Microcystins (MC) are cyanobacterial hepatotoxins responsible for animal-poisoning and human health incidents. Immunoassays provide a sensitive and fast means to detect these toxins, but cross-reactivity (CR) characteristic of different antibodies was variable. Here, we have produced and characterized a monoclonal antibody (Clone MC8C10) with highly specificity against the most frequent and most toxic variant of microcystins, MC-LR. MC8C10 is more specific against MC-LR among the reported antibodies before. The immunogen was synthesized from the modified MC-LR and bovine serum albumin (BSA). An indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) with MC8C10 was established to detect the MCs in waters, which showed highly specificity with MC-LR, and have a detection limit for MC-LR 0.1 μg L−1, the 50% inhibition concentration (IC50) for MC-LR was 1.8 ± 0.1 μg L−1 and the quantitative detection range was from 0.3 to 10 μg L−1. The [4-arginine] microcystins and the nodularin-R showed lower cross-reactivates (CR < 10%), and other MCs such as MC-LF and MC-LW are not recognized (CR < 10−4). The analysis results of real water samples with ic-ELISA showed that all the coefficients of variation were less than 15%, and the recovery was (100.3 ± 5.9)%. So the highly specific ic-ELISA will commendably suit for sensitive analysis for MC-LR in surface water as well as drinking water.  相似文献

A multi-residue method for the determination of organochlorine pesticides in fish feed samples was developed and optimized. The method is based on a cleanup step of the extracted fat, carried out by liquid–liquid extraction on diatomaceous earth cartridge with n-hexane/acetonitrile (80/20, v/v) followed by solid phase extraction (SPE) with silica gel–SCX cartridge, before the identification and quantification of the residues by gas chromatography–triple quadrupole tandem spectrometry (GC–MS/MS). Performance characteristics, such as accuracy, precision, linear range, limits of detection (LOD) and quantification (LOQ), for each pesticide were determined. Instrumental LODs ranged from 0.01 to 0.11 μg L−1, LOQs were in the range of 0.02–0.35 μg L−1, and calibration curves were linear (r2 > 0.999) in the whole range of explored concentrations (5–100 μg L−1). Repeatability values were in the range of 3–15%, evaluated from the relative standard deviation of six samples spiked at 100 μg kg−1 of fat, and in compliance with that derived by the Horwitz's equation. No matrix effects or interfering substances were observed in fish feed analyses. The proposed method allowed high recoveries (92–116%) of spiked extracted fat samples at 100 μg kg−1, and very low LODs (between 0.02 and 0.63 μg kg−1) and LOQs (between 0.05 and 2.09 μg kg−1) determined in fish feed samples.  相似文献

A high-performance liquid chromatography-fluorescence detection method was optimized and validated to determine tetracyclines residues in bovine milk. Post-column derivatization using metal complexation in non-aqueous reagent increased the fluorescence of chelates by a factor up to 2.54 compared to water (signal-to-noise ratio enhancement). Overall recoveries ranged from 61 to 115%, with RSDr from 5 to 15% (n = 54). Detection limits ranged from 5 to 35 μg kg−1. Limits of quantification were established at 50 μg kg−1. Decision limits (CCα) were 109, 108 and 124 μg kg−1 and detection capabilities (CCβ) 119, 117 and 161 μg kg−1 for oxytetracycline, tetracycline and chlortetracycline, respectively. The method was applied successfully in a national monitoring program.  相似文献

A highly sensitive flow analysis system has been developed for the trace determination of reactive phosphate in natural waters, which uses a polymer inclusion membrane (PIM) with Aliquat 336 as the carrier for on-line analyte separation and preconcentration. The system operates under flow injection (FI) and continuous flow (CF) conditions. Under optimal FI conditions the system is characterised by a linear concentration range between 0.5 and 1000 μg L−1 P, a sampling rate of 10 h−1, a limit of detection of 0.5 μg L−1 P and RSDs of 3.2% (n = 10, 100 μg L−1) and 7.7% (n = 10, 10 μg L−1). Under CF conditions with 10 min stop-flow time and sample solution flow rate of 1.32 mL min−1 the flow system offers a limit of detection of 0.04 μg L−1 P, a sampling rate of 5 h−1 and an RSD of 3.4% (n = 5, 2.0 μg L−1). Interference studies revealed that anions commonly found in natural waters did not interfere when in excess of at least one order of magnitude. The flow system, operating under CF conditions, was successfully applied to the analysis of natural water samples containing concentrations of phosphate in the low μg L−1 P range, using the multipoint standard addition method.  相似文献

