神经元轴突外包裹的髓鞘结构对于提高神经元传导速率,维持神经系统稳定性有重要作用。在中枢神经系统中,髓鞘主要由少突胶质细胞形成。成髓鞘过程在内源性和外源性因素的共同调节下进行,神经元轴突信号在这个过程中扮演重要角色。髓鞘发育过程依赖于轴突的促进信号和抑制信号的相互平衡:促进信号包括层粘连蛋白和神经调节素等,神经元电信号能启动并促进髓鞘再生;抑制信号包括细胞黏附分子以及Notch信号。本文综述了一些因子尤其是神经元信号在髓鞘发育中的作用,也讨论了脱髓鞘疾病中神经元如何参与髓鞘再生。这些总结有助于理解髓鞘发育的机制,也有助于脱髓鞘疾病的研究和治疗。  相似文献

少突胶质细胞在中枢神经系统中具有重要和广泛的生理功能。视神经损伤后,出现髓鞘脱失、少突胶质细胞死亡和髓鞘再生等病理改变,产生的髓鞘碎片能抑制视神经轴索再生。少突胶质细胞的抑制特性由特定的抑制分子介导,目前已鉴定的抑制分子主要有Nogo、髓鞘相关糖蛋白(myelin—associated glycoprotein,MAG)、少突胶质细胞髓鞘糖蛋白(oligodendrocyte myelin glycoprotein,OMgp)等,它们通过同一受体复合体传导抑制信号。阻滞抑制分子及其受体,或调整神经元的内在生长状态以克服抑制分子的抑制作用,可以促进视神经损伤后再生。本文就这方面的进展作一综述。  相似文献

肌腱蛋白R(tenascin-R, TN-R)是一种重要的细胞外基质糖蛋白(extracellular matrix,ECM).分布于中枢神经系统, 主要在髓鞘形成早期的少突胶质细胞中表达,成熟的胶质细胞及某些神经元(如脊髓,视网膜, 小脑和海马的中间神经元)也有表达.TN-R具有复杂的结构,由三种不同的结构域组成,从氨基端到羧基端依次为:类似于表皮生长因子的重复片段, 类似于Ⅲ型纤连蛋白重复片段,类似(血)纤维蛋白原片段组成.TN-R具有多种复杂的功能, 对神经元具有排斥作用,促进或抑制神经元突起的生长, 诱导神经元形态的极性化, 并和髓鞘的形成有关,TN-R结构的复杂性和功能的多样性提示,TN-R有多个受体存在,已经发现的受体有F3/F11,MAG,XL1,Xprocan等.  相似文献

LRRC4基因是候选的脑胶质瘤抑制基因,其细胞外区域含有一个保守的LRR和一个IgC2 结构域.本研究结合生物信息学分析,通过一步PCR法构建了不同结构域缺失的突变体(pc ΔLRR, pcΔIgC2或pcΔTm).并将全长LRRC4(pcLRRC4)和各突变体转染至U251细胞,构建了稳定表达的U251细胞系.通过MTT,软琼脂集落形成及Transwell体外侵袭模型检测发现,全长LRRC4能够抑制U251细胞的生长和侵袭;LRR结构域缺失的突变体不再抑制U251细胞的生长和侵袭;而IgC2或Tm区缺失的突变体仍然可以抑制U251细胞的生长和侵袭.该结果表明,LRRC4抑制U251细胞的生长和侵袭依赖于它的LRR结构域,而不是IgC2或Tm结构域.  相似文献

目的观察神经元-小胶质细胞间EphA4/ephrin信号通路在脑缺血后的炎性损伤中的作用及机制。方法建立神经元-小胶质细胞混合培养体系和糖氧剥夺再复氧(oxygen-glucose deprivation and reperfusion, OGD/R)模型,使用预聚集化的EphA4-Fc激动小胶质细胞ephrin配体,检测OGD/R后的细胞凋亡、小胶质细胞增殖和亚型极化以及小胶质细胞功能改变。结果 EphA4受体高表达于原代神经元,与对照组相比,预聚集化EphA4-Fc干预加重OGD/R导致的细胞凋亡,促进小胶质细胞增殖以及向M1型(促炎型)极化(炎症表型)。结论神经元-小胶质细胞间EphA4/ephrin信号通路通过调控小胶质细胞亚型极化参与脑缺血后的炎性损伤的过程。  相似文献

