In a 2 X 2 X 2 factorial experiment, protein intake, selenium supplementation, and intake of phosphorus were related to the incidence of retained placenta in cows fed varying concentrations of these nutrients during the dry period. The population incidence of 38% retained placenta in 26 control cows was independent of phosphorus intake which ranged between 39 and 95 g per day. When a protein supplement was included in the concentrate as soybean meal, the incidence of retained placenta was reduced to 20% for supplemented animals from 50% for the animals that received no soybean meal. This increased protein in the ration was reflective of a mean increase from .02 ppm of selenium to .06 ppm of selenium in the ration. By increasing the mean daily intake of selenium at least 3 wk prepartum from .23 mg to .92 mg daily, overall incidence of retained placenta was reduced from 38% to 0%. A positive prophylactic effect was achieved regardless of whether alpha tocopherol was supplemented as well. These mature dairy cows were deficient in selenium, and supplementation of selenium reduced the incidence of retained placenta.  相似文献

Incidence of clinical mastitis and duration of clinical symptoms for complete lactations were evaluated for 80 cows randomly assigned to one of four groups: vitamin E supplemented- and selenium injected, selenium injected, vitamin E supplemented, and controls. Vitamin E supplementation and selenium injection were during the dry period. Log-linear analysis of incidence data revealed a significant 37% reduction of clinical mastitis by vitamin E. Incidence was not affected by selenium alone, nor was there any evidence for interaction of vitamin E with selenium on incidence. However, duration of clinical symptoms (calendar months clinical/quarter lactating) was reduced by 46% for the selenium group, 44% for the vitamin E group, and 62% for the vitamin E-selenium group as compared to controls. We conclude that dairy cow diets deficient of vitamin E may elevate incidence of clinical mastitis. Selenium deficiency may result in greater duration of clinical symptoms, and selenium may interact with vitamin E. Coliform bacteria and species of streptococcus other than Streptococcus agalactiae were isolated from 70% of the clinical cases.  相似文献

The objective of this study was to evaluate the effect of prepartum intravaginal administration of an anti-Escherichia coli bacteriophage cocktail on uterine health and reproductive performance of lactating dairy cows. The study was conducted on a commercial dairy farm located near Ithaca, New York, from March to September 2012, and 107 cows were enrolled. The cows were divided randomly into 2 groups (treatment and control); treatment group cows were submitted to 3 intravaginal administrations of 20 mL of a bacteriophage cocktail including 10 different phages in a lysate with titration of approximately 109 plaque-forming units, on d 230, 260, and 275 of gestation. Control cows were not treated with a placebo, and physical examination and sample collection was conducted equally for both groups. Vaginal swabs were collected at d 230, 260, and 275 d of gestation, cervical swabs were collected at 3 ± 1 d in milk, and blood was collected at 275 d of gestation for nonesterified fatty acids and interleukin (IL)-6 serum level measurements and at 10 DIM for β-hydroxybutyrate and IL-6 serum level measurements. The bacteriophage cocktail significantly increased the incidence of retained placenta and metritis. Swab samples were cultured for E. coli and no difference was found between treatment and control groups. The lysate contained 6 × 109 IU of lipopolysaccharide/mL and could have caused prepartum overstimulation of toll-like receptor-4 (TLR4)-chemokine pathway, leading to a weaker immune response. However, IL-6 was not affected by treatment, although sample size was insufficient to prove a treatment effect on the suppression of the chemokine cascade and innate immune system. Bacteriophage treatment had an excellent bactericidal effect in previous in vitro trials; however, special attention must be given to production of the cocktail to obtain a purified solution free of E. coli bacterial products and fragments, particularly lipopolysaccharide.  相似文献

Three groups of six calves each were fed a milk replacer and a starter concentrate for 15weeks. Calves of the first group received the basal diet containing selenium (Se) and vitamin E at 0.095-0.128mg and 30-33mg per kg of total solids, respectively. Calves of the second group received the basal diet supplemented with Se-enriched yeast to increase dietary Se concentration to 0.50mg/kg. The third group of calves received the latter diet supplemented with vitamin E to increase its concentration to 100mg/kg. There was no effect of diet on growth rate, digestibility of dry matter and Se, chemical composition of meat (M. Longissimus thoracis et lumborum), meat colour and fatty acid profile of meat lipids. The Se supplementation significantly increased Se concentration in muscle from 0.21 to 0.43mg/kg. The activity of glutathione peroxidase (GSH-Px) in muscle and liver tissue of Se-supplemented animals was increased by 56% and 67%, respectively, compared to the control. The combined supplementation of vitamin E and Se significantly improved the lipid stability of meat compared to the control diet, but not compared to the Se-supplemented diet. It can be concluded that dietary Se supplementation increases the concentration of Se and the GSH-Px activity in meat, but has limited potential for improving meat oxidative stability.  相似文献