The status of pesticide pollution in Tanzania   总被引:1,自引:0,他引:1  
The paper summarises the findings of recent studies carried out to assess the levels of pesticide residues in water, sediment, soil and some biota collected from different parts of Tanzania. Although the intention is to cover the whole country, so far the studies have focused on areas with known large-scale pesticide use (Southern Lake Victoria and its basin, TPC sugar Plantations in Kilimanjaro region, Dar es Salaam coast, Mahonda-Makoba basin in Zanzibar) and a former pesticide storage area at Vikuge Farm in Coast region). Analysis of the cleaned extracts in GC-ECD/NPD revealed the dominance of organochlorines in all samples. Generally, low levels of residues were found in areas associated with agricultural pesticide use but the levels in the former storage areas were substantially high. DDT and HCH were dominant in all the studied areas. In the former areas, levels of ∑DDT in water, sediments and soil were up to 2 μg L−1, 700 μg kg−1 and 500 μg kg−1, respectively, while those of ∑HCH were up to 0.2 μg L−1, 132 μg kg−1 and 60 μg kg−1, respectively. The levels in aquatic biota were much higher than those in the water most likely due to bioaccumulation. In the former storage area at Vikuge the levels of pesticides in the topsoil were alarmingly high. Their concentrations were up to 282,000 mg kg−1 dry weight for ∑DDT and up to 63,000 mg kg−1 for ∑HCH. A herbicide, pendimethalin [N-(1-ethylpropyl)-2,6-dinitro-3,4-xylidine], was also found at concentrations up to 41,000 mg kg−1 dry weight. Thus the total pesticide content in the soil was almost 40%. Following these findings the area is now earmarked to be a demonstration site for a proposed GEF project ‘Bioremediation of POPs impacted soils in East Africa’.  相似文献

A novel technique, high temperature headspace liquid-phase microextraction (HS-LPME) with room temperature ionic liquid (RTIL), 1-butyl-3-methylimidazolium hexafluorophosphate ([C4MIM][PF6]) as extractant, was developed for the analysis of dichlorodiphenyltrichloroethane (p,p′-DDT and o,p′-DDT) and its metabolites including 4,4′-dichlorodiphenyldichloroethylene (p,p′-DDE) and 4,4′-dichlorodiphenyldichloroethane (p,p′-DDD) in water samples by high performance liquid chromatography with ultraviolet detection. The parameters such as salt content, sample pH and temperature, stirring rate, extraction time, microdrop volume, and sample volume, were found to have significant influence on the HS-LPME. The conditions optimized for extraction of target compounds were as follows: 35% NaCl (w/v), neutral pH condition, 70 °C, 800 rpm, 30 min, 10 μL [C4MIM][PF6], and 25 mL sample solutions. Under the optimized conditions, the linear range, detection limit (S/N = 3), and precision (R.S.D., n = 6) were 0.3-30 μg L−1, 0.07 μg L−1, and 8.0% for p,p′-DDD, 0.3-30 μg L−1, 0.08 μg L−1, and 7.1% for p,p′-DDT, 0.3-30 μg L−1, 0.08 μg L−1, and 7.2% for o,p′-DDT, and 0.2-30 μg L−1, 0.05 μg L−1, and 6.8% for p,p′-DDE, respectively. Water samples including tap water, well water, snow water, reservoir water, and wastewater were analyzed by the proposed procedure and the recoveries at 5 μg L−1 spiked level were in the range of 86.8-102.6%.  相似文献

