SQLE在乳腺癌中的表达及其临床意义的生物信息学分析

摘    要:目的:利用生物信息学方法分析角鲨烯环氧化酶(squalene epoxidase,SQLE)在乳腺癌中的表达情况及其临床意义。方法:挖掘Oncomine和GEPIA数据库中 SQLE在乳腺癌和正常乳腺组织中的表达情况,分析CCLE数据库中 SQLE在乳腺癌细胞系中的表达水平,同时利用HPA和UALCAN数据库分析 SQLE蛋白在正常和乳腺癌组织中的表达水平,并利用免疫组化方法通过临床样本进一步验证SQLE蛋白的表达情况。最后用K-M plotter数据库和GEPIA数据库分析 SQLE表达与预后的关系。结果:Oncomine和GEPIA数据分析显示 SQLE基因在乳腺癌组织中高表达,CCLE数据分析表明 SQLE在乳腺癌细胞系中显著高表达,HPA数据库免疫组化结果和UALCAN数据库结果以及临床样本免疫组化结果均显示乳腺癌组织 SQLE蛋白相对于正常乳腺组织高表达;Kapan-Meier Plotter和GEPIA数据库分析显示高表达SQLE的乳腺癌预后不良。结论:SQLE在乳腺癌组织和细胞中均高表达,其可能是乳腺癌中潜在的促癌基因,有望成为乳腺癌治疗的新靶标。

Abstract:Objective:To explore the expression of SQLE and its clinical significance in breast cancer.Methods:The expression of SQLE in breast cancer tissues and normal tissues was analyzed in Oncomine and GEPIA databases,and the expression of SQLE in breast cancer cell lines was evaluated in CCLE database.Meanwhile,HPA database and UALCAN database were used to analyze the expression of SQLE protein in normal and breast cancer tissues.Results:Oncomine and GEPIA databases showed that SQLE gene was highly expressed in breast cancer tissues,and CCLE database analysis showed that SQLE was highly expressed in all breast cancer cell lines.Immunohistochemistry results from the HPA database showed that the expression of SQLE protein was significantly higher in breast cancer tissues compared with normal tissues.UALCAN database showed that SQLE protein was highly expressed in breast cancer tissues.Furthermore,through analyzing the data from oncomine and GEPI database,the prognostic data showed a worse OS in the patients with higher SQLE expression than in those with a lower expression.Conclusion:SQLE is over-expressed in breast cancer,and its expression is associated with the prognosis of breast cancer,which may be used as an important target of medication for breast cancer in the clinic.
