Background aimsWe evaluated hematopoietic stem cells according to CD34 expression and aldehyde dehydrogenase (ALDH) activity in peripheral blood and apheresis product samples from patients after mobilization with granulocyte–colony-stimulating factor (G-CSF) alone or G-CSF after high-dose cyclophosphamide (4 g/m2 once daily, intravenously on day 1). We also investigated the relationship between the number of SSClo CD45dim CD34hi cells, SSClo ALDHbr cells and engraftment.MethodsThirty patients (20 males and 10 females), who were candidates for autologous peripheral blood stem cell transplantation, were included in the study. Cyclophosphamide + G-CSF was used for 17 and G-CSF alone for 24 mobilizations. Primary diagnoses were multiple myeloma (n% = 14), Hodgkin's lymphoma (n% = 7), non-Hodgkin's lymphoma (n% = 2), acute myloid leukemia (n% = 2), chronic lymphocytic leukemia (n% = 1) and germ cell testis tumor (n% = 1).ResultsNumbers of SSClo CD45dim CD34hi cells and SSClo ALDHbr cells were highly correlated in both peripheral blood and apheresis products (P < 0.001). We could not find a relationship between the transplanted SSClo CD45dim CD34hi cell dose or SSClo ALDHbr cell dose and platelet or neutrophil recovery. The optimal thresholds for SSClo CD45dim CD34hi cells were 5.40 × 106/kg for neutrophil recovery and 7.22 × 106/kg for platelet recovery. The optimal thresholds for SSClo ALDHbr cells were 6.53 × 106/kg for neutrophil recovery and 8.72 × 106/kg platelet recovery.ConclusionsAccording to our data, numbers of SSClo ALDHbr cells are in very good agreement with numbers of SSClo CD45dim CD34hi cells and can be a predictor of stem cell mobilization.  相似文献

Emerging data have implicated a critical role for CD4 in the pathogenesis of systemic lupus erythematosus (SLE). This study was designed to delineate the contribution of CD4+ T cells in the pathogenesis of SLE disease. Forty-four patients (3 male: 41 female) and 20 healthy volunteers (4 male: 16 female) were included in the study. CD4+ lymphocytes analysis was done using three-color flow cytometry with antibodies against human-CD95, a prototype cell death receptor, and the chemokine receptor-7 (CCR7) after gating for lymphocytes based on the forward and side scatter. Serum levels of IL-6, IL-12, IL-17, TNF-α and IL-10 cytokines were assayed using ELISA. Disease activity was assessed using the SLE disease activity index (SLEDAI). Based on the expression of CCR7 and CD95, CD4+ lymphocytes were subdivided into three particular subsets; CD4+CD95+CCR7+ cells, CD4+CD95CCR7+ cells and CD4+CD95+CCR7 cells. Percentage of CD4+CD95+CCR7+ cell subset was significantly higher in patients with SLE with active disease (SLEDAI > 6) and inactive (SLEDAI < 6) as compared with controls (P = 0.005), and it showed a significant positive correlation with ANA titer (P = 0.01), and a negative correlation with WBCs count (P = 0.001). CD4+CD95+CCR7 cell subset was significantly higher in active SLE patients in comparison to patients with inactive disease and controls (P = 0.05, P = 0.005 respectively), and it correlates positively with SLEDAI, IL-6 and IL-17 levels (P = 0.001, 0.05, 0.01 respectively), and negatively with blood WBCs counts (P = 0.001). The third CD4+CD95CCR7+cell subset was found significantly lower in SLE patients compared with controls, and it was found negatively correlated with IL-10, IL-6, and IL-17. The results show that CD4+CD95+subset lacking expression of CCR7 is associated with cell mediated inflammatory response as manifested by its correlation with signs of inflammation, inflammatory cytokines and disease activity index. Whereas, CD4+CD95+CCR7+ correlate more with antibody immune responses as manifested by association with serum ANA. These data suggest disparate roles of these cell subsets in the pathophysiology of SLE. A better understanding of the characteristics of CD4 cell subsets may shed light on the pathogenesis of autoimmune diseases, particularly SLE.  相似文献

