Lactic acid was added to batch very high gravity (VHG) fermentations and to continuous VHG fermentations equilibrated to steady state with Saccharomyces cerevisiae. A 53% reduction in colony-forming units (CFU) ml–1 of S. cerevisiae was observed in continuous fermentation at an undissociated lactic acid concentration of 3.44% w/v; and greater than 99.9% reduction was evident at 5.35% w/v lactic acid. The differences in yeast cell number in these fermentations were not due to pH, since batch fermentations over a pH range of 2.5–5.0 did not lead to changes in growth rate. Similar fermentations performed in batch showed that growth inhibition with added lactic acid was nearly identical. This indicates that the apparent high resistance of S. cerevisiae to lactic acid in continuous VHG fermentations is not a function of culture mode. Although the total amount of ethanol decreased from 48.7 g l–1 to 14.5 g l–1 when 4.74% w/v undissociated lactic acid was added, the specific ethanol productivity increased ca. 3.2-fold (from 7.42×10–7 g to 24.0×10–7 g ethanol CFU–1 h–1), which indicated that lactic acid stress improved the ethanol production of each surviving cell. In multistage continuous fermentations, lactic acid was not responsible for the 83% (CFU ml–1) reduction in viable S. cerevisiae yeasts when Lactobacillus paracasei was introduced to the system at a controlled pH of 6.0. The competition for trace nutrients in those fermentations and not lactic acid produced by L. paracasei likely caused the yeast inhibition.  相似文献

Jasmonic acid altered the accumulation of major anthocyanins in Vitis vinifera cell culture. Peonidin 3-glucoside content at day three was increased from 0.3 to 1.7 mg g–1 dry cell wt while other major anthocyanins were increased by smaller increments. By day 14, the content of methylated and acylated anthocyanins (peonidin 3-p-coumaroylglucoside and malvidin 3-p-coumaroylglucoside) was 6.3 mg g–1 DCW, in response to treatment with jasmonic acid, and comprising 45% (w/w) of total anthocyanins. In comparison, the untreated control culture contained 1.2 mg g–1 DCW which made up 32% (w/w) of total anthocyanins. Light further enhanced anthocyanin accumulation induced by jasmonic acid elicitation. The content of peonidin 3-glucoside at day 3 was 6.6 mg g–1 DCW, 22-fold higher than control cultures while the content in response to light irradiation alone was 0.6 mg g–1 DCW. When a highly pigmented cell line was elicited with jasmonic acid total anthocyanins increased from 9.2 to 20.7 mg g–1 DCW, but there was no change in the anthocyanin composition.  相似文献

Continuous and repeat-batch biofilm fermentations using Actinobacillus succinogenes were performed with immobilized and suspended-cell systems. For the immobilized continuous system, plastic composite supports (PCS) containing 50% (w/w) polypropylene (PP), 35% (w/w) ground soybean hulls, 5% (w/w) dried bovine albumin, 2.5% (w/w) soybean flour, 2.5% (w/w) yeast extract, 2.5% (w/w) dried red blood cells, and 2.5% (w/w) peptone, or PP tubes (8.5 cm in length) were arranged around the agitator shaft in a grid formation. Agitation was controlled at 125 rpm and 150 rpm. Samples were taken at dilution rates of 0.2, 0.4, 0.6, 0.8, 1.0, and 1.2 h–1 and analyzed for succinic acid production and glucose consumption (g l–1). For PCS bioreactors, the highest final succinic acid concentrations (10.1 g –1, 10.4 g l–1) and percentage yields (62.6%, 71.6%) occurred at the dilution rate of 0.2 h–1. PCS disks were evaluated in a repeat-batch biofilm reactor. Suspended-cell batch fermentations were performed in flasks and a repeat-batch bioreactor. The maximum concentration of succinic acid produced was 40 g l–1. Peak succinic acid percentage yields in continuous and repeat-batch fermentations of A. succinogenes were observed in suspended-cell continuous fermentations at a dilution rate of 1.0 h–1 (76.2%) and in PCS repeat-batch fermentations with an initial glucose concentration of 40 g l–1 (86.7%).  相似文献

Candida glycerinogenes is an aerobe which does not depend on sulphite for production of glycerol. With a sufficient O2 supply, up to 130 g glycerol l–1 was produced with 2.6 g acetic acid l–1 as by-product. However, with an insufficient O2 supply – with higher volumes of medium or at higher corn steep liquid concentrations – the glycerol concentration was lower because the by-products, ethanol, pyruvate and lactic acid, were produced in greater amounts, up to 45 g l–1, 4.3 g l–1, 1.6 g l–1, respectively, whereas, less acetic acid (0.6 g l–1) was produced. In addition, ethanol decreased to 0.4 g l–1 and the glycerol yield improved from 34 to 50% (w/w) by adding 50 g sulphite l–1, nevertheless, acetic acid increased to 7.8 g l–1.  相似文献

