本文给出了6个籼型杂交稻亲本成熟胚愈伤组织再生体系建立优化措施。采用6个有重要育种价值的杂交籼稻亲本成熟胚盾片诱导愈伤组织作为分化再生的外植体,通过调节2,4-D浓度、培养基成分、接种方式、激素组合和愈伤组织分化途径,建立适合籼稻遗传转化的再生体系。结果表明,MB培养基是较为合适的愈伤组织诱导培养基类型,6个品种在MB培养基上的愈伤组织诱导率均在60%-80%之间。半粒米的接种方式能够明显提高愈伤组织诱导率,提高幅度达到28.2%。通过调节2,4-D浓度和激素组合,愈伤组织诱导率最高可达到97.9%,两步分化法和适当干燥处理能够提高愈伤组织的分化效率,6品种愈伤组织分化率均在50%-90%之间。初步建立了6个杂交籼稻亲本品种成熟胚愈伤组织的再生体系,为以后遗传转化奠定基础。  相似文献

影响小麦成熟胚再生频率因素的研究(简报)   总被引:1,自引:0,他引:1  
以小麦成熟胚为外植体,研究了基本培养基、预处理类型、接种方式、植物激素的浓度和不同组合以及分化培养基中是否添加抗生素对愈伤组织诱导和分化的影响.在此基础上建立了一套高效的小麦成熟胚植株再生系统。经过试验,我们选择在MS培养基上接种经无菌水预处理的纵切成熟胚作为起始的试验条件。在含2mg/L2,4-D的MS培养基上.初级愈伤组织的诱导频率可达80%以上.在继代培养基中添加0.5mg/L6-BA和0.2mg/LNAA可以显著提高胚性愈伤组织的产生。而在再生培养基中加入适当浓度的头孢霉素可以有效提高胚性愈伤组织再生出小植株的比例。利用该再生系统,我们从5个小麦优良主栽品种的成熟胚再生出了可育的植株,再生频率达15.3%-34.5%。  相似文献

对6个籼稻(oryza sativa Subsp.Shien)品种和39个籼×籼杂种的花药在离体条件下进行培养,有5个品种及35个杂种得到了愈伤组织,平均诱导率为2.18%。在3个品种及11个籼×籼杂种中得到了绿苗或绿芽。本文着重报道基本培养基及其附加成分在诱导籼稻花药产生愈伤组织及根芽分化中的作用。 1.试验了几种诱导愈伤组织的培养基,以Miller培养基 2,4—D2毫克/升 酵母浸出液1,000毫克/升 激动素1毫克/升 吲哚乙酸2毫克/升 椰乳15%为最好。诱导率高者可达11—15%,平均诱导率在3%以上。 2.Ms、Nitsch及Miller培养基均可诱导籼稻花药愈伤组织分化出绿色的花粉植株。 3.籼稻花药愈伤组织的分化,随着激动素/生长素比值的增高,绿苗分化率及总分化率均有提高的趋势。而粳稻的这种关系不甚明显。 4.Miller培养基附加2毫克/升的吲哚乙酸对促进具茎、叶而无根的籼稻花粉小植株产生根有很好的作用。在这种培养基上,不仅可以诱导根的发生,而且根系发达,生长较弱的苗转移到这种培养基后,因根系健壮,生势好,转入盆栽,基本可以全部成活。  相似文献

商用马铃薯叶肉原生质体,在不同的液体培养基中浅层培养,耳生细胞分裂,并获得愈伤组织。将愈伤组织转到固体培养基上诱导分化,已从克新四号和68—62两个马铃薯品种的原生质体得到再生的完整植株。再生细胞的分裂频率受原生质体培养密度的影响。细胞团的生氏对液体培养基中的蔗糖浓度敏感。不同的原生质体培养基对愈伤组织的分化频率的影响非常显著。在分比培养基中加入3%的甘露醇,能提高愈伤组织的分化频率,并缩短分化期。  相似文献

