地衣芽孢杆菌16S rRNA基因的TD-PCR扩增及系统发育分析   总被引:1,自引:0,他引:1

运用16SrRNA基因序列分析了中国工业微生物菌种保藏管理中心(CICC)保存的30株地衣芽孢杆菌的系统发育关系,结果显示:24株菌株位于地衣芽孢杆菌系统发育分支;3株菌株位于蜡状芽孢杆菌-苏云金芽孢杆菌系统发育分支;1株菌株位于枯草芽孢杆菌系统发育分支;2株菌株与其它地衣芽孢杆菌菌株间序列同源性为96.4%~97.4%,明显低于其它地衣芽孢杆菌菌株间同源性,分类地位不明确,有待进一步讨论。通过比较分析16SrRNA基因5′端500bp、3′端500bp以及其全基因的系统发育树,表明16SrRNA基因5′端500bp可以很好的代表全基因序列进行系统发育研究,可用于区分地衣芽孢杆菌、枯草芽孢杆菌以及蜡状芽孢杆菌分支。  相似文献

对来自4个不同省份的5条蚯蚓的肠道及体表细菌进行分离,共获得122株细菌。通过脱脂奶粉平板法初筛,纤维蛋白平板法复筛,以透明圈为筛选标记,共筛选出产纤溶酶菌株12株,其中菌株SC-3-W-3的纤溶酶活力较高,达到了538.64 U/mL(相当于尿激酶的活力单位)。通过对其形态、培养、生理生化特征进行研究,发现其与蜡状芽孢杆菌Bacillus cereus Frankland的特征很相符。进一步对SC-3-W-3的16S rDNA序列及系统发育分析表明,该菌株与蜡状芽孢杆菌的同源性高达100%。综合生理生化及16S rDNA序列比对结果,将SC-3-W-3菌株鉴定为蜡状芽孢杆菌。  相似文献

蜡状芽孢杆菌是能产生孢子的革兰氏阳性菌,广泛分布于水体和土壤中,因能引起食物中毒而广为人知。本研究中,从养殖大菱鲆体表分离到一株溶血性细菌,经形态学鉴定和系统发育学分析,鉴定为蜡状芽孢杆菌。根据分离的时间和地点,这株溶血性的菌株被命名为蜡状芽孢杆菌WH2015。溶血试验证实WH2015菌株能够裂解羊红细胞。对蜡状芽孢杆菌WH2015基因组测序和分析的结果显示其基因组大小为5.8 Mb,平均G+C含量为35%,有6 568个基因。与其它蜡状芽孢杆菌株系进行比较基因组分析显示,WH2015菌株基因组几乎拥有所有的核心溶血性基因,提示其溶血机理与其它报导过蜡状芽孢杆菌株系的溶血机理相似。本研究在分离鉴定养殖大菱鲆体表溶血性蜡状芽胞杆菌WH2015的基础上,对其基因组进行了测序和分析并鉴定了其所具有的致病性基因和毒力因子,帮助我们理解由蜡状芽孢杆菌引起的养殖大菱鲆疾病,以及为其防治措施的实施和疾病的治疗提供可靠的理论基础。  相似文献

四株高产磷脂酶C新菌株的鉴定和分类   总被引:4,自引:0,他引:4  
在前面研究高产磷脂酶C(PLC)菌株筛选及其抗血小板功能的基础上,对新筛选的四株高产PLC的754-1、779、970和1107菌株进行了鉴定和分类。通过形态特征、培养特征、生理生化特征以及16S rRNA序列测定及其同源性分析,证实754-1菌株与Bacillus cereus基本一致,因此将754-1菌株分类属于蜡状芽孢杆菌,命名为蜡状芽孢杆菌深圳株754-1,B.cereus shenzhen 754-1 strain。而779、970和1107三株菌则与Bacillus mycoides基本一致,因而将这三株菌分类属于蕈状芽孢杆菌,分别命名为蕈状芽孢杆菌深圳株779,B.mycoides shenzhen 779 strain,草状芽孢杆菌深圳株970,B.mycoides shenzhen 970 strain,和蕈状芽孢杆菌深圳株1107,B.mycoides shenzhen 1107 strain。这将为进一步利用这些菌株及其PLC打下坚实的基础。  相似文献

