a mycotoxin (potentially carcinogenic secondary metabolite derived from fungal contamination), produced by some Aspergillus and Penicillium strains. Although present and legislated in different food sources in the human diet, the regulation for wine intake is still under discussion. The Office International de la Vigne et du Vin (OIV) recommended maximum levels in wine of 2 μg l−1. Some reports refer to OTA contamination in wines up to 15 μg l−1 and a special incidence in red wines from the southern regions of Europe and the north of Africa, but the majority of the data available are below 1 μg l−1. When working at such low concentrations, the problem of the uncertainty of the results becomes decisive towards the implementation of legal limits. In order to assess the global uncertainty associated with OTA determination in wines and widen the data set and knowledge of the situation in Portugal, 340 wines were analysed (189 Port Wine, 85 Vinho Verde and 66 wines from other regions in the country) by a high performance liquid chromatography (HPLC)-fluorescence detection (FD) method using immunoaffinity columns for clean up. OTA was detected in 69 wines by the method used, but in concentrations below 0.5 μg l−1, except for two which showed levels up to a maximum of 2.1 μg l−1. However, the global uncertainty for OTA is close to 37% for concentrations above 0.5 μg l−1, and therefore, such value can be below or exceed the OIV limit. In the vicinity of the limit of detection, 0.084 μg l−1, the global uncertainty rises exponentially to a maximum of about 70%. This can be an obstacle when discussing safety intake limits. Ethanol and glucose content did not interfere in the clean up of OTA by immunoaffinity columns.  相似文献

In this study, the steroid hormone levels in river and tap water samples were determined by using a novel dispersive liquid-liquid microextraction method based on the solidification of a floating organic drop (DLLME-SFO). Several parameters were optimized, including the type and volume of the extraction and dispersive solvents, extraction time, and salt effect. DLLME-SFO is a fast, cheap, and easy-to-use method for detecting trace levels of samples. Most importantly, this method uses less-toxic solvent. The correlation coefficient of the calibration curve was higher than 0.9991. The linear range was from 5 to 1000 μg L−1. The spiked environmental water samples were analyzed using DLLME-SFO. The relative recoveries ranged from 87% to 116% for river water (which was spiked with 4 μg L−1 for E1, 3 μg L−1 for E2, 4 μg L−1 for EE2 and 9 μg L−1 for E3) and 89% to 102% for tap water (which was spiked with 6 μg L−1 for E1, 5 μg L−1 for E2, 6 μg L−1 for EE2 and 10 μg L−1 for E3). The detection limits of the method ranged from 0.8 to 2.7 μg L−1 for spiked river water and 1.4 to 3.1 μg L−1 for spiked tap water. The methods precision ranged from 8% to 14% for spiked river water and 7% to 14% for spiked tap water.  相似文献

In this paper, the conversion of azoxystrobin in a strongly fluorescent degradation product by UV irradiation with quantitative purposes and its fluorimetric determination are reported for the first time. A multicommuted flow injection-solid phase spectroscopy (FI-SPS) system combined with photochemically-induced fluorescence (PIF) is developed for the determination of azoxystrobin in grapes, must and wine. Grape samples were homogenized and extracted with methanol and further cleaned-up by solid-phase extraction on C18 silica gel. Wine samples were solid-phase extracted on C18 sorbent using dichloromethane as eluent. Recoveries of azoxystrobin from spiked grapes (0.5-2.0 mg Kg−1), must (0.5-2.0 μg mL−1) and wine (0.5-2.0 μg mL−1) were 84.0-87.6%, 95.5-105.9% and 88.5-111.2%, respectively. The quantification limit for grapes was 0.021 mg Kg−1, being within European Union regulations, and 18 μg L−1 and 8 μg L−1 for must and wine, respectively.  相似文献

A new high-performance liquid chromatography method using a coulometric array detector to simultaneously analyse 4-ethylphenol, 4-ethylguaiacol, 4-vinylphenol, and 4-vinylguaiacol in wine was established. This procedure offers important advantages as it does not require sample preparation, with the exception of filtration, and performs chromatographic separation in short time, making control of wine production processes easier. The method is linear up to concentrations of 2000 μg L−1 and precise (R.S.D. < 3%). Limits of detection are low (1-3 μg L−1) and suitable for analytical requirements in the oenological field. When compared to gas-chromatography-flame ionisation detection, the proposed method gives similar results with a shorter execution time.  相似文献

