Abstract: The aim of this study is to investigate in vitro antioxidant, antimicrobial, and anticancer activity of the acetone extracts of the lichens Umbilicaria crustulosa, U. cylindrica, and U. polyphylla. Antioxidant activity was evaluated by 5 separate methods: free radical scavenging, superoxide anion radical scavenging, reducing power, determination of total phenolic compounds, and determination of total flavonoid content. Of the lichens tested, U. polyphylla had largest free radical scavenging activity (72.79% inhibition at a concentration of 1 mg/mL), which was similar as standard antioxidants in the same concentration. Moreover, the tested extracts had effective reducing power and superoxide anion radical scavenging. Total content of phenol and flavonoid in extracts was determined as pyrocatechol equivalent, and as rutin equivalent, respectively. The strong relationships between total phenolic and flavonoid contents and the antioxidant effect of tested extracts were observed. The antimicrobial activity was estimated by determination of the minimal inhibitory concentration by the broth microdilution method. The most active was extract of U. polyphylla with minimum inhibitory concentration values ranging from 1.56 to 12.5 mg/mL. Anticancer activity was tested against FemX (human melanoma) and LS174 (human colon carcinoma) cell lines using MTT method. All extracts were found to be strong anticancer activity toward both cell lines with IC50 values ranging from 28.45 to 97.82 μg/mL. The present study shows that tested lichen extracts demonstrated a strong antioxidant, antimicrobial, and anticancer effects. That suggests that lichens may be used as possible natural antioxidant, antimicrobial, and anticancer agents.  相似文献

The aim of this study is to investigate in vitro antioxidant, antimicrobial, and anticancer activities of the acetone extracts of the lichens Cladonia furcata, Cladonia pyxidata and Cladonia rangiferina and their atranorin and fumarprotocetraric acid constituents. Antioxidant activity was evaluated by free radical scavenging, superoxide anion radical scavenging, reducing power, and determination of total phenolic and flavonoid compounds. As a result of the study atranorin had largest free radical scavenging activity with IC50 values 131.48 μg/mL. Moreover, the tested samples had effective reducing power and superoxide anion radical scavenging. Total content of phenol and flavonoid in extracts was determined as pyrocatechol equivalent, and as rutin equivalent, respectively. The strong relationships between total phenolic and flavonoid contents and the antioxidant effect of tested extracts were observed. The antimicrobial activity was estimated by determination of the minimal inhibitory concentration by the broth microdilution method. The most active was fumarprotocetraric acid with minimum inhibitory concentration values ranging from 0.031 to 0.125 mg/mL. Anticancer activity was tested against FemX (human melanoma) and LS174 (human colon carcinoma) cell lines using MTT method. The antioxidant activities, reducing powers, 2,2‐diphenyl‐l‐picrylhydrazyl (DPPH) radical‐scavenging activities, total phenolic compound contents and antimicrobial activities of ether, ethanol and hot water extracts of Polygonum cognatum Meissn were studied in vitro. The highest antioxidant activity was found in the water extract. However, there were no statistically significant differences among 15 µg ml?1 extract‐containing samples in linoleic acid emulsion (0.02 M , pH 7.0) during 120 h of incubation (P > 0.05). The reducing power of the water extract was the highest, but its reducing power was markedly lower than that of ascorbic acid. The highest DPPH radical‐scavenging activity was found in the water extract, with 50% DPPH radical scavenging at a concentration of 100 µg ml?1 dried water extract, while at the same concentration of dried ethanol extract the value was 12%. Surprisingly, no DPPH radical‐scavenging activity was observed in the ether extract. The concentrations of phenolic compounds found were 0.48, 0.50 and 0.01 µg ml?1 gallic