Background aimsImmunomagnetic CD34+ cell selection (ICS) is utilized in autologous and allogeneic transplants. In the first case it is used to reduce the neoplastic contamination of concentrates, while in the second case it is needed to carry out a T-depletion of cell concentrates in order to reduce the incidence of graft-versus-host disease (GvHD) in patients who have undergone haplo-identical transplants.MethodsThe efficacy of CliniMACS technology, after reduction of platelet contamination, incubation of monoclonal antibodies (MAb) and successive washings of concentrates, performed in 16 ICS using the standard method without reducing platelet content, was compared with the use of the automated system CytoMate, which was carried out in 46 ICS.ResultsIn the group of ICS carried out after automatic manipulation, a significant statistical difference in purity was noted (91.39% versus 83.57, P = 0.017) compared with the group of ICS carried out with the standard procedure. The same significant difference was noted in relation to the remaining percentages of CD3+ and CD19+ cells (2.31% versus 5.68%, P = 0.012, and 1.58% versus 2.71%, P = 0.014, respectively). Recovery of CD34+ cells overlapped in the two groups (70.49% versus 68.39%, P = 0.774).ConclusionsImmunomagnetic selection carried out using the automated procedure was more efficient, producing a purer sample, more efficient T-depletion and optimal reduction of B cells, without influencing cell recovery. Furthermore, conforming to good manufacturing practice (GMP) guidelines, the entire procedure with CytoMate took place in a contamination-controlled environment.  相似文献

ObjectiveTo analyze the effect of Argon-Helium cryosurgery (AHCS) combined with transcatheter renal arterial embolization (TRAE) on the differentiation of regulatory CD4+ CD25+ T cell (Treg) and its implication in patients with renal carcinoma.MethodsSeventy seven patients are included in the study, and divided into two groups: TRAE group (n = 45, receiving TRAE only) and TRAE + cryoablation group (n = 32, receiving cryoablation 2–3 weeks after TRAE). The percentage of Treg cells and T lymphocyte subsets (CD4+T, CD8+T, and CD4+T/CD8+T) in the peripheral blood is measured by flow cytometry previous to the therapy and 3 months after therapy. Meanwhile, the extent of tumor necrosis is measured by MRI or CT 1 month after therapy.ResultsThe percentages of Treg cells of patients in TRAE + cryoablation group decrease from (6.65 ± 1.22)% to (3.93 ± 1.16)%, (t = 42.768, P < 0.01), and the percentages of CD4+T and CD4+T/CD8+T increase significantly (P < 0.01). However, the results of patients in TRAE group show that the percentages of Treg, CD4+T, CD8+T and CD4+T/CD8+T increase slightly although the differences had no statistical significance (P > 0.05). The tumor necrosis rate of TRAE + cryoablation group is 57.5%, significantly higher than those of TRAE group, which shows 31.6% (t = 6.784, P < 0.01). The median survival duration of the TRAE + cryoablation group is 20 months, significantly longer than that of the TRAE group (χ2 = 7.368, P < 0.01). The decreasing extent of Treg cells is correlated with tumor necrosis rates (r = 0.90, P < 0.01) and life time (r = 0.67, P < 0.01).ConclusionThe therapy of TRAE combined with cryoablation contributes to reduce the percentage of Treg cells and improve the immune situation of patients with renal cell carcinoma, which consequently increase tumor necrosis rate and prolong the patients‘ survival duration.  相似文献

