Aqueous solutions of fractions of an extracellular linear mannan formed by Rhodotorula rubra yeast have been investigated by hydrodynamic methods (high-speed sedimentation, translation isothermic diffusion and viscometry). The molecular weight was determined according to Svedberg ( ) and the polydispersity parameters of the initial sample were also determined (Mw/Mn = 1·20 and Mz/Mw = 1·21). Relationships between the molecular weight (M) and so, Do and [η] in the range were: [η] = 2·33 × 10−2 M0.75, Do = 1·65 × 10−4 M0·58, so = 2·24 × 10−15 M0·43. The equilibrium rigidity and hydrodynamic diameter of chains representing mannan molecules were evaluated.  相似文献

The formation of nitrite reductase and cytochrome c in Micrococcus denitrificans was repressed by O2. The purified nitrite reductase utilized reduced forms of cytochrome c, phenazine methosulphate, benzyl viologen and methyl viologen, respectively, as electron donors. The enzyme was inhibited by KCN, NaN3 and NH2OH each at 1 mM, whereas CO and bathocuproin, diethyl dithiocarbamate, o-phenanthroline and ,'-dipyridyl at 1 mM concentrations were relatively ineffective. The purified enzyme contains cytochromes, probably of the c and a2 types, in one complex. A Km of 46 μM for NO2 and a pH optimum of 6.7 were recorded for the enzyme. The molecular weight of the enzyme was estimated to be around 130000, and its anodic mobility was 6.8·10−6 cm2·sec−1·V−1 at pH 4.55.

The most highly purified nitrite reductase still exhibited cytochrome c oxidase activity with a Km of 27 μM for O2. This activity was also inhibited by KCN, NaN3 and NH2OH and by NO2.

A constitutive cytochrome oxidase associated with membranes was also isolated from cells grown anaerobically with NO2. It was inhibited by smaller amounts of KCN, NaN3 and NH2OH than the cytochrome oxidase activity of the nitrite reductase enzyme and also differed in having a pH optimum of about 8 and a Km for O2 of less than 0.1 μM. Spectroscopically, cytochromes b and c were found to be associated with the constitutive oxidase in the particulate preparation. Its activity was also inhibited by NO2.

The physiological role of the cytochrome oxidase activity associated with the purified nitrite reductase is likely to be of secondary importance for the following reasons: (a) it accounts for less than 10% of total cytochrome c oxidase activity of cell extracts; (b) the constitutive cytochrome c oxidase has a smaller Km for O2 and would therefore be expected to function more efficiently especially at low concentrations of O2.  相似文献

1. Rate constants for reduction of paraquat ion (1,1′-dimethyl-4,4′-bipyridy-lium, PQ2+) to paraquat radical (PQ+·) by eaq and CO2· have been measured by pulse radiolysis. Reduction by eaq is diffusion controlled (k = 8.4·1010 M−1·s−1) and reduction by CO2· is also very fast k = 1.5·1010 M−1·s−1).

2. The reaction of paraquat radical with oxygen has been analysed to give rate constants of 7.7·108 M−1·s−1 and 6.5·108 M−1·s−1 for the reactions of paraquat radical with O2 and O2·, respectively. The similarity in these rate constants is in marked contrast to the difference in redox potentials of O2 and O2· (− 0.59 V and + 1.12 V, respectively).

3. These rate constants, together with that for the self-reaction of O2·, have been used to calculate the steady-state concentration of O2· under conditions thought to apply at the site of reduction of paraquat in the plant cell. On the basis of these calculations the decay of O2· appears to be governed almost entirely by its self-reaction, and the concentration 5 μm away from the thylakoid is still 90% of that at the thylakoid itself. Thus, O2· persists long enough to diffuse as far as the chloroplast envelope and tonoplast, which are the first structures to be damaged by paraquat treatment. O2· is therefore sufficiently long-lived to be a candidate for the phytotoxic product formed by paraquat in plants.  相似文献

1. Difference spectra, at room and liquid N2 temperatures, of S2O42−-, and NO2-reduced intact cells and cell-free preparations of Nitrobacter agilis demonstrated the presence of cytochromes of the c- and a-types. Reduction of cytochromes by succinate, and to a limited extent, by NADPH also occurred, provided KCN (0.1 mM) was also present.