本文对33株枯草芽孢杆菌群菌株进行β-甘露聚糖酶活性筛选,其中的32株具有β-甘露聚糖酶活性,只有1株无β-甘露聚糖酶活性.通过基因克隆测序的方法获得33株枯草芽孢杆菌群菌株β-甘露聚糖酶基因编码区全序列,对酶基因进行同源性分析并构建系统发育树;在β-甘露聚糖酶基因系统发育树中,33株枯草芽孢杆菌群菌株聚为3个分支,分别是枯草芽孢杆菌分支、地衣芽孢杆菌分支和解淀粉芽孢杆菌分支;枯草芽孢杆菌、地衣芽孢杆菌和解淀粉芽孢杆菌β-甘露聚糖酶基因种内同源性大于91%,而种间同源性为60%69%.  相似文献

从曾侯乙墓中椁室、东椁室、西椁室和北椁室分别取样,经平板划线培养,分离,纯化共获得典型的菌株16株。将16株分别涂片镜检,生理生化分析鉴定,结果证明芽孢杆菌属11株,其中苏云金变种1株,地衣芽孢杆菌1株,巨大芽孢杆菌1株,球形芽孢杆菌5株,蜡状芽孢杆菌2株,多粘芽孢杆菌1株。其余菌株微杆菌属4株,黄色杆菌属黄杆菌1株。  相似文献

从中国农业科学院北京畜牧兽医研究所鸡舍附近土壤中分离到一株芽孢杆菌P-25,并进行了分子鉴定。通过形态鉴定、革兰氏染色、生理生化测定、16SrRNA序列分析和系统发育树构建,确定该菌株为蜡状芽孢杆菌(Bacillus cereus),其16SrRNAGenBank登录号为GU271135。  相似文献

豆豉纤溶酶高产菌株的筛选研究   总被引:5,自引:0,他引:5  
以中国豆豉为材料,取6个不同来源的样品经富集培养后,用自制血纤维蛋白平板进行初筛;利用LB纤维蛋白平板复筛与血琼脂平板进行体外溶血试验相结合的方法,筛选出安全性良好并有一定纤溶活性的菌株。再对复筛得到的菌株进行摇瓶培养,用纤维蛋白平板法测定并比较不同菌株发酵液的纤溶活性。结果成功地筛选出5株纤溶活性高和通过溶血性实验的芽孢杆菌菌株,其中菌株DC-C4粗酶液的酶活稍高,达到280IU/mL。采用16S-23S rDNA序列分析法及传统的生理生化特征鉴定法对该菌株进行鉴定,确定菌株DC-C4为蜡状芽孢杆菌(Bacillus cereus)。本实验为开发新型溶栓药物及保健食品提供了实验参考依据。  相似文献

一株碱性纤维素酶产生菌的分离、鉴定及酶谱分析   总被引:1,自引:0,他引:1  
目的:从土样中分离株碱性纤维素酶高产菌株.方法:利用CMC平板初筛,然后利用摇瓶复筛,筛选酶活力高的菌株,对分离出的一株高产菌株进行了鉴定并对其所产酶进行了酶谱分析.结果:获得一株碱性纤维素酶高产菌株H12,酶活力达到1.96U/ml.结论:该菌株呈长杆状、革兰氏染色为阳性、产芽孢;16S rDNA基因序列为1 419bp,与短小芽孢杆菌16S rDNA基因序列具有最高的同源性,基于16S rDNA基因序列的同源性分析以及系统发育分析等方面的多相分类研究,鉴定菌株H12为为短小芽孢杆菌;碱性纤维素酶的酶谱分析只有一条水解条带,酶分子量在75kD左右.  相似文献

规律成簇间隔短回文重复序列(clustered regularly interspaced short palindromic repeats,CRISPR)是最近发现针对噬菌体等外源遗传物质的获得性和可遗传性的新型原核生物防御系统。通过BLAST、多序列比对、RNA二级结构预测等生物信息学方法对已经完成全基因组测序的蜡状芽孢杆菌群24个菌株进行CRISPR的系统分析,结果表明:42%的菌株含有该结构;8个CRISPR座位的正向重复序列可以形成RNA二级结构,提示正向重复序列可能介导外源DNA或RNA与CAS编码蛋白的相互作用;31%的间区序列与噬菌体、质粒、蜡状芽孢杆菌群基因组序列具有同源性,进一步验证间区序列很可能来源于外源可移动遗传因子。由于大部分蜡状芽孢杆菌群菌株含有多个前噬菌体和质粒,通过对蜡状芽孢杆菌群CRISPR的分析,为揭示其对宿主菌与噬菌体,以及宿主菌与质粒间的关系奠定基础。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. The dichtomously branched stigma in each carpel of the Tricyrteae is supplied by a bifurcated dorsal bundle.  相似文献