Cows were fed diets either supplemented with .2 ppm Se and 70 IU vitamin E/kg diet DM (21 cows) or unsupplemented (40 cows) during the dry period (approximately 60 d). From parturition to 21 d of lactation, cows were fed diets that were either supplemented with .3 ppm Se and 40 IU/kg vitamin E or unsupplemented. At d 21 following parturition, 18 cows fed the unsupplemented diet were switched to diets containing 0 or .3 ppm supplemental Se and 0 or 40 IU/kg supplemental vitamin E arranged factorially. These diets were fed for the next 32 d. The remaining cows continued their respective diets for 32 d. Plasma Se concentrations averaged .1 microgram/ml for supplemented cows but were .05 micrograms/ml for unsupplemented cows. Plasma Se concentration from cows fed supplemental Se from 21 to 53 d postpartum increased rapidly and were not different from long-term supplemented cows. Whole blood glutathione peroxidase activity was lower in unsupplemented than in supplemented cows. Short-term Se supplementation increased glutathione peroxidase activity above that for unsupplemented animals, but activity was still less than that in long-term supplemented animals. Plasma alpha-tocopherol concentrations at parturition and d 21 postpartum were lower in unsupplemented than in supplemented animals. On d 53 postpartum, no differences in plasma alpha-tocopherol concentrations were found between long-term supplemented and unsupplemented cows. Supplementing vitamin E during the dry period increased alpha-tocopherol content of colostrum.  相似文献

Relationships among vitamin E, selenium, and bovine blood neutrophils   总被引:1,自引:0,他引:1  
Effects of vitamin E and selenium supplementation on in vitro phagocytosis and intracellular kill of bacteria by bovine neutrophils were investigated. Diets were not supplemented with vitamin E and selenium during the dry period and first 21 d of lactation. Cows were then assigned to one of four treatment diets for 30 d. Treatment diets were either unsupplemented or supplemented with vitamin E, selenium, or both vitamin E and selenium, in a 2 x 2 factorial arrangement. Peripheral blood neutrophils were isolated from each cow on lactation d 51. Vitamin E supplementation of diets increased intracellular kill of Staphylococcus aureus and Escherichia coli by neutrophils. Intracellular kill of S. aureus was greater in neutrophils isolated from selenium supplemented cows than in neutrophils from cows without supplemental selenium. Intracellular kill of E. coli did not differ between neutrophils from selenium supplemented and selenium unsupplemented cows. Ability of neutrophils to phagocytize either S. aureus or E. coli was independent of vitamin E and selenium.  相似文献

Incidence of retained placenta in dairy cows was evaluated in 627 parturitions. The herd was divided prepartum into three groups: 1) control, no treatment (n = 217 cows); 2) cows injected intramuscularly (n = 190) 21 to 10 d prior parturition with 45 mg Se and 2040 IU of vitamin E; and 3) cows intraruminally administered (n = 220) with two 30-g pellets containing 10% elemental selenium 2 mo prior to expected calving. Incidence of retained placenta (22.1%) was not reduced by Se in combination with vitamin E injection or intraruminal Se pellet nor were other measures of reproduction improved for cows fed a prepartum diet adequate in Se. At parturition the blood plasma Se concentrations were higher in treated postpartum with Se than in untreated cows. No difference in blood plasma Se was observed at parturition between cows with or without placenta retention. Cows dosed intraruminally with Se had a significant increase in milk Se, but this was too small to be a danger to human health. The present results on placenta retention suggest that this disorder is not a Se responsive disease in the dairy cow.  相似文献