A new method for the determination of ochratoxin A and T-2 toxin in alcoholic beverages (wine and beer) by hollow fiber liquid microextraction was optimized. The extraction step was followed by ultra high-pressure liquid chromatography coupled to tandem mass spectrometry (UHPLC-MS/MS). The extraction procedure was based on the extraction of mycotoxins from the sample to the organic solvent (1-octanol) immobilized in the fiber, and afterwards, they were desorbed in a mixture of acetonitrile/water (80:20, v/v) at pH 7 prior to chromatographic determination. Different variables affecting the extraction process such as organic solvent, salt content, extraction time and desorption solution were studied. The developed method was validated in wine and beer, using white wine and alcoholic beer as representative matrices for both types of samples. Relative recoveries higher than 70% were obtained for the selected mycotoxins. Good linearity (R2 > 0.993) was obtained and quantification limits (0.02-0.09 μg L−1) below European regulatory levels were achieved. Repeatability, expressed as relative standard deviation, was always lower than 12%, whereas interday precision was lower than 21%. The proposed method was applied to the analysis of several types of wines and beers and ochratoxin A was detected in a rosé wine at 1.1 μg L−1.  相似文献

This work presents a new methodology to quantify ethyl carbamate (EC) in fortified wines. The presented approach combines the microextraction by packed sorbent (MEPS), using a hand-held automated analytical syringe, with one-dimensional gas chromatography coupled with mass spectrometry detection (GC–MS). The performance of different MEPS sorbent materials was tested, namely SIL, C2, C8, C18, and M1. Also, several extraction solvents and the matrix effect were evaluated. Experimental data showed that C8 and dichloromethane were the best sorbent/solvent pair to extract EC. Concerning solvent and sample volumes optimization used in MEPS extraction an experimental design (DoE) was carried out. The best extraction yield was achieved passing 300 μL of sample and 100 μL of dichloromethane. The method validation was performed using a matrix-matched calibration using both sweet and dry fortified wines, to minimize the matrix effect. The proposed methodology presented good linearity (R2 = 0.9999) and high sensitivity, with quite low limits of detection (LOD) and quantification (LOQ), 1.5 μg L−1 and 4.5 μg L−1, respectively. The recoveries varied between 97% and 106%, while the method precision (repeatability and reproducibility) was lower than 7%. The applicability of the methodology was confirmed through the analysis of 16 fortified wines, with values ranging between 7.3 and 206 μg L−1. All chromatograms showed good peak resolution, confirming its selectivity. The developed MEPS/GC–MS methodology arises as an important tool to quantify EC in fortified wines, combining efficiency and effectiveness, with simpler, faster and affordable analytical procedures that provide great sensitivity without using sophisticated and expensive equipment.  相似文献

Acrolein (propenal) is found in many foods and beverages and may pose a health hazard due to its cytotoxicity. Considerable knowledge gaps regarding human exposure to acrolein exist, and there is a lack of reliable analytical methods. Hydroalcoholic dilutions prepared for calibration purposes from pure acrolein show considerable degradation of the compound and nuclear magnetic resonance (NMR) spectroscopy showed that 1,3,3-propanetriol and 3-hydroxypropionaldehyde are formed. The degradation can be prevented by addition of hydroquinone as stabilizer to the calibration solutions, which then show linear concentration-response behaviour required for quantitative analysis. The stabilized calibration solutions were used for quantitative headspace solid-phase microextraction/gas chromatography–mass spectrometry (HS-SPME/GC–MS) determination of acrolein in alcoholic beverages with a detection limit of 14 μg L−1. Of 117 tested alcoholic beverages, 64 were tested positive with the highest incidence in grape marc spirits and whiskey (100%, mean 252 μg L−1), followed by fruit spirits (86%, mean 591 μg/L−1), tequila (86%, mean 404 μg L−1), Asian spirits (43%, mean 54 μg L−1) and wine (9%, mean 0.7 μg L−1). Acrolein could not be detected in beer, vodka, absinthe and bottled water. Six of the fruit and grape marc spirits had acrolein levels above the World Health Organization (WHO) provisional tolerable concentration of 1.5 mg L−1.  相似文献

