miR-940在非小细胞肺癌中的表达及对肺腺癌A549细胞生物学功能的影响

摘    要:目的:探讨miR-940在非小细胞肺癌中的表达及对肺腺癌A549细胞增殖、迁移、侵袭的影响。方法:选取标本库中2015年10月至2016年12月在我院胸外二科行手术治疗的非小细胞肺癌患者85例,检测癌组织和癌旁组织中miR-940的表达水平。将miR-940 mimics和miR-940 inhibitors转染至A549细胞中,检测转染后各组A549细胞中miR-940的表达水平。根据细胞增殖实验、划痕实验和Transwell实验检测各组细胞的增殖、迁移及侵袭能力变化。双荧光素酶实验证实miR-940的靶基因。结果:miR-940在癌组织中的表达量显著低于癌旁组织中的表达量,差异有统计学意义(P=0.004)。细胞增殖实验结果显示,miR-940 mimics组的细胞增殖率显著低于miR-940 inhibitors组(P<0.05);细胞划痕实验结果显示,miR-940 mimics组的划痕愈合率明显低于miR-940 inhibitors组(P<0.05);Transwell细胞侵袭实验结果显示,miR-940 mimics组的侵袭细胞数显著低于miR-940 inhibitors组的侵袭细胞数,差异有统计学意义(P<0.01)。Cbl-b是miR-940的直接靶基因。结论:miR-940在非小细胞肺癌中呈低表达,miR-940可能通过靶向抑制Cbl-b基因,进而抑制A549细胞的增殖、迁移和侵袭能力。

Abstract:Objective:To investigate the expression of miR-940 in non-small cell lung cancer and its effect on proliferation,migration and invasion of lung adenocarcinoma A549 cells.Methods:From October 2015 to December 2016,85 patients with non-small cell lung cancer who underwent surgical treatment in the second department of thoracic surgery of our hospital were selected from the specimen library,and the expression level of miR-940 in cancer tissues and adjacent tissues was detected.The miR-940 mimics and miR-940 inhibitors were transfected into A549 cells,and the expression level of miR-940 in A549 cells of each group after transfection was detected.The change of proliferation,migration and invasion ability of each group was detected according to cell proliferation test,scratch test and Transwell test.Double luciferase experiment confirmed the target gene of miR-940.Results:The expression of miR-940 in cancer tissues was significantly lower than that in adjacent tissues (P=0.004).The results of cell proliferation experiment showed that the cell proliferation rate of miR-940 mimics group was significantly lower than that of miR-940 inhibitors group (P<0.05).The results of cell scratch test showed that the scratch healing rate of miR-940 mimics group was significantly lower than that of miR-940 inhibitors group (P<0.05).Transwell cell invasion test showed that the number of invasion cells of miR-940 mimics group was lower than that of miR-940 inhibitors group,and the difference was statistically significant (P<0.01).Cbl-b was the direct target gene of miR-940.Conclusion:miR-940 expression is low in non-small cell lung cancer.miR-940 may inhibit the proliferation,migration and invasion of A549 cells by suppressing Cbl-b gene.