In a two-lactation-gestation cycle experiment, 152 Holstein cows with low serum Se and vitamin E were fed total mixed rations and assigned at parturition to four groups (1, control; 2, 500 IU vitamin E/d; 3, 2 mg Se/d; 4, 500 IU vitamin E plus 2 mg Se/d). Supplements were not fed during dry periods. Serum Se and vitamin E were increased within 1 mo by oral supplements. Maximal mean serum Se in cycles 1 (67 ng/ml at 7 mo) and 2 (74 ng/ml at 4 mo) occurred in groups 3 and 4, respectively. Maximal mean serum vitamin E in cycle 1 (3.3 micrograms/ml at mo 8) and 2 (3.03 micrograms/ml at mo 2) occurred in groups 4 and 2, respectively. Selenium treatment of the dams increased Se in colostrum and in serum of presuckle calves. Vitamin E supplementation of dams did not affect vitamin E in serum of presuckled calves. Reproductive performance was not affected by supplement. In an 8-wk study, 24 lactating cows with low serum Se were assigned (6/group) to 0, 2.5, 5, and 10 mg supplemental Se/d. Maximal mean serum Se concentrations of 23, 56, 71, and 79 ng/ml were attained by wk 4 in the above respective groups. These data indicate that 2 to 2.5 mg supplemental Se/cow per d were inadequate for desirable serum Se concentrations and support recent changes in allowed Se supplementation for dairy cattle.  相似文献

Retained placenta (RP), defined as fetal membranes not being expelled within 24 h after calving, is a costly disease in multiparous dairy cows that has been linked to immune suppression, infections, elevated lipid mobilization, and depleted status of antioxidants including α-tocopherol, and that increases the risk of other diseases (OD) in early lactation. Early detection of cows at increased risk of developing RP, OD, or both in early lactation could improve treatment success and result in improved milk production and reproductive performance. To identify risk indicators of RP, OD, or both, we used a nested case-control design and compared multiparous dairy cows that developed RP (n = 32) with cows that remained healthy (H; n = 32) or cows that developed OD (n = 32) in early lactation. We compared peripartal body condition score (BCS) as well as serum concentrations of α-tocopherol, metabolites [β-hydroxybutyrate (BHBA), cholesterol, glucose, nonesterified fatty acids (NEFA), and urea N], haptoglobin, and macrominerals (i.e., calcium, magnesium, and phosphorus) on d −21, −14, −7, −3, −1, 0, 1, 3, 7, 14, 21, 28, 35, 42, and 49 postpartum. In addition, average serum concentrations were calculated for each cow for the last 3 wk prepartum, for 3 and 2 wk prepartum combined, for the last week prepartum, and for the morning after calving and compared between groups. The RP cows had lower BCS than the H or OD cows until 2 wk postpartum. During the prepartal periods, RP and OD cows had lower α-tocopherol concentrations (corrected or not for cholesterol concentration) and higher NEFA and BHBA concentrations than H cows. Thus, lower prepartal BCS could be an early predictor for RP risk, and lower α-tocopherol concentrations and higher NEFA and BHBA concentrations could be early predictors for disease.  相似文献

Twenty-eight Holstein heifer calves were allotted at birth to one of four treatments: 1) 0 mg, 2) 1,400 mg, or 3) 2,800 mg of dl-alpha-tocopherol acetate given orally at weekly intervals, or 4) 1,400 IU of dl-alpha-tocopherol weekly by intramuscular injection in order for us to study their performance and metabolic profile. Calves were fed milk at 8% of birth weight until they were weaned at 6 wk of age and fed a complete calf starter ad libitum from birth. Calves were on experiment for 12 wk. There were no significant differences in weekly weight gains, starter consumption, and fecal scores among treatments. However, there was a trend toward greater starter consumption and weight gains in supplemental calves. Serum alpha-tocopherol concentration measured after 7 d of each administration was significantly higher at wk 4 in calves given the high oral supplementation and at wk 2, 4, 6, and 8 higher in injected calves than in unsupplemented calves. Creatine kinase activity was higher in unsupplemented calves and negatively correlated with serum alpha-tocopherol until wk 8, suggesting preclinical muscular dystrophy. Alkaline phosphatase activity was higher with the high oral supplementation. Serum carbon dioxide values showed a trend toward positive correlation with those for serum tocopherol; however, the values were within normal range. There were no significant differences in creatinine, glucose, phosphorus, calcium, urea nitrogen, chloride, sodium, potassium, albumin, and total protein among treatments. Serum glucose was higher in all calves at wk 10 and 12 than at wk 4, 6, and 8. Calves may not get enough vitamin E with conventional calf starters, and supplementation may be essential to obtain maximum performance.  相似文献

