胃癌中miR-146a和LIN52表达及其与化疗敏感性和预后的关系

摘    要:目的:研究胃癌组织中微小RNA-146a(miR-146a)、LIN52的表达关系,并对其与患者化疗敏感性和预后的关系进行探讨。方法:选取2012年8月至2015年6月进行手术治疗的87例胃癌患者的癌组织标本为胃癌组,另选取距癌组织边缘>5 cm的对应癌旁组织标本为癌旁组织组。采用qRT-PCR法检测miR-146a、LIN52 mRNA相对表达量,免疫组化法检测LIN52蛋白表达,采用MTT法检测胃癌细胞对5-FU、DDP、ADM、MMC敏感性,应用Pearson法对miR-146a与LIN52表达的相关性进行分析,采用Kaplan-Meier对胃癌患者3年的生存情况进行分析。结果:与癌旁组织组相比,胃癌组miR-146a表达水平降低(P<0.05),LIN52 mRNA表达水平升高(P<0.05),LIN52蛋白阳性表达率升高(P<0.05);miR-146a、LIN52表达均与年龄、淋巴结转移、临床分期、组织分化程度、浸润深度相关(P<0.05);药敏试验表明,miR-146a、LIN52表达情况与肿瘤细胞对5-FU、DDP的敏感性有关;Pearson法分析显示miR-146a与LIN52呈显著负相关(r=-0.898,P<0.05);Kaplan-Meier法分析显示miR-146a高表达组3年内总生存率远高于低表达组(58.8%vs 18.9%)。LIN52阴性患者3年内总生存率远高于阳性患者(74.1%vs 16.7%)。结论:胃癌组miR-146a低表达,LIN52高表达,miR-146a、LIN52与胃癌细胞对化疗药物敏感性及预后相关,检测其表达对化疗药物的选择具有一定参考价值。

Abstract:Objective:To study the relationship between the expression of microRNA-146a(miR-146a) and LIN52 in gastric cancer and to investigate their relationships with chemosensitivity and prognosis.Methods:From August 2012 to June 2015,87 patients with gastric cancer underwent surgical treatment were selected as gastric cancer group,and the corresponding para cancerous tissues>5 cm from the edge of the cancer tissue were selected as paracancerous tissues group.The relative expression levels of miR-146a and LIN52 mRNAs were detected by qRT-PCR.The expression of LIN52 protein was detected by immunohistochemistry.MTT method was used to detect the sensitivities of gastric cancer cells to 5-FU,DDP,ADM and MMC.The correlation between miR-146a and LIN52 expressions was analyzed by Pearson.The 3-year survival of gastric cancer patients was analyzed by Kaplan-Meier.Results:Compared with the paracancerous tissues,the expression of miR-146a was lower in gastric cancer group(P<0.05),and the expression of LIN52 mRNA was higher(P<0.05).Compared with the paracancerous group,the positive expression rate of LIN52 protein in gastric cancer group was higher(P<0.05).miR-146a and LIN52 expressions were correlated with age,lymph node metastasis,clinical stage,tissue differentiation and depth of invasion(P<0.05).Drug sensitivity test showed that the expressions of miR-146a and LIN52 were related to the sensitivity of tumor cells to 5-FU and DDP.Pearson analysis showed that miR-146a was negatively correlated with LIN52(r=-0.898,P<0.05).Kaplan-Meier analysis showed that the 3-year overall survival rate in the high expression group was much higher than that in the low expression group(58.8% vs 18.9%).The 3-year overall survival rate of LIN52 negative patients was much higher than that of positive patients(74.1% vs 16.7%).Conclusion:The expression level of miR-146a is decreased and LIN52 mRNA is increased in gastric cancer group.miR-146a and LIN52 are related to chemosensitivity of gastric cancer cells.Detection of its expression has a certain reference value for the selection of chemotherapeutic drugs.