Background aimsMesenchymal stromal cells (MSC) are the most popular cells used in regenerative medicine and biotechnology. The clonogenic potential of these cells is defined by colony-forming unit-fibroblasts (CFU-F). It is well known that there is an interaction between hematopoietic cells and stromal cells in disease formation pathogenesis. Therefore we hypothesized that there should be a quantitative and qualitative relationship between MSC colonies (CFU-F) and hematopoietic stem cell colonies (colony-forming unit-granulocyte-macrophages; CFU-GM) among patients with and without hematologic diseases.MethodsForty-two patients were included in this study. Patients were divided into three groups: group A, patients with hematologic malignancies (n = 20); group B, patients with bone marrow (BM) failure (n = 11); group C, patients without hematologic diseases (n = 11). BM aspirates were plated in different densities for CFU-F culture. The plating density was the same for CFU-GM culture.ResultsCFU-GM colonies grew in 90% of group A cells and all of group B and C cells (P = 0.0001). CFU-F colonies became visible on the ninth day of plating in group A and on the eight day in groups B and C. There was no statistically significant difference between the groups for the duration of CFU-F colony formation (P = 0.12). There were differences in the morphology of the colonies among the groups.ConclusionsThis is the first study that has compared the clonogenic potential of stromal cells and hematopoietic stem cells in the same subjects with and without hematologic diseases. No correlation was shown between the clonogenic potential of stromal cells and hematopoietic cells.  相似文献

Thirty-three Holstein-Friesian cows were followed from 14 days pre partum until the fourth ovulation post partum. Housing conditions and basic ration were identical for all animals. Concentrates were individually supplemented according to the daily milk production level, using two different types of protein rich concentrates: soybean meal and rapeseed meal. Soybean and rapeseed meal are known to be respectively high and low in isoflavones. Cows were randomly divided into three groups and blocked for parity. Group I (n = 11) was supplemented with soybean meal and acted as control group. Groups II (n = 11) and III (n = 11) were respectively supplemented with soybean and rapeseed meal and were subjected to a biopsy sampling of the corpus luteum at day 9 of the first three postpartal estrous cycles.Soybean meal supplementation to lactating dairy cows (1.72 kg on average) induced an increase in the blood concentration of equol, dihydrodaidzein, o-desmethylangolensin in both soy groups and resulted in a reduced area occupied by steroidogenic (P = 0.012) and endothelial cells (P = 0.0007) in the luteal biopsies. Blood concentrations of equol and glycitein were negatively correlated with the areas occupied by steroidogenic (r = −0.410 with P = 0.0002, respectively r = −0.351 with P = 0.008) and endothelial cells (r = −0.337 with P = 0.01, respectively r = −0.233 with P = 0.085) in the 3 first estrous cycles. The latter however did not affect the diestrous peripheral blood progesterone concentration.  相似文献

IntroductionChildhood Idiopathic Nephrotic Syndrome (INS) responds to glucocorticoid therapy, however, 60–80% of patients relapse and some of them become steroid non responsive. INS may occur because of T cell dysfunction, abnormal cytokines and podocytopathies which reverse on steroid treatment. The reason of relapses could be imbalances in T cells phenotypes and respective cytokines. Herein, we hypothesize that relapses in INS may occur due to imbalance in T-regulatory and T-effector cell with their respective cytokines and overexpression of P-gp on lymphocytes.MethodsThe frequency of peripheral blood CD4+CD25+FoxP3+ Treg, CD4+IFN-γ+ Th1 and CD4+IL-4+ Th2 lymphocytes and their respective cytokines and P-gp expression on peripheral blood lymphocytes (PBLs) were analyzed in INS patients at baseline (n = 26), during remission (n = 24) and at relapse (n = 15).ResultsCompared to baseline, the frequency of Tregs was significantly increased at remission and decreased during relapse. In contrast, the frequency of Th1 and Th2 lymphocytes was significantly decreased during remission and increased at the time of relapse. Similarly, expression of P-gp was significantly high at baseline and at the time of relapse as compared to remission. Levels of cytokines IL-10 and TGF-β in the supernatant of stimulated PBMCs was increased during remission and decreased during relapse. In contrast, levels of IFN-γ and IL-4 were decreased during remission and increased at the time of relapse.ConclusionsSteroid therapy in INS induces decreased P-gp expression on PBLs along with increased frequency and cytokine response of T-regulatory cells, and reduced frequency and respective cytokine response of Th1 and Th2 cells during remission. However, reversal in the frequency and respective cytokines of T-regs, Th1 and Th2, and P-gp expression on PBLs occurs during relapses on follow-up.  相似文献