Two experiments were conducted to study the effects of feeding legume silages and providing supplemental vitamin E in concentrates on the oxidative stability of milk. In experiment 1, six multiparous Holstein-Friesian cows were offered 1 of 6 silage treatments in a cyclical changeover-design experiment, with four 4-wk periods. The silages were grass, red clover, white clover, alfalfa, grass and red clover mixture (50:50 on a DM basis), and grass and white clover mixture (50:50 on a DM basis). In experiment 2, 8 cows were used in a changeover-design experiment with three 4-wk periods. The 4 treatments were a factorial combination of forages (grass silage or red clover silage) and supplemental vitamin E in the form of all-rac-alpha-tocopheryl acetate (29 or 290 IU/kg of DM in the concentrate). All forages were offered ad libitum and a flat rate of concentrates (8 kg/d) was fed in both experiments. Red clover silage led to significantly higher forage intakes, milk yields, and milk protein percentage in experiment 2, which was in agreement with results from experiment 1. There was no effect of vitamin E on feed intake, milk production, or milk fat and protein percentage. Red clover silage also led to significant changes in milk fatty acid profiles, particularly increased levels of polyunsaturated fatty acids. Milk samples were stored at 4 degrees C and 20 degrees C and analyzed for alpha-tocopherol and thiobarbituric acid reactive substances at intervals to determine oxidative stability. Diets based on red clover and alfalfa silages were associated with more rapid loss of alpha-tocopherol and increased production of thiobarbituric acid reactive substances during the storage of milk in comparison with diets based on grass silage. The increased oxidative deterioration of milk produced from cows fed red clover silage was avoided by vitamin E supplementation.  相似文献

The effect of parenteral administration of two subcutaneous injections of vitamin E and Se (5 mg and 0.1 mg/kg of body weight, respectively) during the dry period on the mammary health and milk somatic cell counts of 25 dairy ewes was investigated. Supplementation reduced somatic cell counts (5.4 vs. 6.0 log10) during the subsequent lactation but had no effect on the incidence of clinical mastitis (4% vs. 6%) and intramammary infections (9.0% vs. 11.3%). Furthermore, the administration of vitamin E and Se was associated with differences in differential cell counts of milk samples (macrophages, 48.8% vs. 38.4%; polymorphonuclear neutrophils, 40.1% vs. 50.7%; and eosinophils, 0.7% vs. 1.4% for control ewes and ewes receiving supplements, respectively). The administration of these supplements also increased erythrocyte glutathione peroxidase activity (139.5 vs. 86.3 U/ml of packed cell volume) and the percentage of blood neutrophils that reduced nitroblue tetrazolium after bacterial extract stimulation (48.6% vs. 38.7%). Parenteral administration of vitamin E and Se to ewes during the dry period appeared to have influenced mammary gland status during the subsequent lactation and particularly total and differential milk cell counts.  相似文献

The aim of the study was to investigate whether vitamin E supplements in larger amounts than recommended could reduce incidence of disease, improve immune competence, and increase rate of weight gain of conventionally barley-fed beef cattle. Mean daily intake of vitamin E by individual calves in the experimental group was 200 mg during the first 2 mo, 400 mg during the next 2 mo, and 600 mg during the rest of the period. Corresponding daily intakes of vitamin E for the control group were 50, 100, and 150 mg. Mean plasma vitamin E of the experimental group increased from .49 mg/L at the start of the trial to 2.03 mg/L at the end, but that of the control group was lower at the end (.36 mg/L) than at the beginning (.53 mg/L). No significant differences were observed between the groups concerning incidence of disease or magnitude of lymphocyte stimulation. The results indicated that there was a surprisingly poor biological availability of the dietary vitamin. Therefore, a comparison in reality was made between calves with inadequate and normal vitamin E status. The differences in daily BW gain and time to reach slaughter weight thus probably were effects of the low vitamin E status rather than positive effects of additional vitamin in the diet.  相似文献

The aim of this research was to study the effect of milking frequency and supplementation with a vitamin-mineral complex above requirements on intake, body weight (BW), and milk yield and composition in high-yielding Assaf ewes. Sixteen lactating Assaf ewes were used in this study, separated into 4 groups of 4 ewes each. Animals in 2 of the groups (control groups) did not receive any extra vitamin-mineral supplement, whereas animals in the other 2 groups (supplement groups) received daily an oral dose of 1g of vitamin E (1,000 IU, DL-α-tocopherol acetate) and 0.4 mg of selenium (sodium selenite anhydrous). The experiment consisted of 2 consecutive periods of 3 wk (twice-daily milking in both mammary glands) and 8 wk (once-daily milking in one mammary gland and twice-daily milking in the other gland). Intake, BW, and milk composition were controlled weekly, and milk production was recorded 3 times a week. Administration of the vitamin-mineral supplement had no effect on dry matter intake, BW, or milk production and composition. The reduction of milking from twice to once a day caused a decrease in milk production and lactose concentration and a significant increase in protein concentration, total solids, and somatic cell count, without affecting the fat content. Administration of a vitamin E and Se supplement at the doses used in the present study does not seem to exert, in the short term, a noticeable effect on the mammary gland when milking frequency is reduced.  相似文献