2. A particulate, heat-labile nitrite oxidase having an absolute requirement for O2 was prepared from N. agilis cells using sonic oscillation and differential centrifugation. The particles also possessed NADH oxidase, succinoxidase, formate oxidase and traces of NADPH oxidase activity. The stoichiometry of the nitrite oxidase reaction approached the theoretical value of 2 moles of NO2 consumed per mole of O2 consumed. The pH optimum of the nitrite oxidase system shifted to progressively more alkaline values as the NO2 concentration was increased, changing from a pH value of 6.8 at 0.6 mM KNO2 to pH 8.0 at 0.01 M KNO2 with apparent Km's of 0.2 and 1.2 mM NO2, respectively. Computations of the HNO2 concentrations present under the above conditions showed an approx. 500-fold greater affinity for HNO2 which was independent of pH, suggesting the involvement of HNO2 as both a substrate and an inhibitor (at higher concentrations) of the nitrite oxidase system. The marked inhibition by NaN3, NaCN and Na2S, as well the light-reversible inhibition by CO, indicated the presence of cytochrome oxidase which was subsequently characterized. NO2 proved to be a competitive inhibitor of the nitrite oxidase system.

3. The particulate preparation also possessed a heat-labile nitrite-cytochrome c reductase activity which was energy independent and routinely measured under anaerobic conditions. As in the case of nitrite oxidase, the affinity of the enzyme for NO3 increased as the pH was lowered, but the pH optimum remained unaffected. In terms of calculated HNO2 concentration an approximately constant Km of about 0.2 μM was estimated at the several pH's examined. The inhibition by NO3 was shown to be competitive. The marked sensitivity of the reductase to several metal-binding agents implicated a metal component in the electron transport chain at the site prior to cytochrome c.

Static light scattering of high amylopectin waxy maize starch gently dispersed in 90% dimethyl sulfoxide–water yielded a weight average molecular weight Mw and radius of gyration Rg of 560×106 g/mol and 342 nm, respectively. To obtain an independent hydrodynamic characterization of these solutions, we measured the sedimentation coefficient for the main component in an analytical ultracentrifuge. The value of s0, the infinite dilution sedimentation coefficient, was 199 S. The translational diffusion coefficient D0 in very dilute solutions was measured by dynamic light scattering at 90° and found to be 2.33×10−9 cm2/s. An effective hydrodynamic radius Rh was calculated from this diffusion constant using the Stokes–Einstein equation and found to be 348 nm. The structure-related parameter ρ=Rg/Rh was calculated to be 0.98. The weight average molecular weight calculated from the Svedberg equation using the values measured for s0 and D0 was 593×106 g/mol. This result is in reasonable agreement with the light scattering results. As light scattering results are subject to experimental errors due to the possibility of dust contamination, the presence of microgel or aggregates, and the questionable applicability of light scattering theory to interpret results for macromolecular sizes approaching the wave length of light used as a source for scattering, it is advisable to have corroborating hydrodynamic data when possible to further validate light scattering results in this very high molecular weight range.  相似文献

The thermoluminescence band observed in chloroplasts after flash excitation at ambient temperatures has recently been identified as being due to recombination of the electron on the semiquinone form of the secondary plastoquinone acceptor, QB, with positive charges on the oxygen-evolving enzyme, S2 and S3 (Rutherford, A.W., Crofts, A.R. and Inoue, Y. (1982) Biochim. Biophys. Acta 682, 457–465). Further investigation of this thermoluminescence confirms this assignment and provides information on the function of PS II. The following data are reported: (1) Washing of chloroplasts with ferricyanide lowers the concentration of QB in the dark and predictable changes in the extent of the thermoluminescence band are observed. (2) The thermoluminescence intensity arising from S2QB is approximately one half of that arising from S3QB. (3) Preflash treatment followed by dark adaptation results in changes in the intensity of the thermoluminescence band recorded after a series of flashes. These changes can be explained according to the above assignments for the origin of the thermoluminescence and if QB provides an important source of deactivating electrons for the S states. Computer simulations of the preflash data are reported using the above assumptions. Previously unexplained data already in the literature (Läufer, A. and Inoue, Y. (1980) Photobiochem. Photobiophys. 1, 339–346) can be satisfactorily explained and are simulated using the above assumptions. (4) Lowering the pH to pH 5.5 results in a shift of the S2QB thermoluminescence band to higher temperatures while that arising from S3QB does not shift. This effect is interpreted as indicating that QB is protonated and the S2 to S3 reaction involves deprotonation while the S1 to S2 reaction does not.  相似文献

