Coiled morphotype Cylindrospermopsis raciborskii (Woloszynska) Seenayya et Subba Raju that forms a water bloom in a shallow pond in northern Taiwan exhibits a negative correlation between trichome size and temperature. To investigate how temperature influences the growth and trichome size of C. raciborskii, two C. raciborskii strains isolated from the pond in August and February were grown at three temperatures (18, 24 and 30°C). Both strains exhibited the lowest and highest specific growth rates at 18°C and 24°C, respectively, and the trichomes became the largest at 18°C. However, specific growth rates of the strain isolated in August exceeded those of the strain isolated in February, and the trichomes of the strain isolated in February were larger than those of the strain isolated in August regardless of temperature. Moreover, although both strains produced larger numbers of akinetes at higher temperatures, the strain isolated in August produced many more akinetes than did the other. These findings suggest that the two strains are not identical, leading to the conclusion that the C. raciborskii population in the pond consists of at least two ecotypes. Large trichome formation and akinete production are thought to be different types of countermeasure against cold of C. raciborskii, and the patterns of investment in developing these functions seemed to vary between the strains.  相似文献

Two Synechococcus strains from the Culture Collection of the Institute for Marine Sciences of Andalusia (Cádiz, Spain), namely Syn01 and Syn02, were found to be closely related to the model strain Synechococcus sp. PCC7002 according to 16S rDNA (99% identity). Pigment and lipid profiles and crtR genes of these strains were ascertained and compared. The sequences of the crtR genes of these strains were constituted by 888 bp, and showed 99% identity between Syn01 and Syn02, and 94% identity of Syn01 and Syn02 to Synechococcus sp. PCC7002. There was coincidence in photosynthetic pigments between the three strains apart from the pigment synechoxanthin, which could be only observed in Synechococcus sp. PCC7002. Species of sulfoquinovosyl‐diacyl‐glycerol (SQDG), phosphatidyl‐glycerol (PG), mono‐ and di‐galactosyl‐diacyl‐glycerol (MGDG and DGDG) were detected by high performance liquid chromatography‐mass spectrometry analysis of lipid extracts. The most abundant species within each lipid class were those containing C18:3 together with C16:0 fatty acyl substituents in the glycerol backbone of the same molecule. From these results it is concluded that these cyanobacterial strains belong to group 2 of the lipid classification of cyanobacteria.  相似文献

Composition of fatty acids in Boekelovia hooglandii Nicolai et Baas Becking (Chrysophyceae) was investigated as a function of salinity. It was confirmed by gas chromatography that the composition of fatty acids in cells cultured in a 50 mmol L?1 NaCl medium consisted of C14:0, C15:0, C16:0, C16:1, C18:0, C18:1, C18:2, C18:3, C18:4, C20:0, C20:4, C20:5, C22:5 and C22:6, in which C14:0, C16:0, C16:1, C18:4, C20:0, C20:5, C22:5 and C22:6 were main constituents. When the cells were cultured in a medium with different concentrations of NaCl ranging from 50 to 800 mmol L?1, the mole percentage of fatty acids such as C14:0, C16:0 and C16:1 decreased with increases in the salinity, while the mole percentage of highly polyunsaturated fatty acids such as C18:4, C20:5, C22:5 and C22:6 increased. When the cells were transferred from a 200 mmol L?1 NaCl medium to a 600 mmol L?1 NaCl medium, a decrease in mole percentage of C14:0, C16:0 and C16:1, and an increase in C18:4, C20:5, C22:5 and C22:6 were observed within 4 h. However, no change in the compositions of fatty acids was observed within 4 h when the cells were transferred from a 600 mmol L?1 NaCl medium to a 200 mmol L?1 NaCl one. The increase in the content of highly polyunsaturated fatty acids was considered to reflect the rapid response to upshock and to be the characteristic of salt tolerance in B. hooglandii.  相似文献

Representative strains of Gardnerella vaginalis were degraded using both an alkaline and an acid methanolysis and the fatty acid methyl esters released examined by thin-layer and gas chromatography. The profiles obtained were both qualitatively and quantitatively similar and were comprised of straight chain saturated and unsaturated non-hydroxylated fatty acids with hexadecanoic acid (16:0) and octadecenoic acid (18:1) the major components. All of the strains contained very characteristic polar lipid patterns consisting of diphosphatidylglycerol, phosphatidylglycerol, phosphatidylinositol, five partially identified glycolipids and an uncharacterised phospholipid. Analyses of wall amino acid preparations using gas chromatography showed that Gardnerella vaginalis strains contain major amounts of alanine, glycine, glutamic acid and lysine. The chemical data support the integrity of the genus Gardnerella.  相似文献