An on-line retention and preconcentration system based on a sheep wool-packed microcolumn combined with flame atomic absorption spectrometry is proposed for trace level determination of Cd in wine. A chelating reagent 2-(5-bromo-2-pyridylazo)-5-diethylaminophenol was immobilized onto the wool before retention of the analyte. Several factors influencing the preconcentration efficiency of Cd and his subsequent determination, such as pH, eluent type, sample and eluent flow rates, interfering effects, were studied. A preconcentration factor of 39 was obtained with only 20 mL of sample. The relative standard deviation for five determinations of 1 μg L−1 Cd was 3.4%. The calibration graph was linear with a correlation coefficient of 0.998 at levels near the detection limit and up to at least 25 μg L−1. The limit of detection was 37 ng L−1. The accuracy of the proposed methodology was tested by comparison of the results with those obtained by electrothermal atomic absorption spectrometry analysis along with a recovery study. Finally, the method was employed for evaluating Cd levels in different wines including, blank, rose, and red.  相似文献

Phthalates (PAEs) are ubiquitous toxic chemical compounds. During the last few years, some phthalate metabolites (MPAEs) have been proposed as appropriate biomarkers in human urine samples to determine PAE human intake and exposure. So, it is necessary to have fast, easy, robust and validated analytical methods to determine selected MPAEs in urine human samples. Two different instrumental methods based on gas (GC) and ultra-high performance liquid (UHPLC) chromatography coupled to mass spectrometry (MS) have been optimized, characterized and validated for the simultaneous determination of nine primary and secondary phthalate metabolites in urine samples. Both instrumental methods have similar sensitivity (detection limits ranged from 0.03 to 8.89 pg μL−1 and from 0.06 to 0.49 pg μL−1 in GC–MS and UHPLC–MS2, respectively), precision (repeatability, expressed as relative standard deviation, which was lower than 8.4% in both systems, except for 5OH-MEHP in the case of GC–MS) and accuracy. But some advantages of the UHPLC–MS2 method, such as more selectivity and lower time in the chromatographic runs (6.8 min vs. 28.5 min), have caused the UHPLC–MS2 method to be chosen to analyze the twenty one human urine samples from the general Spanish population. Regarding these samples, MEP showed the highest median concentration (68.6 μg L−1), followed by MiBP (23.3 μg L−1), 5cx-MEPP (22.5 μg L−1) and MBP (19.3 μg L−1). MMP (6.99 μg L−1), 5oxo-MEHP (6.15 μg L−1), 5OH-MEHP (5.30 μg L−1) and MEHP (4.40 μg L−1) showed intermediate levels. Finally, the lowest levels were found for MBzP (2.55 μg L−1). These data are within the same order of magnitude as those found in other similar populations.  相似文献

Bismuth and Sb were evaluated as internal standards (IS) to minimize matrix effects on the direct and simultaneous determination of As, Cu, and Pb in cachaça by graphite furnace atomic absorption spectrometry using W-coated platform plus Pd-Mg(NO3)2 as modifier. For 20 μL injected sample, calibration within the 0.5-10 μg L−1 As, 100-1000 μg L−1 Cu and 0.5-30 μg L−1 Pb intervals were established using the ratios As absorbance to Sb absorbance, Cu absorbance to Bi absorbance and Pb absorbance to Bi absorbance versus analytes concentration, respectively. Typical linear correlations of 0.998, 0.999 and 0.999 were, respectively, obtained. The proposed method was applied for direct determination of As, Cu and Pb in 10 commercial cachaça samples and results were in agreement with those obtained by inductively coupled plasma mass spectrometry at 95% confidence level. The found characteristic masses were 30 pg As, 274 pg Cu and 39 pg Pb. The useful lifetime of the graphite tube was around 760 firings. Recoveries of As, Cu and Pb added to cachaça samples varied, respectively, from 98% to 109%, 97% to 108% and 98% to 104% with internal standards and from 48% to 54%, 53% to 92% and 62% to 97% without internal standards. The limits of detection were 0.13 μg L−1 As, 22 μg L−1 Cu and 0.05 μg L−1 Pb. The relative standard deviations (n = 12) for a spiked sample containing 20 μg L−1 As, Pb and 500 μg L−1 Cu were 1.6%, 1.0%, and 1.8% with IS and 4.3%, 5.2%, and 5.5% without IS.  相似文献