以10个大麦优良品种为实验材料,成熟胚为外植体,研究基因型、种子的不同切割方式、培养基、激素等对大麦成熟胚愈伤组织的诱导及植株再生的影响.结果表明,种子纵切后接种出愈率显著高于横切;改良MS培养基能提高出愈率;在愈伤组织诱导过程中,不同品种对激素2,4-D与Dicamba的反应表现不同;初代愈伤组织经过3次继代培养后会转变为两种类型的胚性愈伤组织;不同品种的植株再生在不同浓度有机添加物的分化培养基上表现不同;长时间的继代培养,一些品种在植株再生过程中出现一定数量的白化苗.供试材料均能进行愈伤组织诱导,但是只有部分品种能再生植株.本实验筛选出愈伤组织诱导频率和绿苗分化率均较高,适合于遗传转化的受体材料,如87-3175、87-0053、97-4010、97-6004及208813-509.  相似文献

对国内外17个籼稻品种进行了基因枪转化,研究了有利于籼稻愈伤组织诱导和生长的培养条件和筛选程序,15个品种获得了潮霉素抗性愈伤组织,5个品种再生了植株,包括当前国际上推广的IR系统及国内的优良品种和恢复系,分子生物学证据证明潮霉素基因已经整合入籼稻的基因组。最高植株转化频率接近一般粳稻水平,多数低于粳稻水平。这为建立籼稻的转化系统打下了基础。  相似文献

以Reid、唐四平头和其它种质等3个杂种优势类群共19份玉米自交系为试验材料,以玉米幼胚作为外植体,研究了基因型、培养基和激素对玉米幼胚愈伤组织的诱导及植株再生的影响,结果表明供试材料均能进行愈伤组织的诱导,但是仅有12个自交系能再生植株。N6和改良N6培养基有助于提高愈伤组织的质量及其生长速度,2,4-D在愈伤组织的诱导中起着关键性作用。在诱导培养基中添加0.2mg/L的6-BA或KT会使胚性愈伤组织的诱导频率下降以及降低愈伤组织的质量。在胚状体诱导培养基中添加1mg/L的KT能促进绿苗的分化,但是浓度过高会使丛生苗分化过多。此外,通过对不同杂种优势类群自交系玉米幼胚培养特性的分析,发现在唐四平头类群的4个自交系中,黄早四的绿苗分化率仅为0.5%,其它3个自交系不能再生植株。但是,从Reid和其它种质类群的供试自交系中筛选出了胚性愈伤组织的诱导频率和绿苗分化率均较高的、适合于遗传转化的受体材料,如3189/4380、4380/陕综5、8103、先早17、18-599红、18-599白、501、178和冀53。  相似文献

为了提高籼稻愈伤组织的植株再生频率,研究了影响再生的各种因素,如:在诱导培养基或继代培养基中加细胞分裂素和萘乙酸(KT、BAP、玉米素或Zip1毫克/升),或加Thidiazuron(0.5毫克/升),以及愈及组织的部分干燥处理等。这些措施明显地提高籼稻愈伤组织的再生频率。结合使用这些处理可使TN1、IR72和IR64的愈伤组织再生植株频率较对照提高5-14倍。  相似文献

3年来通过对籼稻(Oryza sativa subsp.hsien)花药培养的研究,愈伤组织的诱导率从0.2%提高到平均为2.85%,最高达18.75%;绿苗分化率从5%以下提高到平均为13.1%最高达到45.5%。在6个籼稻品种、38个籼×籼杂种中成功地诱导出了绿苗。本文着重研究了提高诱导率的有关因素。以单核晚期花粉的花药愈伤组织诱导频率最高。籼稻花粉去分化过程相对地要求较低浓度的培养基,籼稻花药对高浓度的改良 RM-1964培养基完全没反应。用杂种的诱导率较亲本材料为高。以易于诱导愈伤组织或易于分化绿苗的品种作亲本的杂种后代,诱导率与绿苗分化率亦较高。籼稻形成愈伤组织的高峰是在接种后的50—60天之间。愈伤组织分化的高潮是在转移后10—30天之间。籼稻愈伤组织的分化以先分化根的类型最多,在先分化根的愈伤组织中有36.74%能再分化出芽来。  相似文献