Oncorhynchus kawamurae (Osteichthyes: Salmonidae) (common name “Kunimasu”), a species endemic to Lake Tazawa, Akita Prefecture, Japan, was believed to have been extinct since 1940. However, nine specimens were discovered in March and April 2010 in Lake Saiko, Yamanashi Prefecture, one of the lakes to which eyed eggs of the species were introduced in 1935. These were identified as O. kawamurae because of having 47–62 pyloric caeca, 37–43 gill-rakers, a black-colored body, and spawning at 30–40 m depth in early spring, which are unique characteristics within Oncorhynchus. Furthermore, the distinctiveness of Kunimasu from sympatric kokanee (O. nerka) was supported by microsatellite DNA data.  相似文献

Viet nam is known as an endemic area of melioidosis but its etiologic agent originated in Viet nam was not extensively studied. For the first time, we analyzed the cellular lipid and fatty acid compositions of 15 Vietnamese isolates of Burkholderia pseudomallei, 10 from humans and 5 from the environment. Cellular lipid compositions were analyzed by two-dimensional thin-layer chromatography on silica gel G plates. Cellular fatty acid methyl esters were analyzed by gas chromatography (GC) and gas chromatography/mass spectrometry (GC/MS). The major lipids in all the isolates were phosphatidylglycerol (PG), two forms of phosphatidylethanolamine (PE-1 and PE-2), and two forms of ornithine-containing lipid (OL-1 and OL-2). PE-1 contained non-hydroxy fatty acids at both sn-1 and ?2 positions, while PE-2 possessed 2-hydroxy fatty acids and non-hydroxy fatty acids in a ratio of 1: 1. Since snake venom phospholipase A2 digestion of PE-2 liberated 2-hydroxy fatty acids, it was confirmed that these acids are at the sn-2 position of glycerol moiety. In both OL-1 and OL-2, amide-linked fatty acid was 3-hydroxy palmitic acid (3-OH-C16: 0), while ester-linked fatty acids were non-hydroxy acids in OL-1 and 2-hydroxy acids in OL-2. The total cellular fatty acid compositions of the test strains were characterized by the presence of 2-hydroxy palmitic (2-OH-C16: 0), 2-hydroxy hexadecenoic (2-OH-C16: 1), 2-hydroxy octadecenoic (2-OH-C18: 1), 2-hydroxy methylene octadecanoic (2-OH-C19CPA), 3-hydroxy myristic (3-OH-C14: 0) and 3-hydroxy palmitic (3-OH-C16: 0) acids. There were significant differences in the concentration of hexadecenoic (C16: 1), methylene hexadecanoic (C17CPA), octadecenoic (C18: 1) and methylene octadecanoic (C19CPA) acids among the Vietnamese isolates of B. pseudomallei. However, no significant difference was observed in cellular lipid and fatty acid components between strains of human and environmental origins.  相似文献

Fatty acid compositions of rainbow trout Oncorhynchus mykiss (Walbaum, 1792) was determined during embryogenesis, yolk-sac fry, swim up fry and fry stages. Embryonic and yolk-sac development of fertilized eggs were followed until fry weighed 25 g and samples were taken from each stage to analyse fatty acid composition. Analyses of fatty acid composition were performed by gas chromatography mass spectrometry. Throughout all the developmental stages of O. mykiss important changes, such as an increase in the percentage of 18:2, 18:3 and 20:5 and as a decrease in the percentage of 14:0, 16:2, 20:0, 20:2, 20:3, 20:4 and 22:0 were observed in the amounts of fatty acid of developing eggs. During transition from the embryonic to the yolk-sac stage, an important increase in 20:0, 20:3, 20:5, 22:0 and a decrease in 18:3, 22:1 was determined between 45-day old embryo and yolk-sac fry. During transition from the 43 day yolk-sac stage to swim up stage, a decrease in the percentage of 18:0, 20:2 and an increase in the percentage of 14:0, 16:2, 18:2, 18:3 was seen. In all the observed stages, no qualitative but significant quantitative variations were determined in the fatty acid composition of O. mykiss. It may be concluded that 14:0, 16:0, 16:1, 18:0, 18:1 18, 20 and 22 polyunsaturated fatty acids are necessary for normal development of O. mykiss during their life stages.  相似文献