髓鞘来源的中枢神经再生抑制因子——Nogo-A被发现除表达于成熟的少突胶质细胞,还广泛高表达于多种类型的神经元中.目前,神经元中Nogo-A的功能还不明确.为探讨神经元内Nogo-A的功能,以HEK293FT细胞为模型,利用信号途径报告基因系统筛选过表达Nogo-A对多种信号途径的调控作用,发现过表达Nogo-A能特异激活NF-κB信号,利用不同的Nogo-A剪接体和截断体形式研究,证明Nogo-A激活NF-κB信号依赖于其氨基端的proline rich结构域,进一步使用NF-κB信号途径相关分子显性突变体揭示IκBα、TRAF6、 Rac/Cdc42 参与Nogo-A激活NF-κB信号.结果提示,Nogo-A可以显著激活NF-κB信号,且依赖于Nogo-A氨基段的proline rich结构域.  相似文献

既往的研究中,乳酸一直被认为是细胞糖酵解产生的代谢废物。然而,近年的研究表明,乳酸可作为重要的能量代谢底物和信号分子影响多组织器官的生理进程,其运输载体单羧酸转运体及受体G蛋白偶联受体81可能在神经保护过程中发挥关键作用。在中枢神经系统内,乳酸可作用于多种细胞如神经元、星形胶质细胞、小胶质细胞、少突胶质细胞和血管内皮细胞等,参与改善脑能量代谢,增强神经发生,提高突触可塑性,降低神经炎症,缓解神经毒性,促使髓鞘再生,进而影响个体认知功能。适宜的运动有利于脑健康,但运动能否通过调节乳酸及相关生物学机制改善认知功能未见详尽报道。该文通过分析运动如何调控乳酸及其运输载体和受体的生理水平,以及乳酸如何调节认知功能,探讨乳酸作为运动改善认知功能中介分子的可能性,为借助运动疗法缓解认知衰退的相关疾病提供理论支持。  相似文献

中枢神经系统 (CNS)轴突再生的主要障碍之一是存在抑制再生的蛋白 ,迄今 ,已在少突胶质细胞 /髓鞘中相继发现至少三个重要的轴突再生抑制蛋白 ,即髓鞘相关糖蛋白 (MAG)、Nogo A和少突胶质细胞 /髓鞘糖蛋白 (OMgp)。最近的研究又证实 ,这三个不同的抑制成分可能主要通过与一个共同的受体Nogo6 6受体 (NgR)结合而发挥作用。这些研究成果扩充了对CNS损伤后轴突再生障碍的理解 ,也为探讨CNS损伤的治疗新策略提供了新的思路。  相似文献

凋亡相关蛋白中的Bcl-2家族是细胞凋亡的关键调节分子,由抗凋亡和促凋亡成员组成,这些成员之间通过相互协同作用调节了线粒体结构与功能的稳定性,从而在线粒体水平发挥着细胞凋亡的“开关”作用.抗凋亡成员大都分布于线粒体的外膜,与促凋亡成员的BH3结构域相互作用对细胞凋亡发挥抵抗作用.促凋亡成员则主要分布于细胞浆中,细胞接受死亡信号刺激后,促凋亡成员自身受到一系列的调节,如典型的Bax构象改变、BAD和Bik的磷酸化调节以及Bid和Bim的蛋白裂解效应等,使得促凋亡成员在凋亡信号的刺激下整合于线粒体外膜,最终导致线粒体通透转换孔的开放,进而释放包括细胞色素c、凋亡诱导因子、Smac等重要的凋亡因子,随后caspase被激活进而断裂重要的细胞内结构蛋白与功能分子,执行细胞凋亡.  相似文献

在中枢神经系统中,脱髓鞘病变后的机体可以实现一定程度的再髓鞘修复,但尚不足以诱导功能恢复。越来越多的研究表明,神经元电活动作为一种轴突信号,调控着发育性成髓鞘过程并且能够启动及促进轴突再髓鞘。通过激活神经元电活动促进轴突再髓鞘修复的策略已经成为治疗相关疾病的潜在突破口,但该再髓鞘过程中涉及到的少突胶质谱系及星形胶质细胞的变化及机制仍不够清楚。该文拟主要围绕神经元电活动调控轴突再髓鞘的相关机制进行综述,为今后研究提供参考。  相似文献

