本文介绍了测定板栗中氨基酸的高效液相色谱法。样品水解产物中的氨基酸采用离子交换法分离、梯度洗脱、OPA(邻苯二甲醛)柱后衍生,然后再用荧光测法检测(Ex348nm,Em450nm)。17种氨基酸在50分钟内得到完全的分离。方法的标准偏差在0.16%以内,变异系数为2.8—14%(n=4)。  相似文献

研究了反相超高效液相色谱法测定水质中乙酸含量的色谱条件。结果表明:在Waters T3(50mm×3mm×1.8μm)色谱柱上,以NaH2PO4(20mM/L,pH=2.6)/H3PO4缓冲液作为流动相,210nm处紫外检测水质中的乙酸;其流动相流速为0.2mL/min,柱温30℃,进样量1.0μL。该方法简便快捷,样品加标回收率为68.5%~72.1%。  相似文献

建立了檀香叶中γ-氨基丁酸(GABA)含量的高效液相色谱检测方法.以乙醚为溶剂,采用索氏提取除去色素,水提醇沉去除糖类和蛋白质的净化方法,以丹酰氯为衍生剂,高效液相色谱法分离,荧光检测器测定,外标法定量.峰面积与浓度之间线性关系良好,回归方程为y=475 211x+ 69 037,R2=0.999 8,加标回收率为112 %~120%,检测限为7μg/g.该方法操作步骤简单,灵敏度高,用于檀香叶中GABA的检测,方法简便、准确、高效.  相似文献

采用高效液相色谱法(HPLC)同时测定金银花中异绿原酸A、B、C的含量,并对色谱条件进行优化。结果表明,最佳色谱条件为Eclipse plus C18色谱柱(4.6mm×150mm,5μm),流动相为0.1%磷酸溶液(A)-乙腈(B),梯度洗脱,洗脱条件为0-20min,15%~30%B,20-30min,30%~50%B,流速1.0mL/min,检测波长325nm,柱温30℃,进样量10μL。在最佳的色谱条件下,实现了金银花对照品及样品中异绿原酸3种结构化合物的基线分离,为异绿原酸A、B、C含量的同时测定奠定了基础。该方法优化后缩短了测试时间,为金银花质量的快速检测提供参考依据。  相似文献

为了提取白桦三萜炎物质,建立了高效液相色谱法测定白桦(Betula platyphylla Suk.)树皮中白桦酯醇含量的方法、色谱柱为ODS C18;流动相为甲醇:水(88:12),紫外检测波长为210nm,柱温30℃;柱压6.4MPa,流速1mL/min,白桦酯醇的线性范围为15.0~300.0μg,平均回收率为93.93%。对同一样品重复测定5次,相对标准偏差为0.36%,与比色法和气相色谱法相比,本法更准确、简便。  相似文献

以不同引种地不同品种的9个新鲜牡丹花瓣为实验材料,采用高效液相色谱法(HPLC)测定9个不同引种地不同品种牡丹花瓣样品中的化学成分。液相色谱条件为Eclipse plus C18色谱柱(4.6×150mm,5μm),流动相为乙腈-1%乙酸梯度洗脱,流速0.8mL/min,检测波长360nm,柱温为35℃。通过中药色谱指纹图谱相似度评价系统(国家药典委员会)进行相似度分析,建立9个样品牡丹花瓣的指纹图谱,为科学评价与有效控制牡丹花瓣产品质量提供新方法。结果表明,本研究建立的分析方法有较好的重复性,9个引种牡丹花瓣样品指纹图谱共有6个主要共有峰,可作为鉴别引种地牡丹花瓣质量的主要依据。  相似文献