1. 1. The superoxide anion radical (O2) reacts with ferricytochrome c to form ferrocytochrome c. No intermediate complexes are observable. No reaction could be detected between O2 and ferrocytochrome c.

2. 2. At 20 °C the rate constant for the reaction at pH 4.7 to 6.7 is 1.4 · 106 M−1 · s−1 and as the pH increases above 6.7 the rate constant steadily decreases. The dependence on pH is the same for tuna heart and horse heart cytochrome c. No reaction could be demonstrated between O2 and the form of cytochrome c which exists above pH ≈ 9.2. The dependence of the rate constant on pH can be explained if cytochrome c has pKs of 7.45 and 9.2, and O2 reacts with the form present below pH 7.45 with k = 1.4 · 106 M−1 · s−1, the form above pH 7.45 with k = 3.0 · 105 M−1 · s−1, and the form present above pH 9.2 with k = 0.

3. 3. The reaction has an activation energy of 20 kJ mol−1 and an enthalpy of activation at 25 °C of 18 kJ mol−1 both above and below pH 7.45. It is suggested that O2 may reduce cytochrome c through a track composed of aromatic amino acids, and that little protein rearrangement is required for the formation of the activated complex.

4. 4. No reduction of ferricytochrome c by HO2 radicals could be demonstrated at pH 1.2–6.2 but at pH 5.3, HO2 radicals oxidize ferrocytochrome c with a rate constant of about 5 · 105–5 · 106 M−1 · s−1

Eight hemicellulosic fractions were obtained by sequential treatment of dewaxed barley straw with 0.1 M NaOH at 45 °C for 3 h, 0.25, 0.5, 1.0, 1.5, 2.0, and 3.0% H2O2 at 45 °C for 3 h at pH 11.5, and 10% KOH–1% Na2B4O7·10H2O at 28 °C for 15 h under continuous agitation. The yields of the fractions were 8.0, 3.1, 3.3, 3.3, 2.2, 2.0, 2.0, and 9.9%, respectively, of the initial amount of barley straw, corresponding to the dissolution of 21.6, 8.4, 8.9, 8.9, 5.9, 5.4, 5.4, and 26.7% of the original hemicelluloses. Meanwhile, the successive treatment also solubilized 29.1, 15.8, 14.6, 10.8, 4.5, 3.2, 2.7, and 3.7% of the original lignin, respectively. This sequential extraction together resulted in dissolution of 91.1% of the original hemicelluloses and 84.8% of the original lignin. The 0.1 M NaOH-soluble hemicellulosic fraction contained mainly xylose, glucose, and arabinose, 44.2, 15.7, and 15.2%, respectively, while the 10% KOH–1% Na2B4O7·10H2O-soluble fraction predominated in xylose, 75.0%. The six alkaline peroxide-soluble fractions were composed of 50.3–54.4% xylose, 14.7–16.9% arabinose, 6.8–10.7% glucose, 6.8–8.5% glucuronic acid or 4-O-methyl- -glucuronic acid, 0.4–1.5% mannose, and 0.3–1.2% rhamnose. All the hemicellulosic fractions contained substantial amounts of glucuronoarabinoxylans and noticeable quantities of β-glucans. In comparison, the six hemicellulosic fractions, isolated with alkaline peroxide, had much higher molecular weights (56,890–63,810 g mol−1) than those of the two hemicellulosic preparations (28,000–29,080 g mol−1), isolated with alkali in the absence of hydrogen peroxide. The thermal stability of the hemicelluloses increased with an increment of their molar mass.  相似文献