acid equivalent in 10 µg ml?1 water, ethanol and ether extracts respectively. The ether and ethanol extracts showed antimicrobial activity against Staphylococcus aureus and Bacillus subtilis. The present work examines the in vitro antioxidant and antimicrobial properties of the essential oil and various extracts from the herbal parts of Origanum syriacum L var bevanii. Polar subfractions of methanol extracts from both deodorised and non‐deodorised materials showed the highest DPPH (2,2‐diphenyl‐l‐picrylhydrazyl) radical‐scavenging activity, with IC50 values of 21.40 and 26.98 µg ml?1 respectively, whereas the IC50 of the essential oil was 134.00 µg ml?1. The antioxidant potential of the extracts appeared to be closely related to the presence of polar phenolics. However, the inhibitive effect on linoleic acid oxidation might be promoted by the presence of non‐polar phenolics, as both hexane and dichloromethane extracts showed high antioxidant activities. The antimicrobial activity of the essential oil was superior to those of the other extracts. Nineteen compounds representing 962 g kg?1 of the essential oil were identified; carvacrol (669 g kg?1) was the main component. Overall, the results suggest that the essential oil and extracts from the herbal parts of O syriacum could be used as natural preservative ingredients in the food industry. In Turkish kitchen, Cyclotrichium niveum, which is consumed as a dietary supplement, have been extensively used in soup and food for their odor and flavor. The present study examines the possible antioxidant, antimicrobial and radical scavenging capacity of Cyclotrichium niveum (Boiss.) Manden and Scheng. In order to evaluate antioxidant and radical scavenging activity different in vitro methodologies such as 2,2´-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) radical scavenging activity, 1,1-diphenyl-2-picryl-hydrazyl (DPPH·) free radical scavenging, total antioxidant activity by ferric thiocyanate, total reducing power by potassium ferricyanide reduction method, superoxide anion radical scavenging, hydrogen peroxide scavenging, and ferrous ions chelating activities were used. In addition, antimicrobial and antifungal activity of the both extracts tested against twenty five microorganisms. Total phenolic compounds and total flavonoids contents in water extract of Cyclotrichium niveum (Boiss.) Manden. & Scheng (WECN) and ethanol extract of Cyclotrichium niveum (Boiss.) Abstract: We examined the free radical scavenging activity of Sasa veitchii extract (Hoshi's Striped Bamboo Extract®, HSBE), well known in folk medicine as an efficient drug and antioxidant in detail. To evaluate the free radical scavenging activity of HSBE, its reactivity as hydrogen atom donor toward the 1, 1‐diphenyl‐2‐picrylhydrazyl has been measured using stopped‐flow spectrophotometry. It was found that the second‐order rate constant, k2, obtained at 25 °C was 1.4 (g/L)?1s?1 for HSBE. To compare different chain‐breaking antioxidants quantitatively, we obtained the second‐order rate constant, k2′, on the molar basis of active hydroxyl groups in the tested substances. As a result, the k2′ values for HSBE, 6‐hydroxy‐2, 5, 7, 8‐tetramethylchroman‐2‐carboxylic acid (Trolox), caffeic acid, and (+)‐catechin were 2.6 × 103, 2.0 × 103, 2.3 × 102, and 6.0 × 102 M?1s?1, respectively. These results show that HSBE and Trolox exerted the same free radical scavenging activity under these conditions. In addition, HSBE significantly inhibited the oxidation of methyl linoleate micelles in aqueous dispersions at 30 °C and its antioxidant activity (kinh) was more effective than those of caffeic acid and (+)‐catechin. This is the first study on bamboo extracts in the context of radical scavenging activity that reports kinetic results. Practical Application: We determined the stoichiometric number and the rate constants of bamboo extracts using the method that was devised in determining the antioxidant activity of mixtures. This is the first study of bamboo extracts as an antioxidant that reports the stoichiometric and kinetic results.  相似文献