It is unclear why some cows fail to expel the placenta following calving. One theory suggests the fetal placenta must be recognized as "foreign" tissue and rejected by the immune system after parturition to cause expulsion of the placenta. We hypothesized that impaired neutrophil function causes retained placenta (RP). We examined the ability of neutrophils to recognize fetal cotyledon tissue as assessed by a chemotaxis assay, which utilized a placental homogenate obtained from a spontaneously expelled placenta as the chemoattractant. Neutrophil killing ability was also estimated by determining myeloperoxidase activity in isolated neutrophils. Blood samples were obtained from 142 periparturient dairy cattle in two herds. Twenty cattle developed RP (14.1%). Neutrophils isolated from blood of cows with RP had significantly lower neutrophil function in both assays before calving, and this impaired function lasted for 1 to 2 wk after parturition. The addition of antibody directed against interleukin-8 (IL-8) to the cotyledon preparation used as a chemoattractant inhibited chemotaxis by 41%, suggesting that one of the chemoattractants present in the cotyledon at parturition is IL-8. At calving, plasma IL-8 concentration was lower in RP cows (51 +/- 12 pg/ml) than in cows expelling the placenta normally (134 +/- 11 pg/ml). From these data, we suggest that neutrophil function is a determining factor for the development of RP in dairy cattle. Also, depressed production of IL-8 may be a factor affecting neutrophil function in cows developing RP.  相似文献

Sixty Holstein cows were allocated to three groups. Twenty cows had retained placenta. The remaining cows were examined on d 14 postpartum and those with purulent discharges (n = 22) were assigned to one group and the remaining (n = 18) to a control group. Within each group, cows were given randomly either gonadotropin-releasing hormone (i.m., 200 micrograms) or saline on d 15 postpartum to evaluate the effect on changes in ovarian structures and plasma progesterone through 50 d postpartum and fertility. Corpora lutea were found in control cows by d 21, cows with uterine discharge by d 28, and cows with retained placenta by d 27. Maximum progesterone production during the first luteal phase was higher in control cows than in cows with purulent discharge or retained placenta (4.66 ng/ml compared with 3.23 and 3.34 ng/ml, respectively). Duration of the first corpus luteum was affected by clinical condition. Only 6.2% of cows with retained placenta had corpora lutea of normal duration (12 to 16 d), whereas 43.8 and 50.0% of cows with uterine discharge and control cows had normal postpartum luteal phases. Measures of fertility were not affected by gonadotropin-releasing hormone. Control cows had less days to conception (97) and fewer services per conception (1.6) than cows with retained placenta (134 and 2.5, respectively). Clinical group affected reproduction more than gonadotropin-releasing hormone did, possibly by altering ovarian function.  相似文献

To elucidate the effects of dietary vitamin E with or without flaxseed on beef fatty acid composition, 80 feedlot steers were fed 4 diets: Control-E (451 IU dl-α-tocopheryl acetate/head/day), Control+E (1051 IU dl-α-tocopheryl acetate/head/day), Flax-E (10% ground) and Flax+E. Vitamin E had no effect on animal growth or carcass weight (p>0.05), while flaxseed-fed steers had greater average daily gain (p=0.007), final live weight (p=0.005) and heavier carcasses (p=0.012). Feeding flaxseed increased the total n-3 fatty acid content of beef and this response was further accentuated by the inclusion of high levels of vitamin E in the diet. Feeding flax increased levels of some 18:3n-3 partial hydrogenation products including c15- and t13/14-18:1 and several 18:2 isomers (p<0.001) but decreased t10-18:1 (p<0.001). Vitamin E enhanced intramuscular levels of 18:3n-3 and its biohydrogenation products leading to greater accumulations of total n-3 fatty acids in lean ground beef. The consequences of increasing the concentrations of partially hydrogenated products on human health have yet to be investigated.  相似文献