A general and broad class-specific enzyme-linked immunosorbent assay was developed for the O,O-dimethyl organophosphorus pesticides, including malathion, dimethoate, phenthoate, phosmet, methidathion, fenitrothion, methyl parathion and fenthion. Three haptens with different spacer-arms were synthesized. The haptens were conjugated to bovine serum albumin (BSA) for immunogens and to ovalbumin (OVA) for coating antigens. Rabbits were immunized with the immunogens and six polyclonal antisera were produced and screened against each of the coating antigens using competitive indirect enzyme-linked immunosorbent assay for selecting the proper antiserum. The effect of hapten heterology on immunoassay sensitivity was also studied. The antibody-antigen combination with the most selectivity for malathion was further optimized and tested for tolerance to co-solvent, pH and ionic strength changes. The IC50 values, under optimum conditions, were estimated to be 30.1 μg L−1for malathion, 28.9 μg L−1 for dimethoate, 88.3 μg L−1 for phenthoate, 159.7 μg L−1 for phosmet, 191.7 μg L−1 for methidathion, 324.0 μg L−1 for fenitrothion, 483.9 μg L−1 for methyl parathion, and 788.9 μg L−1 for fenthion. Recoveries of malathion, dimethoate, phenthoate, phosmet and methidathion from fortified Chinese cabbage samples ranged between 77.1% and 104.7%. This assay can be used in monitoring studies for the multi-residue determination of O,O-dimethyl organophosphorus pesticides.  相似文献

The development of a direct competitive enzyme-linked immunosorbent assay (ELISA) based on polyclonal antibodies specific for semicarbazide (SEM) is described. Molecular modelling of the hapten mimics and other key components of the assay system was conducted to explain antibody properties in relation to hapten design. The small aliphatic molecule SEM was coupled to 3-carboxybenzaldehyde to produce carboxyphenyl-SEM (CPSEM), for the generation of specific antibodies. Five rabbits produced antibodies against NPSEM (used in direct and indirect ELISA formats) exhibiting a 50% binding inhibition level (IC50 values) of 0.06-2.28 μg L−1 in assay buffer for SEM. The most sensitive indirect assay based on the antibody MVK39 showed a high dynamic range providing a linear readout in the range of 0.01-0.2 μg L−1. Antibody MVK31 (IgG) allowed specific SEM detection at an IC50 = 0.14 μg L−1 in direct ELISA and was evaluated using solvent extracted SEM-spiked porcine and baby food samples. Recovery levels determined from fortified samples (0.5, 1.0, 1.5, 5, 10 and 20 μg kg−1) of porcine and baby food ranged from 82.9 to 105.3%, respectively, with a coefficient of variation less than 15.5%. Respective detection capability and threshold of the assay for porcine muscle, set on the basis of acceptance of no false negative results, was 0.3 and 0.11 μg kg−1.  相似文献

In this study, the steroid hormone levels in river and tap water samples were determined by using a novel dispersive liquid-liquid microextraction method based on the solidification of a floating organic drop (DLLME-SFO). Several parameters were optimized, including the type and volume of the extraction and dispersive solvents, extraction time, and salt effect. DLLME-SFO is a fast, cheap, and easy-to-use method for detecting trace levels of samples. Most importantly, this method uses less-toxic solvent. The correlation coefficient of the calibration curve was higher than 0.9991. The linear range was from 5 to 1000 μg L−1. The spiked environmental water samples were analyzed using DLLME-SFO. The relative recoveries ranged from 87% to 116% for river water (which was spiked with 4 μg L−1 for E1, 3 μg L−1 for E2, 4 μg L−1 for EE2 and 9 μg L−1 for E3) and 89% to 102% for tap water (which was spiked with 6 μg L−1 for E1, 5 μg L−1 for E2, 6 μg L−1 for EE2 and 10 μg L−1 for E3). The detection limits of the method ranged from 0.8 to 2.7 μg L−1 for spiked river water and 1.4 to 3.1 μg L−1 for spiked tap water. The methods precision ranged from 8% to 14% for spiked river water and 7% to 14% for spiked tap water.  相似文献