BackgroundSystemic reactions are related to the pathogenesis of Aspirin Exacerbated Respiratory Disease (AERD). With this work we wanted to study the changes in the systemic levels of inflammatory mediators in both baseline and after oral aspirin challenge in patients with and without AERD.MethodsPatients with nasal polyposis and asthma with AERD (n = 20) and without (n = 18) were orally challenged with aspirin in a single-blind placebo controlled study. Serum samples and urine were collected before and 6 h after placebo and aspirin oral challenges. Serum levels of inflammatory mediators were assayed by using the Luminex technology and ELISA. The concentrations of 9-alpha, 11-beta prostaglandin F2, and leukotriene E4 (uLTE4) were measured in urine samples by ELISA. The expression of T-cell surface markers was analyzed in peripheral blood mononuclear cells isolated before and after the challenges.ResultsAERD patients showed significantly higher baseline levels of s-IL-5R-alpha, uLTE4 and percentage of CD4+CD25+CD127pos and CD4+CD45RACD45RO+ but decreased levels of TGF-β1 and number of CD4+CD25+CD127neg cells. Aspirin challenge induced the release of uLTE4, IL-6 and increased the number of CD4+CD45RACD45RO+ memory T-cells only in AERD patients but failed to reduce the levels of sCD40L as observed in non-AERD subjects. Further, IL-8 and sIL-5R-alpha levels directly correlated with the PD20ASA and the effects of aspirin on IL-6 and number of memory T-cells was more pronounced in subjects showing more strong reaction (bronchial and nasal).ConclusionsAERD patients have a differential baseline inflammatory pattern that supports the role inflammation as underlying mechanism of the disease. Systemic response to oral aspirin challenge was related to an increase in serum IL-6 and the number of circulating memory T-cells in AERD patients.  相似文献

Background aimsThe rising use of allogeneic transplantation in older recipients necessitates considering older related donors. The effect of related donor age for peripheral blood stem cell allografts (PBSC) on graft maintenance and outcomes, independent of CD34+cell dose, has not been well-characterized.MethodsHLA-related donors (98% siblings) underwent a uniform filgrastim-based mobilization regimen aiming to collect and infuse 5 × 106 CD34+ cells/recipient kg. Donor and recipient age were modeled in multiple ways to account for the correlation, and outcomes reported by decade of donor age.ResultsThe median donor and recipient ages were 52 years and 54 years, respectively. The mean CD34+ cell dose infused was 5.6 × 106 CD34+/kg and 75% of patients received a narrow range between 4.4 and 6.6 × 106 CD34+ cells/kg. Neither better PBSC mobilization nor higher CD34+ content of allografts was significantly associated with engraftment or transplant outcomes. After adjusting for recipient age and other prognostic factors, older donor age by decade conferred a lower risk of non-relapse mortality (NRM) [hazard ratio (HR) = 0.64, 95% confidence interval (CI) 0.45–0.91, P = 0.013] and borderline improvement in overall survival (OS) (HR = 0.76, 95% CI 0.58–0.99, P = 0.045) without altering progression-free survival (PFS) (HR = 0.85, 95% CI 0.66–1.07, P = 0.18).ConclusionsOlder donor age does not worsen outcome after matched related donor PBSC transplantation in patients receiving a narrow range CD34+ cells. The relatively small sample size mandates that the finding of similar to improved outcomes for older related donor age must be confirmed in larger studies.  相似文献

BACKGROUNDImportance of androgen receptor (AR) as an independent prognostic marker in Pakistani women with breast cancer (BCa) remains unexplored. Our aim was to identify the expression and potential prognostic value of AR, its upstream regulator (pAkt) and target gene (pPTEN) in invasive BCa.METHODSThis study used a cohort of 200 Pakistani women with invasive BCa diagnosed during 2002-2011. Expression of AR, pAkt and pPTEN was determined on formalin fixed paraffin embedded tissue sections by immunohistochemistry. The association of AR, pAkt and pPTEN with clinicopathological parameters was determined. Survival analyses were undertaken on patients with ≥ 5 years of follow-up (n = 82).RESULTSExpression of AR, pAkt and pPTEN was observed in 47.5%, 81.3% and 50.6% of patients, respectively. AR-expressing tumors were low or intermediate in grade (P < .001) and expressed ER (P = .002) and PR (P = .001). Patients with AR+ tumors had significantly higher OS (Mean OS = 10.2 ± 0.465 years) compared to patients with AR? tumors (Mean OS = 5.8 ± 0.348 years) (P = .047). Furthermore, AR-positivity was associated with improved OS in patients receiving endocrine therapy (P = .020). Patients with AR+ /pAkt+ /pPTEN? tumors, had increased OS (Mean OS = 7.1 ± 0.535 years) compared to patients with AR?/pAkt+/pPTEN? tumors (Mean OS = 5.1 ± 0.738 years).CONCLUSIONAR-expressing tumors are frequently characterized by low or intermediate grade tumors, expressing ER and PR. In addition, expression of AR, pAkt and pPTEN, could be considered in prognostication of patients with invasive BCa.  相似文献