A gas chromatography–mass spectrometric (GC–MS) method has been established for the determination of hydrazine in drinking water and surface water. This method is based on the derivatization of hydrazine with ortho-phthalaldehyde (OPA) in water. The following optimum reaction conditions were established: reagent dosage, 40 mg mL−1 of OPA; pH 2; reaction for 20 min at 70 °C. The organic derivative was extracted with methylene chloride and then measured by GC–MS. Under the established condition, the detection and the quantification limits were 0.002 μg L−1 and 0.007 μg L−1 by using 5.0-mL of surface water or drinking water, respectively. The calibration curve showed good linearity with r2 = 0.9991 (for working range of 0.05–100 μg L−1) and the accuracy was in a range of 95–106%, and the precision of the assay was less than 13% in water. Hydrazine was detected in a concentration range of 0.05–0.14 μg L−1 in 2 samples of 10 raw drinking water samples and in a concentration range of 0.09–0.55 μg L−1 in 4 samples of 10 treated drinking water samples.  相似文献

Ultrasound-assisted extraction (UAE) and direct immersion solid-phase microextraction (DI-SPME) were evaluated for the monoterpenic compounds determination in wine samples. The wine extracts obtained were analyzed by gas chromatography-mass spectrometry (GC-MS). The optimization of the variables affecting UAE and SPME methods was carried out in order to achieve the best extraction efficiency. Both UAE and SPME are quantitative (recoveries in the range 93-97% and 71.8-90.9%, respectively), precise (coefficients of variation below 5.5%), sensitive (limits of detection between 30-39 μg L−1 and 11-25 μg L−1, respectively) and linear over one order of magnitude. The application of both methods to red wine samples showed that UAE provided higher extraction of monoterpenic compounds than SPME. Although SPME remains an attractive alternative technique due to its speed, low sample volume requirements and solvent free character.  相似文献

Two novel methodologies for quantification of C13 norisoprenoids in wines were developed. The first methodology, method A (reference method) was based on the headspace solid-phase microextraction combined with gas chromatography–quadrupole mass spectrometry operating in selected ion monitoring mode (HS-SPME–GC–qMS–SIM). This methodology allowed to select the GC conditions for an adequate chromatographic resolution of wine components. The second methodology, method B (rapid method) was based on the HS-SPME–GC–qMS–SIM, using GC conditions that allowed to obtain a C13 norisoprenoid volatile signature. In the later, the GC capillary column of 30 m at 220 °C was used acting as a transfer line of the components sorbed by the SPME coating fibre to the mass spectrometer, which acts as a sensor for m/z fragments 142 and 192. It does not require any pre-treatment of the sample, and the C13 norisoprenoid composition of the wine was evaluated based on the chromatographic profile and specific m/z fragments, without complete chromatographic separation of its components. For quantification purposes, external calibration curves were constructed with β-ionone chemical standard. Calibration curves with regression coefficient (r2) of 0.9940 and 0.9968, RSD of 1.08% and 12.51%, and detection limits of 1.10 and 1.57 μg L−1 were obtained for methods A and B, respectively. These methodologies were applied to seventeen white and red table wines. Two vitispirane isomers (158–1529 μg L−1) and 1,1,6-trimethyl-1,2-dihydronaphthalene (TDN) (6.42–39.45 μg L−1) were quantified. The data obtained for vitispirane isomers and TDN using the two methods were highly correlated (r2 of 0.9756 and 0.9630, respectively). Associated to the fast and robust character of the proposed rapid method B and considering the extraction time, it is important to focus its selectivity and potential applicability if specific m/z fragments would be established for new analytes.  相似文献