Graphene nanosheets, dispersed in Nafion (Nafion-G) solution, were used in combination with in situ plated bismuth film electrode for fabricating the enhanced electrochemical sensing platform to determine the lead (Pb2+) and cadmium (Cd2+) by differential pulse anodic stripping voltammetry (DPASV). The electrochemical properties of the composite film modified glassy carbon electrode were investigated. It is found that the prepared Nafion-G composite film not only exhibited improved sensitivity for the metal ion detections, but also alleviated the interferences due to the synergistic effect of graphene nanosheets and Nafion. The linear calibration curves ranged from 0.5 μg L−1 to 50 μg L−1 for Pb2+ and 1.5 μg L−1 to 30 μg L−1 for Cd2+, respectively. The detection limits (S/N = 3) were estimated to be around 0.02 μg L−1 for Pb2+ and Cd2+. The practical application of the proposed method was verified in the water sample determination.  相似文献

Clenbuterol has been extracted by mixed solid-phase extraction from two biological matrices (bovine hair and urine) and detected by GC/MS (selected ion monitoring (SIM) and full-SCAN modes). The analytical signal has been modelled with univariate and three-way models, namely DTLD, PARAFAC, PARAFAC2, Tucker3 and trilinear PLS. Since clenbuterol is a banned substance a comparative study of the capability of detection (CCβ, X0=0) has been performed as a function of the sample (hair, 74 μg kg−1 and urine, 0.36 μg l−1), the mode in which the signals are monitored (SCAN, 283 μg kg−1 and SIM, 74 μg kg−1) and the statistical model (univariate, 283 μg kg−1 and trilinear PLS, 20.91 μg kg−1). The capability of detection has been calculated as stated in ISO 11843 and Decision 2002/657/EC setting in all cases the probabilities of false positive and of false negative at 0.05.The identification of the mass spectra must be done to confirm the presence of clenbuterol and has been carried out through PARAFAC. The correlation coefficient between the spectra estimated by PARAFAC and the library spectra is 0.96 (hair, SCAN mode) and 1.00 (hair and urine, SIM mode).The Decision 2002/657/EC advocates the use of independent mass fragments to identify banned compounds. These recommendations together with the effect of the number of ions registered on the capability of detection have lead us to select five uncorrelated fragments (86, 243, 262, 264 and 277) from the data set of 210 ions by hierarchical clustering of variables.  相似文献

Competitive electrochemical enzyme-linked immunosorbent assays based on disposable screen-printed electrodes have been developed for quantitative determination of ochratoxin A (OTA). The assays were carried out using monoclonal antibodies in the direct and indirect format. OTA working range, I50 and detection limits were 0.05-2.5 and 0.1-7.5 μg L−1, 0.35 (±0.04) μg L−1 and 0.9 (±0.1) μg L−1, 60 and 100 μg L−1 in the direct and indirect assay format, respectively. The immunosensor in the direct format was selected for the determination of OTA in wheat. Samples were extracted with aqueous acetonitrile and the extract analyzed directly by the assay without clean-up. The I50 in real samples was 0.2 μg L−1 corresponding to 1.6 μg/kg in the wheat sample with a detection limit of 0.4 μg/kg (calculated as blank signal −3σ). Within- and between-assay variability were less than 5 and 10%, respectively. A good correlation (r = 0.9992) was found by comparative analysis of naturally contaminated wheat samples using this assay and an HPLC/immunoaffinity clean-up method based on the AOAC Official Method 2000.03 for the determination of OTA in barley.  相似文献