A method for callus induction, adventitious bud regeneration, shoot multiplication and rooting of in vitro formed shoots of Helianthus annuus L. var. Argentario is described. Hypocotyl and cotyledon explants formed callus on medium containing 2 mgl–1 naphthalene acetic acid and 0.5 mgl–1 benzyladenine. Adventitious buds were formed on hypocotyl segments on medium containing 0.5–2 mgl–1 benzyladenine. The optimal level of sucrose concentration for shoot regeneration from hypocotyls was 1.5%. Multiplication from shoot apices was promoted by kinetin (2 mgl–1) plus gibberellic acid (5 mgl–1), benzyladenine (2 mgl–1) plus gibberellic acid (10 mgl–1) or at lower frequency by benzyladenine (1 mgl–1). A general feature of the plantlets formed in vitro was the precocious flowering.  相似文献

With the objective of verifying the influence of oleic acid as a nutritional supplement in the production of poly(3-hydroxybutyrate-co-3-hydroxyvalerate) by Ralstonia eutropha, cultures were established with 0.3 g oleic acid l–1 and without this supplement, in 30 g inverted sugar l–1 and 1 g propionic acid l–1. The use of this supplement increased the accumulation of polymer from 18.3% to 28.3% (w/w) although the mass of 3-hydroxyvalerate in the polymer remained constant for both cultures.  相似文献

Production of poly(3-hydroxybutyrate) [P(3HB)] from wheyby fed-batch culture of recombinant Escherichia coli CGSC 4401 harboring a plasmid containing the Alcaligenes latus polyhydroxyalkanoate (PHA) biosynthesis genes was examined in a 30 l fermenter supplying air only. With lactose below 2 g l–1, cells grew to 12 g dry cell l–1 with 9% (w/w) P(3HB) content. Accumulation of P(3HB) could be triggered by increasing lactose to 20 g l–1. By employing this strategy, 51 g dry cell l–1 was obtained with a 70% (w/w) P(3HB) content after 26 h. The productivity was 1.35 g P(3HB) l–1 h–1. The same fermentation strategy was used in a 300 l fermenter, and 30 g dry cell l–1 with 67% (w/w) P(3HB) content was obtained in 20 h.  相似文献

We estimated the secondary production of Rhyacophila minora, Ameletus sp., and Isonychia bicolor in three acidic streams and one circumneutral stream in Randolph County, West Virginia. Quantitative benthic samples were collected monthly from these second-order streams from November 1990 to October 1991. Mean pH values in the acidic streams were 4.5, 4.8, and 4.8, and mean pH in the circumneutral stream was 6.7. Production estimates for Rhyacophia minora in the acidic streams were 49.6, 19.2, and 15.8 mg m–2 y–1. Production of R. minora in the circumneutral stream was 1.0 mg m–2 y–1. Ameletus sp. production estimates for the acidic streams were 144.8, 176.8, and 208.3 mg m–2 y–1. Ameletus sp. production in the circumneutral stream was 7.4 mg m–2 y–1. Secondary production of I. bicolor in the circumneutral stream was 116.6 mg m–2 y–1. No Isonychia were collected from the acidic streams. The higher production of R. minora and Ameletus sp. in the acidic streams may be associated with differences in macroinvertebrate community structure.  相似文献

Phototrophic bacterial cells in the effluent from a lighted upflow anaerobic sludge blanket reactor supplied with a medium containing 142 mg S (as SO4 2–) l–1 accumulated a 6.8% w/w oleic acid content in cells and 19 mg cell-bound oleic acid l–1 in the effluent. Pure cultures of Rhodopseudomonas palustris and Blastochloris sulfoviridis isolated from the effluent also accumulated 5.1 and 6.4% w/w oleic acid contents in cells, respectively. The oleic acid content in the cells recovered from the LUASB reactor effluent was related to the phototrophic bacterial population in the LUASB reactor. The inverse relationship was observed in the LUASB reactor between phototrophic bacterial growth and sulfate concentration in the influent.  相似文献