大车前不仅有很高的药用价值,在生态学研究方面也是重要模式植物。关于大车前的组培,目前报道很少。我们通过不定芽直接再生和愈伤组织诱导两种途径,建立了大车前(Plantago major L. ‘Giant Turkish.’)的快速高效组培再生系统。完整的成熟种子培养在添加IAA和TDZ的MS培养基中,不经过愈伤的分化阶段,从子叶节的部位产生不定芽,直接不定芽的诱导频率达到100%。在0.2mg/L IAA和1.0mg/L TDZ作用下,培养4~5周后平均每个外植体产生再生芽的数目达到14.6个。对同一个外植体诱导得到的9株再生植株进行的RAPD检测表明,部分植株在DNA水平上发生了变异。以叶片作为外植体,在添加1.0mg/L NAA的MS固体培养基中培养3周后,伤口处形成愈伤组织,产生愈伤的频率平均为98%。愈伤组织在添加4.0mg/L 6BA的MS固体培养基中分化得到再生芽,分化频率为25%,平均每块愈伤产生再生芽2.8个。两种途径得到的再生芽转到1/2 MS培养基上均可生根、长成完整植株,小苗移栽到温室90%能够存活。  相似文献

在栽培稻的籼粳亚种间,花药培养力存在显著差异,这一差异主要是由遗传因素引起的。以适合籼粳稻杂种花药培养的SK_3培养基,经花药培养获得了一个籼粳交F_1代的加倍单倍体(DH)群体,对该群体的110个株系用同一种培养基进行花药培养,利用该群体构建的分子图谱进行有关水稻花药培养力的数量性状基因座位(QTLs)的分析。结果表明,与水稻花药培养力有关的4个性状在DH群体中均表现为连续分布,愈伤组织诱导率与绿苗分化率之间不存在相关性,而绿苗产率与愈伤组织诱导率和绿苗分化率均显著相关。在第6、7、8、10和12 5条染色体上分别检测到与愈伤组织诱导率有关的5个QTLs,其加性效应均为正。在第1和第9染色体上检测到与绿苗分化率有关的2个QTLs,这两个性状间的QTs不存在连锁。在第9染色体上有一个主效基因与白苗分化率有关,对绿苗产率则没有检测到特有的QTL。  相似文献

Interactive effects of genotypes with callus induction and regeneration media combinations on green plantlet regeneration response were studied for three indica rice (Oryza sativa L.) cultivars, IR-72, IR-54 and Karnal Local. Isolated mature-embryos were used to derive scutellar callus and fifteen media combinations involving MS, N6, R2, SK1 and some modifications were tested. Regeneration percentage as well as the shoot-bud induction frequency were influenced by genotype, callus induction medium, regeneration medium, interaction between genotype and the two media (callus induction and regeneration) as well the interaction between the callus induction medium and regeneration medium. Basal media combination of SK1m (callusing) and MS (regeneration) was found to be the best for cv. Karnal Local in which regeneration frequency of 88% and shoot-bud induction of 233% was observed. In IR-72, the highest regeneration frequency of 47.5% and shoot-bud induction frequency of 77% was obtained on MS-MS combination. In IR-54, highest regeneration frequency (25%) was recorded on MMS(N)-MMS(N) combination, whereas, highest frequency of shoot-bud induction (50%) was observed on MMS(S)-MS combination. Although genotype and the composition of the callus induction basal medium were the major determinants of regeneration response, an overall analysis of variation also revealed a significant interaction between the media used for de-differentiation (callusing) and re-differentiation (plantlet regeneration). Anther culturability of rice is significantly different between indica and japonica varieties. A doubled haploid (DH) population was established via anther culture of an indica/japonica hybrid on SK3 medium, which had been shown particularly suitable for anther culture of indica/japonica hybrids. For analyzing the quantitative trait loci (QTLs) responsible for anther culturability, anthers of the DH lines were again cultured with SK3 medium and parameters for four traits representing the anther culturability were surveyed and analyzed with the molecular map constructed from the same DH population. The parameters for four major traits were as follows: callus induction frequency (CI), green plantlet differentiation frequency (GPD), albino plantlet differentiation frequency (APD), and green plantlet yield frequency (GPY). All four traits displayed continuous distributions among the DH lines. The correlation coefficients between these traits were also tested and showed that there was no relationship between callus induction and green plantlet differentiation frequencies, but both showed strong positive correlation with the frequency of green plantlet yield. For callus induction frequency, five QTLs were identified on chromosomes 6, 7, 8, 10 and 12. Two QTLs for green plantlet differentiation frequency were located on chromosomes 1 and 9. There was a major QTL for albino plantlet differentiation frequency on chromosome 9. No independent QTL was found for green plantlet yield frequency. The results may be useful in the selection of parents with high response to anther culture for rice haploid breeding and in the establishment of permanent DH populations for molecular mapping.  相似文献

