小檗碱通过上调miR-145-5p抑制宫颈癌细胞的增殖、侵袭、迁移和谷氨酰胺代谢

摘    要:目的:探索小檗碱(berberine,BBR)对宫颈癌(cervical cancer,CC)细胞增殖、侵袭、迁移和谷氨酰胺代谢的影响。方法:qRT-PCR法检测miR-145-5p表达水平;MTT法和集落形成实验检测细胞增殖;划痕愈合实验和Transwell实验检测细胞的迁移和侵袭;Western blot检测基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)、上皮钙黏蛋白(E-cadherin)、神经钙黏蛋白(N-cadherin)、波形蛋白(Vimentin)蛋白水平;使用相应的试剂盒测量谷氨酰胺消耗量、α-酮戊二酸(α-ketoglutarate,α-KG)产量和三磷酸腺苷(adenosine triphosphate,ATP)产量。初步在小鼠体内,探究BBR对异种移植物及miR-145-5p表达水平的影响。结果:BBR能够抑制CC细胞增殖、迁移、侵袭和谷氨酰胺代谢,且可显著下调MMP-9、N-cadherin和Vimentin水平,上调miR-145-5p、E-cadherin水平(P<0.05)。抑制miR-145-5p表达能够逆转BBR对CC细胞的影响(P<0.05);另外,在小鼠体内,BBR可显著降低肿瘤组织的重量及体积,同时上调miR-145-5p表达水平(P<0.05)。结论:BBR通过上调miR-145-5p抑制CC细胞的增殖、迁移、侵袭和谷氨酰胺代谢。

Abstract:Objective:To explore the effects of berberine (BBR) on proliferation,invasion,migration and glutamine metabolism of cervical cancer (CC) cells.Methods:QRT-PCR method was used to detect the expression level of miR-145-5p.MTT method and colony formation test were used to detect cell proliferation.Scratch healing test and Transwell test were used to detect cell migration and invasion.Western blot was used to detect the protein levels of matrix metalloproteinase-9 (MMP-9),epithelial cadherin (E-cadherin),neural cadherin (N-cadherin),and Vimentin and the corresponding kits were used to measure glutamine consumption,α-ketoglutarate (α-KG) production and adenosine triphosphate (ATP) production.Preliminarily in mice,to explore the effect of BBR on xenograft and miR-145-5p expression.Results:BBR was able to inhibit proliferation,migration,invasion and glutamine metabolism of CC cells,significantly down-regulate the levels of MMP-9,N-cadherin and Vimentin,and up-regulate the levels of miR-145-5p and E-cadherin (P<0.05).Inhibiting the expression of miR-145-5p was able to reverse the effect of BBR on CC cells (P<0.05).In addition,in mice,BBR could significantly reduce the weight and volume of tumor tissues,and at the same time up-regulate the expression level of miR-145-5p (P<0.05).Conclusion:BBR inhibits the proliferation,migration,invasion and glutamine metabolism of CC cells by up-regulating miR-145-5p.