In this study, the antioxidant capability, total phenolic content and antimicrobial activity of ethanolic extracts of seven fruits from the Brazilian Atlantic Forest were evaluated. The conditions for the extraction of crude phenolics from the fruits were determined using an experimental factorial design. Total phenolic content, 1,1‐diphenyl‐2‐picrylhydrazyl radical (DPPH?) scavenging activity and β‐carotene‐linoleic acid couple oxidation assays were used to evaluate the antioxidant properties of the extracts. In addition, antimicrobial activity was screened using two Gram‐negative bacteria (Escherichia coli and Klebsiella pneumoniae) and one Gram‐positive bacterium (Staphylococcus aureus). All native fruits assayed in this study have high potential as natural antioxidant sources. Among the seven fruits evaluated, Jabuticaba and Uvaia had the highest antioxidant activity in the DPPH? and of β‐carotene‐linoleic acid coupled oxidation assays. In the biological assay, K. pneumoniae was the most sensitive microorganism to the fruit extracts, and the Jabuticaba extract had a slight inhibitory effect against this Gram‐positive bacterium.  相似文献

BACKGROUND: Total phenolics content, antioxidant activity and cytotoxicity of the methanol extracts from leaf parts of 13 Korean traditional salad plants were investigated in order to determine their properties. RESULTS: The highest phenolics content (mg ferulic acid equivalents kg?1 dry weight (d.w.), omit one) was found in methanol extracts from Polygonum aviculare, at 293.7 ± 6.0, followed by Euonymus alatus, at 250.7 ± 3.3, Saxifraga stolonifera, at 125.0 ± 8.1 and Ligularia fischeri, at 122.5 ± 5.9. The methanol plant extracts dose‐dependently increased free radical scavenging activity. Methanol extracts of Polygonum aviculare, Euonymus alatus and Saxifraga stolonifera, at 31 mg kg?1, exhibited the highest 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH) radical scavenging activity (%) by 90.8 ± 4.2, 85.7 ± 3.9 and 64.1 ± 3.2, respectively. According to 3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide (MTT) assay, the methanol extracts from Portulaca oleracea (IC50 < 25.0 µg mL?1) showed the highest cytotoxicity against Calu‐6, followed by Plantago asiatica (49.2 µg mL?1) and Osmunda japonica (89.6 µg mL?1). CONCLUSION: Total phenolics content of the tested plant extracts was correlated with the DPPH radical scavenging activity, suggesting the phenolics compounds are contributing to the antioxidant properties of Korean salad plants. The leaf parts of the 13 Korean traditional salad plants described here that are currently used as foods may also provide some benefit to human health, and research into their potential benefits as preventative and/or therapeutic agents is warranted. Three different parameters (temperature, solvent, and extraction time) were studied regarding to pressure liquid extraction (PLE) of antioxidant and antimicrobial compounds from Spirulina platensis. Two different antioxidant methods, β-carotene bleaching method and DPPH (2,2-Diphenyl-1-picrylhydrazyl hydrate) free radical scavenging assay, were used to determine the optimal PLE conditions for antioxidants extraction. The selected conditions were as follows: extraction temperature equal to 115 °C, extraction time equal to 15 min and ethanol as extracting solvent. The main antioxidant compounds found in this extract were identified as zeaxanthin, a myxoxanthophyll-like compound and very polar phenolic compounds. Moreover, antimicrobial activity of different PLE fractions was tested against Staphylococcus aureus ATCC 25923, Escherichia coli ATCC 11775, Candida albicans ATCC 60193, and Aspergillus niger ATCC 16404. Data obtained showed the hexane and petroleum ether extracts were slightly more active than ethanolic extracts. As for water extracts, none of them were active against the microorganisms tested. Data indicated that both 115 and 170 °C were the best extraction temperatures conditions in order to optimize the extraction of antimicrobial compounds, whereas 9 min was the optimal extraction time. Besides, C. albicans was the most sensitive microorganism to all Spirulina PLE extracts.  相似文献

The methanolic extracts of 16 Helichrysum species were investigated for their in vitro antioxidant, radical scavenging and antimicrobial activities. All the extracts showed strong antioxidant and radical scavenging activity. The highest total antioxidant capacity as ascorbic acid equivalents (AAE) of 194.64 mg/g dry extract was obtained for Helichrysum noeanum in the phosphomolybdenum assay. The highest IC50 value (7.95 μg/ml) was observed for the extract of Helichrysum stoechas subsp. barellieri in the DPPH assay. The total phenolic contents of the extracts ranged from 66.74 to 160.63 mg gallic acid equivalents (GAE)/g dry extract. The major component present in the extracts was identified as chlorogenic acid followed by apigenin-7-glucoside and apigenin by HPLC analysis. All the extracts showed significant antimicrobial activity against microorganisms containing 13 bacteria and two yeasts in the agar diffusion method.  相似文献

This study was performed to evaluate the anticancer, antioxidant, and antimicrobial activities of fractions and isolated compounds from anemone (Anemone cathayensis). Fourteen compounds were isolated from extracts. Anticancer activities of fractions and compounds were determined by MTT assay, and all tested fractions showed inhibition activity on human breast cancer cells (MDA-MB-231). The fraction 6 displayed the strongest anticancer activity, and inhibition percent was 50.32%. The antioxidant effect of fractions was evaluated by using DPPH scavenging assays. Fraction 5 had a higher DPPH radical scavenging activity with low IC50 value of 30.578 μg/mL. The antimicrobial activity of the fractions was evaluated against 3 microorganisms using the agar well diffusion method. The fractions also showed moderate antimicrobial activity. These results suggest that anemone could hold a good potential source for human health.  相似文献