Twenty four strains representing eight species of gram positive yellow-pigmented rods (Oerskovia turbata, Oerskovia xanthineolytica, CDC Coryneform groups A-3, A-4, A-5, Listeria denitrificans, Corynebacterium aquaticum and Brevibacterium acetylicum) were divided into two major groups based on the relative amounts of 12 methyltetradecanoate (15:0a) obtained by capillary gas liquid chromatography. O. turbata, O. xanthineolytica, CDC groups A-3 and A-4, L. denitrificans and C. aquaticum were placed in the first group due to the presence of a higher percentage (29–47%) of 15:0a, than CDC group A-5 and B. acetylicum. The latter contained 2–6% of this fatty acid, and were placed in the second group.All species in the two groups except C. aquaticum and CDC group A-4, were further separated from each other based on the qualitative and quantitative differences in their fatty acid compositions. In addition, the eight strains of CDC group A-5 revealed four different patterns and were further divided into four subgroups. This study supports the importance of the composition of cellular fatty acids in differentiating some closely related organisms.  相似文献

Aim: To verify a possible correlation between cell lipid composition, expression of key genes in lipid metabolism and fermentative behaviour of Saccharomyces cerevisiae wine strains. Methods and Results: The fermentative abilities of two commercial wine strains of S. cerevisiae were tested under stressful conditions. Cell number, glucose and fructose concentrations, expression of ACS1, ACS2, ACC1, OLE1, ERG9, ERG10, ARE1 and ARE2 and lipid content were evaluated. The strain that failed to complete the fermentation had lower amounts of C16:1 and C16:0 fatty acids at the beginning of fermentation (0 h) and late logarithmic phase (72 h). While the amount of C18:1 in this strain was lower than that in the strain that completed the fermentation at 0 h, same levels were observed for both strains at 72 h. The sterol levels were generally higher in the strain that failed to complete the fermentation. Gene expression generally increased from the beginning of the fermentation to the late logarithmic phase in both strains. Conclusion: A positive correlation between good fermentative ability, elevated fatty acid content and ACC1 gene expression has been identified. Significance and Impact of the Study: The cell lipid content at the time of inoculum and expression of ACC1 gene of starter strains should be carefully considered in order to identify the possible stuck/sluggish fermentations.  相似文献

Several strains of Pseudomonas were screened for lipase production using the rhodamine agar diffusion test. On the basis of the diameter of the halo produced, P. fragi CRDA 323 and P. putida ATCC 795 were considered to be good and weak lipase producers respectively. P. fragi, cultured in a 2-1 fermenter, produced a maximal amount of lipase after 3–4 days of incubation at 27°C. The lipase extract of P. fragi was obtained by acidification of culture supernatant at pH 4.0 and partially purified with ammonium sulphate precipitation. The majority of lipase activity (42%) was located in fraction IV, precipitated at 20%–40% of saturation, with a 19-fold enzyme purification. The K m and V max values for the partially purified enzymatic extract (fraction IV) were 0.70 mg/ml and 0.97 × 10–3 U/min respectively. Fraction IV, which showed and optimum activity at pH 8.5, was used for the interesterification of butter fat in a microemulsion free co-surfactant system containing Span 60 (sorbitol monostearate) and Tween 60 (polyoxyethylene sorbitan monostearate) in the ratio 48:52 (v/v). The results showed that the interesterification of butter fat resulted in a considerable decrease in long-chain saturated fatty acids (C12:0, C14:0 and C16:0) with a concomitant increase in C18:0 and C18:1 at the sn-2 position of selected triacylglycerols. In addition, the results demonstrated an increase in the fatty acids (C12:0, C14:0 and 16:0) among the 1 and 3 positions of the triacylglycerol molecules of modified buffer fat accompanied by a decrease in C18:0 and C18:1.  相似文献