Our aim was to determine the role of opioidergic processes in the effects of nutrition on the secretion of LH pulses in the mature male sheep. In the first of three experiments, adult Merino rams were acclimatised to a maintenance diet and then allocated to one of three dietary groups (n = 5): continuation of the maintenance diet (Group M); reduction to half of the maintenance allocation (Group HM); or supplementation of the maintenance diet with lupin grain (Group HD). An initial administration of naloxone (2 mg/kg body weight, i.v.) was followed at 40-min intervals by three further administrations (1 mg/kg). Blood was sampled every 20 min for 12 h before the initial naloxone administration and then for a further 6 h. LH pulse frequency after naloxone treatment was significantly higher in Group HD than in Group HM (P < 0.05). The second study tested whether the response to naloxone depended on calcium status. We used 22 adult Merino rams in two consecutive experiments, one in which the rams were fed a maintenance diet, and one in which the rams were fed with the maintenance diet plus 1 kg lupin grain for 5 weeks. In both experiments, rams were allocated to groups that received one of the following treatments: (a) 0.02 g/kg calcium borogluconate + 0.2 mg/kg naloxone hydrochloride (Nal + Ca2+; n = 6); (b) 0.2 mg/kg naloxone hydrochloride (Nal; n = 6); (c) 0.02 g/kg calcium borogluconate (Ca2+; n = 5); (d) 0.1 ml/kg NaCl 0.9% (Saline; n = 5). All treatments were given as a single i.v. administration daily for 5 days. Blood was sampled every 20 min for 24 h during the acclimatization period (Day 0) and on the last day (Day 5) of treatment. In the first study (under maintenance), none of the treatments affected LH pulse frequency. In the second study (the lupin-supplemented rams), LH pulse frequency was significantly increased (P < 0.05) by the administration of naloxone + Ca2+, naloxone alone and Ca2+ alone. Overall, rams on a low plane of nutrition showed the smallest response to naloxone, suggesting that an opioidergic mechanism is not involved in the suppressive effect of restricted nutrition on the gonadotrophic axis. Rather, because testosterone secretion was increased on the high plane of nutrition, the LH responses to naloxone are better explained by the effects of testosterone on opioidergic mechanisms. Finally, we failed to observe any interaction between opioids and calcium in the control of LH secretion.  相似文献

Background aimsThe aim of this study was to evaluate the efficacy and safety of cytokine-induced killer (CIK) cell therapy for solid carcinomas.MethodsWe performed a computerized search of phase II/III clinical trial databases of CIK cell-based therapy using a combination of the terms ‘cytokine-induced killer cells’, ‘tumor’ and ‘cancer’.ResultsTreatment with CIK cells was associated with a significantly improved half-year survival (P = 0.003), 1-year survival (P = 0.0005), 2-year survival (P  < 0.01) and mean survival time (MST) (P  < 0.001). Patients in the CIK group showed a prolonged half-year progression-free survival (PFS) (P  < 0.01), 1-year PFS (P < 0.01) and median time to progression (MTTP) (P < 0.001). A favored disease control rate (DCR) was observed in patients receiving CIK cell therapy, while the objective response rate (ORR) was not altered (P = 0.05) compared with the non-CIK group (P = 0.007). CIK cell therapy could also reduce the adverse effects of grade III and IV leukopenia caused by chemotherapy (P = 0.002) and diminish hepatitis B virus (HBV)-DNA content (P < 0.01). However, the incidence of fever in the CIK therapy group was significantly higher than in the non-CIK group (P = 0.02). The percentage of CD3+, CD4+, CD4+ CD8+, CD3? CD56+ and CD3+ CD56+ T-lymphocyte subsets in the peripheral blood of cancer patients was significantly increased, whereas the percentage of CD8+ T-lymphocyte cells was significantly decreased in the CIK group compared with the non-CIK group (P < 0.01).ConclusionsCIK cell therapy has demonstrated a significant superiority in prolonging the MST, PFS, DCR and quality of life (QoL) of patients.  相似文献