BACKGROUND: This study aimed to evaluate the antioxidant and antimicrobial activity of extracts (aqueous, ethanolic and essential oil) from Satureja montana and to characterise the chemical composition of its essential oil. RESULTS: Satureja montana L. essential oil had relatively high antimicrobial activities against the seven species of bacteria tested. In contrast, aqueous extracts did not reveal antibacterial activity, and the ethanol extract was not effective against Salmonella typhimurium. The major volatile constituents of the essential oil were carvacrol (306 g L?1), thymol (141 g L?1), and carvacrol methyl ether (63 g L?1). The strongest antioxidant capacity was obtained with the hot water extracts of S. montana, whereas the plant essential oil revealed the highest phenolic content. CONCLUSION: The findings indicate that the bioactive extracts of S. montana have strong potential for use as natural antimicrobials and antioxidants in the preservation of processed food. The abilities of broccoli seed extracts and purified sulforaphane (SF) to scavenge 2,2′‐azinobis [3‐ethylbenzothiazoline‐6‐sulphonate] (ABTS?+), 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH?) and superoxide anions were studied. The free radical scavenging activities of broccoli seed extracts had no exact correlation with SF content, indicating that SF was not the main antioxidant component. The broccoli seed extracts were separated by high‐speed countercurrent chromatography to obtain purified SF, which showed dose‐dependent free radical scavenging activities in the DPPH? and ABTS?+ assays, but its activities were weaker to those of ascorbic acid. The same trend was observed in the chemiluminescence assay. The results showed that broccoli seed extracts had strong free radical scavenging activities that were not attributable to SF.  相似文献

Plant samples from several species and populations of the genus Sideritis (Labiatae) grown in Bulgaria (S scardica, S syriaca and S montana) were extracted with different solvents. Their antioxidant activities were determined by the β‐carotene bleaching test (BCBT), 2,2′‐diphenyl‐1‐picrylhydrazyl (DPPH?) radical scavenging method and static headspace gas chromatography (HS‐GC) and compared with the antioxidant activity of two reference compounds of different polarity, viz butylated hydroxytoluene (BHT) and rosmarinic acid. The pure reference compounds were applied in a ten‐times lower concentration than the plant extracts. The highest antioxidant activity in the BCBT, close to that of BHT, was observed for the more apolar extracts. The inhibitory effect on β‐carotene bleaching of the polar extracts and rosmarinic acid was much lower than that of BHT. The inhibition of hexanal formation in bulk safflower oil by most of S syriaca and S scardica extracts was as effective as BHT but less so than rosmarinic acid. S montana extracts showed weak antioxidant or even pro‐oxidant properties. Extracts from butanol and from ethyl acetate and the total methanol extracts from all Sideritis plants studied showed a strong radical scavenging activity against DPPH?, close to that of rosmarinic acid. S montana extracts were, as a whole, slightly weaker radical inhibitors than the extracts from the other two species. Barks of various plants have been reported to possess both in vitro and in vivo antioxidant activity. In this study, antioxidant activity of the extract from the barks of Dillenia indica was evaluated by various in vitro methods. The bark of D. indica was extracted with 70% aqueous acetone. The total phenolic content was determined by Folin-Ciocalteu method and the antioxidant activity was assayed through various in vitro methods such as antioxidant capacity by phosphomolybdenum method, radical scavenging activity using α, α-diphenyl-β-picrylhydrazyl method, hydroxyl radical (?OH) scavenging activity by deoxyribose method, and superoxide anion (O2 ??) scavenging activity by phenazine methosulphate/NADH-nitroblue tetrazolium system. The total phenolic content of the extract as tannic acid equivalents was 54%. The total antioxidant capacity of the extract was found to be 3.12 mmoles/g as equivalent to ascorbic acid at 50 ppm concentration. At 25 ppm concentration, the radical scavenging activity of butylated hydroxyanisole and extract showed 90.9% and 91.0%, respectively. The ?OH scavenging activity of the extract was shown to be 53.9% at 100 ppm concentration. At a concentration of 50 μg, the O2 ?? scavenging activity of the extract was 31.7% as compared to 47.7% by gallic acid. These results indicated that Dillenia indica barks contained large amount of phenolics and possessed potent antioxidant property.  相似文献