Complexes of type A4[VO(tart)]2·nH2O, where A = Rb or Cs and tart =d,l-tartrate(4−) (n = 2) or d,d-tartrate(4−) (n = 2 for Rb and n = 3 for Cs), were prepared from an aqueous mixture of V2O5, AOH and H4tart. These complexes were studied by single-crystal X-ray diffraction methods: Rb4[VO(d,l-tart)]2·2H2O, space group P1 with a = 8.156(1),b = 8.246(1),c = 8.719(1)Å, = 66.09(1)°, β = 65.07(1)°, γ = 82.40(1)°,Z = 2, 1917 observed reflections, and final Rw = 0.035; Cs4[VO(d,l-tart)]2·2H2O, space group P21/c with a = 9.350(1),b = 13.728(2),c = 8.479(1)Å, β = 106.77(1)°,Z = 4, 2235 observed reflections, and final Rw = 0.054; Rb4[VO(d,d-tart)]2·2H2O, space group P4122 with a = 8.072(1),c = 32.006(3)Å,Z = 8, 1014 observed reflections and final Rw = 0.038; Cs4[VO(d,d-tart)]2·3H2O, space group P122 with a = 8.184(1),c = 33.680(5)Å,Z = 8, 1310 observed reflections, and final Rw = 0.063. Bulk magnetic susceptibility data (1.5–300 K) for these compounds and A4[VOl,l-tart)]2·nH2O (A = Rb, Cs) were obtained on polycrystalline samples. These data were analyzed in terms of a Van Vleck exchange coupled S = 1/2 model which was modified to include an interdimer exchange parameters Θ. Analysis of the low-temperature (1.5–20 K) susceptibility data gave 2J = +1.30 cm−1 and Θ = −1.86 K for Rb4[VO(d,l-tart)]2·2H2O, 2J = +1.16 cm−1 and Θ = −1.69 K for Cs4[VO(d,l-tart)]2·2H2O, 2J = +1.90 cm−1 and Θ = −0.82 K for Rb4[VO(d,d-tart)]2·2H2O, 2J = +2.04 cm−1 and Θ = −0.80 K for Rb4[VO(l,l-tart)]2·2H2O, 2J = +1.52 cm−1 and Θ = −0.25 K for Cs4[VO(d,d-tart)]2·3H2O, and 2J = +1.64 cm−1 and Θ = −0.31 K for Cs4[VO(l,l-tart)]2·3H2O. These results suggest the magnitudes of intradimer (ferromagnetic and interdimer (antiferromagnetic) exchange interactions are similar in these complexes, as observed for the analogous Na salts.  相似文献

Hydrogen peroxide, produced by inflammatory and vascular cells, induces oxidative stress that may contribute to endothelial dysfunction. In smooth muscle cells, H2O2 induces production of O2 by activating NADPH oxidase. However, the mechanisms whereby H2O2 induces oxidative stress in endothelial cells are poorly understood. We examined the effects of H2O2 on O2 levels on porcine aortic endothelial cells (PAEC). Treatment with 60 μmol/L H2O2 markedly increased intracellular O2 levels (determined by conversion of dihydroethidium to hydroxyethidium) and produced cytotoxicity (determined by propidium iodide staining) in PAEC. Overexpression of human manganese superoxide dismutase in PAEC reduced O2 levels and attenuated cytotoxicity resulting from treatment with H2O2. L-NAME, an inhibitor of nitric oxide synthase (NOS), and apocynin, an inhibitor of NADPH oxidase, reduced O2 levels in PAEC treated with H2O2, suggesting that both NOS and NADPH oxidase contribute to H2O2-induced O2 in PAEC. Inhibition of NADPH oxidase using apocynin and NOS rescue with L-sepiapterin together reduced O2 levels in PAEC treated with H2O2 to control levels. This suggests interaction-distinct NOS and NADPH oxidase pathways to superoxide. We conclude that H2O2 produces oxidative stress in endothelial cells by increasing intracellular O2 levels through NOS and NADPH oxidase. These findings suggest a complex interaction between H2O2 and oxidant-generating enzymes that may contribute to endothelial dysfunction.  相似文献