Sulphides in water samples were determined by stripping chronopotentiometry in a computer controlled flow system with a flow-through electrochemical cell. The working electrode was a porous glassy carbon electrode coated with Nafion and mercury. The sample was diluted with 0.1 mol L−1 NaOH and analysed. Sulphides in the sample were collected in the porous electrode as mercury sulphide and then stripped by a current of −500 μA. The limit of detection was found to be 1.6 μg L−1 and 0.5 μg L−1 for 1 mL and 5 mL of preconcentrated sample, respectively. The linear range for 1 mL sample was found to be 5-400 μg L−1. The repeatability and reproducibility was found to be 2.6% and 4.8%, respectively. The method was applied to analyses of waste water samples from a tannery.  相似文献

A cost-effective sequential injection system incorporating with an in-line UV digestion for breakdown of organic matter prior to voltammetric determination of Zn(II), Cd(II), Pb(II) and Cu(II) by anodic stripping voltammetry (ASV) on a hanging mercury drop electrode (HMDE) of a small scale voltammetric cell was developed. A low-cost small scale voltammetric cell was fabricated from disposable pipet tip and microcentrifuge tube with volume of about 3 mL for conveniently incorporated with the SI system. A home-made UV digestion unit was fabricated employing a small size and low wattage UV lamps and flow reactor made from PTFE tubing coiled around the UV lamp. An in-line single standard calibration or a standard addition procedure was developed employing a monosegmented flow technique. Performance of the proposed system was tested for in-line digestion of model water samples containing metal ions and some organic ligands such as strong organic ligand (EDTA) or intermediate organic ligand (humic acid). The wet acid digestion method (USEPA 3010a) was used as a standard digestion method for comparison. Under the optimum conditions, with deposition time of 180 s, linear calibration graphs in range of 10-300 μg L−1 Zn(II), 5-200 μg L−1 Cd(II), 10-200 μg L−1 Pb(II), 20-400 μg L−1 Cu(II) were obtained with detection limit of 3.6, 0.1, 0.7 and 4.3 μg L−1, respectively. Relative standard deviation were 4.2, 2.6, 3.1 and 4.7% for seven replicate analyses of 27 μg L−1 Zn(II), 13 μg L−1 Cd(II), 13 μg L−1 Pb(II) and 27 μg L−1 Cu(II), respectively. The system was validated by certified reference material of trace metals in natural water (SRM 1640 NIST). The developed system was successfully applied for speciation of Cd(II) Pb(II) and Cu(II) in ground water samples collected from nearby zinc mining area.  相似文献

A sensitive method for the determination of chromium ion(VI) in complex matrices such as crude oil and sludge is presented based on the decreasing effect of Cr(VI) on cathodic adsorptive stripping peak height of Cu-adenine complex. Under the optimum experimental conditions (pH 7.5 Britton-Robinson buffer, 5 × 10−5 M copper, 8 × 10−6 M adenine and accumulation potential −250 mV versus Ag/AgCl), a linear decrease of the peak current of Cu-adenine was observed, when the chromium(VI) concentration was increased from 5 μg L−1 to 120 μg L−1. Detection limit of 2 μg L−1 was achieved for 120 s accumulation time. The relative standard deviations (R.S.D., %) were 1.8% and 4% for chromium(VI) concentrations of 18 μg L−1 and 100 μg L−1, respectively. The method was applied to the determination of chromium(VI) in the presence of high levels of chromium(III), in various real samples such as crude oil, crude oil tank button sludge, waste water and tap water samples. Effects of foreign ions and surfactants on the voltammetric peak and the influences of instrumental and analytical parameters were investigated in detail. The accuracy of the results was checked by ICP and/or AA.  相似文献