为了使中纤板板面具有更好的色泽,增加销量,文章就如何提高中纤板表面白度进行了试验性研究。采用以草酸、漂白粉、VBL型荧光增白剂、双氧水为主剂的处理配方分别用涂刷的方法对小样中纤板表面进行增白试验,通过测定和分析处理效果发现,草酸、双氧水是适合中纤板板面增白的处理剂,综合考虑处理结果,以及成本和环保要求等因素,最后确定H2O2(27.5%)、NaOH(5%)、Na2SiO3(5%)以适当比例混合和浓度为5%的草酸为最佳中纤板板面增白配方。最后采用喷雾法用最佳处理配方对大样中纤板进行增白试验。结果表明,采用这两个配方对中纤板表面进行喷雾增白处理,白度分别可提高42.6%和17.5%,处理成本分别为127.11元/m^3和38.35元/m^3。  相似文献

综述了果蔬抗氧化作用的可能机理和果蔬抗氧化作用的评价方法;重点介绍了Rancimat法、β-胡萝卜-亚油酸乳化液氧化法、DPPH·法、硫代巴比妥酸反应物(TBAS)法和FRAP法5种目前常用的果蔬抗氧化作用评价方法及其采用这些方法所取得的研究结果.指出有必要加强在同一条件下用不同的测定方法对同一来源的果蔬进行抗氧化活性比较的研究,这对于选择抗氧化活性测定的方法具有指导意义.  相似文献

山杏叶桦木酸、齐墩果酸和熊果酸的研究   总被引:1,自引:0,他引:1  
我国山杏资源十分丰富,是绿化荒山的优良树种。采用HydersirBDSC18色谱柱(150×4.6mm,5μm),流动相体积比为90:10的0.3‰硫酸甲醇溶液一3‰硫酸水溶液,检测波长210nm,建立了山杏叶中三萜酸测定的高效液相色谱法。结果表明:桦木酸、齐墩果酸与熊果酸的浓度在30~2000μg/mL(r桦=0.9971,r齐=0.9986,r熊=0.9978)范围内与色谱峰面积呈良好线性关系;平均加标回收率分别为99.9%、101.1%和98.9%。以山杏叶为材料,采用高效液相色谱法和紫外可见分光光度法分别对山杏叶中的三萜酸成分进行分析,结果表明,山杏叶富含三萜酸,落叶期山杏叶中总三萜酸(17078#g/g)、桦木酸(1842μg/g)、齐墩果酸(2536μg/g)和熊果酸(5706μg/g)含量均高于非落叶期。本研究首次建立了山杏叶中桦木酸、齐墩果酸和熊果酸同时测定的反向高效液相色谱法(HPLC),发现落叶期山杏叶富含三萜酸,为山杏叶综合开发利用提供了基础数据。  相似文献

用高效液相色谱法同时测定广西5地互叶白千层中槲皮素和山奈酚含量。样品用盐酸-甲醇(体积比5∶100)混合溶液水浴加热回流提取,C18柱色谱分离,以甲醇-乙腈-0.4%磷酸(体积比21∶21∶58)水溶液为流动相,检测波长370 nm,柱温为室温,流速1 mL/min。测定了不同产地互叶白千层中的槲皮素和山奈酚质量分数,其中邕宁南晓的槲皮素和山奈酚的质量分数最高,分别为5.151和2.530 mg/g,邕宁的槲皮素和山奈酚的质量分数为最低,分别为2.386和1.658 mg/g。  相似文献

Wood treated with CCB (chromated copper borate) has an expected service life of 20–50 years, depending on conditions of service and method of treatment. After this period, treated wood is discarded as special waste. Due to toxic elements (Cu, Cr, and B) in such treated wood, burning and landfill disposal are not considered as environmentally sound solutions. Chemical extraction seems more promising. The principle of the method is to convert fixed biocides in wood into soluble form and leach them out of the wood. In order to elucidate this process, copper(II) sulfate, potassium dichromate, and CCB-treated samples were leached using five different agents, namely, water, an aqueous solution of oxalic or acetic acid, and an aqueous solution of oxalic or acetic acid with ammonia. Afterwards, the amounts of leached copper and chromium were determined. In a parallel experiment, electron paramagnetic resonance measurements of wet- and dry-leached and unleached samples were performed in order to determine the paramagnetic complexes that were formed. The highest chromium leaching was achieved with the aqueous solution of oxalic acid, as chromium was converted into water-soluble chromium oxalate. While the mixture of oxalic acid and ammonia resulted in the highest copper leaching, EPR observation revealed that in this case water-soluble complexes of copper, oxalic acid, and ammonia were formed. We found out that oxalates formation is not the only mechanism responsible for copper or chromium leaching. High acidity may play an important role too.  相似文献