The preparation and reaction chemistry of 1,3- and 1,2-diene and related complexes derived from metal carbonyl containing anions and allenic electrophiles are addressed. The preparation of some CpFe(CO)2 η1-diene complexes and their conversion into CpFe(CO) η3-diene complexes is presented followed by reactions of CpMo(CO)3, CpW(CO)3 and CpMo(CO)2PR3 anions with allenic electrophiles which produce metal complexed cyclobutenones (via CO and alkene insertions from the initially formed product) and 1,2-diene complexes, respectively. Lastly, the reactions of PPh3(CO)3Co anions with allenic electrophiles are outlined which result in several different coordination geometries depending on the reaction conditions used.  相似文献

Resonance Raman measurements have been performed with solutions of iodine-complexed synthetic amyloses (DP 25–200), malto-oligomers (DP 3–18, and -cylodextrin. Interest was focused on the minimum chain length for helical complex formation and a possible preferred length for the polyiodine chain. Four fundamental vibrations are observed at 164, 112, 52 and 24 cm−1. The 112 cm−1 Raman line was shown to arise both from free I3 (enhanced at 363.8 nm excitation) and from bound iodine (relatively most intense at 457.9 nm excitation). The main signal of complexed iodine at 164 cm−1is enhanced at an excitation wavelength close to the long wavelength absorption maximum. This signal is observed firt with malto-octaose and -cyclodextrin. The less intense signals at 52 and 24−1 are only detected at DP 15 and higher. Raman spectra give no evidence for a preferred length of the polyiodine chain. Significantly identical Raman spectra are obtained when using different molar ratios of I2/KI solution or I2 solution initially free of I ions. The results are discussed in view of previous assignments of the Raman lines to I2, I3/I2, and I5 subunits. Our findings are incompatible with I3 units as the only bound species. They are compatible with both I3/I2 and I3 subunits under certain conditions. In the case of I2 solution used for complexation we favour the polyiodine chain model proposed previously by Cramer35,36. The I3 ions formed could function mainly as chain initiators, as has been suggested by Cesàro and Brant30.  相似文献

A procedure is described for preparing particles from cells of Micrococcus denitrificans which were broken osmotically after treatment with lysozyme.

1. 1. The preparations catalysed ATP synthesis coupled to O2 uptake or NO3 reduction. With NADH or succinate as the electron donors the P:O ratios were about 1.5 and 0.5, respectively; and the P:NO3 ratios were about 0.9 and 0.06, respectively.

2. 2. Addition of ADP or Pi to the reaction mixture increased the rates of NADH-dependent O2 uptake and NO3 reduction. Addition of 1 mM 2,4-dinitrophenol, which inhibited phosphorylation by 50–60%, increased the basal rates of electron transport.

3. 3. Evidence derived from spectrophotometry and from the differential inhibition by antimycin A of O2 and NO3 reduction leads to the conclusion that the nitrate reductase interacted with the respiratory chain in the region of the b-type cytochrome, and that the c-type cytochrome present was not involved in the reduction of NO3 to NO2.

The perchlorate (ClO4)-respiring organism, strain perc1ace, can grow using nitrate (NO3) as a terminal electron acceptor. In resting cell suspensions, NO3 grown cells reduced ClO4, and ClO4 grown cells reduced NO3. Activity assays showed that nitrate reductase (NR) activity was 1.31 μmol min−1 (mg protein)−1 in ClO4 grown cells, and perchlorate reductase (PR) activity was 4.24 μmol min−1 (mg protein)−1 in NO3 grown cells. PR activity was detected within the periplasmic space, with activities as high as 14 μmol min−1 (mg protein)−1. The NR had a pH optimum of 9.0 while the PR had an optimum of 8.0. This study suggests that separate terminal reductases are present in strain perclace to reduce NO3 and ClO4.  相似文献

