以人工合成的微卫星序列 (GTG) 5,(GT) 8,(CAC) 5和人源小卫星 33 1 5作引物 ,扩增纵纹腹小的基因组DNA ,产生多态性DNA片段 ,回收了 8个表现个体特异性的片段。当用小的基因组总DNA探针与它们杂交时 ,其中 2个表现阳性 ,说明PCR方法扩增出的高变异产物含有重复序列。用含重复序列的个体特异性PCR产物作探针 ,与无关个体小基因组DNA的HaeⅢ酶切产物进行DNA印迹 ,获得了变异性较高的DNA指纹图谱。且通过对京白鸡家系分析表明 ,用小基因组DNA的PCR产物分离制备的探针所获得的DNA指纹图带能够稳定的遗传。因此 ,高变异的PCR产物可以有效地用作DNA指纹探针。  相似文献

外源DNA导入小麦的RAPD验证及遗传分析   总被引:7,自引:2,他引:5  
将外源DNA特别是牛胸腺DNA转移给普通小麦,对在其后代中选育出的矮杆变异体进行了RAPD分子验证及其遗传学分析研究,获得了3个主要结果:(1)对变异体D111,受体814527,供体矮孟牛I型进行的RAPD验证中,通过137个引物由5个引物检测出了DNA的多态性,表明矮秆变异体D111的出现可能是供体的片段DNA进入了受体并得到稳定遗传。(2)对变异体D011,D1453,受体814527,供体牛胸腺DNA进行的RAPD验证中,在供试的180个引物中,变异体,受体扩增产物的带型绝大多数一致而与供体则完全不一致,其中有很少引物对变异体和受体扩增产物的带型表现不同,还有个别引物对变异体扩增产物的带型与供体的个别带一致,由此说明亲缘关系极远的牛胸腺DNA导入受体引起的变异更为广泛和复杂;(3)3个矮秆变异体的遗传分析表明,控制其株高的基因位于核基因组,在杂种后代中表现了明显降低株高的作用,其杂种后代的某些农艺性状也表现良好,因此,利用它们进行杂交育种或在杂种优势的利用上,都将是有较好应用价值前途的新矮源。  相似文献

RAPD条件优化及天麻基因组DNA多态性分析   总被引:7,自引:0,他引:7  
建立了RAPD扩增条件快速优化程序与方法.并应用于天麻基因组DNA扩增条件的优化及多态性的测定:获得了天麻基因组DNA的RAPD扩增优化条件和DNA指纹图谱;分析了模板DNA、引物、dNTP、Taq DNA聚合酶等的浓度和退火温度对RAPD扩增的影响.结果表明:天麻基因组DNA用引物S1扩增的片段具有更明显的多态性,这种指纹图谱更适合于天麻遗传分化研究;而用引物S12扩增的DNA指纹图谱具有更大的相似性,这种指纹图谱更适合于天麻真伪鉴别.该方法使RAPD扩增条件优化过程实现了程序化和数量化,是获得RAPD优化条件的简便快速、经济实用方法.应用该方法进行RAPD扩增,可获得图谱清晰、稳定可靠的实验结果.  相似文献

通过在稻瘟病菌Pyricularia oryza。基因组文库中的筛选,找到一个散布的并具有基因组特异性的层重复顺序POR6。本文报道用这一株针对6个日本菌株和26个中国北方菌株进行DNA指纹作图的结果。其中22个中国北方菌株按其杂交带型百分相似率被分成8个株系。一些在我们实验室保存的菌株用传统方法鉴定发现在转管过程中会发生致病性变异。当用POR6作探针与这些菌株NNA的EcoRV酶切片段杂交时,检测出它们的无性世代中出现数条EcoRV多态性片段。  相似文献

大仓鼠DNA 指纹谱探针的筛选   总被引:1,自引:0,他引:1  
采用一种简便提取高质量DNA 的方法, 从大仓鼠肝脏组织中提取其总DNA , 分别以人工合成的微卫星核心序列(GTG)5和(CA)8做单一引物, 进行特异引物PCR 反应。电泳检测后回收15 条特异性片段。与被标记过的大仓鼠基因组DNA 反向杂交结果表明, 15 个片段中(GTG)5-8 、(CA)8-1b和(CA) 8-5b产生了较强的阳性信号。我们依据3 个片段的测序结果设计适合DIG标记的探针, 该探针得到的大仓鼠不同地理种群个体的指纹图谱有较高的个体特异性和种群多态性, 而且与传统的来源于其它生物重复序列的探针如33.6 和33.15 形成的指纹图谱相比得到的变异适中, 便于统计。  相似文献

采用RAPD技术,对洋葱细胞质雄性不育系70及其保持系71的基因组DNA的多态性进行分析。结果表明,在被检测的100条随机引物中,46条引物具有扩增产物,5条引物扩增出多态性条带。对这5条引物进行重复筛选和单株验证,只有G02扩增出的多态性片段,且表现稳定。推测该标记与育性有关。  相似文献

目的获得东方田鼠的特异DNA序列.方法Aβ基因使用PCR,基因克隆,斑点杂交,DNA序列分析,生物信息学技术.结果根据小鼠MHCⅡ外显子2及其两侧序列,合成引物并扩增东方田鼠基因组DNA,将PCR产物回收、测序后,分别设计内引物扩增东方田鼠基因组DNA,其中一对引物可得到特异性扩增带,将得到的DNA片段插入PGEM-Teasy载体,进行序列分析.用这对引物扩增人、昆明小鼠、BALB/c小鼠及C57BL/6J小鼠基因组DNA,均无扩增产物.以东方田鼠特异性扩增产物为探针进行斑点杂交,除东方田鼠基因组DNA外,其他几种动物基因组DNA均为阴性结果.进一步对该DNA片段进行了BLAST同源性搜索和外显子预测,在Genbank中没有发现高度同源序列,并且找到一个可能的外显子,该外显子由69个氨基酸组成.结论获得的DNA片段为东方田鼠的特异片段,这将为从分子水平深入研究东方田鼠的遗传背景、生物进化规律以及东方田鼠抗日本血吸虫的机理奠定基础.  相似文献

根据鱼类外周血细胞都有核的特点,采用从冷冻和低渗双重处理分离的细胞核提取基因组DNA.以此法获得的白鲢和鳙鱼的基因组DNA为模板,和Operon公司生产的OPN和OPM两个组共40个随机引物,对这两种鱼进行了随机扩增多态DNA(RAPD)分析;确定了对这两种鱼基因组相关区域可进行随机PCR扩增的有效引物,特别是哪些可产生种群内或群体的RAPD遗传标记,即可产生个体特异性和群体特异性RAPD带谱的引物.讨论了RAPD遗传分子标记在鱼类遗传,特别是遗传多样性研究,和鱼类种质资源评估和管理中的应用前景问题.  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. The dichtomously branched stigma in each carpel of the Tricyrteae is supplied by a bifurcated dorsal bundle.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献