Our objective was to determine the influence of short-term exposure to endophyte-infected tall fescue on reproductive function of ram lambs. Rams (214 days of age) were fed a diet free of endophyte-infected fescue seed (EF; n = 8) or endophyte-infected fescue seed (EI; n = 9; 34% of diet; 4.8 μg g−1 ergovaline) for 6 weeks. Feed offered to EF rams, individually fed, was reduced to the average intake of EI lambs from previous day so that intake was similar between treatments and averaged 2.4% BW (DM basis), leading to daily intake of 33.7 μg ergovaline kg−1 BW for the EI fed lambs. Daily high ambient temperature for the trial ranged between 16 and 26 °C. Respiration rate and rectal temperature were measured at 14:00 daily. Blood was collected for serum concentrations of prolactin (weekly) and testosterone (twice weekly). Body weight and body condition scores (BCS; 1 = thin; 5 = fat) were determined every 14 days. Scrotal circumference, scrotal skin temperature, and semen characteristics were determined weekly. Rams were slaughtered after 6 weeks of feeding. Signs of fescue toxicosis in EI fed rams included increased rectal temperature (P < 0.001, R2 = 0.11) and respiration rate (day, P < 0.001, R2 = 0.25) when high ambient temperature exceeded 22 °C and reduced serum concentrations of prolactin (diet × day, P < 0.001). Body weight of EI fed rams tended to decrease after 36 days of feeding compared with EF fed rams (−3.0 kg versus 0.51 kg; P < 0.07) and BCS was similar between treatment throughout the trial. Serum concentrations of testosterone were greater in EI compared with EF fed rams (diet × day, P < 0.005, R2 = 0.08). Scrotal skin temperature, scrotal circumference, semen volume, percent sperm motility, and percent abnormal sperm were similar between treatments. Spermatozoa concentration tended to be greater in EF compared with EI fed rams after 43 days of feeding (P < 0.10; R2 = 0.15). Rate of forward movement of spermatozoa tended to increase at a greater rate between Days 8 and 29 in EF compared with EI fed rams (diet × day, P < 0.08). Feeding endophyte-infected fescue seed to ram lambs was associated with potential decreased fertility and increased serum concentrations of testosterone. Short term exposure of endophyte toxins to male ruminants may negatively impact reproductive responses. Feeding for longer periods may further reduce fertility and merits further research.  相似文献