An improved LC-MS/MS method for the determination of semicarbazide in whole egg is described. Waters OASIS-MCX cation exchange purification cartridges increased the sensitivity for analysis by LC-MS/MS. The validation study was carried out according to criteria and requirements of Commission Decision 2002/657/EC for confirmatory analysis and provided the data as follows: The correlation coefficient for the matrix calibration curve, in the range of 0–5 μg kg−1, was r = 0.9968. The detection capability and decision limit, measured according to ISO11843-2, were CCα = 0.20 μg kg−1 and CCβ = 0.25 μg kg−1. Repeatability (CVSr) and within-laboratory reproducibility (CVSwr) determined for the concentration levels of 0.2, 0.5 and 1.0 μg kg−1 SEM ranged from 11.9 to 5.7% and 11.8 to 6.3%, respectively. The validated method was applied to investigate SEM stability in incurred materials (egg homogenates) during long-term storage at −20 °C and 4 °C. The study proved by a two-sampling test that SEM at levels of 17. 7, 1.2, 10.6 and 0.47 μg kg−1 was stable for up to 12 months.  相似文献

Fipronil, a phenylpyrazole insecticide introduced for pest control on a broad range of crops, can also affect non-target insects such as honeybees. More widely, non-target environment such as milk produced by dairy cows fed with maize silage from treated seeds (=silage T) can be affected. To assess the potential transfer of fipronil residues (sulfone, sulfide, fipronil, desulfinyl and amide), a methodology including gas chromatography coupled with tandem mass spectrometry (GC-MS/MS) analysis was developed and validated according to the 2002/657/EC decision, in order to reach a level of quantification below 0.1 μg L−1 in milk and 0.1 μg kg−1 in plants. Twelve dairy cows were fed with silage T during 4 months. Concentration of fipronil in treated seeds was estimated at 1 g kg−1, whereas silage from these seeds contained 0.30 ± 0.05 μg kg−1 of dry material of fipronil, 0.13 ± 0.03 μg kg−1 of dry material of sulfone. Sulfide residues were below the limit of quantification. Silage from untreated seeds (=silage U) presented traces of fipronil and sulfone, respectively at 0.04 ± 0.06 and 0.02 ± 0.03 μg kg−1 of dry material. Contribution of fipronil residues from supplies was insignificant. During administration of silage T, only sulfone residues were quantified in milk. The average concentration was 0.14 ± 0.05 μg L−1. Before and after administration, sulfone residues were detected but not quantifiable (<0.025 μg L−1). Our results suggest a transfer of fipronil from feed to milk under its sulfone form. Moreover, traces of fipronil residues in maize U, soya, wheat and straw show a diffuse contamination of this pesticide in the environment.  相似文献

Streptomycin (STR) and dihydrostreptomycin (DHSTR) are two of the most common aminoglycoside antibiotics used in veterinary medicine. The physicochemical properties of both substances, make their determination challenging. In the present study the development of methods based on ion-pair chromatography (IPC) and on hydrophilic interaction chromatography (HILIC), for the determination of the above mentioned aminoglycosides in the range of 100–1000 μg L−1 is described. The two methods were validated according to EU requirements for residues in food. The recoveries for the IPC method were 69.3% and 56.5% of STR and DHSTR, respectively, and for HILIC method 85.5% and 72.3%, respectively. The intra- and inter-day precision, studied at 100, 200 and 300 μg kg−1 levels in milk samples, gave %RSD ≤ 13 for both methods. LOQs for the HILIC method were 14 μg kg−1 for both analytes and for the IPC method were 109 and 31 μg kg−1, for STR and DHSTR, respectively. The sensitivity of the HILIC method is 80 and 210 times greater than that of the ICP method, for STR and DHSTR, respectively.  相似文献

In this article, we report a new method that involves headspace single-drop microextraction and ion chromatography for the preconcentration and determination of fluoride. The method lies in the in situ hydrogen fluoride generation and subsequent sequestration into an alkaline microdrop (15 μL) exposed to the headspace above the stirred aqueous sample. The NaF formed in the drop was then determined by ion chromatography. The influences of some crucial single-drop microextraction parameters such as the extraction temperature, extraction time, sample stirring speed, sulphuric acid concentration and ionic strength of the sample, on extraction efficiency were investigated. In the optimal condition, an enrichment factor of 97 was achieved in 15 min. The calibration working range was from 10 μg L−1 to 2000 μg L−1 (R2 = 0.998), and the limit of detection (signal to noise ratio of 3) was 3.8 μg L−1 of fluoride. Finally, the proposed method was successfully applied to the determination of fluoride in different milk samples. The recoveries of fluoride (at spiked concentrations of 200 μg L−1 and 600 μg L−1 into milk) in real samples ranged from 96.9% to 107.7%. Intra-day precision (N = 3) in terms of peak area, expressed as relative standard deviation, was found to be within the range of 0.24-1.02%.  相似文献