The composition of fatty acids in 12 strains of the genera Thermus, Meiothermus, Geobacillus and Alicyclobacillus was analyzed by gas chromatography–mass spectrometry. Major FAs found in the profiles included i-15:0, i-17:0, ai-15:0, i-16:0, 16:0, ai-17:0, together with some minor components. Branched FAs were predominant, forming more than 80% of all FAs measured. Fast atom bombardment-mass spectrometry was used for analysis of unusual glycophospholipids, i.e., acylglycosylcardiolipins from genera Geobacillus and Alicyclobacillus and 1-(hydroxy(2-(O-acylglycosyl-oxy)hexadecyloxy)phosphoryloxy) hexadecan-2-yl esters of C15–C17 acids from genera Thermus and Meiothermus. Cloning and preliminary sequence analysis of 16S rDNA showed that these isolates belong to the genera Thermus, Meiothermus, Geobacillus and Alicyclobacillus.  相似文献

  • The Arabidopsis trichome is a polyploid epidermal cell resulting from multiple rounds of endocycles. The CYCLIN‐DEPENDENT KINASE INHIBITOR (CKI) family proteins are core cell cycle regulators that promote the endocycle. CONSTITUTIVE EXPRESSION OF PR GENES 5 (CPR5) is a plant‐specific nucleoporin. It has been found that two Arabidopsis CKI, SIAMESE (SIM) and SIAMESE‐RELATED 1 (SMR1), function downstream of CPR5 to activate plant effector‐triggered cell death. The sim smr1 double mutants form multicellular and clustered trichomes, while the cpr5 mutants produce dead and branchless trichomes. This study explored roles of the CPR5‐CKI signalling pathway in trichome cell cycle transition.
  • To examine the underlying mechanism of how cell cycle transition is regulated in plant trichomes, Trypan blue staining, flow cytometry, scanning electron microscopy (SEM) and nuclear DNA measurement were conducted.
  • The native promoter‐driven CKI and GUS fusion reporter showed that both SIM and SMR1 proteins were preferentially expressed in trichomes. The cpr5‐induced dead and branchless trichomes were fully suppressed by the sim smr1 double mutant, suggesting that SIM and SMR1 function downstream of CPR5 in trichome development. Flow cytometry analysis showed that as compared to the number of 2C (C = DNA content in a haploid nucleus) cells, the number of 4C cells significantly increased, whereas that of polyploidy cells (8C and 16C) dramatically decreased in the cpr5 mutant. The elevated 4C/2C ratio in the cpr5 mutant is consistent with de‐repression of pro‐endocycle regulators SIM and SMR1. The polyploidy cells (8C and 16C) may be selectively targeted to cell death, which is therefore attributed to the branchless trichomes in the cpr5 mutant. Nuclear DNA content analysis demonstrated that the nuclear DNA content of trichomes in the cpr5 sim mutant was significantly higher than in the sim mutant, indicating that CPR5 is a negative endocycle regulator in trichomes.
  • This study reveals that the CPR5‐CKI signalling pathway controls trichome cell cycle transition and excessive endocycles are required for cell death in plant trichomes.

Abstract The changes of fatty acids in larvae of the wheat blossom midge, Sitodiplosis mosellana (Gehin) at different periods were examined by gas choromatography. There were 10–16 kinds of fatty acids, of which the predominant ingredients were palmitic (C16:0), oleic (C18:1) and linoleic (C18:2) acids which were more than 95% in total fatty acids, stearic acid (C18:0) about 2%‐3.5% and any of the others was less than 1%. The fatty acid compositions increased from mid‐May, when larvae of the wheat blossom midge left the wheat‐ears and fallen on the ground, to April of next year before pupating and emerging. No arachidic acid (C20.0) was discovered in over‐summering, over‐wintering as well as inactive over‐wintered larvae. The content of saturated fatty acids in over‐summering, overwintering as well as inactive over‐wintered larvae were less than those of in active over‐wintered larvae and wheat‐ear larvae. Therefore, changes of the arachidic acid and the proportions of saturated fatty acids/unsaturated fatty acids could be used as one of the biochemical criteria to determine the active state and the degree of diapause in larvae of the wheat blossom midge.  相似文献