An improved procedure has been developed for high frequency androgenesis in indica × Basmati rice hybrids using a liquid culture medium. Anthers from fourteen genotypes comprising of indica × Basmati rice F1 hybrids, F2 plants and the parental rice cultivars, were floated in liquid RZM, N6M, and Heh5M media. Anther culture frequencies (percentage of anthers forming calluses) in most of the genotypes were significantly higher in RZM medium (16–75%) compared to those obtained in N6M (7–29%) and Heh5M (7–41%) media. Agarose (1.0% w/v)-solidified MSR1 medium containing 3.0% (w/v) maltose, 1 mg l−1 kinetin, 1 mg l−1 6-benzyladenine (BA) and 0.5 mg l−1α-naphthalene acetic acid (NAA) induced green shoot regeneration at high frequencies compared to the medium (MSR2) lacking BA. In all the genotypes, microspore calluses initiated in RZM medium regenerated green shoots with over tenfold higher frequencies compared to the calluses initiated in other two media. High plant regeneration frequencies (up to 270 green plants/1000 anthers) were obtained from microspore-derived calluses of some of the F1 hybrids (Gobind × Basmati 370, Gobind × Taraori Basmati) and F2 plants (Gobind × Basmati 370, Gobind × Taraori Basmati, HKR86-3 × Taraori Basmati) as compared to their actual parents. Cytological analysis of the root tips of the progeny seedlings of the microspore-derived plants revealed haploids at a frequency of about 50%; 22% of the microspore- derived plants had > 5% spikelet fertility and were diploid. Use of RZM liquid and MSR1 media, respectively for anther culture and plant regeneration resulted in several fold increase in the recovery of green plants from recalcitrant indica × Basmati rice F1 hybrids/F2 plants which were comparable to those reported for japonica rice varieties/hybrids leading to the improved feasibility of using doubled haploids in genetic, breeding and mapping research with indica rice. The effects of amino acid cysteine to culture systems of microspore-derived callus induction as well as plantlet regeneration were studied. Isolated pollen along with anther walls of basmati cultivars, Pusa basmati 1, Basmati 370 and Basmati 386 were cultured in a medium based on N6 salts supplemented with or without cysteine following pollen embedment in agarose. The induction and regeneration medium with cysteine gave twice as effective androgenesis and plantlet regeneration in recalcitrant basmati rice cultivars as compared with medium lacking cysteine. Unlike the highly responsive model systems, most of the indica cultivars responded rather poorly in anther culture. So the study may accelerate the introgression of desirable genes into basmati rice using anther culture as a breeding tool. Response of microspores in androgenesis, plant regeneration and albinism was genotype specific. Regeneration of Indica rice varieties remains a limiting factor for researchers undertaking transformation experiments.  相似文献

药用野生稻TAC克隆转化籼稻的体系初探   总被引:2,自引:0,他引:2  
采用已构建的药用野生稻TAC文库,比较研究4个籼稻品种在不同培养基的诱导率及愈伤组织对潮霉素抗性。通过2种不同诱导方法产生的愈伤组织,将携带药用野生稻大片段DNA的TAC克隆转化到籼稻品种中的结果表明,除了‘粤香占’以外,其它3个品种在心诱导培养基中添加微量B,后,其愈伤诱导率最高。不同品种的潮霉素筛选适合浓度存在差异,其中‘华粳籼74’的适合筛选浓度为50mg.L-1,‘粤香占’为40mg.L-1。用预培养5d的愈伤组织进行遗传转化,在潮霉素筛选之前先以头孢拉定抑菌处理的,容易获得转基因植株。4个品种中以‘粤香占’的抗性愈伤组织筛选率和分化率最高,分别为22.58%和14.86%。分子检测11株转基因水稻的结果表明,其中8株含有抗性标记基因。据此认为,药用野生稻TAC文库在水稻创新育种中可能有一定的利用前景。  相似文献