LINGO-1 has been critically implicated in the central regulation of CNS axon regeneration and oligodendrocyte maturation. We have recently demonstrated that pretreatment with LINGO-1 antagonist (LINGO-1-Fc) inhibited low potassium-induced cerebellar granular neurons (CGNs) apoptosis. In the present study, we examined the neuroprotective mechanism of LINGO-1-Fc by Western blot and in situ GST pull-down assay. CGN cultures were preincubated in medium with LINGO-1-Fc or control protein at the concentration of 10 mug/ml for 2 h and then switched to low potassium medium in the presence of corresponding proteins. Cultures were harvested at indicated time intervals for successive analysis. Several apoptosis-associated signaling factors, GSK-3beta, ERK1/2, and Rho GTPases, were observed to be activated in response to potassium deprivation and the activation/dephosphorylation of GSK-3beta was suppressed by LINGO-1-Fc pretreatment compared with control group. Besides, the endogenous LINGO-1 expression level of CGN cultures was augmented by low potassium stimuli and restrained by LINGO-1 antagonist treatment. Although the protein level of p75(NTR) and Nogo-A were down-regulated in different patterns during apoptosis, neither of them was affected by LINGO-1-Fc application. Taken together, these results suggest a new mechanism of LINGO-1 antagonist regulated neuronal survival involving protein synthesis of LINGO-1 and inactivation of GSK-3 pathway.  相似文献

The effects of L1-Fc and CHL1-Fc fusion proteins on neuronal survival were investigated. Cerebellar granule neurons of mouse and hippocampal neurons of rat embryo undergo apoptosis when cultured in serum-free medium. Treatment with chimeric proteins containing the extracellular domains of the neural adhesion molecules L1 or CHL1 fused to the Fc region of human immunoglobulin significantly enhanced the survival of neurons. Compared to the control, the percentage of surviving neurons increased about 60% and 45% with L1 and CHL1 fusion proteins, respectively. A fusion protein containing the extracellular domain of NCAM had no effect on survival. The L1 and CHL1 fusion proteins were effective both in soluble form or when offered as a substrate, with the maximal effect at about 1 microg/mL. To explore the intracellular events related to the neuronal survival effects of L1-Fc fusion protein, Bcl-2 and c-Jun expression were analyzed by Western blotting. The level of Bcl-2 in cerebellar granule neurons was increased by treatment with L1-Fc at both 1 and 5 days of culture. The level of c-Jun was not significantly affected at the early time point and was reduced by L1-Fc fusion protein after long-term culture. The results demonstrate that the neural adhesion molecule L1 and its relative CHL1 are potential neuronal survival factors for neurons of the central nervous system. Bcl-2 may serve as one of the intracellular mediators of the neuronal survival effects of L1.  相似文献

A major obstacle for successful axon regeneration in the adult central nervous system (CNS) arises from inhibitory molecules in CNS myelin, which signal through a common receptor complex on neurons consisting of the ligand-binding Nogo-66 receptor (NgR) and two transmembrane coreceptors, p75 and LINGO-1. However, p75 expression is only detectable in subpopulations of mature neurons, raising the question of how these inhibitory signals are transduced in neurons lacking p75. In this study, we demonstrate that TROY (also known as TAJ), a TNF receptor family member selectively expressed in the adult nervous system, can form a functional receptor complex with NgR and LINGO-1 to mediate cellular responses to myelin inhibitors. Also, both overexpressing a dominant-negative TROY or presence of a soluble TROY protein can efficiently block neuronal response to myelin inhibitors. Our results implicate TROY in mediating myelin inhibition, offering new insights into the molecular mechanisms of regeneration failure in the adult nervous system.  相似文献

In mouse cerebellar granule neurons (CGNs) the marine neurotoxin domoic acid (DomA) induces neuronal cell death, either by apoptosis or by necrosis, depending on its concentration, with apoptotic damage predominating in response to low concentrations (100 nM). DomA-induced apoptosis is due to selective activation of AMPA/kainate receptors, and is mediated by DomA-induced oxidative stress, leading to mitochondrial dysfunction and activation of caspase-3. The p38 MAP kinase and the c-Jun NH2-terminal protein kinase (JNK) have been shown to be preferentially activated by oxidative stress. Here we report that DomA increases p38 MAP kinase and JNK phosphorylation, and that this effect is more pronounced in CGNs from Gclm (-/-) mice, which lack the modifier subunit of glutamate-cysteine ligase, have very low glutathione (GSH) levels, and are more sensitive to DomA-induced apoptosis than CGNs from wild-type mice. The increased phosphorylation of JNK and p38 kinase was paralleled by a decreased phosphorylation of Erk 1/2. The AMPA/kainate receptor antagonist NBQX, but not the NMDA receptor antagonist MK-801, prevents DomA-induced activation of p38 and JNK kinases. Several antioxidants (GSH ethyl ester, catalase and phenylbutylnitrone) also prevent DomA-induced phosphorylation of JNK and p38 MAP kinases. Inhibitors of p38 (SB203580) and of JNK (SP600125) antagonize DomA-induced apoptosis. These results indicate the importance of oxidative stress-activated JNK and p38 MAP kinase pathways in DomA-induced apoptosis in CGNs.  相似文献