Diets supplemented with long chain, n  3 polyunsaturated fatty acids (PUFA) have improved the health and performance of neonatal and growing animals. This study was conducted with lambs that were orphaned at approximately 1 day of age to determine whether supplementing milk replacer fed lambs with oils rich in long chain n  3 or n  6 PUFA would alter plasma lipid profiles and affect growth characteristics and immune functions. From days 1 to 28 of age, lambs had ad libitum access to commercial milk replacer. From days 7 to 28 of age, lambs received twice daily either 1 g of soybean oil, 1 g of fish oil, or 1 g of safflower oil per os in a gelatin capsule (n = 60 pens; 20 pens/treatment; one ewe and one ram with similar initial body weights/pen). On days 7, 14, 21, and 28 of age, lambs were weighed, and jugular blood was collected from ram lambs. Lymphocyte proliferation in vitro, differential white blood cell (WBC) counts, and weight gains were quantified. Plasma from days 7 and 28 was used for fatty acid analyses. Fish oil increased (P < 0.001) plasma total n  3 fatty acid concentration and total n  3:total n  6 fatty acid ratio. Pen body weight (i.e., total lamb weight per pen) increased (P < 0.001) with day (day 7, 11.9 kg; day 14, 15.1 kg; day 21, 18.2 kg; and day 28, 21.2 kg), but oil treatment did not affect pen body weight. Neither oil treatment, day, nor oil treatment × day interaction were significant for pen body weight gains (3.5 kg), pen average daily gains (0.5 kg), pen milk intakes (19.0 kg), or pen gain:feed ratio (0.18) measured during three intervals: days 7–14; days 14–21; and days 21–28. Day, but not oil treatment, affected (P < 0.001) unstimulated, concanavalin A stimulated, and lipopolysaccharides stimulated lymphocyte proliferation: days 14, 21, and 28 proliferation > day 7 proliferation. For neutrophils per 100 WBC, the treatment × day interaction was significant (P < 0.05). Oil treatment and day affected (P < 0.01 and <0.05, respectively) lymphocyte numbers per 100 WBC. For monocytes, eosinophils, and basophils, neither oil treatment, day, nor the oil treatment × day interaction were significant. Fish oil altered plasma fatty acid profiles, but it did not seem to improve measures of the performance or immune function of healthy, milk replacer fed lambs.  相似文献

Dorset (D) rams heterozygous for the Callipyge gene were single—sire mated to non-carrier ewes to produce Callipyge heterozygous (CLPG, n = 49) and normal (D, n = 33) lambs. Suffolk (S) and Texel (T) rams were mated to similar ewes to produce non-carrier crossbred S (n = 55) and T (n = 52) lambs. Lambs were finished on a high-energy diet to a target live weight of 57 kg. Pre-slaughter weight was recorded for each lamb prior to its transfer and slaughter through a commercial facility. Hot carcass weight and kidney and pelvic fat (KPF) were recorded at slaughter. Chilled carcasses were measured then fabricated into trimmed retail cuts by plant personnel. Each cut was weighed, and two loin chops were collected from each carcass for later cooking. CLPG lambs had the highest dressing % (53.6 versus 49.8–50.6; P < 0.05). At the same cold carcass weight, CLPG lambs had larger longissimus muscle areas (19.5 cm2 versus 14.0–15.2 cm2 for the rest; P < 0.05), less KPF (0.9 kg versus 1.04–1.13 kg; P < 0.05), less carcass fat (P < 0.05 for all measures), shorter carcasses (60.7 cm versus 61.8–64.7 cm; P < 0.05), and heavier trimmed sirloins, legs, and shoulders than any other group (a11 P < 0.05). They were similar to S lambs in receiving the lowest mean USDA yield grade. CLPG carcasses had the highest proportion of carcass weight represented by trimmed cuts (70% versus 65.7–67.8% for the rest; P < 0.05), the highest proportion of trimmed cuts (62.2% versus 59.7–60.6% for the rest; P < 0.05) represented by the most valuable cuts (leg + loin + rack + sirloin), and the highest composite carcass value ($135.8 versus $125–129 for the rest; P < 0.05). CLPG lambs also produced loin chops with the highest mean Warner–Bratzler shear values (5.4 kg versus 2.8–2.9 kg for the rest; P < 0.05) and the highest % cooking loss (31% versus 29–29.6% for the rest; P < 0.05).  相似文献