The regeneration response of mature and immature rice embryos from 15 rice varieties (Oryza sativa L. var. indica) was separately measured in terms of regeneration per cent and total number of regenerated plantlets obtained for a fixed sample size per variety, when cultured on standard MS media. The response of the rice varieties was classed as high, medium or poor, based on the above criteria. The regeneration per cent of genotypes showing high, medium and poor response ranged from 67 to 97, 38 to 63 and 0 to 36 respectively. The variation in the regeneration response amongst the defined groups was found to be statistically significant in terms of regeneration per cent and total plantlets for a constant sample size when subjected to one way analysis of variance and Duncan's multiple range test. Only small influences on regeneration frequency were found when the most and least responsive genotypes, Binnatoa and BR-23 were cultured on N6 and MS media with additional or different supplements.  相似文献

Genetic transformation of most indica rice (Oryza sativa) cultivars is hampered by poor in vitro culture performance and low regeneration potential. Histological study of primary calli can provide substantial information on their regeneration potential and can be used for early grading of calli expected to develop plantlets on regeneration media. The study was aimed to undertake histological analysis of primary calli derived from mature seeds of five indica rice cultivars viz. KSK-133, KS-282, Shaheen Basmati, Super Basmati, and DilRosh in order to assess their regeneration potential on different media combinations supplemented with various hormone concentrations (N6 + 2 mg/L 2,4-Dichlorophenoxyacetic acid; N6 + 2 mg/L 2–4 D + 2 mg/L Benzylaminopurine and MS + 2 mg/L 2,4-D). Calli with regeneration capability were subjected to histological assays by examining toulidine blue stained 5–8 μm thin sections for the presence of meristematic zones exhibiting embryogenic callus features. Based on our observations, formation of embryoids or embryoid-like structures was pronounced in KSK-133 and KS-282 calli. However, DilRosh, Super Basmati and Shaheen Basmati did not show these characteristic features. Three-week-old calli of all rice cultivars were transferred into regeneration medium (MS + 2 mg/L BAP + 1 mg/L Naphthaleneacetic acid). KSK-133 and KS-282 showed the highest regeneration potential (81% and 76%, respectively). These data were supported by histological observations where characteristic embryogenic units (EU) were noticed in these genotypes. These meristematic regions displayed high mitotic activity and stained relatively dark. The embryogenic calli cells were found heavily cytoplasmic with prominent nuclei and were located on the callus surface or inside surrounded by parenchymal cells.

Effects of four culture media on callus induction, regeneration and number of plants per unit culture were studied with mature seeds from five indica rice genotypes as explants. Based on the morphology, the calli were classified into four types as I to IV. Type I and type II are most suited to initiate suspension cultures or as target material for transformation. Number of plants regenerated per unit culture, formation of easily dissociating cell clusters and frequency of type I and type II calli were highest on NBKNB medium. Thus NBKNB medium is suitable for in vitro culture of even the hitherto recalcitrant indica genotypes.  相似文献

A plant regeneration system through multiple adventitious shoot differentiation from callus cultures has been established in slash pine (Pinus elliottii). Influences of seven different basal media on callus induction, adventitious shoot formation, and rooting were investigated. Among the different basal media, B5, SH, and TE proved to be suitable for callus induction and plantlet regeneration. Multiple adventitious shoot formation was obtained from callus cultures of slash pine on B5, SH, and TE media containing indole-3-butyric acid, N6-benzyladenine, and thidiazuron. Scanning electron microscopy demonstrated the early development of adventitious shoots derived from callus cultures. These results indicate that an efficient plant regeneration protocol for micropropagation of slash pine had been established. This protocol could be most useful for future studies on genetic transformation of slash pine.  相似文献