A gas chromatography–mass spectrometric (GC–MS) method has been established for the determination of hydrazine in drinking water and surface water. This method is based on the derivatization of hydrazine with ortho-phthalaldehyde (OPA) in water. The following optimum reaction conditions were established: reagent dosage, 40 mg mL−1 of OPA; pH 2; reaction for 20 min at 70 °C. The organic derivative was extracted with methylene chloride and then measured by GC–MS. Under the established condition, the detection and the quantification limits were 0.002 μg L−1 and 0.007 μg L−1 by using 5.0-mL of surface water or drinking water, respectively. The calibration curve showed good linearity with r2 = 0.9991 (for working range of 0.05–100 μg L−1) and the accuracy was in a range of 95–106%, and the precision of the assay was less than 13% in water. Hydrazine was detected in a concentration range of 0.05–0.14 μg L−1 in 2 samples of 10 raw drinking water samples and in a concentration range of 0.09–0.55 μg L−1 in 4 samples of 10 treated drinking water samples.  相似文献

Bismuth and Sb were evaluated as internal standards (IS) to minimize matrix effects on the direct and simultaneous determination of As, Cu, and Pb in cachaça by graphite furnace atomic absorption spectrometry using W-coated platform plus Pd-Mg(NO3)2 as modifier. For 20 μL injected sample, calibration within the 0.5-10 μg L−1 As, 100-1000 μg L−1 Cu and 0.5-30 μg L−1 Pb intervals were established using the ratios As absorbance to Sb absorbance, Cu absorbance to Bi absorbance and Pb absorbance to Bi absorbance versus analytes concentration, respectively. Typical linear correlations of 0.998, 0.999 and 0.999 were, respectively, obtained. The proposed method was applied for direct determination of As, Cu and Pb in 10 commercial cachaça samples and results were in agreement with those obtained by inductively coupled plasma mass spectrometry at 95% confidence level. The found characteristic masses were 30 pg As, 274 pg Cu and 39 pg Pb. The useful lifetime of the graphite tube was around 760 firings. Recoveries of As, Cu and Pb added to cachaça samples varied, respectively, from 98% to 109%, 97% to 108% and 98% to 104% with internal standards and from 48% to 54%, 53% to 92% and 62% to 97% without internal standards. The limits of detection were 0.13 μg L−1 As, 22 μg L−1 Cu and 0.05 μg L−1 Pb. The relative standard deviations (n = 12) for a spiked sample containing 20 μg L−1 As, Pb and 500 μg L−1 Cu were 1.6%, 1.0%, and 1.8% with IS and 4.3%, 5.2%, and 5.5% without IS.  相似文献

This study examines the application of solid-phase microextraction coupled with high performance liquid chromatography combined with post-column photochemically induced fluorimetry derivatization and fluorescence detection (SPME-HPLC-PIF-FD) for the determination of four phenylurea herbicides (monolinuron, diuron, linuron and neburon) and propanil in groundwater. Direct immersion (DI) SPME was applied using a 60 μm polydimethylsiloxane/divinylbenzene (PDMS/DVB) fiber for the extraction of the pesticides from groundwater samples. An AQUASIL C18 column (150 mm × 4.6 mm i.d., 5 μm) was used for separation and determination in HPLC. The method was evaluated with respect to the limits of detection (LODs) and the limits of quantification (LOQs) according to IUPAC. The limits of detection varied between 0.019 μg L−1 and 0.034 μg L−1. Limits of quantification ranged between 0.051 μg L−1 and 0.088 μg L−1. These values meet the recommended limits for individual pesticides in groundwater (0.1 μg L−1) established by the EU. Recoveries ranged between 86% and 105% and relative standard deviation values between 2% and 8%.  相似文献