The in vitro antimicrobial and antioxidant activities of the essential oil and methanolic extract of Micromeria fruticosa ssp serpyllifolia as well as the composition of the essential oil were examined. The essential oil exhibited activity against 14 bacteria, three fungi and a yeast, with minimal inhibitory concentration (MIC) values ranging from 31.25 to 125 µl ml?1, whilst the methanolic extract was inactive. Antioxidant activity was measured by two methods, namely scavenging of the free radical DPPH and inhibition of linoleic acid oxidation. The methanolic extract exhibited significant antioxidant activity in both assays, providing 50% inhibition at 70.9 ± 0.5 µg ml?1 concentration in the DPPH assay and inhibiting linoleic acid oxidation to 59% at 2 mg ml?1 concentration, whilst the essential oil showed activity only at higher concentrations. The gallic acid equivalent total phenolic content of the methanolic extract was found to be 55.2 ± 2.00 µg mg?1 dry weight extract (5.5% w/w). The chemical composition of the hydrodistilled essential oil was analysed by means of GC/MS. Twenty‐nine constituents were identified, the main ones being piperitenone (50.61%) and pulegone (29.19%). Pawpaw (Asimina triloba [L.] Dunal) possesses antioxidant compounds and strong inhibitors of cancer cells, and is widely cultivated in North America, Canada, and Korea. We analyzed the total phenolic and total flavonoid contents (TPC and TFC, respectively) of pawpaw plants grown in Korea and the antioxidant activities of their roots, twigs, leaves, and fruit with respect to 1,1‐diphenyl‐2‐picryl‐hydrazyl (DPPH) radical scavenging activity, 2,2′‐azino‐bis diammonium salt (ABTS) radical scavenging activity, ferrous (Fe2+) chelating ability, and nitrite scavenging activity. Pearson's correlation analyses revealed a linear correlation between TPC and antioxidant activities (r2 >0.69). Root methanol extracts had higher TPC and antioxidant activities than other extracts, which was also consistent with those from the phenolic compounds found in those extracts. Therefore, antioxidant activities seem to depend on the TPC of each pawpaw tissue and pawpaw roots might be useful as a natural source of natural antioxidants.  相似文献

Antioxidant and antimicrobial activity of the ethyl acetate and water subfractions of methanolic extracts of three Spanish onion varieties were assayed. Flavonoids were mainly present in ethyl acetate subfraction being 34.92 ± 0.75, 7.95 ± 0.16, 0.38 ± 0.01 μmol of rutin eq. g?1 D.W. and its antioxidant capacity was 74.86 ± 1.77, 24.59 ± 0.67, 4.55 ± 0.44 μmol Trolox g?1 D.W. of Grano de Oro, Fuentes de Ebro and Calçot de Valls varieties, respectively. The antimicrobial activity of flavonol standards and onion extracts was evaluated against some food spoiler microorganisms. Quercetin and kaempferol were inhibitory against gram positive bacteria such as Bacillus cereus, Staphylococcus aureus, Microcroccus luteus and Listeria monocytogenes. Gram negative bacteria Escherichia coli and Pseudomonas aeruginosa were less sensible to the antimicrobial effect of both flavonol standards and Candida albicans was totally resistant. Among the onion extracts tested only ethyl acetate subfraction showed antimicrobial inhibition.  相似文献

Dill (Anethum graveolens L.) have been extensively used in salads, soups, and pickles for its aromatic odor and flavor. Recently, interest in plant-derived food additives has grown. In this study, the possible antioxidant properties of water, ethanol, and acetone extracts of dill leaves were investigated. In order to evaluate antioxidant activities of all extracts, different antioxidant tests were used, such as total antioxidant activity by ferric thiocyanate method, reducing power, DPPH (1,1-diphenyl-2-picryl-hydrazyl) free radical scavenging, hydrogen peroxide scavenging, and ferrous ions chelating activities. The content of phenolic compounds was also determined to be the gallic acid equivalent. Among the three extracts, the water extract of dill leaf showed the most potent antioxidative capacity in each assay, showing 79.66% (at 1 mg/mL) in the DPPH? radical scavenging activity, 63% (at 800 μg/mL) in the metal chelating effect, 60% (at 400 μg/mL) in the H2O2 scavenging activity, and 0.61 absorbance (at 1 mg/mL) in the reducing power.  相似文献

Antibacterial and antioxidant potential of essential oil, extract and its fractions of Bidens frondosa Linn were evaluated. Sixty‐one components representing 95.41% of the total oil were identified. The essential oil (7.5 μL disc?1), methanol extract and its different organic subfractions (0.5 μg disc?1) of B. frondosa displayed a great potential of antibacterial activity against Staphylococcus aureus (ATCC 6538 and KCTC 1916), Listeria monocytogenes ATCC 19116, Bacillus subtilis ATCC 6633, Pseudomonas aeruginosa KCTC 2004, Salmonella enteritidis KCTC 12021 and Enterobacter aerogenes KCTC 2190. Antioxidant activity was evaluated by using 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH) assay. The free radical scavenging activity of ethyl acetate (EtOAc) fraction was superior to all other fractions (IC50 = 11.96 μg mL?1), which was higher than synthetic antioxidant butylated hydroxyanisole, (IC50 = 18.27 μg mL?1). Furthermore, the amount of total phenolic compounds was determined and its content in EtOAc fraction was the highest as compared to methanol extract or other fractions. The results indicate that the oil and extracts of B. frondosa could serve as an important bio‐resource of antimicrobial agents and antioxidants for using in the food industries.  相似文献