The reactions of Ru(NH3)5py2+, Ru(NH3)4bpy2+, Ru2(NH3)10pz5+, RuRh(NH3)10pz5+ and Ru(NH3)5pz2+ with bromine are first-order in ruthenium and first-order in bromine. The rates decrease with increasing bromide ion concentration and, except for Ru(NH3)5pz2+, are independent of hydrogen ion concentration. The reactions are postulated to proceed via outer-sphere, one-electron transfer from Ru(II) to Br2 with the formation of Br2 as a reactive intermediate. The bromide inhibition is ascribed to the formation of Br3 which is unreactive in outer-sphere reactions because of the barrier imposed by the need to undergo reductive cleavage. The reaction of Ru(NH3)5pz2+ is inhibited by hydrogen ions. The hydrogen ion dependence shows that Ru(NH3)5pzH3+ has a pKa of 2.49 and is at least 500 times less reactive than Ru(NH3)5pz2+. The reaction of Ru2(NH3)10pz4+ with bromine is biphasic. The second phase has a rate identical to that of the Ru2(NH3)10pz5+-Br2 reaction. A detailed analysis shows that the reaction of Ru2(NH3)10pz4+ with bromine proceeds by a sequence of one-electron steps, Br2 being produced as an intermediate. A linear free energy relationship between rate constants and equilibrium constants, obeyed for all the reactions studied, provides an estimate of 1.5 × 102 M−1 s−1 for the self-exchange rate constant of the Br2/Br2 couple.  相似文献

The mechanism of the charge separation and stabilization of separated charges was studied using the femtosecond absorption spectroscopy. It was found that nuclear wavepacket motions on potential energy surface of the excited state of the primary electron donor P* leads to a coherent formation of the charge separated states P+BA, P+HA and P+HB (where BA, HB and HA are the primary and secondary electron acceptors, respectively) in native, pheophytin-modified and mutant reaction centers (RCs) of Rhodobacter sphaeroides R-26 and in Chloroflexus aurantiacus RCs. The processes were studied by measurements of coherent oscillations in kinetics at 890 and 935 nm (the stimulated emission bands of P*), at 800 nm (the absorption band of BA) and at 1020 nm (the absorption band of BA) as well as at 760 nm (the absorption band of HA) and at 750 nm (the absorption band of HB). It was found that wavepacket motion on the 130–150 cm−1 potential surface of P* is accompanied by approaches to the intercrossing region between P* and P+BA surfaces at 120 and 380 fs delays emitting light at 935 nm (P*) and absorbing light at 1020 nm (P+BA). In the presence of Tyr M210 (Rb. sphaeroides) or M195 (C. aurantiacus) the stabilization of P+BA is observed within a few picosseconds in contrast to YM210W. At even earlier delay (40 fs) the emission at 895 nm and bleaching at 748 nm are observed in C. aurantiacus RCs showing the wavepacket approach to the intercrossing between the P* and P+HB surfaces at that time. The 32 cm−1 rotation mode of HOH was found to modulate the electron transfer rate probably due to including of this molecule in polar chain connecting PB and BA and participating in the charge separation. The mechanism of the charge separation and stabilization of separated charges is discussed in terms of the role of nuclear motions, of polar groups connecting P and acceptors and of proton of OH group of TyrM210.  相似文献

Nitrogen dioxide (NO2) is a key biological oxidant. It can be derived from peroxynitrite via the interaction of nitric oxide with superoxide, from nitrite with peroxidases, or from autoxidation of nitric oxide. In this study, submicromolar concentrations of NO2 were generated in < 1 μs using pulse radiolysis, and the kinetics of scavenging NO2 by glutathione, cysteine, or uric acid were monitored by spectrophotometry. The formation of the urate radical was observed directly, while the production of the oxidizing radical obtained on reaction of NO2 with the thiols (the thiyl radical) was monitored via oxidation of 2,2′-azino-bis-(3-ethylthiazoline-6-sulfonic acid). At pH 7.4, rate constants for reaction of NO2 with glutathione, cysteine, and urate were estimated as 2 × 107, 5 × 107, and 2 × 107 M−1 s−1, respectively. The variation of these rate constants with pH indicated that thiolate reacted much faster than undissociated thiol. The dissociation of urate also accelerated reaction with NO2 at pH > 8. The thiyl radical from GSH reacted with urate with a rate constant of 3 × 107 M−1 s−1. The implications of these values are: (i) the lifetime of NO2 in cytosol is < 10 μs; (ii) thiols are the dominant ‘sink’ for NO2 in cells/tissue, whereas urate is also a major scavenger in plasma; (iii) the diffusion distance of NO2 is 0.2 μm in the cytoplasm and < 0.8 μm in plasma; (iv) urate protects GSH against depletion on oxidative challenge from NO2; and (v) reactions between NO2 and thiols/urate severely limit the likelihood of reaction of NO2 with NO• to form N2O3 in the cytoplasm.  相似文献