Although the role of the microtubule-binding domain of the tau protein in the modulation of microtubule assembly is widely established, other possible functions of this protein have been poorly investigated. We have analyzed the effect of adenovirally mediated expression of two fragments of the N-terminal portion - free of microtubule-binding domain - of the tau protein in cerebellar granule neurons (CGNs). We found that while the expression of the tau (1-230) fragment, as well as of full-length tau, inhibits the onset of apoptosis, the tau (1-44) fragment exerts a powerful toxic action on the same neurons. The antiapoptotic action of tau (1-230) is exerted at the level of Akt-mediated activation of the caspase cascade. On the other hand, the toxic action of the (1-44) fragment is not prevented by inhibitors of CGN apoptosis, but is fully inhibited by NMDA receptor antagonists. These findings point to a novel, physiological role of the N-terminal domain of tau, but also underlay that its possible proteolytic truncation mediated by apoptotic proteases may generate a highly toxic fragment that could contribute to neuronal death.  相似文献

The neuroprotective properties of bis(7)-tacrine, a novel dimeric acetylcholinesterase (AChE) inhibitor, on glutamate-induced excitotoxicity were investigated in primary cultured cerebellar granule neurons (CGNs). Exposure of CGNs to 75 mum glutamate resulted in neuronal apoptosis as demonstrated by Hoechst staining, TUNEL, and DNA fragmentation assays. The bis(7)-tacrine treatment (0.01-1 mum) on CGNs markedly reduced glutamate-induced apoptosis in dose- and time-dependent manners. However, donepezil and other AChE inhibitors, even at concentrations of inhibiting AChE to the similar extents as 1 mum bis(7)-tacrine, failed to prevent glutamate-induced excitotoxicity in CGNs; moreover, both atropine and dihydro-beta-erythroidine, the cholinoreceptor antagonists, did not affect the anti-apoptotic properties of bis(7)-tacrine, suggesting that the neuroprotection of bis(7)-tacrine appears to be independent of inhibiting AChE and cholinergic transmission. In addition, ERK1/2 and p38 pathways, downstream signals of N-methyl-d-aspartate (NMDA) receptors, were rapidly activated after the exposure of glutamate to CGNs. Bis(7)-tacrine inhibited the apoptosis and the activation of these two signals with the same efficacy as the coapplication of PD98059 and SB203580. Furthermore, using fluorescence Ca(2+) imaging, patch clamp, and receptor-ligand binding techniques, bis(7)-tacrine was found effectively to buffer the intracellular Ca(2+) increase triggered by glutamate, to reduce NMDA-activated currents and to compete with [(3)H]MK-801 with an IC(50) value of 0.763 mum in rat cerebellar cortex membranes. These findings strongly suggest that bis(7)-tacrine prevents glutamate-induced neuronal apoptosis through directly blocking NMDA receptors at the MK-801-binding site, which offers a new and clinically significant modality as to how the agent exerts neuroprotective effects.  相似文献

Aberrant activation of the Rb/E2F1 pathway in cycling cells, in response to mitogenic or nonmitogenic stress signals, leads to apoptosis through hyperphosphorylation of Rb. To test whether in postmitotic neurons the Rb/E2F1 pathway can be activated by the nonmitogenic stress signaling, we examined the role of the p38 stress-activated protein kinase (SAPK) in regulating Rb phosphorylation in response to Fas (CD95/APO1)-mediated apoptosis of cultured cerebellar granule neurons (CGNs). Anti-Fas antibody induced a dramatic and early activation of p38. Activated p38 was correlated with the induction of hyperphosphorylation of both endogenous and exogenous Rb. The p38-selective inhibitor, SB203580, attenuated such an increase in pRb phosphorylation and significantly protected CGNs from Fas-induced apoptosis. The cyclin-dependent kinase-mediated Rb phosphorylation played a lesser role in this neuronal death paradigm, since cyclin-dependent kinase inhibitors, such as olomoucine, roscovitine, and flavopiridol, did not significantly prevent anti-Fas antibody-evoked neuronal apoptosis. Hyperphosphorylation of Rb by p38 SAPK resulted in the release of Rb-bound E2F1. Increased E2F1 modulated neuronal apoptosis, since E2F1-/- CGNs were significantly less susceptible to Fas-mediated apoptosis in comparison with the wild-type CGNs. Taken together, these studies demonstrate that neuronal Rb/E2F1 is modulated by the nonproliferative p38 SAPK in Fas-mediated neuronal apoptosis.  相似文献
