摘    要:目的:探讨RAB3C表达对非小细胞肺癌细胞A549及NCI-H1299细胞侵袭转移的影响及其机制。方法:构建siR-RAB3C慢病毒载体转染A549及NCI-H1299细胞,使用qRT-PCR及Western blotting法验证每种细胞中RAB3C的mRNA及蛋白表达情况。应用Transwell实验及细胞划痕试验检测下调RAB3C后细胞侵袭情况的变化。Western blotting法比较对照组及siR-RAB3C组细胞中E-cadherin、N-cadherin及Vimentin的表达水平。结果:siR-RAB3C转染组细胞RAB3C的mRNA及蛋白表达量均下调。划痕试验结果显示,与对照组细胞相比,siR-RAB3C组细胞迁移率显著减少。Western blotting实验结果显示,下调RAB3C表达促进E-cadherin,抑制N-cadherin及Vimentin蛋白的表达。结论:调控RAB3C表达通过抑制上皮间质转化抑制非小细胞肺癌A549及H1299细胞侵袭转移。

Abstract:Objective:To explore the effect of RAB3C expression on cell invasion in human non-small cell lung cancer.Methods:A549 and NCI-H1299 cells were transfected with si-NCAPG or control vectors.The mRNA and protein level expression of RAB3C in A549 and NCI-H1299 cells were then verified by qRT-PCR and Western blotting.Transwell experiments were conducted to detect the cell invasion in the control group or si-RAB3C group.Western blotting was used to compare the expression levels of E-cadherin,N-cadherin and Vimentin in the cells of control group and siR-RAB3C group.Results:The expression of RAB3C in both mRNA and protein level was down-regulated in the cells transfected with si-RAB3C.Compared to the control cells,down-regulation of RAB3C expression inhibited the cell invasion in both A549 and NCI-H1299 cells.The expression of E-cadherin was promoted,but N-cadherin and Vimentin expressions were inhibited in the cells transfected with siR-RAB3C.Conclusion:Down-regulated expression of RAB3C regulated epithelial mesenchymal transformation and inhibited cell invasion in human non-small cell lung cancer.