The reactions of the polysulfur and selenium cationic clusters S82+ and Se82+ with various iron carbonyls were investigated. Several new chalcogen containing iron carbonyl cluster cations were isolated, depending on the nature of the counteranion. In the presence of SbF6 as a counterion, the cluster [Fe3(E2)2(CO)10] [SbF6]2·SO2 (E = S, Se) could be isolated from the reaction of E82+ and excess iron carbonyl. The cluster is a picnic-basket shaped molecule of two iron centers linked by two Se2 groups, with the whole fragment capped by an Fe(CO)4 group. Crystallographic data for C10O12Fe3Se4Sb2F12S (I): space group monoclinic P21/c, A = 11.810(9), b = 24.023(6), c = 10.853(7) Å, β = 107.15(5)°, V = 2942(3) Å3, Z = 4, R = 0.0426, Rw = 0.0503. When Sb2F11 is present as the counterion, or Se4[Sb2F11]2 is used as the cluster cation source, a different cluster can be isolated, which has the formula [Fe4(Se2)3(CO)12] [SbF6]2·3SO2. The dication contains two Fe2Se2 fragments bridged by an Se2 group. Crystallographic data for C12O18Fe4Se6Sb2F12S3 (III): space group triclinic , b = 18.400(9), C = 10.253(4) Å, = 93.10(4), β = 103.74(3), γ = 93.98(3)°, V = 1995(1) Å3, Z = 2, R = 0.0328, Rw = 0.0325. The CO stretches in the IR spectrum all show a large shift to higher wavenumbers, suggesting almost no τ backbonding from the metals. This also correlates with the observed bond distances. All the compounds are extremely sensitive to air and water, and readily lose SO2 when removed from the solvent. Thus all the crystals were handled at −100°C. The clusters seem to be either insoluble or unstable in all solvents investigated.  相似文献

Using defatted and SH-blocked bovine serum albumin (BSA), measurements of differential scanning calorimetry (d.s.c.) have been made mainly in NaSCN solution. BSA undergoes a heat-induced conformational transition in a particular range of pH and ionic strength and is separated into two thermally independent units, each of which has different thermostability in acidic and alkaline pH regions. Comparisons were made of the pH dependencies of the enthalpy of thermal denaturation (ΔH) and the temperature of thermal denaturation (Td) in 0.01 NaSCN with those in 0.01 NaCl. It has been found that the stabilizing effect of NaSCN on BSA is larger than that of NaCl at pH 3.5–8, and that the heat-induced transition occurs by the electrostatic repulsive forces among the positively charged amino acid residues in a segment Arg 184–Arg 216 containing Trp 212 and the primary binding sites of anions. At ionic strength 0.01, the relative effectiveness of anions in suppressing the heat-induced transition and increasing the thermostability of BSA follows the order ClO4 − SCN > I > SO42− > Br > Cl. At ionic strength 0.1, the heat-induced transition is suppressed in all the salt solutions, and a Td increase follows the order ClO4 SCN > I > Br > Cl SO42−. Thus, the highly chaotropic ions thermostabilize BSA more markedly than kosmotropic ions in the low and moderate salt concentrations. In contrast, chaotropic ions destabilize BSA and kosmotropic ions stabilize BSA at the higher concentrations. An adequate amount of NaCl or NaSCN prevents the destruction of the environment of the binding site in the segment containing Trp 212 in 4 urea solution at pH 7.0.  相似文献
