采用分子印迹技术,以芹菜素(API)为模板分子,2-乙烯基吡啶(2-Vpy)为功能单体,乙二醇二甲基丙烯酸酯(EDMA)为交联剂,合成了芹菜素分子印迹聚合物(API-MIP).通过紫外光谱研究了API与2-Vpy的非共价作用,采用平衡结合试验评价了API-MIP对底物分子的识别能力和吸附性能,进行了吸附动力学研究.结果表明:API-MIP通过静电作用对API产生分子识别,并对API表现出很高的亲和力;Scatchard分析表明,API-MIP中存在对API有不同亲和力的两类结合位点;与对照物在API-MIP上的吸附行为比较表明,API-MIP对API具有良好的分子识别性能.API-MIP对API的选择性吸附基本在1.5 h内完成. 相似文献

以乙酸镍和2,2′-联吡啶配合物为模板、4-乙烯吡啶为功能单体、乙二醇二甲基丙烯酸酯为交联剂,合成了具有类似金属螯合抗体结合位点的金属配合物印迹聚合物,系统研究了金属离子对模板聚合物选择性结合2,2′-联吡啶的调节作用。结果表明:印迹聚合物对N i(Ⅱ)-2,2′-联吡啶配合物有选择性识别能力。采用Scatchard模型评价分子印迹聚合物的结合特性,得到高亲和力结合位点的平衡离解常数Kd1=0.082 mm o l/L,表观最大结合量Qm ax1=82.2μm o l/g,低亲和力结合位点的Kd2=0.400 mm o l/L,Qm ax2=91.3μm o l/g。 相似文献

Targeted recombination in mammalian cells is rare compared with non-homologous integration. In Saccharomyces cerevisiae the reverse is true. Differences in targeting efficiency could arise because a target of unique DNA is 200 times more dilute in mammalian genomes than it is in yeast. We tested this possibility by measuring gene targeting in normal CHO cells with two copies of the dihydrofolate reductase (DHFR) gene and in amplified CHOC 400 cells, which carry 800 copies. If the concentration of the target gene is critical, amplified cells should show an enhanced frequency of targeted recombination relative to non-homologous integration. Using a positive/negative selection protocol, we demonstrated that the efficiency of targeting into DHFR genes is indistinguishable in normal and amplified CHO cells. As targeting does not depend on the number of targets, the search for homology is not a rate-limiting step in the mammalian pathway of gene targeting. Thus, the difference in genome size is not the basis for the different outcomes of targeting experiments in S. cerevisiae and mammals. 相似文献

Proteomic studies have yielded detailed lists of the proteins present in a cell. Comparatively little is known, however, about how these proteins interact and are spatially arranged within the 'functional modules' of the cell: that is, the 'molecular sociology' of the cell. This gap is now being bridged by using emerging experimental techniques, such as mass spectrometry of complexes and single-particle cryo-electron microscopy, to complement traditional biochemical and biophysical methods. With the development of integrative computational methods to exploit the data obtained, such hybrid approaches will uncover the molecular architectures, and perhaps even atomic models, of many protein complexes. With these structures in hand, researchers will be poised to use cryo-electron tomography to view protein complexes in action within cells, providing unprecedented insights into protein-interaction networks. 相似文献

In this study, GFP mRNA in COS-7 cell and GFP-transfected COS-7 cell was detected in real time using phosphorothioate-modified
molecular beacon based on living cell imaging method. Results showed that phosphorothioate-modified molecular beacon still
kept the advantages of molecular beacon, such as, excellent selectivity, high sensitivity, and no separation detection. In
addition, this modification could significantly increase the nuclease resistance of molecular beacon. Phosphorothioate-modified
molecular beacon can efficiently reduce the false positive signal and improve the accuracy of living cell mRNA detection.

Diabetes-specific microvascular disease is a leading cause of blindness, renal failure and nerve damage, and diabetes-accelerated atherosclerosis leads to increased risk of myocardial infarction, stroke and limb amputation. Four main molecular mechanisms have been implicated in glucose-mediated vascular damage. All seem to reflect a single hyperglycaemia-induced process of overproduction of superoxide by the mitochondrial electron-transport chain.
