The reaction of hydrazine derivatives with α,β-unsaturated ketones, such as cyclohexenyl(phenyl)methanone and (E)-2-benzylidenecyclohexanone, were investigated.The reaction between methylhydrazine and e.g., cyclohexenyl(phenyl)methanone was particularly interesting as 3a,4,5,6,7,7a-hexahydro-1-methyl-3-phenyl-1H-indazole was obtained as the major product together with 4,5,6,7-tetrahydro-2-methyl-3-phenyl-2H-indazole as a minor product.  相似文献

The interaction of cyclopentanone, 2-cyclopentenone, 1,3-cyclopentanedione, 3-methyl-1,2-cyclopentanedione, γ-butyrolactone, 2(5H)-furanone, ascorbic acid, and 5,6-O-isopropylidenyl-2,3-O-dimethylascorbic acid with α-hydroxyethyl radicals (α-HER) generated during the radiolysis of deaerated ethanol has been studied in the continuous irradiation mode. The test compounds, except γ-butyrolactone, oxidize α-HER. 2(5H)-Furanone and 2-cyclopentenone give hydroxyethylation products via the free-radical chain mechanism. In contrast to 2(5H)-furanone and 2-cyclopentenone, ascorbic and 5,6-O-isopropylidenyl-2,3-O-dimethyl-L-ascorbic acids are weaker oxidants for α-HER and attach these radicals at the multiple carbon-carbon bonds.  相似文献

Michael reactions of levoglucosenone and its α-bromo and α-iodo derivatives with α,ω-dinitroalkanes were studied under conditions of chemical and electrochemical generation of base. Procedures were developed for stereospecific fusion of a cyclopentane ring to α-bromo- and α-iodolevoglucosenones, 2-iodocyclopent-2-en-1-one, and 2-iodocyclohex-2-en-1-one by the action of 2,2-dimethyl-1,3-dinitropropane.  相似文献

A conformational investigation of -methylthioacetone, -methylthiocyclohexanone, -phenylthioacetone, and -phenylthiocyclohexanone was undertaken in the MM2(87) force field. In all cases theac-G rotamers with unidirectional orientation of the C=O and S-R bonds are preferred in accordance with the experimental investigations. In all cases the energy of dipole-dipole interaction makes a contribution to the stabilization of these forms. For the eclipsedsp conformers the relatively high calculated values of the energies contradict the data of the experimental investigations, which take account of their participation in the equilibrium. X-ray crystallographic analysis of -phenylthioacetone (3) showed that thesp-T structure with a planar orientation of the thioanisole fragment is realized in the solid phase.A. E. Reactions of α-nitrocinnamic acids esters with indole and its derivatives lead to the formation of the products of alkylation at the C3-reaction center of the heterocycle. The hydrogenation of the adduct of 1-methylindole and α-nitro-β-phenylacrylate on the Raney nickel catalyst afforded indolylaminopropanoate, that was used for the synthesis of ethyl 2-acetylamino-3-(1-methylindol-3-yl)-3-phenylpropanoate, a precursor of the methylated in the indole ring phenyl-substituted tryptophan.  相似文献

A series of cyclodextrin/scutellarin inclusion complexes were prepared from α-cyclodextrin, β-cyclodextrin and 2-hydroxypropyl-β-cyclodextrin with scutellarin (SCU), and their inclusion complexation behaviors, such as stoichiometry, complex stability constants and inclusion mode, were investigated by means of UV/Vis spectroscopy, 1H NMR and 2D NMR. The results showed that the SCU could be efficiently encapsulated in the cyclodextrin cavity in aqueous solution to produce complexes that were more soluble than free SCU. The enhanced binding ability of cyclodextrins towards SCU was discussed from the viewpoint of the size/shape-fit and multiple recognition mechanism between host and guest.  相似文献

We report herein a method that allows for the formation of a C–N bond between the C–3 carbon of α-lactams and the nitrogen atom of indoles. A general procedure for the coupling of indoles and α-lactams in only 25 min with high yield is reported. The scope of the reaction was extended by the development of a method for the in situ generation of less stable phenyl-substituted α-lactams. The developed method provides an atom-economical method for the formation of substituted α-amino amides that are found in a variety of biologically-active compounds.  相似文献