The feeding of propionic acid for production of poly(3-hydroxybutyrate-co-3-hydroxyvalerate) [P(3HB-co-3HV)] by Alcaligenes eutrophus ATCC17697 was optimized using a fed-batch culture system. The concentration of propionic acid was maintained at 3 g l–1 as growth was inhibited by propionic acid in the broth. A pH-stat substrate feeding system was used in which propionic acid was fed automatically to maintain a pH of the culture broth at 7.0. By feeding a substrate solution containing 20% (w/v) propionic acid, 4.9% (w/v) ammonia water [at a molar ratio of carbon to nitrogen (C/N molar ratio) of 10] in cell growth phase, the concentration of propionic acid in the broth was maintained at 3 g l–1 giving a specific growth rate of 0.4 h–1. To promote P(3HB-co-3HV) production, two stage fed-batch culture which consisted of the stage for the cell growth and the stage for the P(3HB-co-3HV) accumulation was carried out. When the substrate solution whose C/N molar ratio was 50 was fed in P(3HB-co-3HV) accumulation phase, the cell concentration and the P(3HB-co-3HV) content in the cells reached 64 g l–1 and 58% (w/w) in 55.5 h, respectively.  相似文献

An isolate of Isochrysis galbana rich in eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) has been grown as a chemostat culture at 20° C and pH 8.00 controlled by CO2 injection. From a low dilution rate (D) of 0.0024 h–1 to 0.0377 h–1, close to maximum growth, a decrease in EPA content from 5.21% dry weight (d.w.) to 2.80% d.w. was observed, although the percentage of EPA in the total fatty acids increased. Lipids were fractionated, EPA being the major fatty acid found in the glycolipid fraction, whereas in the neutral lipid fraction were mainly palmitic and palmitoleic acids. At the same time, the biomass concentration also decreased from 1015 mg·l–1 to 202 mg·l–1 over the range of Ds mentioned. Summary Citric acid was produced with immobilized Yarrowia lipolytica yeast in repeated batch-shake-flask and air-lift fermentations. In active and passive immobilization methods calcium alginate, -carrageenan, polyurethane gel, nylon web and polyurethane foams were tested as carriers in repeated-batch fermentations. The highest citric acid productivity of 155 mg l–1 h–1 was reached with alginate-bead-immobilized cells in the first batch. A decrease in bead diameter from 5–6 mm to 2–3 mm increased the volumetric citric acid productivity threefold. In an air-lift bioreactor the highest citric acid productivity of 120 mg l–1 h–1 with a product concentration of 16.4 g l–1 was obtained with cells immobilized in -carrageenan beads. A new method was established for somatic embryogenesis and plant regeneration from callus cultures of Dioscorea zingiberensis C.H. Wright. Primary callus was induced by culturing stems, leaves and petioles on Murashige and Skoog (MS) medium supplemented with 0.5–2.0 mg l–1 N6-benzyladenine (BA) and 0–2.0 mg l–1 -naphthaleneacetic acid (NAA) or 2,4-dichlorophenoxyacetic acid (2,4-D) for 1 month. The highest frequency (87%) of callus formation was achieved from stem explants treated with 0.5 mg l–1 BA and 2.0 mg l–1 2,4-D. Somatic embryos were obtained by subculturing embryogenic calli derived from stem explants on MS medium supplemented with 2.0–4.0 mg l–1 BA and 0–0.4 mg l–1 NAA or 2,4-D for 3 weeks. The optimum combination of 4.0 mg l–1 BA and 0.2 mg l–1 NAA promoted embryo formation on one-third of the calli. After a further month of subculture on the same medium, mature embryos were transferred to MS medium supplemented with 0–4.0 mg l–1 BA, NAA or indole-3-butyric acid (IBA) for further development of plantlets and tuber formation. Hairy roots of Rauvolfia micrantha were induced from hypocotyl explants of 2–3 weeks old aseptic seedlings using Agrobacterium rhizogenes ATCC 15834. Hairy roots grown in half-strength Murashige & Skoog (MS) medium with 0.2 mg indole 3-butyric acid l–1 and 0.1 mg -naphthaleneacetic acid l–1 produced more ajmaline (0.01 mg g–1 dry wt) and ajmalicine (0.006 mg g–1 dry wt) than roots grown in auxin-free medium. Ajmaline (0.003 mg g–1 dry wt) and ajmalicine (0.0007 mg g–1 dry wt) were also produced in normal root cultures. This is the first report of production of ajmaline and ajmalicine in hairy root cultures of Rauvolfia micrantha.  相似文献