Twenty-six axenic strains of planktonic Anabaena with coiled trichomes belonging to 13 species were investigated by analyzing the pattern and content of their fatty acid composition, and by comparing their fatty acid composition with their morphological properties. In general, the planktonic Anabaena with coiled trichomes contained 14:0, 16:0, 16:1(cis-), 18:0, 18:1, 18:2, and 18:3() as their major fatty acid component, and were classified as Type 2 according to the Kenyon-Murata System. The Type 2 was further divided into two subtypes: Type 2A with 16:2 and 16:3, and Type 2B without 16:2 and 16:3. Among these Anabaena strains with coiled form, A. oumiana (NIES-73 and Ana Kas1) and A. eucompacta (Ana Chiba) contained Type 2B fatty acid composition, and other strains contained Type 2A. Among the strains with the latter type, A. circinalis (Ana Da) and A. curva (Ana Ao) had low levels of 18:3(). Most Anabaena strains with coiled trichomes showed a strong correlation between morphological characteristics and fatty acid composition.  相似文献

1. The composition of fatty acids (FA) from C14 to C18 was measured for edible seston and for individual Daphnia galeata , Diaphanosoma brachyurum and Eodiaptomus japonicus from Lake Biwa using a pyrolysis–gas chromatography (Py–GC) method. Based on the relative abundance of the FA, inter-and intraspecific differences in composition were examined.
2. Among the FA, C16 : 0 was the most abundant, both in the three zooplankton species and in edible seston smaller than 20 μm, suggesting that the composition of the zooplankton was roughly reflected by their diet. However, the abundance of C18 : 1 relative to C16 : 1 and C18 : 0 was much higher in each zooplankton species than in the diet.
3. Comparison of the relative abundance of FA among the three zooplankton species revealed that intraspecific differences in FA composition are greater than interspecific differences. These results indicate that variability in FA composition is not necessarily species-specific, and can be obscured by variation in composition between individuals.  相似文献

As understanding of the evolutionary relationships between strains and species of root nodule bacteria increases the need for a rapid identification method that correlates well with phylogenetic relationships is clear. We have examined 123 strains ofRhizobium: R. fredii (19),R. galegae (20),R. leguminosarum (22),R. loti (17),R. meliloti (21), andR. tropici (18) and six unknowns. All strains were grown on modified tryptone yeast-extract (TY) agar, as log phase cultures, scraped from the agar, lysed, and the released fatty acids derivatized to their corresponding methyl esters. The methyl esters were analysed by gas-chromatography using the MIDI/Hewlett-Packard Microbial Identification System. All species studied contained 16:0, 17:0, 18:0 and 19cyclow9C fatty acids but onlyR loti andR tropici produced 12:0 3 OH,13:0 iso 3 OH,18:1w9C and 15:0 iso 3 OH,17:0 iso 3 OH and 20:2w6,9C fatty acids respectively. Principal component analysis was used to show that strains could be divided into clusters corresponding to the six species. Fatty acid profiles for each species were developed and these correctly identified at least 95% of the strains belonging to each species. A dendrogram is presented showing the relationships betweenRhizobium species based on fatty acid composition. The data base was used to identify unknown soil isolates as strains ofRhizobium lacking a symbiotic plasmid and a bacterium capable of expressing a symbiotic plasmid fromR. leguminosarum asSphingobacterium spiritovorum.  相似文献

The effect of different solvents and pollutants on the cellular fatty acid composition of three bacterial strains: Thauera aromatica, Geobacter sulfurreducens and Desulfococcus multivorans, representatives of diverse predominant anaerobic metabolisms was investigated. As the prevailing adaptive mechanism in cells of T. aromatica and G. sulfurreducens whose cellular fatty acids patterns were dominated by palmitic acid (C16:0) and palmitoleic acid (C16:1cis), the cells reacted by an increase in the degree of saturation of their membrane fatty acids when grown in the presence of sublethal concentrations of the chemicals. Next to palmitic acid C16:0, the fatty acid pattern of D. multivorans was dominated by anteiso-branched fatty acids which are characteristic for several sulfate-reducing bacteria. The cells responded to the solvents with an increase in the ratio of straight-chain saturated (C14:0, C16:0, C18:0) to anteiso-branched fatty acids (C15:0anteiso, C17:0anteiso, C17:1anteisoΔ9cis). The results show that anaerobic bacteria react with similar mechanisms like aerobic bacteria in order to adapt their membrane to toxic organic solvents. The observed adaptive modifications on the level of membrane fatty acid composition can only be carried out with de novo synthesis of the fatty acids which is strictly related to cell growth. As the growth rates of anaerobic bacteria are generally much lower than in the so far investigated aerobic bacteria, this adaptive response needs more time in anaerobic bacteria. This might be one explanation for the previously observed higher sensitivity of anaerobic bacteria when compared with aerobic ones.  相似文献