As one of the most investigated flavonoids, apigenin, is considered to be a strong α-glucosidase inhibitor. However, the clinical utility of apigenin is limited due to its low solubility. It was reported that the solubility and biological activity can be improved by introducing sole carboxyalkyl group into apigenin, especially the 7′-substitution. With the increase of length of the alkyl chain in carboxyalkyl group, B ring of the apigenin derivative is embedded much more deeply into the binding cavity while the carboxyalkyl stretches to the neighboring cavity. All of the terminal carboxyl groups form hydrogen bonding interactions easily with the surrounding polar amino acids, such as His239, Ser244, Arg312 and Asp349. Thus, the electron density values of the carbonyl in the carboxyl group become higher than the solution status due to the strong molecular interactions. In fact, electron densities of most of the chemical bonds are decreased after molecular docking procedure. On compared with the solution phase, however, dipole moments of most of these molecules are increased, and their vectors are reoriented distinctly in the active sites. It is noticed that all of the Highest Occupied Molecular Orbital (HOMO) and Lowest Unoccupied Molecular Orbital (LUMO) are distributed throughout the whole parent apigenin ring in solution phase, whereas the disappeared situation happened on the B rings of some molecules (II–IV) in the active site, leading to higher energy gaps.  相似文献

Incorporation reactions of carbon dioxide (CO(2)) with N-Boc-α-amido and α-acetoxy stannanes were developed using CsF as a mild tin activator. Monoprotected α-amido stannanes could be used, and the corresponding arylglycine derivatives were obtained in moderate-to-high yields under 1 MPa (10 atm) of CO(2) pressure. α-Acetoxy stannanes also underwent carboxylation to afford mandelic acid derivatives in excellent yields under ambient CO(2) pressure. Both transformations enabled the synthesis of α-tertiary and α-quaternary carboxylic acid derivatives. In addition, the chirality of (S)-N-tert-butylsulfonyl-α-amido stannanes was transferred with up to 90% inversion of configuration at 100 °C.  相似文献

Abstract

α-mangostin, a polyphenol xanthone derivative, was mainly isolated from pericarps of the mangosteen fruit (Garcinia mangostana L.). In present investigation, a series of derivatives were designed, synthesised and evaluated in vitro for their inhibitory activity of acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE). Among the synthesised xanthones, compounds 1, 9, 13 and 16 showed AChE selective inhibitory activity, 15 was a BuChE selective inhibitor while 2, 3, 5, 6, 7, 12 and 14 were dual inhibitors. The most potent inhibitor of AChE was 16 while 5 was the most potent inhibitor of BuChE with IC50 values of 5.26?μM and 7.55?μM respectively.  相似文献

Inclusion complexation of ciprofloxacin (CIP), sparfloxacin (SPA), and ofloxacin (OFL) drugs with α-CD and β-CD was studied by UV-visible, fluorescence, time-resolved fluorescence, Fourier transform infrared spectroscopy (FTIR), hydrogen nuclear magnetic resonance (1HNMR), scanning electron microscopy (SEM), and molecular modelling techniques. Changes in the absorbance and fluorescence intensities and fluorescence lifetime of the drugs in the cyclodextrin (CD) solutions suggest the formation of inclusion complexes. Carbonyl stretching frequency moved to higher wave numbers and broadening of the N–H stretching band indicated the formation of inclusion complex. Cyclohexane ring protons of the drugs show remarkable upfield or downfield shift in the 1HNMR spectrum, indicating that the cyclohexane part of the guest molecule is entrapped inside CD cavities. SEM images of CIP/CD, SPA/CD, and OFL/CD complexes have a crystal structure with different morphology from the isolated CIP, SPA, OFL, and CDs. Investigations of the energetic, thermodynamic, and electronic properties of parametric model number 3 computational calculations confirmed the stability of the inclusion complex.  相似文献

In the course of our studies on the regioselective carbon-oxygen bond cleavage of the benzylidene acetal group of hexopyranosides with a reducing agent, we found that a combination of a Lewis acid and a reducing agent triggered a ring-opening reaction of the pyranose ring of methyl α-D-allopyranosides. The formation of an acyclic boronate ester by the attachment of a hydride ion at C-1 indicated that the unexpected endocyclic cleavage of the bond between the anomeric carbon atom and the pyranose ring oxygen atom proceeded via an oxacarbenium ion intermediate produced by the chelation between O5/O6 of the pyranoside and the Lewis acid, followed by nucleophile substitution with a hydride ion at C1.  相似文献