Candida bombicola (ATCC 22214) and C. apicola (ATCC 96134), grown on glucose (100 g l–1) and arachidonic acid (5Z, 8Z, 11Z, 14Z-eicosatetraenoic acid; AA), 1.25 g l–1, synthesized sophorolipid up to 0.93 g l–1. Acid hydrolysis of sophorolipid yielded 19-hydroxy-5Z, 8Z, 11Z, 14Z-eicosatetraenoic acid (19-HETE) and 20-hydroxy-5Z, 8Z, 11Z, 14Z-eicosatetraenoic acid (20-HETE) which were identified by TLC and GC-MS; the ratio of synthesis was 73:27, respectively. Conversion of AA by immobilized Candida bombicola, suspended in beads of 2% (w/v) calcium alginate for 96 h, gave an 83% conversion of 1 g AA l–1 to 19- and 20-HETE. There was no significant loss in the efficiency of the immobilized cells after ten uses.  相似文献

A mutant strain of Yarrowia lipolytica was developed which produced 8.0 g l--hydroxybutyric acid l–1 from butyric acid in a batch culture. The optimum culture conditions in the fermenter for maintenance of a high cell activity, determined by chemostat analyses, were a specific growth rate of 0.06 h–1, a glucose concentration of 2.0 g l–1, and a butyric acid concentration of 8.1 g l–1. Summary The effect of doubling the saturated fatty acid content on the electrophysiology ofNeurospora crassa membranes was studied. Intracellular membrane input resistance (R m ) and potential (E m ) were measured for wild-type (w/t) andcel (Tween 40) organisms as a function of temperature. Over the 0 to 40°C temperature range studied, meanE m values of bothw/t andcel (Tw 40) organisms increased from –160 to –210 mV. This difference is greater than that expected from Nernst potential considerations, indicating an active component ofE m . This active component is insensitive to a doubling of the saturated fatty acid content.R m exhibits a temperature dependence and hysteresis. Averaged data indicate an increase inR m with decreased temperature. The slope of the temperature dependence varies among individual hyphae. Above 17.5°Ccel (Tw 40) hyphae averaged greater than 70% higher values ofR m thanw/t. Below 17.5°Cw/t R m data divided into low and high temperature dependence groups, whilecel data exhibited a low temperature dependence. The results are discussed in relation to gel-liquid crystal phase transitions, membrane fluidity, and the contribution of fatty acid structure to membrane electrical properties.  相似文献

The growth and survival of juvenile Haliotis rubra, when fed with the diatom Navicula sp. cultured in f/2 medium containing combined nitrogen at 24.71 mg NO3-N L–1 (high), 12.35 mg NO3-N L–1 (standard) or 2.47 mg NO3-N L–1 (low), were compared in a 33-day trial. The alga in the low nitrogen medium contained 37% less total amino acid than that in the high and standard nitrogen media. There was a slightly greater reduction in essential amino acids (40%) compared to non-essential amino acids (35%). Juvenile abalone feeding on Navicula grown in medium with low nitrate and lower total amino acid content grew more slowly than when fed on the same species grown in standard or higher nitrogen medium with a higher amino acid content. The growth rate of juveniles was highest (43 m d–1) in the high nitrate treatment followed (40 m d–1) by the standard nitrate treatment and lowest (31 m d–1) in the low nitrate treatment. The survival of the juveniles was also effected by the diet. Survival was better in the high and standard nitrogen media (88%) than the low nitrogen medium (75%). The results suggest that in order to achieve uniformity in nutritional quality of diatoms and good growth of abalone juveniles in commercial abalone nurseries, the nitrogen concentration in tanks should be monitored and additional nitrate added to provide an optimum concentration of between 2 and 12 mg NO3-N L–1.  相似文献

In autumn 1986, six small lakes at different stages of acidification were stocked with one-summer-old whitefish, Coregonus pallasi Valenciennes 1848, in order to see whether whitefish stocking would be a suitable method for the mitigation of acidification effects. In two of the lakes the introduction was a complete failure: the whitefish did not survive, evidently due to high acidity and high aluminium concentrations of the lake waters. In one of the most acidified lakes (pH 4.3–4.8, Allab 29–125 g 1–1) and in two less acidic lakes (pH 5.0–5.2 and 5.4–6.4), introduction was successful. Three years after the introduction, the mean weights of the fish in those three lakes were 580, 250 and 360 g respectively, with the weight and also the condition factor of stocked whitefish being highest in the most acidified lake. In that lake there were few or no fish present during the introduction, whereas in the less acid lakes there were dense populations of perch and therefore a potential interspecific competition for food. Different availability of food in the lakes was presumed to be the main reason for the growth differences. Plasma Na+ and Cl concentrations of whitefish were lower in the acidic lakes than in the lake with pH around 6 three years after stocking. This suggests that, despite the good growth and highest condition factor of whitefish in the most acid lake, the fish still experienced some acid stress.  相似文献