There has been considerable public interest and a growing number of scientific studies linking certain phenolic compounds in grapes and wines, particularly trans-resveratrol (trans-3,5,4′-trihydroxystilbene, TRA), to human health benefits. Typical TRA concentrations in wine are very low. It is a polar compound with very low volatility, which makes it difficult to extract and to separate on a gas chromatography (GC) column without derivatization. In this study, a new method for trace analysis of TRA was developed using solid-phase microextraction (SPME) with on-fiber silylation derivatization. Multidimensional GC equipped with a heartcut valve and cryogenic focusing was coupled with a mass-selective detector and used for improved separations and analysis. The effects of SPME fiber selection, extraction time, temperature, and desorption time were investigated. The derivatization conditions, time/temperature and the volume of derivatization reagent were also optimized. The calibration curve was linear over the concentration range from 10 ng L−1 to 5 mg L−1, with a correlation coefficient of 0.9996. The average recovery of TRA in red wine was 83.6 ± 5.6%. The method detection limit (MDL) for TRA in ethanol:water (12.5:87.5, v/v) solution in this study was 7.08 ng L−1 whereas the MDL for TRA in pure water was 2.85 ng L−1. The new method was used to test the TRA content in six selected Iowa red wine samples. Measured concentrations varied from 12.72 to 851.9 μg L−1.  相似文献

A stir bar sorptive extraction with liquid desorption followed by large volume injection coupled to gas chromatography-quadrupole mass spectrometry (SBSE-LD/LVI-GC-qMS) was evaluated for the simultaneous determination of higher alcohol acetates (HAA), isoamyl esters (IsoE) and ethyl esters (EE) of fatty acids. The method performance was assessed and compared with other solventless technique, the solid-phase microextraction (SPME) in headspace mode (HS). For both techniques, influential experimental parameters were optimised to provide sensitive and robust methods. The SBSE-LD/LVI methodology was previously optimised in terms of extraction time, influence of ethanol in the matrix, liquid desorption (LD) conditions and instrumental settings. Higher extraction efficiency was obtained using 60 min of extraction time, 10% ethanol content, n-pentane as desorption solvent, 15 min for the back-extraction period, 10 mL min−1 for the solvent vent flow rate and 10 °C for the inlet temperature. For HS-SPME, the fibre coated with 50/30 μm divinylbenzene/carboxen/polydimethylsiloxane (DVB/CAR/PDMS) afforded highest extraction efficiency, providing the best sensitivity for the target volatiles, particularly when the samples were extracted at 25 °C for 60 min under continuous stirring in the presence of sodium chloride (10% (w/v)). Both methodologies showed good linearity over the concentration range tested, with correlation coefficients higher than 0.984 for HS-SPME and 0.982 for SBES-LD approach, for all analytes. A good reproducibility was attained and low detection limits were achieved using both SBSE-LD (0.03-28.96 μg L−1) and HS-SPME (0.02-20.29 μg L−1) methodologies. The quantification limits for SBSE-LD approach ranging from 0.11 to 96.56 μg Land from 0.06 to 67.63 μg L−1 for HS-SPME. Using the HS-SPME approach an average recovery of about 70% was obtained whilst by using SBSE-LD obtained average recovery were close to 80%. The analytical and procedural advantages and disadvantages of these two methods have been compared. Both analytical methods were used to determine the HAA, IsoE and EE fatty acids content in “Terras Madeirenses” table wines. A total of 16 esters were identified and quantified from the wine extracts by HS-SPME whereas by SBSE-LD technique were found 25 esters which include 2 higher alcohol acetates, 4 isoamyl esters and 19 ethyl esters of fatty acids. Generally SBSE-LD provided higher sensitivity with decreased analysis time.  相似文献

Two non-electroactive biomarkers methylglyoxal (MGo) and glyoxal (Go) in urine and environmental water samples were determined for the first time by capillary electrophoresis with amperometric detection (CE-AD) after derivatizing with an electroactive compound 2-thiobarbituric acid. Experimental conditions of derivatization and CE-AD detection were optimized. Highly linear response was obtained for these two biomarkers over three orders of magnitude with good correlation (r2 > 0.999). The limits of detection (LODs) and limits of quantitation (LOQs) of MGo and Go were 0.2 μg L−1 and 1.0 μg L−1, 0.5 μg L−1 and 2.0 μg L−1, respectively. The average recovery and relative standard deviation (RSD) were within the range of 90.9–101.3% and 0.7–2.2%, respectively. The proposed CE-AD method provides a reliable and sensitive quantitative evaluation of MGo and Go in real sample matrices by employing relatively simple and inexpensive instrument.  相似文献