Background aimsEnumeration of viable CD34+ cells provides critical information for the bone marrow (BM) transplant physician. The single-platform ISHAGE protocol is the most reliable method currently available to quantitate accurately this important subset of cells. Previous studies have shown that 5 CD34+ cells/µL blood predicts the collection of at least 0.5 × 106 CD34+ cells/kg patient weight. From the apheresis product, infusion of 2.5 × 106 viable CD34+ cells (measured pre-cryopreservation)/kg patient weight will reliably permit engraftment of the hematopoietic system (as measured by the time to 20000 platelets/µL) by day 12–14 post-infusion.MethodsWe compared the CD34+ cell numbers derived from Flow Count-based Stem-Kit?; (Beckman Coulter) and Trucount? tube-based stem cell enumeration (SCE) kit (BD Biosciences) ISHAGE templates on BD FACSCalibur? and BD FACSCanto? cytometers on 12 granulocyte–colony-stimulating factor (G-CSF)-mobilized peripheral blood (MPB) and 10 peripheral blood stem cell (PBSC) samples.ResultsComparison of results showed that there was no statistical difference between samples run with Stem-Kit on the FACSCalibur versus SCE kit-based assays on either the FACSCalibur or FACSCanto. Mean results for the Stem-Kit/Calibur combination were 137, for SCE kit/Calibur 140 and for SCE kit/Canto 137 cells/µL. Pair-wise comparison of data based on rank order showed no statistically significant difference and all correlation coefficients had an R2>0.98.ConclusionsThe two kits generated very similar data on a range of fresh samples regardless of instrument platform. These results confirm and extend the utility of the single-platform ISHAGE protocols with a variety of reagent kits and instrument platforms.  相似文献

Background aimsVaricella zoster virus (VZV) causes life-long latent infection in healthy individuals, which reactivates in 10–68% of stem cell transplant patients. Reconstituting immunity through adoptive transfer of T cells specific for VZV may aid in the prophylaxis and treatment of VZV infections. The potential for generating T cells specific for VZV using a clinically approved VZV vaccine strain was investigated.MethodsThe Varivax® vaccine was used to stimulate peripheral blood mononuclear cells from healthy donors. Only reagents approved for clinical manufacture were used. Monocyte-derived dendritic cells pulsed with Varivax (R) were used to stimulate autologous mononuclear cells at a responder to stimulator ratio of 10:1. On day 7, a second stimulation was performed; 20 U/mL interleukin (IL)-2 were added from day 7 and 50 U/mL IL-2 from day 14 onwards. Cell phenotype and functionality were assessed after 21 days of culture.ResultsA mean increase of 11-fold in cell number was observed (n = 18). Cultures were mainly T cells (mean CD3 + 89.7%, CD4 + 54.2%, CD8 + 28.7%) with effector and central memory phenotypes. Cells produced one or more T helper (Th)1 cytokine (interferon-γ, tumor necrosis factor-α and IL-2), and CD4 + (but not CD8 +) cells expressed the cytoxicity marker CD107 when restimulated with VZV antigens.ConclusionsWe have demonstrated a clinically applicable method that yields high numbers of highly reactive T cells specific for VZV. We propose that reconstructing host immunity through adoptive transfer of VZV-specific T cells will reduce the frequency of clinical VZV infection in the period of severe immune suppression that follows allogeneic stem cell transplantation.  相似文献

BackgroundBariatric surgery is widely performed to improve obesity-related disorders, but can lead to nutrient deficiencies. In this study we examined serum trace element concentrations before and after bariatric surgery.MethodsWe obtained serum trace element concentrations by inductively coupled plasma-mass spectrometry (ICP-MS) method in 437 patients (82% women, median preoperative body-mass index 46.7 kg/m2 [interquartile range 42–51]) undergoing either gastric banding (22.7%), sleeve gastrectomy (20.1%), or gastric bypass (57.3%) procedures. Trace element data were available for patients preoperatively (n = 44); and 3 (n = 208), 6 (n = 174), 12 (n = 122), 18 (n = 39), 24 (n = 44) and 36 months (n = 14) post-operatively. All patients were recommended to take a multivitamin-mineral supplement after surgery.ResultsCopper deficiency was found in 2% of patients before surgery; and after surgery deficiency rates ranged from 0 to 5% with no significant change in median concentrations during follow-up (p = 0.68). Selenium deficiency was reported in 2% of patients before surgery; and after surgery deficiency rates ranged from 11 to 15% with a near-significant change in median concentrations (p = 0.056). Zinc deficiency was reported in 7% before surgery; and after surgery deficiency rates ranged from 7 to 15% with no significant change in median concentrations (p = 0.39).ConclusionsIn bariatric surgery patients recommended to take multivitamin-mineral supplements, serum copper, zinc and selenium concentrations were mostly stable during the first years after bariatric surgery. There was a possible tendency for selenium concentrations to decline during the early postoperative period.  相似文献