A capillary zone electrophoresis (CZE) method for simple and rapid determination of ellagic acid (EA) in Eucalyptus globulus wood and in the filtrate from unbleached kraft pulp has been developed. This is the first application of CZE for the detection of EA in industrial streams from cellulosic pulp production. The EA determinations in wood extractives and in pulp filtrates were succeeded only after sample acidification. This new CZE analytical procedure allowed reliable determinations of EA in E. globulus wood (1.1 ± 0.6 g kg?1 of dry wood) and in the filtrates from unbleached kraft pulp (98 ± 0.7 mg L?1). Gas chromatography–mass spectrometry was used as a reference method for the quantification of EA in industrial samples.  相似文献

采用Olsen法测定土壤速效磷,采用土壤无机磷分级测定法测定土壤无机磷,分别测得在不同浓度草酸作用下哈尔滨市保护地土壤的磷含量的动态变化。试验结果表明,草酸对保护地土壤不同形态无机磷含量的影响分别是:草酸处理的Ca2-P含量、Ca8-P含量和Al-P含量随着草酸浓度的增加而增加;草酸处理的Fe-P含量和O-P含量随着培养时间的增加而增加;草酸处理的Ca10-P含量在24 h和96 h有2次的峰值,高浓度草酸处理的Fe-P、O-P、Ca10-P含量反而比低浓度处理的Fe-P、O-P、Ca10-P含量低。  相似文献

To investigate the bleaching mechanism, a lignincarbohydrate complex (LCC) model compound, a vinyl ether-type lignin model dimer, and a hexeneuronic acid model compound were treated with dilute sulfuric acid of different pHs. Beech kraft pulp and red pine kraft pulp were also treated with dilute sulfuric acid and then extracted with aqueous alkali. The amount of hexeneuronic acid degradation products in acid effluents and lignin dissolved in alkali effluents were determined. It was found that the benzyl ether-type LCC bond and the vinyl ether bond in lignin were effectively cleaved under the pH where sulfuric acid bleaching of kraft pulp was effective. Hexeneuronic acid group was also effectively degraded during sulfuric acid bleaching. In beech kraft pulp bleaching, both lignin removal and hexeneuronic acid removal contributed to the kappa number reduction. In red pine bleaching, the contribution of hexeneuronic acid removal was negligible, and most of the kappa number reduction was achieved by the lignin removal.Part of this report was presented at the 9th International Symposium on Wood and Pulping Chemistry, Montreal, July 1997  相似文献

A method of fractionation, quantitation and identification of abscisic acid (ABA) in poplar flowers is presented in the context of intra- and interspecific pollination studies in Populus nigra x P. nigra and P. nigra x P. deltoides, respectively. Poplar flowers were dissected into stigma, ovary and pedicel. Abscisic acid concentrations were measured at receptivity and 5 days after pollination in each part of the flower. Abscisic acid was fractionated by reverse-phase HPLC followed by an enzyme-linked immunosorbent assay (ELISA) with anti-ABA polyclonal antibody and GC-MS identification of the immunoreactive ABA fractions. Samples were fractionated and ABA was identified by the presence of a cross-reactive metabolite of ABA in the extracts. The interspecific cross (P. nigra x P. deltoides) had a high concentration of ABA in the pedicel, where abscission occurred, whereas in the intraspecific cross, ABA concentration in the pedicel was low. The results are discussed with reference to the control ABA exerts on the abscission of poplar flowers after interspecific incompatible pollinations.  相似文献
